Leptin physiology and pathophysiology in energy homeostasis, immune function, neuroendocrine regulation and bone health.

Since its discovery and over the past thirty years, extensive research has significantly expanded our understanding of leptin and its diverse roles in human physiology, pathophysiology and therapeutics. A prototypical adipokine initially identified for its critical function in appetite regulation and energy homeostasis, leptin has been revealed to also exert profound effects on the hypothalamic-pituitary-gonadal, thyroid, adrenal and growth hormone axis, differentially between animals and humans, as well as in regulating immune function. Beyond these roles, leptin plays a pivotal role in signi

Neuroendocrine adaptations to starvation.

Famine and starvation have punctuated the evolutionary past of the human species. As such, we have developed hormonal responses to undernutrition that minimize energy expenditure on processes that are not critical for the survival of the individual, such as reproduction. In this review, we discuss neuroendocrine adaptations to starvation including hypogonadotropic hypogonadism, growth hormone resistance, hypercortisolemia, and the downregulation of the hypothalamic-pituitary-thyroid axis. We review the time-course of these adaptations by describing studies involving the short-term fasting of h

New Perspectives on the Pathogenesis of PCOS: Neuroendocrine Origins.

Polycystic ovary syndrome (PCOS) is the most common endocrine condition in reproductive-aged women. It is characterized by reproductive, endocrine, metabolic, and psychological features. The cause of PCOS is unknown, thus there is no cure and its management remains suboptimal because it relies on the ad hoc empirical management of symptoms only. We review here the strong support for PCOS having a neuroendocrine origin. In particular, we focus on the role of aberrant hypothalamic-pituitary function and associated hyperandrogenism, and their role as major drivers of the mechanisms underpinning t

The role of leptin in regulating bone metabolism.

Leptin was initially best known for its role in energy homeostasis and regulation of energy expenditure. In the past few years we have realized that leptin also plays a major role in neuroendocrine regulation and bone metabolism. Here, we review the literature the indirect and direct pathways through which leptin acts to influence bone metabolism and discuss bone abnormalities related to leptin deficiency in both animal and human studies. The clinical utility of leptin in leptin deficient individuals and its potential to improve metabolic bone disease are also discussed. We are beginning to un

The darkness at the end of the tunnel: summary and evaluation of an international symposium on light, endocrine systems and cancer.

Research on light at night and cancer is evolving at an accelerating pace, fueled largely by exciting results in rodent toxicology and basic human biology. Epidemiologic research is at a relatively early stage of development in which the exposure surrogates such as shift work and blindness predominate. Causal graphs for shift work, light at night and breast cancer illustrate some of the subtleties that can arise in the use of exposure surrogates of different kinds. Baseline data on circadian rhythms and melatonin cycles among human populations living at different latitudes are needed. Epidemio

Aspirin-sensitive asthma due to diffuse neuroendocrine system pathology.

Available clinical data on aspirin-sensitive asthma (ASA) indicate that ASA patients have certain disturbances in the nervous, endocrine, immune and other body systems. It has been found that such patients have a lower melatonin (MT) production in daytime, a pathology of the platelet membrane-receptor complex, and a pathological response to exogenic MT and acetylsalicylic acid. A hypothesis has been suggested in which ASA is considered as apudopathy caused by dysfunction of MT-producing cells. The decreased MT production and the disturbed cell sensitivity to MT lead to pathological changes in

Bronchial carcinoid tumors.

Bronchial carcinoid tumors, termed (incorrectly) "bronchial adenomas" in the past, are uncommon pulmonary neoplasms. These tumors are currently classified as neuroendocrine in origin because of their potential to form and sometimes secrete a variety of chemical substances. Overall, approximately 75% of bronchial carcinoid tumors arise in the lobar bronchi, 10% occur in the main-stem bronchi, and 15% originate in the periphery of the lung. Well-differentiated carcinoid tumors constitute almost 90% of all bronchial carcinoids. Atypical carcinoid tumors have a higher malignant potential than do t

Psychoendocrine aspects of coping with distress.

The psychoendocrine aspects of coping with distress were studied under mental and physical laboratory strain as well as in response to naturalistic stress. In spite of the common underlying endocrine responses to distress, gender differences in psychological response appeared to modify endocrine responses to the experience of pain. If the aspiration level task in the laboratory was well controlled by the subject hypothalamo-pituitary stress hormones were no longer secreted. Furthermore, heavy, physical work elevated blood levels of stress hormones (dopamine, noradrenaline, adrenaline, prolacti

Discrete lesions of the area postrema abolish radiation-induced emesis in the dog.

Studies carried out in several mammalian species during the 1950's led to the concept of a 'vomiting center' located in the dorsolateral reticular formation and a 'chemoreceptor trigger zone' (CTZ) within or near the area postrema (AP). This early work suggested that the AP was essential for vomiting induced by a variety of chemical emetics and by ionizing radiation. However, the lesion techniques used often produced significant damage to neural tissue underlying the AP, as well as to the AP itself, making localization of function very difficult. In the present study, electrolytic lesions conf

Neural mechanisms underlying the pubertal increase in LHRH release in the rhesus monkey.

Puberty is triggered by an increase in pulsatile release of luteinizing hormone-releasing hormone (LHRH) from the hypothalamus. Although the LHRH neurosecretory system is mature well before the onset of puberty, a central inhibitory mechanism restrains LHRH release in juvenile primates. Recent studies suggest that this central inhibition is primarily because of GABAergic neurotransmission. A reduction of GABAergic restraint appears to be essential for the initiation of puberty, but the mechanism that underlies the disinhibition process remains to be elucidated. Future research into the regulat

The vasopressin system as an immunoneuroendocrine therapeutic target in inflammatory conditions.

Arginine vasopressin (AVP), a nonapeptide produced mainly in hypothalamic neurons, is known for regulating osmotic balance and blood vessel tone. Recent evidence shows it also plays key roles in the immunoneuroendocrine axis, reproduction, and cellular stress. This review explores its role as an immune regulator involving hypothalamic signals, organ responses, and inflammation. We cover AVP's structure, receptor types (V1a, V1b, V2), pharmacological analogs, and their interactions with cytokines and immune cells. Experimental and translational data indicate that AVP exerts context-dependent ef

Neuroendocrine timekeepers: changes in normal and premature aging.

The hypothalamus is a central regulator of circadian rhythms, metabolism, and endocrine function, integrating internal and external cues to maintain physiological homeostasis. Aging impairs hypothalamic function, leading to metabolic changes, sleep disturbances, and a higher risk of age-related disease. Laminopathies - rare genetic disorders marked by premature aging - exhibit profound neuroendocrine and circadian rhythm dysfunction, offering insights into mechanisms of hypothalamic aging. The complex interplay between the hypothalamus, circadian rhythms, and systemic aging highlights the crit

[Pathophysiological mechanisms linking chronic stress and cervical spondylosis of vertebral artery type: A theoretical framework of the neuroendocrine-immune network].

Stress is a critical inducer in the onset and progression of many chronic diseases. Prolonged or intense stress can disrupt the overall balance between the nervous, immune, and endocrine systems. The resulting biological signals may act on corresponding receptors in the cervical spine region, leading to adverse pathological changes. The vertebral artery and the surrounding muscular and connective tissues are influenced by biomechanical abnormalities and inflammatory cascades associated with cervical spondylosis of vertebral artery type (CSA), which promotes the release of various hormones. The

Galanin family peptides: Molecular structure, expression and roles in the neuroendocrine axis and in the spinal cord.

Galanin is a neurohormone as well as a neurotransmitter and plays versatile physiological roles for the neuroendocrine axis, such as regulating food intake, insulin level and somatostatin release. It is expressed in the central nervous system, including hypothalamus, pituitary, and the spinal cord, and colocalises with other neuronal peptides within neurons. Structural analyses reveal that the human galanin precursor is 104 amino acid (aa) residues in length, consisting of a mature galanin peptide (aa 33-62), and galanin message-associated peptide (GMAP; aa 63-104) at the C-terminus. Gut microbiome in neuroendocrine and neuroimmune interactions: The case of genistein.

The healthy and diverse microbes living in our gut provide numerous benefits to our health. It is increasingly recognized that the gut microbiome affects the host's neurobehavioral state through production of metabolites, modulation of intestinal immunity (e.g., cytokines) and other mechanisms (e.g., gut neuropeptides). By sending the sensed information (e.g., metabolic and immunologic mediators) about the state of the inner organs to the brain via afferent fibers, the vagus nerve maintains one of the connections between the brain and GI tract, and oversees many critical bodily functions (e.g.

Neuroendocrinology of amphibian metamorphosis.

The timing of metamorphosis is a central amphibian life history trait and is controlled by the interplay of developmental progression, body size and condition, and environmental signals. These different processes and signals are integrated by the neuroendocrine system to regulate production of hormones by the thyroid gland. Thyroid hormone (TH) is the primary morphogen controlling metamorphosis, while corticosteroids (CSs) produced by the interrenal glands synergize with TH to promote metamorphic changes. The actions of TH are modulated by monodeiodinase enzymes expressed in TH target tissues.

A melatonin-independent seasonal timer induces neuroendocrine refractoriness to short day lengths.

The duration of nocturnal pineal melatonin secretion transduces effects of day length (DL) on the neuroendocrine axis of photoperiodic rodents. Long DLs support reproduction, and short DLs induce testicular regression, followed several months later by spontaneous recrudescence; gonadal regrowth is thought to reflect development of tissue refractoriness to melatonin. In most photoperiodic species, pinealectomy does not diminish reproductive competence in long DLs. Turkish hamsters (Mesocricetus brandti) deviate from this norm: elimination of melatonin secretion in long-day males by pinealectomy

Neuroendocrine regulation of sebocytes -- a pathogenetic link between stress and acne.

A causative link between emotional stress and acne has long been postulated. There is mounting evidence that the molecular mechanism underlying this observation is related to the expression of receptors for several neuroendocrine mediators by the sebaceous gland. Recent and ongoing studies have indicated that human sebocytes express functional receptors for corticotropin-releasing hormone, melanocortins, beta-endorphin, vasoactive intestinal polypeptide, neuropeptide Y and calcitonin gene-related peptide. After ligand binding, these receptors modulate the production of inflammatory cytokines,
