Aberrant Neurovascular Coupling in Diabetic Retinopathy Using Arterial Spin Labeling (ASL) and Functional Magnetic Resonance Imaging (fMRI) methods.

BACKGROUND: Previous imaging studies have demonstrated that diabetic retinopathy (DR) is linked to structural and functional abnormalities in the brain. However, the extent to which DR patients exhibit abnormal neurovascular coupling remains largely unknown. METHODS: Thirty-one patients with DR and 31 sex- and age-matched healthy controls underwent resting-state functional magnetic resonance imaging (rs-fMRI) to calculate functional connectivity strength (FCS) and arterial spin-labeling imaging (ASL) to calculate cerebral blood flow (CBF). The study compared CBF-FCS coupling across the entire

Hemodynamic responses in the rat hippocampus are simultaneously controlled by at least two independently acting neurovascular coupling mechanisms.

We combined electrical perforant pathway stimulation with electrophysiological and fMRI recordings in the hippocampus to investigate the effects of neuronal afterdischarges (nAD) on subsequent fMRI BOLD signals in the presence of isoflurane and medetomidine. These two drugs already alter basal hemodynamics in the hippocampus, with isoflurane being mildly vasodilatory and medetomidine being mildly vasoconstrictive. The perforant pathway was stimulated once for 8 seconds with either continuous 20 Hz pulses (continuous stimulation) or 8 bursts of 20 high-frequency pulses (burst stimulation). Burs

Partial wavelet coherence as a robust method for assessment of neurovascular coupling in neonates with hypoxic ischemic encephalopathy.

In neonates with hypoxic ischemic encephalopathy, the computation of wavelet coherence between electroencephalogram (EEG) power and regional cerebral oxygen saturation (rSO2) is a promising method for the assessment of neurovascular coupling (NVC), which in turn is a promising marker for brain injury. However, instabilities in arterial oxygen saturation (SpO2) limit the robustness of previously proposed methods. Therefore, we propose the use of partial wavelet coherence, which can eliminate the influence of SpO2. Furthermore, we study the added value of the novel NVC biomarkers for identificat

Cocaine attenuates blood flow but not neuronal responses to stimulation while preserving neurovascular coupling for resting brain activity.

Cocaine affects neuronal activity and constricts cerebral blood vessels, making it difficult to determine whether cocaine-induced changes in cerebral blood flow (CBF) reflect neuronal activation or its vasoactive effects. Here we assessed the effects of acute cocaine on both resting-state and stimulation responses to investigate cocaine's effects on neurovascular coupling and to differentiate its effects on neuronal activity from its vasoactive actions. We concurrently measured cortical field potentials via thinned-skull electroencephalography recordings and CBF with laser Doppler flowmetry in

Real-time monitoring of wavelet-based neurovascular coupling in neonates with hypoxic ischemic encephalopathy using an hourly time window.

SIGNIFICANCE: Real-time monitoring of neurovascular coupling (NVC) is crucial for early diagnosis and effective treatment strategies in neonates with hypoxic ischemic encephalopathy (HIE). In our previous studies, the NVC of neonates with HIE was determined using wavelet transform coherence (WTC) between the amplitude-integrated electroencephalogram (aEEG) and regional cerebral oxygen saturation ( SctO 2 ) using a post-acquisition analysis. AIM: We propose a time-resolved WTC analysis, providing a real-time analysis tool that facilitates immediate and continuous evaluation of cerebral he

Impairments of neurovascular coupling after stroke lower glymphatic system function and lead to depressive symptom: A longitudinal cohort study.

BACKGROUND: Respective changes in neurovascular coupling (NVC) and glymphatic function have been reported in post-stroke depression (PSD). Recent studies have found a link between NVC and waste clearance by the glymphatic system, which has not been illustrated in PSD. METHOD: We prospectively recruited ninety-six stroke patients and forty-four healthy controls (HC), with fifty-nine patients undergoing a second MRI scan. NVC metrics were investigated by exploring Pearson correlation coefficients and ratios between cerebral blood flow (CBF) and BOLD-derived quantitative maps (ALFF, fALFF, REHO

Do mean values tell the full story? Cardiac cycle and biological sex comparisons in temporally derived neurovascular coupling metrics.

Previous reports have noted cerebrovascular regulation differs across the cardiac cycle, with greater regulation occurring within systole. However, this methodological notion has not been meticulously scrutinized during temporally deduced neurovascular coupling (NVC) metrics with additional respect to biological sex. Analyses of 111 healthy individuals (40 females/71 males) were performed where participants engaged in the "Where's Waldo?" paradigm. All NVC parameters were quantified in the posterior and middle cerebral arteries at 310 unique timepoints. Several individuals completed repeat tes

Anomalous neurovascular coupling in patients with generalized anxiety disorder evaluated by combining cerebral blood flow and functional connectivity strength.

Coupling between neuronal activity and blood perfusion is termed neurovascular coupling, and it provides a new mechanistic perspective into understanding numerous brain diseases. Although abnormal brain activity and blood supply have been separately reported in generalized anxiety disorder (GAD), whether anomalous neurovascular coupling would still be presented in such disease is hitherto unknown. In this study, the neuronal activity and blood supply were measured using the functional connectivity strength (FCS) and cerebral blood flow (CBF). The voxel-wise CBF-FCS correlations and CBF/FCS rat

Alterations in resting cerebrovascular regulation do not affect reactivity to hypoxia, hyperoxia or neurovascular coupling following a SCUBA dive.

NEW FINDINGS: What is the central question of this study? What are the characteristics of cerebral blood flow (CBF) regulation following a single SCUBA dive to a depth of 18 m sea water with a 47 min bottom time. What is the main finding and its importance? Acute alterations in CBF regulation at rest, including extra-cranial vasodilatation, reductions in shear patterns and elevations in intra-cranial blood velocity were observed at rest following a single SCUBA dive. These subtle changes in CBF regulation did not translate into any functional changes in cerebrovascular reactivity to hypoxia or

Study of neurovascular coupling functions for transient focal cerebral ischemia in rats using electrocorticography functional photoacoustic microscopy (ECoG-fPAM).

Recently, the functional photoacoustic microscopy (fPAM) system has been proven to be a reliable imaging technique for measuring the total hemoglobin concentration (HbT), cerebral blood volume (CBV) and hemoglobin oxygen saturation (SO2) in single cerebral blood vessels of rats. In this study, we report for the first time the combination of electrocorticography (ECoG) recordings and fPAM (ECoG-fPAM) to investigate functional hemodynamic changes and neuro-vascular coupling in single cortical arterioles of rats with electrical forepaw stimulation after photothrombotic stroke. Treatment with the poly(ADP-ribose) polymerase inhibitor PJ-34 improves cerebromicrovascular endothelial function, neurovascular coupling responses and cognitive performance in aged mice, supporting the NAD+ depletion hypothesis of neurovascular aging.

Adjustment of cerebral blood flow (CBF) to neuronal activity via neurovascular coupling (NVC) plays an important role in the maintenance of healthy cognitive function. Strong evidence demonstrates that age-related cerebromicrovascular endothelial dysfunction and consequential impairment of NVC responses contribute importantly to cognitive decline. Recent studies demonstrate that NAD+ availability decreases with age in the vasculature and that supplemental NAD+ precursors can ameliorate cerebrovascular dysfunction, rescuing NVC responses and improving cognitive performance in aged mice. The mec

Variation within the visually evoked neurovascular coupling response of the posterior cerebral artery is not influenced by age or sex.

Neurovascular coupling (NVC) is the temporal and spatial coordination between local neuronal activity and regional cerebral blood flow. The literature is unsettled on whether age and/or sex affect NVC, which may relate to differences in methodology and the quantification of NVC in small sample-sized studies. The aim of this study was to 1) determine the relative and combined contribution of age and sex to the variation observed across several distinct NVC metrics (n = 125, 21-66 yr; 41 males) and 2) present an approach for the comprehensive systematic assessment of the NVC response using trans

Real-Time and High-Resolution NIR-II-L Imaging of Netrin-1-Mediated Neurovascular Coupling Driven by Dose-Dependent Electroacupuncture.

Accumulating evidence implicates that disrupted ovarian neurovascular coupling is a vital driver for polycystic ovary syndrome. Electroacupuncture (EA), as a form of peripheral neuromodulation, offers a potential therapeutic avenue to restore this coupling and improve ovarian function. However, how neural regulation leads to vascular changes remains unclear. Conventional histological and imaging methods cannot capture either immediate or long-term effects of EA on neurovascular dynamics, as they are limited by the lack of real-time and continuous in vivo visualization. Here, core-shell lanthan

Direct Evidence for the Feedforward Neurovascular Coupling Mechanism in Humans During Task Onset: An EEG-fNIRS-TCD Multimodal Imaging Study.

This investigation assessed the neurovascular coupling response through integrated assessments of neuronal function [electroencephalography (EEG)], microvascular oxygenation concentrations [functional near-infrared spectroscopy (fNIRS)], and arterial responses [transcranial Doppler ultrasound (TCD)]. The NVC response was assessed in 113 participants (86 females, aged 19-40 years) during visual ("Where's Waldo?") and motor (finger tapping) tasks. Block-averaged, time-frequency power was computed from the EEG data, while hemodynamic response functions were obtained from the fNIRS and TCD metrics

Changes in neurovascular coupling of cerebral blood flow and functional connectivity strength in first-episode schizophrenia patients after antipsychotic treatment.

Previous studies have identified abnormalities in neurovascular coupling in chronic schizophrenia patients based on functional connectivity strength (FCS) and cerebral blood flow (CBF). Pharmacological treatment remains the cornerstone of schizophrenia management. However, it remains unclear whether antipsychotic treatment influences these findings. This study aimed to investigate potential abnormalities in neurovascular coupling in patients with first-episode schizophrenia (FES) and to explore changes before (FES-pre) and after antipsychotic treatment (FES-post). Here, we recruited 133 FES pa

Neurovascular coupling is altered in women who have a history of brain injury from intimate partner violence: a preliminary study.

INTRODUCTION: Intimate partner violence (IPV) is a global health crisis with 30% of women over the age of 15 experiencing at least one event in their lifetime. Brain injury (BI) due to head impacts and/or strangulation is a common but understudied part of this experience. Previous research has shown BI from other injury mechanisms can disrupt neurovascular coupling (NVC). To gain further insight into whether similar changes occur in this population, we assessed NVC responses in women with a history of IPV-BI. METHODS: NVC responses were measured for the middle and posterior cerebral arteries
