A recombination chamber-based measurement system for radiation protection in neutron medical and research centers.

Recombination chambers, which operate based on ion recombination in gas and exhibit LET-dependent response, have been successfully used for many years in radiation protection to assess radiation quality (Q) and relative biological effectiveness (RBE). The REM-type chambers are a family of high-pressure, tissue-equivalent detectors designed for such applications. REM-2 and its successors, REM-3, represent successive generations of these chambers, with improvements in construction, electrode materials, gas filling, signal acquisition and mechanical design. The potential application of these dete

Cell death following BNCT: a theoretical approach based on Monte Carlo simulations.

In parallel to boron measurements and animal studies, investigations on radiation-induced cell death are also in progress in Pavia, with the aim of better characterisation of the effects of a BNCT treatment down to the cellular level. Such studies are being carried out not only experimentally but also theoretically, based on a mechanistic model and a Monte Carlo code. Such model assumes that: (1) only clustered DNA strand breaks can lead to chromosome aberrations; (2) only chromosome fragments within a certain threshold distance can undergo misrejoining; (3) the so-called "lethal aberrations"

Development of a switching-type fluorescence sensor for the detection of boronic acid-containing agents.

Since the therapeutic effect of boron neutron capture therapy is influenced by the intracellular distribution profile of boronoagents containing 10B atoms, it is necessary to establish a method that can determine the intracellular distribution profile of boronoagents. We aimed to develop a small molecule-based fluorescence sensor that changes its fluorescence properties upon complexation with the boronic acid moiety of a boronoagent. Thus, we designed a 2-(2-pyridyl)phenol derivative PPN-1 by introducing a N,O ligand substructure into a zinc sensor probe with excellent fluorescence properties.

Investigation on the efficacy of a tissue equivalent material bolus for BNCT.

BACKGROUND: In radiation therapy, the use of bolus is an effective technique for improving the surface dose. This irradiation technique is also used in BNCT. However, since BNCT uses neutron irradiation, it is important to evaluate both the neutron moderation characteristic and the radioactivation of the bolus. PURPOSE: This study aimed to evaluate the tissue equivalence and activation of commercially available boluses for use in BNCT. METHODS: Two types of commercially available boluses were evaluated. The boluses were placed on a water phantom and irradiated using the NeuCure BNCT system.

Boron tracedrug design for neutron dynamic therapeutics for LDL.

We describe our solution for removal of the low-density lipoprotein (LDL) depot contained in proteins and lipids as a 'druggable' target for atherosclerotic cardiovascular diseases by neutron dynamic therapy (NDT), which we developed using boron tracedrugs for NDT against bovine serum albumin as a model protein. Thus, we examined, among our developed boron tracedrugs, a boron-containing curcuminoid derivative UTX-51, to destroy freshly isolated human LDL dynamically under irradiated thermal neutron to obtain a decreased intensity of band of LDL treated with UTX-51 and thermal neutron irradiati

'Cell knife' for cancer: the clinician's perspective.

Boron Neutron Capture Therapy (BNCT), often referred to as the 'cell knife,' represents a binary, tumor-selective therapeutic modality that minimizes damage to surrounding healthy tissues. This review provides a comprehensive clinical perspective on BNCT, addressing the radiobiological mechanisms and summarizing related clinical trials, with a particular emphasis on glioma and head and neck cancers. Furthermore, the paper touches upon the synergistic potential of BNCT when integrated with other treatment modalities, such as proton and carbon ion radiotherapy, alternative neutron capture therap

High power accelerator-based boron neutron capture with a liquid lithium target and new applications to treatment of infectious diseases.

A new conceptual design for an accelerator-based boron neutron capture therapy (ABNCT) facility based on the high-current low-energy proton beam driven by the linear accelerator at SARAF (Soreq Applied Research Accelerator Facility) incident on a windowless forced-flow liquid-lithium target, is described. The liquid-lithium target, currently in construction at Soreq NRC, will produce a neutron field suitable for the BNCT treatment of deep-seated tumor tissues, through the reaction (7)Li(p,n)(7)Be. The liquid-lithium target is designed to overcome the major problem of solid lithium targets, nam

First experimental demonstration of Compton imaging of10B at clinical concentrations for real-time dosimetry in BNCT.

Objective.Boron neutron capture therapy (BNCT) requires accurate knowledge of the boron distribution during treatment to enable reliable dosimetry and treatment verification. Compton imaging of the 478 keV prompt gamma rays emitted following neutron capture byB has been proposed as a promising technique for real-time boron monitoring. This work aims to experimentally evaluate the feasibility of Compton imaging under clinically relevant boron concentrations and realistic neutron-induced background conditions.Approach.A dedicated experimental campaign was performed at the Institut Laue-Langevin

The role of radiolabeling in BNCT tracers for enhanced dosimetry and treatment planning.

Positron emission tomography (PET) and single photon emission computed tomography (SPECT) are potent technologies for non-invasive imaging of pharmacological and biochemical processes in both preclinical and advanced clinical research settings. In the field of radiation therapy, boron neutron capture therapy (BNCT) stands out because it harnesses biological mechanisms to precisely target tumor cells while preserving the neighboring healthy tissues. To achieve the most favorable therapeutic outcomes, the delivery of boron-enriched tracers to tumors must be selective and efficient, with a substa

The hydroboration reaction as a key for a straightforward synthesis of new MRI-NCT agents.

In this study the hydroboration reaction has been exploited to produce in only four steps a new lipophilic GdBNCT/MRI agent (PB01). As a matter of fact, the formation of a new B–C bond to link the decaborane with the lipophilic moiety greatly simplifies the synthesis of PB01 with respect to the previously reported dual agents. The complexes obtained (PB01a and PB01b) have been fully characterised from the relaxometric point of view and, after disaggregation with HPβCD, both isomers display high affinity for low density lipoproteins (LDLs) that can be exploited as specific carriers of these the

Uptake and Retention of Laser-synthesized Boron Nanoparticles in Tumor Cells and Fibroblasts.

Boron Neutron Capture Therapy (BNCT) is one of the innovative methods for treating oncological diseases. Its selectivity is based on the targeted delivery of the boron-10 isotope to tumor cells, followed by neutron irradiation, the 10B(n, α)7Li reaction occurs with a local release of 2.79 MeV of energy. Budding boron delivery agents are nanoscale systems. This study evaluated in vitro cytotoxicity, accumulation, and retention of elemental boron nanoparticles, synthesized by laser ablation and laser fragmentation, in U87 and BT474 tumor cells and BJ-5ta fibroblasts. It was shown that both types

EGFR-Targeted Cellular Delivery of Therapeutic Nucleic Acids Mediated by Boron Clusters.

New boron carriers with high boron content and targeted cancer-cell delivery are considered the first choice for boron neutron capture therapy (BNCT) for cancer treatment. Previously, we have shown that composites of antisense oligonucleotide and boron clusters are functional nanoparticles for the downregulation of expression of epidermal growth factor receptor (EGFR) and can be loaded into EGFR-overexpressing cancer cells without a transfection factor. In this study, we hypothesize that free cellular uptake is mediated by binding and activation of the EGFR by boron clusters. Proteomic analysi

Development of a thick CdTe detector for BNCT-SPECT.

As well known, it is difficult to know the exact treatment effect of boron neutron capture therapy (BNCT). It depends on the irradiation time, which is changed rather flexibly. At present, it is once fixed before BNCT. Then the actual stopping time is adjusted during BNCT by some means like activation foils. The author's group hence started development of a single-photon emission computed tomography (SPECT) system for BNCT to know the effect of BNCT in real time. By adopting a side surface (1×2 mm(2)) of a CdTe detector (1×2×20 mm(3)) as radiation entrance window, acceptable spatial resolution

Relative biological effectiveness of an accelerator-based BNCT system coupled to a solid-state lithium target: two different approaches for neutron beams.

The relative biological effectiveness (RBE) of neutrons in neutron beams is crucial for the clinical implementation of accelerator-based boron neutron capture therapy (BNCT) systems. The RBE was quantified by comparing the doses required to achieve a 10 % cell survival fraction (D10) between reference radiation (photons) and neutrons. However, in accelerator-based BNCT, the neutron beam includes not only neutrons but also contaminating gamma rays, making it essential to calculate the RBE of neutrons while accounting for the gamma-ray dose. The RBE of neutrons was calculated using a recently pr

Inhibition of growth of human breast cancer cells in culture by neutron capture using liposomes containing 10B.

Cell destruction in boron neutron capture therapy is effected by nuclear reaction between 10B and thermal neutrons with the release of alpha-particles (4He) and lithium-7 ions (7Li). 4He kills cells within 10 microm of the site of 4He generation, therefore it is theoretically possible to destroy tumour cells without affecting adjacent healthy tissue, given selective delivery of compounds containing 10B. Liposomes wore prepared by vortex dispersion of solutions containing 10B compounds with dried lipid films and the effects of those compounds on human breast cancer cells in culture were examine

Investigating the Toxicity and Biodistribution of Boron-loaded Liposomes in Canine and Feline Models.

Boron neutron capture therapy (BNCT) is a targeted cancer treatment that relies on the selective accumulation of boron-containing agents in tumor cells, followed by irradiation with low-energy neutrons to produce a cytotoxic reaction. Liposome-mediated delivery systems have shown promise in improving the biodistribution and tumor specificity of boron agents in mice. This study aimed to evaluate the toxicity of boron-loaded liposomes (MAC/TAC formulations) in canine and feline models and to measure biodistribution in dogs with naturally occurring tumors. Boron-loaded liposomes were prepared usi

Preliminary clinical outcomes of accelerator-based BNCT for head and neck cancer.

Boron Neutron Capture Therapy (BNCT) is a type of particle beam therapy that uses particles produced by the boron neutron capture reaction to selectively destroy cancer cells. The high LET radiation produced by the nuclear reaction between thermal neutrons and boron atoms that has been taken up in cancer tissue can selectively destroy cancer cells and is expected to achieve a considerable antitumor effect even on solid tumors with low radiation sensitivity. In recent years, progress has been made in the development of accelerators specifically for use as a neutron source in BNCT, and accelerat

Combining BNCT with carbonic anhydrase inhibition for mesothelioma treatment: Synthesis, in vitro, in vivo studies of ureidosulfamido carboranes.

Mesothelioma is a malignant neoplasm of mesothelial cells caused by exposure to asbestos. The average survival time after diagnosis is usually nine/twelve months. A multi-therapeutic approach is therefore required to treat and prevent recurrence. Boronated derivatives containing a carborane cage, a sulfamido group and an ureido functionality (CA-USF) have been designed, synthesised and tested, in order to couple Boron Neutron Capture Therapy (BNCT) and the inhibition of Carbonic Anhydrases (CAs), which are overexpressed in many tumours. In vitro studies showed greater inhibition than the refer
