Understanding the Crucial Role of Phosphate and Iron Availability in Regulating Root Nodule Symbiosis.

The symbiosis between legumes and nitrogen-fixing bacteria (rhizobia) is instrumental in sustaining the nitrogen cycle and providing fixed nitrogen to the food chain. Both partners must maintain an efficient nutrient exchange to ensure a successful symbiosis. This mini-review highlights the intricate phosphate and iron uptake and homeostasis processes taking place in legumes during their interactions with rhizobia. The coordination of transport and homeostasis of these nutrients in host plants and rhizobia ensures an efficient nitrogen fixation process and nutrient use. We discuss the genetic

Continuous Cropping Differentially Regulates Nitrogen Metabolism and Nodulation in Soybean Cultivars with Contrasting Continuous Cropping Tolerance Under Varied Nitrogen Levels.

Continuous cropping obstacles severely restrict soybean production; nitrogen application can alleviate continuous cropping obstacles. However, it remains unclear how nitrogen metabolism and nodulation differ between soybean cultivars with contrasting continuous cropping tolerance under varied nitrogen levels. In this study, two soybean cultivars, Liaodou 14 (L14, continuous-cropping-tolerant) and Liaodou 10 (L10, continuous-cropping-sensitive), were cultivated in a sand culture system. Three nitrogen levels, low (LN), medium (MN), and high (HN), were set with three soybean soil extract treatme

Ca(H2PO4)2 and MgSO4 activated nitrogen-related bacteria and genes in thermophilic stage of compost.

This study was conducted to investigate the effects of Ca(H2PO4)2 and MgSO4 on the bacterial community and nitrogen metabolism genes in the aerobic composting of pig manure. The experimental treatments were set up as control (C), 1% Ca(H2PO4)2 + 2% MgSO4 (CaPM1), and 1.5% Ca(H2PO4)2 + 3% MgSO4 (CaPM2), which were used at the end of composting for potting trials. The results showed that Ca(H2PO4)2 and MgSO4 played an excellent role in retaining nitrogen and increasing the alkali-hydrolyzed nitrogen (AN), available phosphorus (AP), and available potassium (AK) contents of the composts. Adding Ca

The Trichodesmium consortium: conserved heterotrophic co-occurrence and genomic signatures of potential interactions.

The nitrogen (N)-fixing cyanobacterium Trichodesmium is globally distributed in warm, oligotrophic oceans, where it contributes a substantial proportion of new N and fuels primary production. These photoautotrophs form macroscopic colonies that serve as relatively nutrient-rich substrates that are colonized by many other organisms. The nature of these associations may modulate ocean N and carbon (C) cycling, and can offer insights into marine co-evolutionary mechanisms. Here we integrate multiple omics-based and experimental approaches to investigate Trichodesmium-associated bacterial consorti

A novel recombinant PHB production platform in filamentous cyanobacteria avoiding nitrogen starvation while preserving cell viability.

During the past decades, the importance of developing sustainable, carbon dioxide (CO2)-neutral and biodegradable alternatives to conventional plastic has become evident in the context of global pollution issues. Therefore, heterotrophic bacteria such as Cupriavidus sp. have been intensively explored for the synthesis of the biodegradable polymer polyhydroxybutyrate (PHB). PHB is also naturally produced by a variety of phototrophic cyanobacteria, which only need sunlight and CO2, thereby allowing a CO2 negative, eco-friendly synthesis of this polymer. However, a major drawback of the use of cy

Ridge with no-tillage facilitates microbial N2 fixation associated with methane oxidation in rice soil.

Aerobic methane-oxidizing bacteria (MOB) play a crucial role in mitigating the greenhouse gas methane emission, particularly prevalent in flooded wetlands. The implementation of ridge with no-tillage practices within a rice-rape rotation system proves effective in overcoming the restrictive redox conditions associated with waterlogging. This approach enhances capillary water availability from furrows, especially during periods of low rainfall, thereby supporting plant growth on the ridges. However, the microbe-mediated accumulation of soil organic carbon and nitrogen remains insufficiently und

Regulatory mechanisms of CH4 : air volume ratios on metabolic flux partitioning in methane-oxidizing bacteria and their impact on single cell protein biosynthetic efficiency.

INTRODUCTION: C1 gas bioconversion for single-cell protein (SCP) production offers dual environmental benefits by mitigating greenhouse gases and generating protein resources. This study systematically determined optimal methane-to-air ratios (CH4:air, v/v) for enhancing methane-oxidizing bacteria (MOB) growth and SCP yield under three distinct nitrogen assimilation modes: nitrate-driven pMMO expression, ammonium-driven sMMO expression, and nitrogen-fixing sMMO expression. METHODS: Experiments were conducted under three nitrogen assimilation regimes: nitrate-fed pMMO expression, ammonium-fed

Rhizobia block Cd absorption in peanut via rhizosphere microbiome assembly and N-regulated host defense.

Cadmium (Cd) contamination severely threatens peanut (Arachis hypogaea L.) production and symbiotic nitrogen fixation (SNF). Although rhizobia inoculation can alleviate heavy metal toxicity in plants, how nitrate (NO3-) regulates Cd translocation and the defense of the "rhizobia-root-nodule" system remains unclear. Here, we investigated the mechanisms by which the peanut rhizobium, Rhizobium sp. HM13, mitigates Cd toxicity and sustains SNF under varying NO3- levels (N- and N+) via field and pot trials. Field trials showed that HM13 reduced rhizosphere bioavailable Cd and decreased seed Cd accu

Metagenomic Insights into the Functional Profiles of Carbon, Nitrogen, and Phosphorus Cycles in Yuncheng Salt Lake Under Different Salinity Gradients.

Salinity is a key driver of microbial community structure and function in salt lake ecosystems, yet how it shapes functional genes involved in carbon (C), nitrogen (N), and phosphorus (P) cycling remains poorly understood. We collected metagenomic samples along a natural salinity gradient in Yuncheng Salt Lake and examined how salinity was associated with microbial taxonomic and functional diversity and with C, N, and P cycling genes. Both diversity metrics decreased significantly with increasing salinity and were positively correlated with each other. The composition and abundance of C, N, an

A denitrifying community associated with a major, marine nitrogen fixer.

The diazotrophic cyanobacterium, Trichodesmium, is an integral component of the marine nitrogen cycle and contributes significant amounts of new nitrogen to oligotrophic, tropical/subtropical ocean surface waters. Trichodesmium forms macroscopic, fusiform (tufts), spherical (puffs) and raft-like colonies that provide a pseudobenthic habitat for a host of other organisms including marine invertebrates, microeukaryotes and numerous other microbes. The diversity and activity of denitrifying bacteria found in association with the colonies was interrogated using a series of molecular-based methodol

Holed up, but thriving: Impact of multitrophic cryoconite communities on glacier elemental cycles.

Cryoconite holes (water and sediment-filled depressions), found on glacier surfaces worldwide, serve as reservoirs of microbes, carbon, trace elements, and nutrients, transferring these components downstream via glacier hydrological networks. Through targeted amplicon sequencing of carbon and nitrogen cycling genes, coupled with functional inference-based methods, we explore the functional diversity of these mini-ecosystems within Antarctica and the Himalayas. These regions showcase distinct environmental gradients and experience varying rates of environmental change influenced by global clima

Anion inhibition studies of two α-carbonic anhydrases from Lotus japonicus, LjCAA1 and LjCAA2.

The model organism for the investigation of symbiotic nitrogen fixation in legumes Lotus japonicus encodes two carbonic anhydrases (CAs, EC 4.2.1.1) belonging to the α-class, LjCAA1 and LjCAA2. Here we report the kinetic characterization and inhibition of these two CAs with inorganic and complex anions and other molecules interacting with zinc proteins, such as sulfamide, sulfamic acid, and phenylboronic/arsonic acids. LjCAA1 showed a high catalytic activity for the CO2 hydration reaction, with a k(cat) of 7.4∗10(5) s(-1) and a k(cat)/K(m) of 9.6∗10(7) M(-1) s(-1) and was inhibited in the low

Novel pennate diatom symbionts support high N2 fixation rates.

Diazotrophy is the most important nitrogen source in the oligotrophic surface ocean, but the organisms involved and their contributions are incompletely understood due to limited observations. Only diazotrophic organisms possess the nifH gene to reduce dinitrogen to ammonium, but their distribution and activity can only be quantified through sampling and experiments during research cruises. Some recent studies document small diatoms with symbionts able to fix nitrogen, a new source of biologically available nitrogen in addition to the well-known cyanobacterial species such as Trichodesmium or

Calcite precipitation by the nitrogen-fixing cyanobacterium Anabaena sp. ATCC 33047.

Microbiologically induced calcium carbonate precipitation (MICP) is an ecologically vital process where the metabolic activity of microorganisms causes the precipitation of calcium carbonate. In cyanobacteria, MICP is thought to occur primarily due to photosynthesis. During this process, cyanobacteria consume carbon dioxide, raising the local pH, which leads to conditions favorable to calcium carbonate precipitation. However, how individual cells contribute to MICP is poorly understood. Here, we use quantitative microscopy to obtain direct evidence of MICP caused by the filamentous cyanobacter

Effect of flumetsulam alone and coexistence with polyethylene microplastics on soil microbial carbon and nitrogen cycles: Elucidation of bacterial community structure, functional gene expression, and enzyme activity.

Flumetsulam (FLU) is a new class of broad-spectrum herbicides. With the widespread use of plastic products, polyethylene (PE) microplastics (MPs) may remain in the soil. It is possible for these two novel contaminants to co-exist in the soil environment. In the present study, we used brown soil as the test soil and determined the toxicity of FLU at 0.05, 0.5 and 2.5 mg kg-1 alone and in combination with PE MPs (1%) on soil microorganisms. The obtained results demonstrated that the exposure of FLU and FLU+MPs had an inhibitory effect on the numbers of bacteria and fungi. In addition, FLU and FL

Nitrogenase Cofactor Maturase NifB Isolated from Transgenic Rice is Active in FeMo-co Synthesis.

The engineering of nitrogen fixation in plants requires assembly of an active prokaryotic nitrogenase complex, which is yet to be achieved. Nitrogenase biogenesis relies on NifB, which catalyzes the formation of the [8Fe-9S-C] metal cluster NifB-co. This is the first committed step in the biosynthesis of the iron-molybdenum cofactor (FeMo-co) found at the nitrogenase active site. The production of NifB in plants is challenging because this protein is often insoluble in eukaryotic cells, and its [Fe-S] clusters are extremely unstable and sensitive to O2. As a first step to address this challeng

De Novo Designed Porphyrin-Binding Proteins Photocatalyze the Nitrogen Reduction Reaction.

In this work, we de novo designed a two-helix α-helical protein capable of forming dimers, which features a central cavity that accommodates porphyrin molecules. The resulting porphyrin-binding protein, formed by mixing the porphyrin with the dimer protein, exhibits visible-light-driven catalytic activity for the nitrogen reduction reaction. To further optimize the protein, we selected three mutation layers and predicted their structures. From these, we identified seven mutants that retained the original protein folding structure, which are used for the photocatalytic ammonia production from n
