Spatial variation and availability of nutrients at an oyster reef in relation to submarine groundwater discharge.

Submarine groundwater discharge (SGD) is often an overlooked component of the solute budgets in coastal embayments and is not considered in oyster restoration, conservation, and management plans. A combination of spatial and temporal geochemistry (nutrients, trace metals, alkalinity components, stable isotopes, and major ions) of porewater and surface water combined with SGD-derived solute fluxes and turnover times was used to examine the significance of SGD in delivering nutrients to paleovalley systems in coastal embayments, thus sustaining the health and productivity of oyster reefs. A 1-km

The NO+O3 reaction: a triple oxygen isotope perspective on the reaction dynamics and atmospheric implications for the transfer of the ozone isotope anomaly.

Atmospheric nitrate shows a large oxygen isotope anomaly (Delta 17 O), characterized by an excess enrichment of 17 O over 18 O, similar to the ozone molecule. Modeling and observations assign this specific isotopic composition mainly to the photochemical steady state that exists in the atmosphere between ozone and nitrate precursors, namely, the nitrogen oxides (NOx=NO+NO2). However, this transfer is poorly quantified and is built on unverified assumptions about which oxygen atoms of ozone are transferred to NO(x), greatly weakening any interpretation of the nitrate oxygen isotopic composition

Defying Dietary Homogeneity: Stable Isotopes in Hair Reveal Persistent Geographic Heterogeneity in Argentina's Modern Diet.

OBJECTIVES: The main objective of the study is to characterize and test the hypothesis of a continuing trend toward homogenization in contemporary Argentine diet through stable isotope analysis, exploring regional variation in the context of globalized food systems. It is expected that the distribution of stable isotopes will exhibit a limited range of values but still display some heterogeneity that allows differentiation of dietary patterns by region. METHODS: A total of 204 hair samples were collected from volunteer donors in Argentina and analyzed for the stable isotopes δ13C, δ15N, and δ

Reactions of nitrogen oxides with heme models. Spectral and kinetic study of nitric oxide reactions with solid and solute Fe(III)(TPP)(NO3).

The reaction(s) of nitric oxide (nitrogen monoxide) gas with sublimed layers containing the nitrato iron(III) complex Fe(III)(TPP)(eta(2)-O(2)NO) (1, TPP = meso-tetraphenyl porphyrinate(2)(-)) leads to formation of several iron porphyrin species that are ligated by various nitrogen oxides. The eventual products of these low-temperature solid-state reactions are the nitrosyl complex Fe(TPP)(NO), the nitro-nitrosyl complex Fe(TPP)(NO(2))(NO), and 1 itself, and the relative final quantities of these were functions of the NO partial pressure. It is particularly notable that isotope labeling experi

Patterns and causes of ammonia nitrogen enrichment in groundwater of a typical agricultural irrigation area in the northern Henan Plain, China.

Groundwater ammonium (NH4+) enrichment in agricultural plains is often attributed to anthropogenic pollution, yet geogenic contributions remain poorly understood. This study investigates NH4+ distribution and genesis in the North Henan Plain, China, using hydrochemical, isotopic, and sediment analyses. Results show that NH4+ concentrations increase with depth in the unconfined aquifer, while confined aquifers exhibit localized enrichment within clay lenses. Dual-nitrate isotopes (δ15N-NO3 and δ18O-NO3) reveal significant anthropogenic nitrogen inputs that are effectively attenuated by denitrif

In situ monitoring of internal water storage reveals nitrogen first flush phenomena, intermittent denitrification, and seasonal ammonium flushing.

Internal water storage (IWS) can be included in bioretention practices to increase storage capacity or promote denitrification-the microbial reduction of nitrate to nitrogen gas. IWS and nitrate dynamics are well studied in laboratory systems. However, the investigation of field environments, consideration of multiple nitrogen species, and determination between mixing versus denitrification is lacking. This study employs in situ monitoring (∼24 h duration) of water level, dissolved oxygen (DO), conductivity, nitrogen species, and dual isotopes of a field bioretention IWS system for nine storms

In vivo urea cycle flux distinguishes and correlates with phenotypic severity in disorders of the urea cycle.

Urea cycle disorders are a group of inborn errors of hepatic metabolism that result in often life-threatening hyperammonemia and hyperglutaminemia. Clinical and laboratory diagnosis of partial deficiencies during asymptomatic periods is difficult, and correlation of phenotypic severity with either genotype and/or in vitro enzyme activity is often imprecise. We hypothesized that stable isotopically determined in vivo rates of total body urea synthesis and urea cycle-specific nitrogen flux would correlate with both phenotypic severity and carrier status in patients with a variety of different en

Continuous Diffusion Model for Concentration Dependence of Nitroxide EPR Parameters in Normal and Supercooled Water.

Electron paramagnetic resonance (EPR) spectra of radicals in solution depend on their relative motion, which modulates the Heisenberg spin exchange and dipole-dipole interactions between them. To gain information on radical diffusion from EPR spectra demands both reliable spectral fitting to find the concentration coefficients of EPR parameters and valid expressions between the concentration and diffusion coefficients. Here, we measured EPR spectra of the 14N- and 15N-labeled perdeuterated TEMPONE radicals in normal and supercooled water at various concentrations. By fitting the EPR spectra to

Grazing reshapes plant nitrogen use trade-off between soil uptake and internal remobilization.

Perennial plants rely on two key ecological strategies to supply nitrogen (N) for photosynthetic tissue construction: root N remobilization (internal cycling) and soil N uptake. How plants trade-off these strategies determines the economy of N use and is likely context-dependent. Yet, it is rarely quantified in situ across growth stages and under varying management, limiting the mechanistic understanding of how plants rebalance N-use trade-offs to adapt to grazing disturbance. To address this gap, we conducted an in situ experiment using 15N isotope labelling techniques in a semiarid grassland

The Assimilation of Diazotroph-Derived Nitrogen by Scleractinian Corals Depends on Their Metabolic Status.

UNLABELLED: Tropical corals are associated with a diverse community of dinitrogen (N2)-fixing prokaryotes (diazotrophs) providing the coral an additional source of bioavailable nitrogen (N) in oligotrophic waters. The overall activity of these diazotrophs changes depending on the current environmental conditions, but to what extent it affects the assimilation of diazotroph-derived N (DDN) by corals is still unknown. Here, in a series of 15N2 tracer experiments, we directly quantified DDN assimilation by scleractinian corals from the Red Sea exposed to different environmental conditions. We sho

6Li and 7li solid-state NMR spectroscopy of nitrogen ceramic phases.

Two nitrogen ceramic phases, the oxynitride LiSiON and the nitride LiSi2N3, have been studied by 6Li and 7Li NMR. Magic angle spinning (MAS) NMR experiments have been carried out at two magnetic field strengths (7.05 and 14.1 T). The spectra give evidence of the relative effects of the quadrupolar and chemical shift interactions. The electric field gradient tensor of both phases has been determined accurately by iterative fitting of the 6Li and 7Li MAS NMR line shapes at the two magnetic field strengths. Due to the fact that for 7Li the quadrupolar interaction is much larger than the chemical

Nitrogen in surface aquifer - Coastal lagoons systems: Analyzing the origin of eutrophication processes.

Coastal lagoons can act as sinks and sources of a wide range of substances, including nutrients, and pollutants. In these ecosystems, primary production is limited more by nitrogen than by phosphorus. For this reason, they are significantly vulnerable to nitrate pollution. In this study, a joined analysis of surface and ground water was conducted to determine the origin, occurrence and processes affecting nitrogen fate in a Mediterranean coastal aquifer-lagoon system. This included the analysis of water levels, as well as hydrochemical and isotopes data evolution for a 4-year period, which rev

Stable Isotope Analysis (δ13C, δ15N and δ18O) of Killer Whale Dentine: Evaluating Treatment Methods and Establishing Best Practices.

RATIONALE: Stable isotope measurements in odontocete teeth can be used to reconstruct lifetime ecology, but some pre-treatments may introduce bias. Isolating dentine collagen with HCl decalcification, removing the organic matrix or drying dentine prior to the analysis of structural carbonates can introduce unintended variability, limiting comparability among studies and populations. This study tested pre-treatment methods to quantify their effects. METHODS: Killer whale teeth were sectioned, annual growth layers microdrilled, and powder allocated to treatments. For collagen δ13C and δ15N, 75

Comparison of the Dual Isotope Tracer Approach with Oro-Ileal Balance Method for Determination of Amino Acid Digestibility in Cannulated Pigs.

BACKGROUND: The dual isotope tracer approach was developed as a less invasive alternative for the measurement of ileal amino acid (AA) digestibility. OBJECTIVES: This study aimed to validate the dual isotope tracer approach with the standardized oro-ileal balance method in growing pigs. METHODS: Eight pigs were fitted with jugular catheters and ileal T-cannulas. On the test day, feed containing intrinsically labeled 15N-milk protein and 13C-spirulina was provided every half hour for 240 min. Ileal digesta and 10 jugular blood samples were collected between 0 min and 540 min. Digesta samples

Discerning animal-sourced food in diet using isotope analysis of human scalp hair and fingernails.

PURPOSE: Diet-related diseases are advancing as the leading cause of death globally. As self-reporting of diet by patients can be associated with errors, stable isotopes of human tissues can be used to diagnose diseases, understand physiology, and detect change in diet. This study investigates the effect of type and amount of food on the nitrogen and carbon concentration (Nconc and Cconc) and isotopic composition (δ15N and δ13C) in human scalp hair and fingernails. METHODS: A total of 100 residents participated in the study whereas only 74 individuals provided complete diet history. Sixty-six

13C and 15N NMR identification of product compound classes from aqueous and solid phase photodegradation of 2,4,6-trinitrotoluene.

Photolysis is one of the main transformation pathways for 2,4,6-trinitrotoluene (TNT) released into the environment. Upon exposure to sunlight, TNT is known to undergo both oxidation and reduction reactions with release of nitrite, nitrate, and ammonium ions, followed by condensation reactions of the oxidation and reduction products. In this study, compound classes of transformation products from the aqueous and solid phase photodegradation of 2,4,6-trinitrotoluene (TNT) have been identified by liquid and solid state 13C and 15N NMR. Aqueous phase experiments were performed on saturated soluti

Quantitation of the relative amounts of anhydrous carbamazepine (C15H12N2O) and carbamazepine dihydrate (C15H12N2O.2H2O) in a mixture by solid-state nuclear magnetic resonance (NMR).

The application of solid state nuclear magnetic resonance (NMR) for the quantitation of the relative amounts of carbamazepine anhydrate (I) and carbamazepine dihydrate (II) in a mixture is presented. The techniques of cross polarization, dipolar decoupling, and magic angle spinning have been used to obtain high-resolution NMR spectra of the samples in the solid state. Although the chemical shifts of I and II were similar, the proton spin lattice relaxation time of II was much shorter than that of I. A delay time of 10 sec between pulses resulted in saturation of the signal from I and in a spec

Tryptophanol enhances nitrogen assimilation in marine diatoms.

Diatoms exhibit high competitive capacity in nitrogen assimilation, but the underlying mechanisms remain unclear. Here, we identify a non-ribosomal peptide synthase-like gene (PtNRPS1) with an atypical domain structure (A-T-R1-R2) in the marine diatom Phaeodactylum tricornutum, crucial for short-term nitrogen assimilation. In vitro enzyme assays show PtNRPS1 catalyzes conversion of L-tryptophan to tryptophanol, a tryptophan-derived indole compound that promotes diatom growth at concentrations far lower than indole-3-acetic acid. Transcriptomic, metabolomic analyses, and stable-isotope analyses

Micro-aeration unlocks endogenous nitrogen removal in anaerobic digesters via metabolically versatile fermentative bacteria.

Ammonia inhibition represents a significant bottleneck in the anaerobic digestion (AD) of high-nitrogen organic wastes. This study employed a batch reactor system subjected to alternating carbon-sufficient and carbon-limited phases to investigate whether pulsed micro-aeration, with a peak oxidation-reduction potential ranging from -150 to -50 mV, could promote the conversion of total ammonia nitrogen (TAN) to N2 in anaerobic digesters maintained under bulk anaerobic conditions, while simultaneously elucidating the competitive interplay between this nitrogen removal process and methanogenesis.
