Effects of annetocin, an oxytocin-related peptide isolated from the earthworm Eisenia foetida, and some putative neurotransmitters on gut motility of the earthworm.

Annetocin, an oxytocin-related peptide recently isolated from the lumbricid earthworm Eisenia foetida, and putative transmitter substances were examined for their effects on rhythmic, spontaneous contractions of isolated gut preparations of the earthworm. Significant, dose-dependent effects of the following substances were observed: acetylcholine (ACh), gamma-aminobutyric acid (GABA), and dopamine were excitatory, while serotonin (5-HT) and octopamine were inhibitory. Annetocin, oxytocin, and vasotocin stimulated spontaneous contraction of the earthworm gut, annetocin being approximately 10-fo

Serotonin-containing neurons in lobsters: origins and characterization of inhibitory postsynaptic potentials.

1. The serotonin-containing neurons in the A1 ganglion of the lobster have been shown to act as "gain setters" in neuronal circuits that control the adoption of behaviorally relevant postures. These neurons are subject to tonic inhibition, which has been proposed as an important regulator of their activity. This study explores the pharmacological nature and anatomic location of the neurons responsible for inhibition of these A1 cells; the role played by inhibitory inputs in controlling the firing rates of these neurons is also examined. 2. Three classes of inhibitory postsynaptic potentials (I

Pharmacological rescue of isolation-induced social and neurochemical deficits in Drosophila melanogaster.

Social isolation (ISO) is a major risk factor for mood disorders, reflected in disrupted neurochemistry and social behavior. To test whether five days of ISO induce a depression-like state in male Drosophila melanogaster, we measured brain monoamines and social interaction networks (SINs), focusing on dopamine (DA) and octopamine (OA), key regulators of motivation and reward. ISO reduced DA, OA, and tyramine (TA) while increasing acetylcholine, and produced disorganized SINs marked by weaker, shorter interactions, reduced clustering and centrality, and hyperactive, uncoordinated locomotion. Quantitative analysis of neurotransmitters in fish brain: A tool to assess neurochemical effects of psychoactive pollutants.

Psychoactive drugs in fish act through mechanisms similar to those in humans because their main neurotransmitter (NT) systems are analogous. Psychoactive drugs are ubiquitous in aquatic environments, having negative effects on fish and other aquatic organisms. To understand the neurochemical changes underlying the effects, studying neurotransmitter levels is essential. The detection of NTs and their metabolites in biological samples may be challenging. A new analytical method using hydrophilic interaction liquid chromatography coupled to high-resolution mass spectrometry was developed. The met

Vasoactive factors in the mechanism of renal sodium handling in cirrhotics: the effect of acute plasma expansion.

Twenty cirrhotic patients with ascites, divided into two groups of 10 each, according to their daily urinary sodium excretion (sodium retainers and sodium excretors) and given a diet of 75 mEq of sodium daily, underwent acute plasma volume expansion with 1,000 ml of 10% dextran in saline, infused through a catheter located in the right atrium. Even if a significant increase in sodium excretion was observed in both groups (p less than 0.001 in sodium excretors and p less than 0.05 in sodium retainers), plasma expansion did not reverse sodium retention in sodium retainers. A significant increase

Optogenetic Control of Activity in Descending Tdc2+ Neurons Modulates Motor Program Bias in the Drosophila Larval Locomotor System.

Motor systems must flexibly select between competing outputs while preserving stability of rhythmic outputs. In Drosophila larvae, the isolated central nervous system is capable of maintaining rhythmicity by generating multiple different fictive motor programs. The biogenic amines octopamine and tyramine are known to regulate larval locomotion, however, how the tdc2+ octopaminergic/tyraminergic system regulates motor program competition is not well understood. Here, we combine dual-colour calcium imaging and optogenetic manipulation to explore how tdc2+ neurons track, permit, and bias fictive

[Clonidine for the treatment of heroin withdrawal syndrome].

In an open study of 50 patients with heroin addiction, clonidine was efficacious by mouth in the treatment of acute heroin-withdrawal syndrome. Mean administration of clonidine (on an "as-needed" basis) was 5 days (maximum 7 days), whilst the mean daily dosage ranged from 0.112 mg to 0.468 mg, the maximum requirement occurring on day 2 of withdrawal and sinking thereafter. Since insomnia was not influenced by clonidine, we offered 100 mg doxepine and/or 10 mg nitrazepam (the latter only until day 4 of treatment). Under this medication a sudden, dramatic decrease in blood pressure was not seen,

High-dimensional mediation analysis to elucidate the role of metabolites in the association between PFAS exposure and reduced SARS-CoV-2 IgG in pregnancy.

We previously found that per- and polyfluoroalkyl substances (PFAS) mixture exposure is inversely associated with SARS-CoV-2 IgG (IgG) antibody levels in pregnant individuals. Here, we aim to identify metabolites mediating this relationship to elucidate the underlying biological pathways. This cross-sectional study included 59 pregnant participants from a US-based pregnancy cohort. Untargeted metabolomic profiling was performed using Liquid Chromatography-High Resolution Sass spectrometry (LC-HRMS), and weighted Quantile Sum (WQS) regression was applied to assess the PFAS and metabolites mixtu

An Overview of Natural Plant Products in the Treatment of Hepatocellular Carcinoma.

BACKGROUND: Liver cancer is the fifth most commonly diagnosed cancer and the second leading cause of cancer-related deaths worldwide. Among the liver cancers, hepatocellular carcinoma has been reported to be responsible for 85-90% of primary liver cancer and it is the second most common cause of cancer mortality with 700,000 deaths documented annually. The major risk factors of HCC include chronic infections with the hepatitis B (HBV) or hepatitis C (HCV) virus, chronic liver diseases, alcoholism as well as dietary carcinogens, such as aflatoxins. Highest incidence rates are estimated to occur

Possible mechanisms of weight loss of Siberian hamsters (Phodopus sungorus sungorus) exposed to short photoperiod.

Several weeks of short day photoperiod (SD) exposure promote a dramatic decrease of white adipose tissue (WAT) mass in Siberian hamsters(Phodopus sungorus sungorus). This slimming effect is accompanied by changes in the adipocyte responsiveness to adrenergic stimulation that are still under debate. We investigated whether possible changes in the antilipolytic responses, and/or lipogenic activities could be involved in such lipid deposition/mobilisation imbalance. Male Siberian hamsters were exposed for 11 weeks to SD or long day photoperiod and basal or stimulated lipolytic and lipogenic activ

Exposure to field-realistic levels of flupyradifurone and thiamethoxam reduces nursing behavior and accelerates foraging onset in Apis mellifera.

Honey bees are essential pollinators in agricultural and natural ecosystems. As a eusocial insect, the honey bee colony is composed of individuals performing different tasks according to their age-dependent caste, with younger nurses caring for the brood and older foragers collecting food outside the hive. Recently, insecticide-induced disruptions in this division of labor have been implicated as factors contributing to colony collapse, but the behavioral and physiological effects of insecticides on nurses remain poorly understood. We investigated the effects of two nicotinic acetylcholine rec

Dietary administration of polyethylene glycol-formulated tyramine enhances immunity and disease resistance in Litopenaeus vannamei.

This study evaluated whether polyethylene glycol (PEG) formulation could prolong the biological efficacy of dietary tyramine (TA) and provide sustained oral immunostimulation in Litopenaeus vannamei by examining growth performance, immune responses, disease resistance, and biogenic amine regulation during a 120-day feeding trial. Shrimp were assigned to four dietary treatments: basal diet (BD), TA, PEG, and TA + PEG. Results showed that TA significantly enhanced growth performance, exhibiting higher weight gain than all other treatment groups at 56 days and improved feed efficiency at 56 and 1

Genetic Polymorphisms Affect Mouse and Human Trace Amine-Associated Receptor 1 Function.

Methamphetamine (MA) and neurotransmitter precursors and metabolites such as tyramine, octopamine, and β-phenethylamine stimulate the G protein-coupled trace amine-associated receptor 1 (TAAR1). TAAR1 has been implicated in human conditions including obesity, schizophrenia, depression, fibromyalgia, migraine, and addiction. Additionally TAAR1 is expressed on lymphocytes and astrocytes involved in inflammation and response to infection. In brain, TAAR1 stimulation reduces synaptic dopamine availability and alters glutamatergic function. TAAR1 is also expressed at low levels in heart, and may re

Defective responsiveness of adenylate cyclase to forskolin in the Drosophila memory mutant rutabaga.

The Drosophila memory mutant rutabaga (rut) has been previously shown to have a defective subpopulation (or functional state) of the enzyme adenylate cyclase. We report here that the reduced adenylate cyclase activity is also associated with a defective responsiveness of the enzyme to forskolin. Forskolin activation isotherms of the enzyme in normal membranes reveal low- and high-affinity forskolin-interacting components; the residual enzyme in the mutant shows a smaller proportion of the high-affinity response. In addition, in mutant membrane preparations, forskolin fails to shift the Km of t

Cerebrospinal fluid monoamine levels in central disorders of hypersomnolence.

STUDY OBJECTIVES: Whether the cause of daytime sleepiness in narcolepsy type 1 (NT1) is a direct consequence of the loss of orexin (ORX) neurons or whether low orexin reduces the efficacy of the monoaminergic systems to promote wakefulness is unclear. The neurobiology underlying sleepiness in other central hypersomnolence disorders, narcolepsy type 2 (NT2), and idiopathic hypersomnia (IH), is currently unknown. METHODS: Eleven biogenic amines including the monoaminergic neurotransmitters and their metabolites and five trace amines were measured in the cerebrospinal fluid (CSF) of 94 drug-free

Astrocyte-specific regulation of hMeCP2 expression in Drosophila.

Alterations in the expression of Methyl-CpG-binding protein 2 (MeCP2) either by mutations or gene duplication leads to a wide spectrum of neurodevelopmental disorders including Rett Syndrome and MeCP2 duplication disorder. Common features of Rett Syndrome (RTT), MeCP2 duplication disorder, and neuropsychiatric disorders indicate that even moderate changes in MeCP2 protein levels result in functional and structural cell abnormalities. In this study, we investigated two areas of MeCP2 pathophysiology using Drosophila as a model system: the effects of MeCP2 glial gain-of-function activity on circ

Activating properties of cocaine and cocaethylene in a behavioral preparation of Drosophila melanogaster.

The use of Drosophila as a model to study the behavioral consequences of stimulant drugs was analyzed in an active preparation of decapitated Drosophila. Application of cocaine and cocaethylene to discrete nerve cord cells regulating motor programs of behavior produced striking patterns of behavioral activity in a concentration-related manner. In general, intense circling behavior and significant wing buzzing activity were distinguishable behavioral markers in flies treated with mM concentrations of cocaine or cocaethylene. The significant changes in motor behavior induced by stimulant drugs i

Coordination and modulation of locomotion pattern generators in Drosophila larvae: effects of altered biogenic amine levels by the tyramine beta hydroxlyase mutation.

Forward locomotion of Drosophila melanogaster larvae is composed of rhythmic waves of contractions that are thought to be produced by segmentally organized central pattern generators. We present a systematic description of spike activity patterns during locomotive contraction waves in semi-intact wild-type and mutant larval preparations. We have shown previously that Tbetah(nM18) mutants, with altered levels of octopamine and tyramine, have a locomotion deficit. By recording en passant from the segmental nerves, we investigated the coordination of the neuronal activity driving contraction wave
