Rapid dimethyl sulfoxide-modified acid-fast stain of Cryptosporidium oocysts in stool specimens.

A rapid dimethyl sulfoxide modification of an acid-fast technique was applied to direct fecal smears to monitor cryptosporidiosis in nonhuman primates. Brilliantly stained pink oocysts against a pale green background demonstrated well-preserved internal morphology and facilitated rapid, simple, noninvasive diagnosis without fluorescent or phase-contrast microscopy.

Oocyst: knowns and unknowns about the lengthiest life stage of the malaria parasite.

The oocyst is the longest life stage of Plasmodium, the causative agent of malaria, one of the most persistent and devastating infectious diseases of humankind. Following ingestion during blood feeding, parasites reproduce sexually and traverse the mosquito midgut epithelium to differentiate into oocysts on the basal lamina, where they undergo prolonged development, ultimately giving rise to thousands of sporozoites capable of infecting a new human host. Oocyst formation represents a severe population bottleneck, resulting in the lowest parasite numbers observed across the parasite life cycle.

The Synergistic Effect of Tanabesol and Calcium Hydroxide on Cryptosporidium parvum Oocyst Inactivation.

BACKGROUND: Cryptosporidiosis is a major diarrheal disease with high mortality, primarily in young animals, and poses a zoonotic threat worldwide. Currently, no available drugs are fully effective against Cryptosporidium, and most commercial disinfectants show limited efficacy against its oocysts, underscoring the need for improved control strategies. AIMS: We aimed to investigate the ability of Tanabesol (an ortho-agent), calcium hydroxide solution, and their combination to inactivate Cryptosporidium parvum oocysts. MATERIALS AND METHODS: Oocyst morphology was examined using scanning electr

Morphological and molecular characteristics of swine coccidia based on single oocyst isolation.

Swine coccidiosis is a host-specific protozoan disease caused by Cystoisospora suis and various Eimeria species, leading to diarrhea or subclinical signs in pigs. In this study, 3296 fecal samples from 55 farms across six provinces in China were collected and examined to determine the prevalence and molecular characteristics of swine coccidia. The single oocyst isolation technique (SOIT) and molecular characterization identified nine coccidian species, with an overall infection prevalence of 13.83%. Infection rates varied by locations, host age groups, and sampling seasons. Among the positive

Examination of artificial oocyst excystation methods in the murine coccidium, Eimeria krijgsmanni.

Eimeria spp. cause coccidiosis characterized by diarrhea and induce serious economic losses in livestock industries. Although several anti-coccidial drugs are currently available, the emergence of resistant strains and drug residues is problematic; therefore, the development of new drugs is needed. Since sporozoites of Eimeria spp. invade host intestinal epithelial cells and numerous merozoites are formed, drugs that target sporozoites are expected to be useful. We previously used murine Eimeria krijgsmanni as a model to examine anti-coccidial drug susceptibility; however, few studies have con

Evaluation of a vaccine candidate isolated from Cryptosporidium parvum oocyst in mice.

BACKGROUND AND AIM: Cryptosporidiosis is a leading cause of diarrheal disease worldwide and is an animal and public health burden. This study aimed to evaluate the protective potential of affinity-purified Cryptosporidium parvum oocyst antigen as a vaccine candidate according to fecal oocyst shedding, humoral and cellular immune responses, histopathological changes, and the number of parasite developmental stages in ileal and hepatic tissues. MATERIALS AND METHODS: We isolated oocysts from naturally infected buffalo calves and identified them molecularly as C. parvum isolates (GenBank: ON7307

The impact of maturity on the ability of Eimeria acervulina and Eimeria meleagrimitis oocysts to sporulate.

The sporulation of oocysts of Eimeria that infect poultry is known to be under the influence of environmental conditions, including temperature, oxygen supply, and moisture. However, even when these conditions are optimal, the level of sporulation can remain low. The effect of oocyst maturity on their ability to sporulate was investigated for two species of Eimeria: E. acervulina of chickens, and E. meleagrimitis of turkeys. After oral infection of birds, oocysts were collected at their production site in the intestine at different times around the prepatent period. The percentage of sporulati

Dynamics and epidemiology of Toxoplasma gondii oocyst shedding in domestic and wild felids.

Oocyst shedding in domestic and wild felids is a critical yet understudied topic in Toxoplasma gondii ecology and epidemiology that shapes human and animal disease burden. We synthesized published literature dating from the discovery of felids as the definitive hosts of T. gondii in the 1960s through March 2021 to examine shedding prevalence, oocyst genotypes, and risk factors for shedding. Oocyst shedding prevalence in many geographic regions exceeded the commonly accepted 1% reported for domestic cats; crude prevalence from cross-sectional field studies of domestic cat shedding ranged from 0

Detection of Hammondia heydorni-like oocysts in feces of a dog with recurrent diarrhea.

We report a male, 15-month-old American Canadian White Shepherd dog suffering from recurrent diarrhea and anorexia. Fecal analysis revealed coccidian oocysts of small size. Their morphology was similar to those of Hammondia (H.) heydorni and Neospora (N.) caninum isolates. Unsporulated oocysts had a mean length of 11.8 ± 0.96 µm and a mean width of 11.9 ± 0.96 µm. H. heydorni infection was suggested based on the larger size compared with N. caninum oocysts. Additionally, real time PCR analysis for N. caninum from the fecal sample was negative. After a single anti-coccidian treatment (0.45 mg/k

Schizogony and gametogony of oocyst-deficient T-263 strain of Toxoplasma gondii.

Oocysts are important stage for the spread of Toxoplasma gondii because they are environmentally resistant. Among all hosts of T. gondii, only felids can excrete oocysts. Cats that have excreted T. gondii oocysts after primary infection develop immunity to re-excretion of oocysts, and this immunity appears to be long-lasting. It would be desirable to have a non-infectious vaccine for the prevention of T. gondii infection in cats and to understand mechanism of immunity to excretion of oocysts. An initial step will be to indentify stage/stages of the parasite for induction of immunity. A chemica

Seasonal retention and release of Cryptosporidium parvum oocysts by environmental biofilms in the laboratory.

Cryptosporidium is a genus of waterborne protozoan parasites that causes significant gastrointestinal disease in humans. These parasites can accumulate in environmental biofilms and be subsequently released to contaminate water supplies. Natural microbial assemblages were collected each season from an eastern Pennsylvania stream and used to grow biofilms in laboratory microcosms in which influx, efflux, and biofilm retention were determined from daily oocyst counts. For each seasonal biofilm, oocysts attached to the biofilm quickly during oocyst dosing. Upon termination of oocyst dosing, the p

Evaluation of recombinant oocyst protein CP41 for detection of cryptosporidium-specific antibodies.

Cryptosporidium is an important cause of diarrhea in developed and developing countries, and its epidemiology is of interest. The methodologies used in the detection of Cryptosporidium-specific antibodies vary widely, which complicates comparison of results. This study assesses the performance of a Cryptosporidium recombinant protein (rCP41) in a serological assay compared to that of a crude antigen preparation. The 41-kDa protein from the oocyst wall was previously cloned and expressed in Escherichia coli. Sera from 192 healthy adults from the Texas Medical Center (Houston) were tested for an

Evaluation of two commercial disinfectants on the viability and infectivity of Cryptosporidium parvum oocysts.

Cryptosporidiosis is mainly a problem in neonatal ruminants. Not only do Cryptosporidium spp. spread ubiquitously in our environment, but the protozoa are highly resistant to harsh environmental conditions and disinfectants, and a control measure is urgently required. This study investigated the potential biocidal activity on Cryptosporidium parvum oocysts of two commercial disinfectants developed originally to be used in farms and food-processing industries. The products, containing formaldehyde and hydrogen peroxide respectively, both had some anticryptosporidial effects. The viability and i

Infectivity and oocyst excretion patterns of Cryptosporidium muris in slightly infected mice.

To determine the infectivity of Cryptosporidium to hosts in slight infections, we examined the infectivity and oocyst output patterns of Cryptosporidium muris in mice inoculated with small numbers of oocysts. One of the 25 ICR mice inoculated with 2.4 x 10(1) oocysts and 19 of the 25 mice inoculated with 2.4 x 10(2) oocysts shed oocysts in the feces after inoculation. Four of the 50 mice inoculated with 2.4 x 10(1) oocysts for 10 consecutive days also shed oocysts and their OPG values were similar to that of the mice which received 2.4 x 10(2) oocysts. Consequently, it is clear that less than

Prevalence of Cryptosporidium parvum infection and pattern of oocyst shedding in calves in Japan.

Cryptosporidium parvum infection and the pattern of oocyst shedding were observed in calves. A total of 480 fecal samples were collected from 30 calves (age, < or =30 days) over a period of 10 months from June 1998 to March 1999. A sucrose centrifugal flotation technique revealed 28/30 (93%) calves were passing Cryptosporidium oocysts. Oocyst shedding was first detected on the sixth day after birth, with 8% of the calves testing positive. This rate increased day by day and reached approximately 80% by day 15. Oocyst shedding varied from 1 to 13 days, with a mean of 7 days. Calves infected with

Immunogenicity of three oocyst extracts of Cryptosporidium baileyi in experimentally infected chickens.

The immunogenic properties of water-soluble and detergent-extracted components of Cryptosporidium baileyi oocysts were studied. Oocyst cytosol antigen (OCA) containing hydrophilic proteins was obtained by freeze-thaw cycles in liquid nitrogen. This was followed by Triton X-114 extraction of remaining oocyst fragments to dissolve membrane-bound proteins (TRE). The remainder of the pellet was solubilized with sodium dodecyl sulfate and treated with 2-mercaptoethanol to reduce disulfide-linked oocyst wall proteins (BME). The immune recognition of these three extracts was evaluated during the cour

Method for detection and enumeration of Cryptosporidium parvum oocysts in feces, manures, and soils.

Eight concentration and purification methods were evaluated to determine percentages of recovery of Cryptosporidium parvum oocysts from calf feces. The NaCl flotation method generally resulted in the highest percentages of recovery. Based on the percentages of recovery, the amounts of fecal debris in the final oocyst preparations, the relatively short processing time (<3 h), and the low expense, the NaCl flotation method was chosen for further evaluation. Extraction efficiency was evaluated by using oocyst concentrations of 25, 50, 10(2), 10(3), 10(4), and 10(5) oocysts g of bovine feces-1. Th

Cryptosporidium parvum: intensity of infection and oocyst excretion patterns in healthy volunteers.

Data about human Cryptosporidium parvum infection have originated from travelers, community and day care center outbreaks, and persons infected with the human immunodeficiency virus. In addition, experimental infection in 29 antibody-negative, healthy, adult volunteers generated information on the dose-infection response of C. parvum (Iowa strain). In that report, low inocula were sufficient to cause infection in 18 and illness in 7 persons. To further define the duration and intensity of infection in this population, oocyst shedding patterns were investigated in the 18 subjects infected with

[Oocysts of Cryptosporidium sp in feces: comparison of the modified Kinyoun and Heine methods].

The diagnosis of intestinal infection by Cryptosporidium sp is crucial today; with the progression of the AIDS epidemic, many cases of cryptosporidiosis have appeared in this setting and in other immunodeficiency diseases. We compared the advantages and disadvantages of Heine's method and modified Kinyoun's method in the following parameters: morphology of the parasite, quantitation of cysts, stability of the staining characteristics timewise on the slides and time spend at staining. All positive fecal smears were obtained from patients with AIDS. The sensitivity of these two techniques was th

Multicriteria analysis of water quality models for simulating Cryptosporidium oocysts at the watershed scale.

Cryptosporidium is an environmentally robust pathogen responsible for severe waterborne disease. As a result, the emergence of Cryptosporidium in surface water has become an increasing concern for drinking water providers. However, simulating Cryptosporidium with available water quality models remains challenging due to its environmental persistence. With limited work available on modeling Cryptosporidium loading, a systematic review is warranted to examine the potential use of water quality models. This review considered twelve water quality models for simulating Cryptosporidium loading, incl
