Benzothiazole and 2-mercaptobenzothiazole impair reproduction by disrupting sperm function in Caenorhabditis elegans.

Benzothiazoles (BTs), derived primarily from tire wear particles, are ubiquitous contaminants that pose significant environmental hazards and health risks. This study evaluated the effects of benzothiazole (BTH) and 2-mercaptobenzothiazole (MBT) on reproductive toxicity by using the model organism Caenorhabditis elegans. Synchronized L1 larvae were exposed to BTH or MBT at 0, 0.1, 1, 10, and 50 μg/L for 60 h. Germline apoptosis, oogenesis, sperm activation, and sperm migration were tested in parental worms, and some reproductive endpoints were further examined in non-exposed F1 offspring. Our

Reorganization of E-cadherin into apical spot junctions mediates interlineage adhesion between epithelial and germline cells.

During development, epithelia must coordinate morphogenesis with neighboring cell lineages to drive structural remodeling of organ systems. How adhesion between epithelial and other cell types is established and maintained remains poorly understood. Using the Drosophila ovary as an in vivo model, we show that anterior follicle cells (AFCs) undergo epithelial plasticity to establish and maintain adhesion with germline nurse cells during late oogenesis. As AFCs spread over the nurse-cell compartment, adherens junctions disassemble, and E-cadherin, together with junctional partners, reorganizes i

Environmental contaminants, fertility, and multioocytic follicles: a lesson from wildlife?

The overall contribution of environmental exposures to infertility is unknown, but a growing scientific database suggests that exposure to various environmental factors, both in utero and neonatally, could dramatically affect adult fertility. Studies of various contaminant-exposed wildlife populations suggest that multiple mechanisms contribute to changes in gonadal development, maturation of germ cells, fertilization, and pregnancy; specifically, the endocrine processes supporting these events. Although great debate and extensive research has occurred during the last decade surrounding fertil

Population dynamics in echinococcosis and cysticercosis: biological parameters of Echinococcus granulosus in dogs and sheep.

The numerical distributions of Echinococcus granulosus in an experimental dog population are described. At all dose rates of protoscoleces from 10 to 175000 the distribution of worms was over-dispersed. Host age had no effect. There was a direct proportionality between the infective-stage density and rate of infection, and between the latter and the index of clumping. The worm burdens were significantly higher in the proximal than distal portions of the small intestine. Lengths of the 3- and 4-segmented worms increased from 4 to 10 and 4 to 8 weeks of age, respectively. Thereafter apolysis was

Wolbachia infection in Aedes aegypti mosquitoes alters blood meal excretion and delays oviposition without affecting trypsin activity.

Blood feeding in Aedes aegypti is essential for reproduction, but also permits the mosquito to act as a vector for key human pathogens such as the Zika and dengue viruses. Wolbachia pipientis is an endosymbiotic bacterium that can manipulate the biology of Aedes aegypti mosquitoes, making them less competent hosts for many pathogens. Yet while Wolbachia affects other aspects of host physiology, it is unclear whether it influences physiological processes associated with blood meal digestion. To that end, we examined the effects of wMel Wolbachia infection in Ae. aegypti, on survival post-blood

Molecular Insights into Female Hybrid Sterility in Interspecific Crosses between Drosophila melanogaster and Drosophila simulans.

Species of the genus Drosophila have served as favorite models in speciation studies; however, genetic factors of interspecific reproductive incompatibility are under-investigated. Here, we performed an analysis of hybrid female sterility by crossing Drosophila melanogaster females and Drosophila simulans males. Using transcriptomic data analysis and molecular, cellular, and genetic approaches, we analyzed differential gene expression, transposable element (TE) activity, piRNA biogenesis, and functional defects of oogenesis in hybrids. Premature germline stem cell loss was the most prominent d

Maternal Germinal Trisomy 21 in Down Syndrome.

It has now been over 50 years since it was discovered that Down syndrome is caused by an extra chromosome 21, i.e., trisomy 21. In the interim, it has become clear that in the majority of cases, the extra chromosome is inherited from the mother, and there is, in this respect, a strong maternal age effect. Numerous investigations have been devoted to clarifying the underlying mechanism, most recently suggesting that this situation is exceedingly complex, involving both biological and environmental factors. On the other hand, it has also been proposed that germinal trisomy 21 mosaicism, arising

Maternal exposure to Aristolochic Acid I affects meiotic I progression by impairing DNA damage repair in fetal oocytes.

Aristolochic acid I (AAI), the predominant compound in Aristolochiaceae plants and Asarum species, is a widespread environmental contaminant capable of accumulating in soil, contaminating water and crops, ultimately entering the human body. Its nephrotoxic, carcinogenic, and reproductive toxic effects pose significant health concerns. This study investigates the impact of maternal AAI exposure on meiotic prophase I (MPI) during early fetal oogenesis. Pregnant mice were orally administered AAI at doses of 0.03125, 0.125, and 1 mg/kg from 14.5 to 16.5 dpc, with fetal ovaries collected at 17.5 dp

Molecular genetics of meiotic initiation in mammals.

Mammalian fertility is dependent upon meiosis, a specialized cell division by which diploid progenitors undergo one round of DNA replication followed by two rounds of chromosomal segregation to produce haploid gametes. The germ line in the testis and ovary undergoes several rounds of mitotic divisions before ultimately transitioning to the meiotic cell cycle. This transition is achieved by replacing the mitotic cell cycle program with the meiotic one. Here, we discuss the molecular players that regulate the transition from mitosis to meiosis. In spermatogenesis, MEIOC, YTHDC2, and RBM46 form a

Behavioural and physiological responses to prey-related cues reflect higher competitiveness of invasive vs. native ladybirds.

Understanding the traits that might be linked with biological invasions represents a great challenge for preventing non-target effects on local biodiversity. In predatory insects, the ability to exploit habitats for oviposition and the physiological response to prey availability differs between species. Those species that respond more readily to environmental changes may confer to their offspring a competitive advantage over other species. Here, we tested the hypothesis that the invasive Harmonia axyridis (Coleoptera: Coccinellidae) makes better use of information from a plant-prey (Vicia faba

Regulation of broad by the Notch pathway affects timing of follicle cell development.

During Drosophila oogenesis, activation of Notch signaling in the follicular epithelium (FE) around stage 6 of oogenesis is essential for entry into the endocycle and a series of other changes such as cell differentiation and migration of subsets of the follicle cells. Notch induces the expression of zinc finger protein Hindsight and suppresses homeodomain protein Cut to regulate the mitotic/endocycle (ME) switch. Here we report that broad (br), encoding a small group of zinc-finger transcription factors resulting from alternative splicing, is a transcriptional target of Notch nuclear effector

The somatic sex determines the requirement for ovarian tumor gene activity in the proliferation of the Drosophila germline.

Gametogenesis in Drosophila requires sex-specific interactions between the soma and germline to control germ cell viability, proliferation, and differentiation. To determine what genetic components are involved in this interaction, we examined whether changes in the sexual identity of the soma affected the function of the ovarian tumor (otu) and ovo genes. These genes are required cell autonomously in the female germline for germ cell proliferation and differentiation. Mutations in otu and ovo cause a range of ovarian defects, including agametic ovaries and tumorous egg cysts, but do not affec

Detection of de novo mutations and analysis of their origin in families with X linked hypohidrotic ectodermal dysplasia.

Hypohidrotic ectodermal dysplasia (EDA) has been localised to the q12-q13.1 region of the X chromosome by both physical and genetic mapping methods. Although linkage analysis using closely linked flanking markers can clarify the carrier status for many females at risk for the disorder, knowledge of the origin of the mutation in instances of possible de novo mutation is critical for accurate genetic counselling of families. Two methods have been used to confirm de novo mutation in families with EDA and to trace their origin. Direct detection of three de novo molecular deletions, one arising dur

Plasminogen activators in tissue remodeling and invasion: mRNA localization in mouse ovaries and implanting embryos.

To assess in vivo the postulated participation of urokinase-type (u-PA) and tissue-type (t-PA) plasminogen activators in processes involving tissue remodeling and cell migration, we have studied the cellular distribution of u-PA and t-PA mRNAs during mouse oogenesis and embryo implantation. By in situ hybridizations, we detected t-PA mRNA in oocytes and u-PA mRNA in granulosa and thecal cells from preovulatory follicles. These findings are compatible with a role for plasminogen activators in oogenesis and follicular disruption. We demonstrated the presence of u-PA mRNA in the invasive and migr

The Role of N6-methyladenosine Modification in Gametogenesis and Embryogenesis: Impact on Fertility.

The most common epigenetic modification of messenger RNAs (mRNAs) is N6-methyladenosine (m6A), which is mainly located near the 3' untranslated region of mRNAs, near the stop codons, and within internal exons. The biological effect of m6A is dynamically modulated by methyltransferases (writers), demethylases (erasers), and m6A-binding proteins (readers). By controlling post-transcriptional gene expression, m6A has a significant impact on numerous biological functions, including RNA transcription, translation, splicing, transport, and degradation. Hence, m6A influences various physiological and

Conservative character of the germ-line cyst organization within enchytraeids (Annelida: Clitellata) ovary - New proofs based on two Achaeta species.

Ovaries in a minute and mainly aquatic clitellates, collectively termed microdriles, are usually tiny paired gonads localized, depending on the family/subfamily, in segments VI, XI, XII, or XIII. Sometimes, two pairs of ovaries are present in consecutive segments, or ovaries are unpaired. Usually, accumulating yolk (vitellogenic) oocytes detach from ovaries and float in the coelom cavity. This study presents the microscopical analyses of unpaired ovaries found in two representatives of the genus Achaeta (family Enchytraeidae). The ovary is an inconspicuous organ composed of germ-line cysts uni

Frequency and Distribution of Crossovers in Caenorhabditis elegans Meiosis by SNP Genotyping using Real-time PCR.

Caenorhabditis elegans is an excellent model organism for studying meiosis. In addition to general advantages, such as a short reproductive cycle, many progenies, and a transparent body for imaging, there are six pairs of homologous holocentric chromosomes. There is strong crossover interference and regulation of crossover distribution. To measure the crossover frequency, visible marker mapping, snip-SNP mapping, and quantification of cytological markers as precursors of crossover formation have been developed. Here, we introduce a modified SNP genotyping method to measure the crossover freque

Modeling Obesity-Associated Ovarian Dysfunction in Drosophila.

We perform quantitative studies to investigate the effect of high-calorie diet on Drosophila oogenesis. We use the central composite design (CCD) method to obtain quadratic regression models of body fat and fertility as a function of the concentrations of protein and sucrose, two major macronutrients in Drosophila diet, and treatment duration. Our results reveal complex interactions between sucrose and protein in impacting body fat and fertility when they are considered as an integrated physiological response. We verify the utility of our quantitative modeling approach by experimentally confir

A chitin-like component in Aedes aegypti eggshells, eggs and ovaries.

An insoluble white substance was prepared from extracts of eggshells of Aedes aegypti, the yellow fever mosquito and dengue vector. Its infrared and proton NMR spectra were similar to that of standard commercial chitin. This putative chitin-like material, also obtained from ovaries, newly laid and dark eggs, was hydrolyzed in acid and a major product was identified by HPLC to be glucosamine. The eggshell acid hydrolysate was also analyzed by ESI-MS and an ion identical to a glucosamine monoprotonated species was detected. The presence of chitin was also analyzed during different developmental
