Ufasomal nanocarriers for enhanced cardiac delivery of nifedipine: a novel strategy to mitigate doxorubicin-induced cardiotoxicity in breast cancer therapy.

The cardiotoxicity of doxorubicin significantly restricts its effectiveness, even though it remains a crucial element of breast cancer chemotherapy. Nifedipine (NFP) offers multi-mechanistic therapeutic benefits, including vasodilation, anti-proliferative activities, suppression of cellular apoptosis, and potent anti-inflammatory and antioxidant properties. However, the translation of oral NFP into an effective cardioprotective adjuvant is hindered by its poor solubility, first-pass hepatic metabolism, and poor bioavailability. Hence, this study aimed to develop a nasal NFP-loaded ufasomes (NL

Changing the paradigm in treatment-resistant depression: A review of long-term efficacy and tolerability of esketamine nasal spray.

Treatment-resistant depression (TRD) is a major public health problem worldwide. There are currently five strategies for its treatment: optimization, change, combination, augmentation, and somatic therapies. However, many of these therapies are not fully effective, are not accessible, or have a significant number of side effects. Esketamine nasal spray has recently been approved, in combination with another oral antidepressant, for TRD, with excellent results in short-term studies. We consider in this review whether this therapy is also safe and effective in the long-term treatment of patients

Consensus statement. Corticosteroid therapy in ENT in the context of the COVID-19 pandemic.

This consensus statement about the indications and modalities of corticosteroid treatment in the context of the COVID-19 pandemic was jointly written by experts from the French Association of Otology and Oto-Neurology (AFON) and from the French Society of Otorhinolaryngology, Head and Neck Surgery (SFORL). There is currently not enough data in favour of danger or benefit from corticosteroids in COVID-19, so until this matter is resolved it is advisable to limit their indications to the most serious clinical pictures for which it is well established that this type of treatment has a positive im

Development of polymer-encapsulated microparticles of a lipophilic API-IL and its lipid based formulations for enhanced solubilisation.

Active Pharmaceutical Ingredient-Ionic liquids (API-ILs) have the potential to improve the bioavailability of BCS Class IV Drugs. However, the problematic physical handling properties of room temperature API-ILs have impaired clinical and commercial exploitation to date. Lipid-based formulations (LBFs) are used to improve the absorption of drugs with limited bioavailability. Nonetheless, LBFs face limitations such as low drug loading capacity and sub-par physical stability. A platform for transforming API-ILs into solid forms at high loadings via spray encapsulation with polymers has been deve

Development and evaluation of an in situ gelling flurbiprofen throat spray for enhanced mucosal drug delivery and throat inflammation treatment.

The aim of this study was to develop and assess an in situ gelling-based throat spray formulation. containing flurbiprofen, designed to transform into a gel upon contact with the mucosal surface of the throat. Oral administration of medications during throat inflammation can be challenging, prompting the need for alternative delivery methods such as throat sprays. Conventional throat sprays are limited by rapid drug clearance due to excessive secretions. In. situ gelling systems aim to address this limitation by forming a protective gel layer upon application, thereby enhancing drug residence

Oral calcitonin.

Both injectable and nasal spray calcitonins have been utilized in the treatment of postmenopausal osteoporosis for over 25 years. More widespread use of calcitonin in the treatment of osteoporosis has been hampered in part due to poor patient acceptability and compliance and the inability of patients to take this medication as an oral pill. In recent years, an oral preparation of calcitonin has been developed that combines the active peptide hormone with a caprylic acid derivative to enhance bioavailability. Clinical trials with oral calcitonin in patients with osteoarthritis are currently bei

Comparison of Corticosteroids by 3 Approaches to the Treatment of Chronic Rhinosinusitis With Nasal Polyps.

PURPOSE: Corticosteroids are regarded as the mainstay of medical treatment of eosinophilic chronic rhinosinusitis with nasal polyps (ECRSwNP). To date, a head-to-head comparison of the efficacy and safety of glucocorticoid preparations administered via different routes for the treatment of chronic rhinosinusitis with nasal polyps has not been reported. To compare the efficacy and safety of steroids administered via the oral, intranasal spray and transnasal nebulization routes in the management of ECRSwNP over a short course. METHODS: Overall, 91 patients with ECRSwNP were recruited prospectiv

[Rhinitis in old age].

The chronic rhinitis is a frequent problem with age. In the nose, normal physiologic changes of aging include loss of nasal tip support, atrophy of mucus-producing mucosal glands, and decreased olfaction. The fragmentation and weakening of the cartilage of the septum also causes airflow changes contributing to nasal stuffiness. These changes contribute to geriatric rhinitis, the symptoms of which are often attributed by the older patient to "allergies" or "sinus trouble". An understanding of these anatomic changes, linked with a thorough history and physical examination, allows the physician t

Pyrethroid Ingestion With Brain and Heart Involvement: A Case Report.

Insecticides have great efficacy and are used in homes and agricultural fields across the globe, including pyrethrins and their synthetic equivalents, or pyrethroids. Among their compounds are deltamethrin and imiprothrin, which are present in many insecticide sprays. Humans can become toxically exposed to them through ingestion, skin contact, or inadvertent inhalation. In humans, gastrointestinal and neurotoxic side effects are frequently observed in cases of acute oral intoxication. Although they have not been well documented up to this point, pyrethroids have occasionally caused cardiotoxic

Rhinitis during pregnancy and rhinitis medicamentosa.

Vasomotor rhinitis is a nonspecific disorder that is caused neither by infection nor allergy but rather by an imbalance of the autonomic nervous system with a preponderant action of parasympathetic fibers on nasal blood vessels. Rhinitis during pregnancy appears to result from the increased production of estrogen; increased estrogen levels caused by treatment, puberty, or liver disease may also cause rhinitis. Nasal saline mist, antihistamines, and topical corticosteroids are recommended; intranasal corticosteroid injections are also useful but must be administered under expert care. Rhinitis
