Ouabain-induced cochlear nerve degeneration: synaptic loss and plasticity in a mouse model of auditory neuropathy.

Ouabain application to the round window can selectively destroy type-I spiral ganglion cells, producing an animal model of auditory neuropathy. To assess the long-term effects of this deafferentation on synaptic organization in the organ of Corti and cochlear nucleus, and to ask whether surviving cochlear neurons show any post-injury plasticity in the adult, we quantified the peripheral and central synapses of type-I neurons at posttreatment times ranging from 1 to 3 months. Measures of normal DPOAEs and greatly reduced auditory brainstem responses (ABRs) confirmed the neuropathy phenotype. Co

Animal model of mania induced by ouabain: Evidence of oxidative stress in submitochondrial particles of the rat brain.

The intracerebroventricular (ICV) administration of ouabain (a Na(+)/K(+)-ATPase inhibitor) in rats has been suggested to mimic some symptoms of human bipolar mania. Clinical studies have shown that bipolar disorder may be related to mitochondrial dysfunction. Herein, we investigated the behavioral and biochemical effects induced by the ICV administration of ouabain in rats. To achieve this aim, the effects of ouabain injection immediately after and 7 days following a single ICV administration (at concentrations of 10(-2) and 10(-3)M) on locomotion was measured using the open-field test. Effects of mood stabilizers on hippocampus and amygdala BDNF levels in an animal model of mania induced by ouabain.

There is a body of evidence suggesting that BDNF is involved in bipolar disorder (BD) pathogenesis. Intracerebroventricular (ICV) injection of ouabain (OUA), a specific Na(+)/K(+) ATPase inhibitor, induces hyperlocomotion in rats, and has been used as an animal model of mania. The present study aims to investigate the effects of the lithium (Li) and valproate (VPT) in an animal model of mania induced by ouabain. In the reversal model, animals received a single ICV injection of OUA or cerebrospinal fluid (aCSF). From the day following the ICV injection, the rats were treated for 6 days with int

CD69 expression induced by thapsigargin, phorbol ester and ouabain on thymocytes is dependent on external Ca2+ entry.

In the present work murine thymocytes exposed to Thapsigargin (TG 10, 20 and 50 nM), Phorbol-12,13,20-triacetate (TPA16 nM) and Ouabain (OUA100 nM) exhibited an increased expression of CD69, a molecule related to cellular activation and associated to Ca(++) influx in other systems. The kinetics of CD69 appearance depended on the stimuli and dose used. TG 50 nM induced an increased expression by 6 h whereas with lower doses (10 and 20 nM) an increase was detected at 18 h. TPA maximal increase was evident at 6 h. OUA lead to an observable increase at 18 h. However, in the case of TPA or TG the p

Effect of adenosine on the ouabain-insensitive Na+-ATPase activity from basolateral membrane of the proximal tubule.

The regulation of the furosemide-sensitive Na+-ATPase activity and ouabain-sensitive (Na+ + K+)ATPase activities from proximal tubules by adenosine was investigated. When the concentration of adenosine was increased the furosemide-sensitive ATPase activity decreased with maximal inhibition at 10(-8) M (56% of inhibition). However, the (Na+ + K+)ATPase activity was not affected by adenosine. Theophylline, an antagonist of P1 adenosine receptor, completely reversed the effect of adenosine on the furosemide-sensitive ATPase activity in a dose-response manner. The adenosine effect was mimicked by

Normalization of high plasma level of ouabain-like immunoreactivity in primary aldosteronism after removal of adenoma.

Plasma ouabain-like immunoreactivity, which has been supposed to be associated with hypertension, was significantly higher in five patients with primary aldosteronism than in age-matched normotensive subjects. High plasma levels of ouabain-like immunoreactivity decreased to normal after removal of adenoma. Extracts of adenoma tissue did not contain any apparent ouabain-like immunoreactivity. Anti-ouabain antibody used in this study did not cross-react with aldosterone, cortisol, corticosterone, arachidonic acid or lysophosphatidylcholine. Hypertension, hypokalemia, a high plasma aldosterone le

The effects of scorpion venom tityustoxin and ouabain on the release of acetylcholine from incubated slices of rat brain.

The in vitro release of acetylcholine in slices of rat brain tissue was followed in the presence of tityustoxin and ouabain. At low ouabain concentrations, the release of acetylcholine caused by both ouabain and tityustoxin, was additive. At higher ouabain concentrations the additive effect of tityustoxin and ouabain on the release of ACh was no longer observed.

Effects of metabolic inhibitors and ouabain on static afferent discharges from the isolated frog muscle spindle.

Effects of metabolic inhibitors and ouabain on the rate of afferent discharges from isolated muscle spindle of the bullfrog were examined in vitro to elucidate the mechanism for generating afferent action potentials. Dinitrophenol, NaCN, iodoacetic acid, and ouabain gradually reduced the rate of discharges. These drugs were without direct effect on the nerve axon. The rate of discharges was also reduced by Li+, which is known to be minimally extruded from the interior of cells by an active transport mechanism. The rate of discharges remained approximately 50% in the presence of NaCN or in the

Regional Crosstalk Between the Amygdala, Hippocampus, and Prefrontal Cortex Following Na+,K+-ATPase Inhibition by Ouabain.

The Na⁺,K⁺-ATPase plays a fundamental role in memory formation by regulating neuronal excitability and NMDA receptor-dependent synaptic plasticity. While this enzyme's dysfunction is broadly linked to cognitive disorders, its specific contribution to fear memory consolidation, particularly within the key brain structure of the basolateral amygdala (BLA), remains undefined. We investigate whether Na+,K+-ATPase inhibition disrupts contextual fear conditioning (CFC) memory consolidation and examine downstream molecular, oxidative, and inflammatory responses in interconnected brain regions. Modulation of cardiac Na+,K+-ATPase cell surface abundance by simulated ischemia-reperfusion and ouabain preconditioning.

Na(+),K(+)-ATPase and cell survival were investigated in a cellular model of ischemia-reperfusion (I/R)-induced injury and protection by ouabain-induced preconditioning (OPC). Rat neonatal cardiac myocytes were subjected to 30 min of substrate and coverslip-induced ischemia followed by 30 min of simulated reperfusion. This significantly compromised cell viability as documented by lactate dehydrogenase release and Annexin V/propidium iodide staining. Total Na(+),K(+)-ATPase α(1)- and α(3)-polypeptide expression remained unchanged, but cell surface biotinylation and immunostaining studies reveal

Ouabain activates the Na-K-ATPase signalosome to induce autosomal dominant polycystic kidney disease cell proliferation.

The Na-K-ATPase is part of a cell signaling complex, the Na-K-ATPase signalosome, which upon activation by the hormone ouabain regulates the function of different cell types. We previously showed that ouabain induces proliferation of epithelial cells derived from renal cysts of patients with autosomal dominant polycystic kidney disease (ADPKD cells). Here, we investigated the signaling pathways responsible for mediating the effects of ouabain in these cells. Incubation of ADPKD cells with ouabain, in concentrations similar to those found in blood, stimulated phosphorylation of the epidermal gr

Exocytotic release of [3H]-acetylcholine by ouabain involves intracellular Ca2+ stores in rat brain cortical slices.

1. The effect of ouabain on the release of [3H]acetylcholine ([3H]ACh) in rat brain cortical slices was investigated. 2. The ouabain-induced release of [3H]ACh was calcium-independent and not blocked by EGTA. 3. BAPTA-AM, a chelator of intracellular calcium, inhibited the ouabain effect suggesting the involvement of intracellular calcium stores. 4. Vesamicol, a drug that blocks the storage of acetylcholine in synaptic vesicles inhibited by 73% the ouabain-induced release of [3H] ACh, suggesting exocytotic release of the neurotransmitter. 5. Honokiol alleviates oxidative stress, neuroinflammation, and apoptosis in excitotoxicity induced by ouabain, a Na + /K + -ATPase inhibitor.

Excitotoxicity, oxidative stress, and neuroinflammation play significant roles in the pathophysiology of neurodegenerative disorders. This study aims to examine the impact of honokiol (HNK) on antioxidant, anti-inflammatory, and anti-apoptotic mechanisms in a model of excitotoxicity induced by intracerebroventricular (icv) ouabain (OUA) in rats. Rats were allocated to five equal groups (n = 10): (I) Animals given intraperitoneal isotonic saline solution for seven days (Control); (II) animals given a single dose of 5 μl OUA solvent (artificial cerebrospinal fluid-icv) on the first day (Sham); (

Ouabain Relieves Sleep Deprivation-Induced Anxiety-Like Behavior in Mice by Suppressing Hippocampal Neuroinflammation and Oxidative Stress.

BACKGROUND: Sleep insufficiency has become a global public health challenge and is closely associated with the onset of mood and anxiety disorders. Neuroinflammation and oxidative stress are considered key pathological substrates underlying these conditions. Ouabain is a prototypical cardiotonic glycoside and an endogenous ligand of Na+/K+-ATPase. In recent years, ouabain has been reported to exert anti-inflammatory and neuroprotective effects; however, whether it can ameliorate sleep deprivation (SD)-associated affective abnormalities remains unclear. METHODS: Using a 72-h modified multiple-

Ouabain regulates kidney metabolic profiling in rat offspring of intrauterine growth restriction induced by low-protein diet.

AIMS: Intrauterine growth restriction (IUGR) can increase the risk of hypertension and kidney disease at adulthood due to fetal programming. In our previous study, we found that supplementation with low concentration of ouabain during pregnancy could restore glomerulus numbers at birth, rescuing kidney development. However, the metabolic pattern of kidney in IUGR offspring and the effect of ouabain have not been evaluated. MAIN METHODS: In this study, based on GC-MS and LC-MS platforms, we used the protein restriction rat model to explore the molecular mechanisms of kidney damage induced by I

How does pressure overload cause cardiac hypertrophy and dysfunction? High-ouabain affinity cardiac Na+ pumps are crucial.

Left ventricular hypertrophy is frequently observed in hypertensive patients and is believed to be due to the pressure overload and cardiomyocyte stretch. Three recent reports on mice with genetically engineered Na+ pumps, however, have demonstrated that cardiac ouabain-sensitive α2-Na+ pumps play a key role in the pathogenesis of transaortic constriction-induced hypertrophy. Hypertrophy was delayed/attenuated in mice with mutant, ouabain-resistant α2-Na+ pumps and in mice with cardiac-selective knockout or transgenic overexpression of α2-Na+ pumps. The latter, seemingly paradoxical, findings

Ouabain, a cardiac glycoside, inhibits the Fanconi anemia/BRCA pathway activated by DNA interstrand cross-linking agents.

Modulation of the DNA repair pathway is an emerging target for the development of anticancer drugs. DNA interstrand cross-links (ICLs), one of the most severe forms of DNA damage caused by anticancer drugs such as cisplatin and mitomycin C (MMC), activates the Fanconi anemia (FA)/BRCA DNA repair pathway. Inhibition of the FA/BRCA pathway can enhance the cytotoxic effects of ICL-inducing anticancer drugs and can reduce anticancer drug resistance. To find FA/BRCA pathway inhibitory small molecules, we established a cell-based high-content screening method for quantitating the activation of the F
