An improved electrophysiological cellular assay to unlock the pharmacological modulation of the NALCN channelosome.

The sodium leak channel non-selective (NALCN) is a key regulator of resting membrane potential and cellular excitability in neurons and endocrine cells. Gain-of-function de novo pathogenic variants in NALCN cause severe neurodevelopmental disorders with a broad and heterogeneous clinical spectrum. Partial inhibition of NALCN has been proposed as a therapeutic strategy; however, progress has been limited by the absence of selective pharmacological modulators. This gap largely reflects the lack of a robust heterologous expression system suitable for high-throughput screening, as functional NALCN

Cardiac α4β2 nicotinic receptors as a therapeutic target for fatal ventricular arrhythmias.

BACKGROUND AND AIMS: Fatal ventricular tachyarrhythmias (FVTs) are a major cause of sudden cardiac death globally. Despite their clinical importance, current antiarrhythmic therapies remain constrained by limited efficacy and proarrhythmic risks. Although endogenous cardiac cholinergic signalling contributes to electrophysiological regulation, the therapeutic potential of targeting ventricular α4β2 nicotinic acetylcholine receptors (nAChRs), a pivotal component of this system, for FVT treatment remains to be elucidated. METHODS: A pharmacological screen identified positive allosteric modulato

Organic solvent-induced changes in membrane geometry in human SH-SY5Y neuroblastoma cells - a common narcotic effect?

Exposure to organic solvents may cause narcotic effects. At the cellular level, these narcotic effects have been associated with a reduction in neuronal excitability caused by changes in membrane structure and function. In order to critically test whether changes in membrane geometry contribute to these narcotic effects, cultured human SH-SY5Y neuroblastoma cells have been exposed to selected organic solvents. The solvent-induced changes in cell membrane capacitance were investigated using the whole-cell patch clamp technique for real-time capacitance measurements. Exposure of SH-SY5Y cells to

Zebrafish model for congenital myasthenic syndrome reveals mechanisms causal to developmental recovery.

Mutations in muscle ACh receptors cause slow-channel syndrome (SCS) and Escobar syndrome, two forms of congenital myasthenia. SCS is a dominant disorder with mutations reported for all receptor subunits except γ. Escobar syndrome is distinct, with mutations located exclusively in γ, and characterized by developmental improvement of muscle function. The zebrafish mutant line, twister, models SCS in terms of a dominant mutation in the α subunit (α(twi)) but shows the behavioral improvement associated with Escobar syndrome. Here, we present a unique electrophysiological study into developmental i

Effects of clozapine and N-desmethylclozapine on synaptic transmission at hippocampal inhibitory and excitatory synapses.

Clozapine is the first atypical antipsychotic, and improves positive and negative symptoms of many patients with schizophrenia resistant to treatment with other antipsychotic agents. Clozapine induces minimal extrapyramidal side effects, but is more often associated with seizures. A large number of studies have been conducted to elucidate pharmacological profiles of clozapine and its major active metabolite, N-desmethylclozapine (NDMC). However, there are only a limited number of electrophysiological studies examining their effects on synaptic transmission. In this study, we examined effects o

Interaction of intravenous anesthetics with human neuronal potassium currents in relation to clinical concentrations.

BACKGROUND: Neuronal voltage-dependent potassium (K) currents are crucial for various cellular functions, such as the integration of temporal information in the central nervous system. Data for the effects of intravenous anesthetics on human neuronal K currents are limited. It was the authors' aim to evaluate the concentration-related effects of three opioids (fentanyl, alfentanil, sufentanil) and seven nonopioids (thiopental, pentobarbital, methohexital, propofol, ketamine, midazolam, droperidol) used in clinical anesthesia on neuronal voltage-dependent K currents of human origin. METHOD: K

Membrane potassium channels and human bladder tumor cells. I. Electrical properties.

These experiments were conducted to determine the membrane K+ currents and channels in human urinary bladder (HTB-9) carcinoma cells in vitro. K+ currents and channel activity were assessed by the whole-cell voltage clamp and by either inside-out or outside-out patch clamp recordings. Cell depolarization resulted in activation of a Ca(2+)-dependent outward K+ current, 0.57 +/- 0.13 nS/pF at -70 mV holding potential and 3.10 +/- 0.15 nS/pF at 30 mV holding potential. Corresponding patch clamp measurements demonstrated a Ca(2+)-activated, voltage-dependent K+ channel (KCa) of 214 +/- 3.0 pS. Sco

Totally 3-dimensional endoscopic ventricular septal defect closure and aortic valve replacement via a periareolar incision.

There are no prior reports of totally endoscopic minimally invasive cardiac surgery for combined ventricular septal defect closure and aortic valve replacement. We utilized a periareolar incision as the main access, inserting a cardioplegia line, vent tube and the aortic cross-clamp entirely through this incision. Only three ports (main incision, camera port and left-hand port) were used. A three-dimensional endoscope facilitated precise techniques. The type I ventricular septal defect was approached and closed from the aortic side using a Gore-Tex patch, with mattress sutures for the lower ed

Antivenom Evaluation by Electrophysiological Analysis.

Scorpion stings on humans are medically relevant because they may contain toxins that specifically target ion channels. During antivenom production, pharmaceutical companies must use a large number of experimental animals to ensure the antivenom's efficacy according to pharmacopeia methods. Here we present an electrophysiological alternative for the evaluation of horse antivenoms produced against two species of Moroccan scorpions: Buthus mardochei and Androctonus mauretanicus. Human sodium and potassium channels and acetylcholine nicotinic receptors were analyzed by standard patch-clamp techni

K+-channel opening as a mechanism for relaxing airways smooth muscle.

The properties and physiological roles of K+-channels in the plasmalemma of airways smooth muscle are currently being clarified by the use of techniques such as patch-clamp recording. K+-channel activity plays an important role in determining both the resting membrane potential and the electrical excitability of the airways smooth muscle cell. Established bronchodilators such as agonists at beta-adrenoceptors and methylxanthines may provide K+-channel opening but this is not crucial for their relaxant action. The actions of other smooth muscle relaxants such as BRL 34915 and pinacidil (Fig.) m

Functional heterogeneity of periglomerular cells in the rat olfactory bulb.

The periglomerular cells of the rat olfactory bulb, a virtually unknown population of interneurons, have been studied applying the whole-cell patch-clamp technique to thin slices. A prominent result, obtained under current-clamp conditions, is that these cells appear to be functionally heterogeneous, and show distinct excitability profiles. Voltage-clamp analysis allows the identification of the ionic basis of these differences and suggests a division into at least two classes, based on the characteristics of the K+ conductances. The first group displays two K+ conductances (delayed rectifier,

Characterization of potassium and chloride channels in the basolateral membrane of bovine nonpigmented ciliary epithelial cells.

PURPOSE: The pigmented epithelial (PE) and the nonpigmented epithelial (NPE) cells of the ciliary body may function as a syncytium for secretion of the aqueous humor, with solute and water entering through the PE and leaving through the NPE cell. Secretion across the basolateral membrane of the NPE cell was postulated to depend on coupling sodium extrusion through the NaK-ATPase to efflux through Cl channels. In the current study, the authors used single-channel patch clamp techniques to characterize the ion channels present in the basolateral membrane of the NPE cell. METHODS: Mild enzymatic

Intravenous administration of lidocaine directly acts on spinal dorsal horn and produces analgesic effect: An in vivo patch-clamp analysis.

Intravenous lidocaine administration produces an analgesic effect in various pain states, such as neuropathic and acute pain, although the underlying mechanisms remains unclear. Here, we hypothesized that intravenous lidocaine acts on spinal cord neurons and induces analgesia in acute pain. We therefore examined the action of intravenous lidocaine in the spinal cord using the in vivo patch-clamp technique. We first investigated the effects of intravenous lidocaine using behavioural measures in rats. We then performed in vivo patch-clamp recording from spinal substantia gelatinosa (SG) neurons.

Calcium near the release site is essential for basal ACh release in Xenopus.

Extracellular calcium is essential for neurotransmitter release, but the detailed mechanism by which Ca2+ regulates basal synaptic release has not yet been fully explored. In this study, calcium imaging and the whole-cell patch-clamp technique were used to investigate the role of Ca2+ in basal acetylcholine (ACh) release in the Xenopus neuromuscular junction and in isolated myocytes exogenously loaded with ACh. Carried out in normal and Ca2+-free extracellular solution, the results indicate that Ca2+ near the release site is essential for basal neurotransmitter release.

Human muscle voltage-gated ion channels and hereditary disease.

Insights in the field of ion channels were made possible by the Nobel-prize-winning patch-clamp technique that enables characterization of channel function, and have greatly been inspired by associated diseases pointing to regions of functional significance. These so-called ion channelopathies have common clinical features, recurrent patterns of mutations, and almost predictable mechanisms of pathogenesis. In skeletal muscle, disorders are associated with mutations in Na+, K+, Ca2+, and Cl- channels that lead to hypoexcitability (causing periodic paralysis) and to hyperexcitabilty (causing myo

Spider venom of Araneus opens and desensitizes glutamate channels in chick spinal cord neurones.

Whole cell currents induced by the excretions of venom glands from the spider Araneus diadematus (ASV) were tested in neurones from the embryonic spinal cord of the chick in culture by the patch clamp technique. ASV in a dose of one bite in 1 ml induced a large membrane current of the same polarity as the excitatory amino acids, which decreased during long-lasting application. The effects were, to a large extent, rapidly reversible. It is suggested that ASV activates and desensitizes the receptor channels for excitatory amino acids.

Phenotypic Severity of SCN5A-Related Bradycardia Is Independent of Dominant-Negative and Coupled Gating Effects.

BACKGROUND: Pathogenic SCN5A variants are associated with inherited arrhythmias such as long-QT syndrome, Brugada syndrome, and sick sinus syndrome. While Nav1.5, an α-subunit of the cardiac sodium channel encoded by SCN5A, has been considered to function as a monomer, recent studies reveal that a reduction of sodium current in wild-type Nav1.5 can be caused by dimerization with loss-of-function mutated Nav1.5 through dominant-negative effects. However, the clinical significance of the dominant-negative effect remains unclear. METHOD: We genetically screened a family who presented with sick s

Telethonin variants found in Brugada syndrome, J-wave pattern ECG, and ARVC reduce peak Nav 1.5 currents in HEK-293 cells.

BACKGROUND: Telethonin (TCAP) is a Z-disk protein that maintains cytoskeletal integrity and various signaling pathways in cardiomyocytes. TCAP is shown to modulate α-subunit of the human cardiac sodium channel (hNav 1.5) by direct interactions. Several TCAP variants are found in cardiomyopathies. We sought to investigate whether TCAP variants are associated with arrhythmia syndromes. METHODS: Mutational analyses for TCAP were performed in 303 Japanese patients with Brugada syndrome, arrhythmogenic right ventricular cardiomyopathy, and J-wave pattern ECG. Using patch-clamp techniques, electrop
