Cellular cartography of the organ of Corti based on optical tissue clearing and machine learning.

The highly organized spatial arrangement of sensory hair cells in the organ of Corti is essential for inner ear function. Here, we report a new analytical pipeline, based on optical clearing of tissue, for the construction of a single-cell resolution map of the organ of Corti. A sorbitol-based optical clearing method enabled imaging of the entire cochlea at subcellular resolution. High-fidelity detection and analysis of all hair cell positions along the entire longitudinal axis of the organ of Corti were performed automatically by machine learning-based pattern recognition. Application of this

Innate immunity and disorders of the female reproductive tract.

Sexually transmitted infections, and their associated sequelae, such as tubal infertility, ectopic pregnancy and preterm labour, are a major worldwide health problem. Chlamydia trachomatis infection is thought to be the leading global cause of tubal infertility and tubal ectopic pregnancy. Preterm birth occurs in around 10% of all deliveries, and nearly 30% of preterm deliveries are associated with intrauterine infection. The mucosal innate immune system of the female reproductive tract has evolved to eliminate such sexually transmitted pathogens whilst maintaining its ability to accommodate s

Innate immune receptors in platelets and platelet-leukocyte interactions.

Platelets are chief cells in hemostasis. Apart from their hemostatic roles, platelets are major inflammatory effector cells that can influence both innate and adaptive immune responses. Activated platelets have thromboinflammatory functions linking hemostatic and immune responses in several physiological and pathological conditions. Among many ways in which platelets exert these functions, platelet expression of pattern recognition receptors (PRRs), including TLR, Nod-like receptor, and C-type lectin receptor families, plays major roles in sensing and responding to pathogen-associated or damag

Respiratory distress in small animals: Pathophysiology and clinical approach.

BACKGROUND: Respiratory distress is a common reason for animals to present to the emergency room. An understanding of respiratory physiology is helpful to facilitate accurate localization of the patient's source of respiratory distress. This knowledge will aid implementation of more appropriate therapies. PHYSIOLOGY/PATHOPHYSIOLOGY SUMMARY: Respiratory distress can occur secondary to lesions at any location of the respiratory system. Diseases affecting different portions of the respiratory track (eg, upper airway, lower airways, parenchymal, or the pleural space) lead to distinguishing clinic

Hypericum perforatum chronic treatment affects cognitive parameters and brain neurotrophic factor levels.

OBJECTIVE: To evaluate the effects of Hypericum perforatum (hypericum) on cognitive behavior and neurotrophic factor levels in the brain of male and female rats. METHODS: Male and female Wistar rats were treated with hypericum or water during 28 days by gavage. The animals were then subjected to the open-field test, novel object recognition and step-down inhibitory avoidance test. Nerve growth factor (NGF), brain-derived neurotrophic factor (BDNF), and glial cell-line derived neurotrophic factor (GDNF) levels were evaluated in the hippocampus and frontal cortex. RESULTS: Hypericum impaired t

Retinal adaptation to object motion.

Due to fixational eye movements, the image on the retina is always in motion, even when one views a stationary scene. When an object moves within the scene, the corresponding patch of retina experiences a different motion trajectory than the surrounding region. Certain retinal ganglion cells respond selectively to this condition, when the motion in the cell's receptive field center is different from that in the surround. Here we show that this response is strongest at the very onset of differential motion, followed by gradual adaptation with a time course of several seconds. Different subregio

Acute exacerbation of idiopathic pulmonary fibrosis as the initial presentation of the disease.

The clinical course of patients with idiopathic pulmonary fibrosis (IPF) is generally marked by a decline in pulmonary function over time, although recently there is increasing recognition that fatal deterioration from acute exacerbation can occur at any stage. The patient described in the present case study was a 65-yr-old male who presented with exertional dyspnoea and fever of 2 weeks' duration. He had no history of chronic lung disease or physiological or radiological hallmarks of pre-existing disease. He underwent surgical lung biopsy and the histological examination showed a backg

Humans have nasal respiratory fingerprints.

Long-term respiratory patterns are generated by remarkably complex brain networks. Because brains are unique, we hypothesized that their dependent respiratory patterns may be similarly unique. To test this hypothesis, we developed a wearable device that precisely measures and logs nasal airflow in each nostril separately for up to 24-h periods. We found that we could identify members of a 97-participant cohort at a remarkable 96.8% accuracy from nasal airflow patterns alone. In other words, humans have individual nasal airflow fingerprints. Moreover, in test-retest experiments, we found that t

Sex-Based Differences in Coronary Physiological Assessment in Patients With Severe Aortic Stenosis.

BACKGROUND: Assessing coronary artery disease in severe aortic stenosis (AS) presents unique challenges. Fractional flow reserve (FFR) may yield unreliable results due to altered coronary hemodynamics, leading to discordance with non-hyperemic indices. Women with AS develop distinct myocardial remodeling patterns that may predispose to coronary microvascular dysfunction. We investigated whether sex influences the diagnostic performance and agreement of invasive physiological indices in severe AS. METHODS: This prospective single-center study enrolled 221 patients with severe AS and intermedia

Automated detection of congestive heart failure from electrocardiogram signal using Stockwell transform and hybrid classification scheme.

BACKGROUND AND OBJECTIVE: The congestive heart failure (CHF) is a life-threatening cardiac disease which arises when the pumping action of the heart is less than that of the normal case. This paper proposes a novel approach to design a classifier-based system for the automated detection of CHF. METHODS: The approach is founded on the use of the Stockwell (S)-transform and frequency division to analyze the time-frequency sub-band matrices stemming from electrocardiogram (ECG) signals. Then, the entropy features are evaluated from the sub-band matrices of ECG. A hybrid classification scheme is

Auditory and audio-visual processing in patients with cochlear, auditory brainstem, and auditory midbrain implants: An EEG study.

There is substantial variability in speech recognition ability across patients with cochlear implants (CIs), auditory brainstem implants (ABIs), and auditory midbrain implants (AMIs). To better understand how this variability is related to central processing differences, the current electroencephalography (EEG) study compared hearing abilities and auditory-cortex activation in patients with electrical stimulation at different sites of the auditory pathway. Three different groups of patients with auditory implants (Hannover Medical School; ABI: n = 6, CI: n = 6; AMI: n = 2) performed a speeded

Tone identification behavior in Rattus norvegicus: muscarinic receptor blockage lowers responsiveness in nontarget selective neurons, while nicotinic receptor blockage selectively lowers target responses.

Learning to associate a stimulus with reinforcement causes plasticity in primary sensory cortex. Neural activity caused by the associated stimulus is paired with reinforcement, but population analyses have not found a selective increase in response to that stimulus. Responses to other stimuli increase as much as, or more than, responses to the associated stimulus. Here, we applied population analysis at a new time point and additionally evaluated whether cholinergic receptor blockers interacted with the plastic changes in cortex. Three days of tone identification behavior caused responsiveness

Subcortical representation of musical dyads: individual differences and neural generators.

When two notes are played simultaneously they form a musical dyad. The sensation of pleasantness, or "consonance", of a dyad is likely driven by the harmonic relation of the frequency components of the combined spectrum of the two notes. Previous work has demonstrated a relation between individual preference for consonant over dissonant dyads, and the strength of neural temporal coding of the harmonicity of consonant relative to dissonant dyads as measured using the electrophysiological "frequency-following response" (FFR). However, this work also demonstrated that both these variables correla

Bacterial Physiology in the Context of Algal Partners.

Marine bacteria display diverse lifestyles, many of which are shaped by close associations with microalgal partners. In the photic zone, where microalgae fix carbon through photosynthesis, bacteria consume algal-derived organic matter and often exhibit growth patterns that mirror algal activity and abundance. These interactions expose bacteria to strong fluctuations, with bursts of algal exudates during high productivity alternating with periods of scarcity. To cope, algal-associated bacteria have evolved strategies that balance rapid growth with the ability to endure starvation. This review h

The Mechanisms of miRNAs on Target Regulation and their Recent Advances in Atherosclerosis.

miRNAs are crucial regulators in a variety of physiological and pathological processes, while their regulation mechanisms were usually described as negatively regulating gene expression by targeting the 3'-untranslated region(3'-UTR) of target gene miRNAs through seed sequence in tremendous studies. However, recent evidence indicated the existence of non-canonical mechanisms mediated by binding other molecules besides mRNAs. Additionally, accumulating evidence showed that functions of intracellular and intercellular miRNAs exhibited spatiotemporal patterns. Considering that detailed knowledge

Types of intrapartum hypoxia on the cardiotocograph (CTG): do they have any relationship with the type of brain injury in the MRI scan in term babies?

Electronic foetal monitoring using cardiotocography is aimed at the timely recognition and management of foetal hypoxia. The primary objective of this study was to examine whether a relationship exists between the types of foetal hypoxia (acute, subacute, evolving, chronic), as identified on cardiotocography and the nature of hypoxic ischaemic encephalopathy, as observed on MRI scans after birth. We conducted a retrospective study of 16 babies born (out of 52,187 births) at St George's Hospital in London during 2006-2017 with a postnatal diagnosis of HIE. Of the 16 babies, only 11 had both MRI

The 1.8-A crystal structure of a matrix metalloproteinase 8-barbiturate inhibitor complex reveals a previously unobserved mechanism for collagenase substrate recognition.

The individual zinc endoproteinases of the tissue degrading matrix metalloproteinase (MMP) family share a common catalytic architecture but are differentiated with respect to substrate specificity, localization, and activation. Variation in domain structure and more subtle structural differences control their characteristic specificity profiles for substrates from among four distinct classes (Nagase, H., and Woessner, J. F. J. (1999) J. Biol. Chem. 274, 21491-21494). Exploitation of these differences may be decisive for the design of anticancer or other drugs, which should be highly selective

Graph Regularized Nonnegative Matrix Factorization for Data Representation.

Matrix factorization techniques have been frequently applied in information retrieval, computer vision, and pattern recognition. Among them, Nonnegative Matrix Factorization (NMF) has received considerable attention due to its psychological and physiological interpretation of naturally occurring data whose representation may be parts based in the human brain. On the other hand, from the geometric perspective, the data is usually sampled from a low-dimensional manifold embedded in a high-dimensional ambient space. One then hopes to find a compact representation, which uncovers the hidden semanti
