Effect of the antimicrobial peptide BmKn2-7 in inhibiting Vibrio parahaemolyticus and its role in improving microbiota, immunity and Vibrio resistance in Litopenaeus vannamei.

Antimicrobial peptide BmKn2-7, a rationally optimized derivative of the scorpion venom peptide BmKn2, shows strong potential as an antibiotic alternative for controlling Vibrio parahaemolyticus in aquaculture. This study evaluated its antibacterial mechanisms and in vivo efficacy in Litopenaeus vannamei through biochemical assays, transcriptomic profiling, and challenge experiments. BmKn2-7 exhibited potent in-vitro activity (MIC = 125 µg/mL) and disrupted bacterial homeostasis by increasing membrane permeability, inducing oxidative stress, and inhibiting P-type ATPase. RNA-seq analysis reveal

Genome Sequencing and Genome-Wide Analysis Uncover Candidate Genes and Loci Linked to Key Production Traits in the Philippine Black Tiger Shrimp (Penaeus monodon).

Penaeus monodon (black tiger shrimp) is a key species in aquaculture globally and in the Philippines. The selection and development of stocks with desirable production traits, such as enhanced growth and disease resistance, are critical for improving production. In this study, we leveraged a newly assembled, high-quality genome sequence from a Philippine population to perform genome-wide scans for SNPs associated with key production traits. Using cohort-based extreme phenotype GWAS (XP-GWAS), we identified variants linked to resistance against white spot syndrome virus (WSSV), body weight, and

scRNA-seq of Penaeus japonicus hemocytes under environmentally-induced restriction of sand-diving behavior.

Penaeus japonicus is an economically important marine shrimp species, but its sand-diving habit affects the further improvement of their culture yield. To investigate the molecular response to the restriction of sand-diving behavior, we designed three culture systems: the sandy group, the sandless group, and the sandless stress group (transfer the individuals to a sandless pond after a week in the sandy group). First, we analyzed the expression differences of stress-related (HSP60) and circadian rhythm genes (period, timeless) at different time points after stress induction, identifying 6 h po

Pollution characterization, adsorption, and SERS-based detection of enrofloxacin adsorbed on microplastics in aquaculture water.

This study investigates the pollution of microplastics (MPs) in aquaculture and their adsorption of enrofloxacin (ENR), and develops a SERS detection method for ENR adsorbed on the surface of MPs based on hydrophilic membrane enrichment. Surface water samples from three South American white shrimp (Litopenaeus vannamei) ponds were analyzed for MP abundance, distribution, morphology, particle size, and type. Results revealed that MP pollution was widespread, with fibers smaller than 1 mm being predominant, and polypropylene was the main polymer type. The adsorption behavior of ENR onto polyprop

Investigating the effect of acute toxicity exposure to combined FeSO4 and FeCl3 in Litopenaeus vannamei through analysis of survival, metal accumulation, oxidative stress, and intestinal flora.

Marine shrimp have been studied as bioindicators in environmental monitoring of heavy metal pollution. High levels of heavy metal iron (Fe) are potentially toxic and readily accumulate in organisms, causing damage. Up to now, there are few studies about the toxicity of Fe in the water on shrimp. In this study, the acute toxic effects of different concentrations of combined FeSO4 and FeCl3 on Pacific white shrimp (Litopenaeus vannamei) were examined from the aspects of survival, metal bioaccumulation, oxidative stress, and changes in intestinal microbes. The median lethal concentration (LC50) o

Effect of organic selenium on performance and gill transcriptome of whiteleg shrimp Penaeus vannamei.

BACKGROUND: Shrimp gill health is an indicator of their overall health and environmental stress, as gills are in direct contact with the external environment. Organic selenium (OS), known for its high bioavailability and antioxidant properties, potentially enhances shrimp growth, survival, and immune responses. This study investigated the physiological features through comprehensive gill gene expression profiling and growth parameters by incorporating OS into Penaeus vannamei diets. METHODS AND RESULTS: Two OS-containing products, AlkoSel (AS) and SelPlex (SL), were supplied to the diet of P.

Transcriptome-wide identification and analysis reveals m6A regulation of metabolic reprogramming in shrimp (Marsupenaeus japonicus) under virus infection.

BACKGROUND: It has been reported that the most common post-transcriptional modification of eukaryotic RNA is N6-methyladenosine (m6A). Previous studies show m6A is a key regulator for viral infection and immune response. However, whether there is a pathogen stimulus-dependent m6A regulation in invertebrate shrimp has not been studied. RESULTS: In this study, we performed a transcriptome-wide profiling of mRNA m6A methylation in shrimp (Marsupenaeus japonicus) after white spot syndrome virus (WSSV) infection by methylated RNA immunoprecipitation sequencing (MeRIP-seq). A total of 15,436 m6A pe

Effects of polyphenolic compounds on the structure, allergenicity and gel properties of shrimp myofibrillar proteins.

BACKGROUND: Shrimp represents an important aquatic resource, whereas shrimp paste products enjoy widespread popularity for their distinctive flavor and nutritional benefits. However, the overall quality of these products is often limited by potential allergenic risks and heavy reliance on the gel-forming capacity of myofibrillar proteins (MPs). Polyphenolic compounds, specifically epigallocatechin-3-gallate (EGCG), epigallocatechin (EGC) and catechin (CC), are known to interact with proteins and modify their functional properties, yet their effect on the allergenicity of shrimp MPs remains une

Hemocyanin modulates ferroptosis through a HSP90-dependent antioxidant function in shrimp.

Ferroptosis, an iron-dependent programed cell death, is essential for maintaining homeostasis and responding to environmental and pathogenic stressors in mammals. In shrimp, ferroptosis is also vital in various biological processes, including stress response and immunity. However, the specific regulatory mechanisms of ferroptosis in shrimp and other marine animals remains unclear. In this study, we demonstrate that hemocyanin actively modulates ferroptosis in the hepatopancreas of Penaeus vannamei, exhibiting a significant inhibitory effect through its small subunit. During ferroptosis, shrimp

Effect of static magnetic field-assisted freezing at different temperatures on the structural and functional properties of pacific white shrimp (Litopenaeus vannamei) myofibrillar protein.

The effects of static magnetic field-assisted freezing (MF) on the structural and functional characteristics of Litopenaeus vannamei myofibrillar protein (MP) at various temperatures (-35 ∼ -20 °C) were examined to assess its influence on MP and its energy-saving potential. The results indicated that -35 °C MF (MF-35) exhibited greater solubility and lower turbidity than -35 °C immersion freezing (IF-35), suggesting that MF-35 inhibited MP aggregation. MF-35 prevented the reduction in fluorescence intensity and α-helix content, protecting the MP tertiary and secondary structures. The emulsifyi

Phytochemical Screening and Computational Insights into VP26 Inhibitors for Mitigating White Spot Syndrome Virus in Shrimp Aquaculture.

White spot syndrome disease (WSSD) is a contagious disease caused by white spot syndrome virus (WSSV) in shrimp aquaculture. Given its high degree of contagiousness, controlling its rapid spread has proven to be challenging, causing significant economic loss to farmers. To prevent these losses, farmers often resort to the use of large doses of general aquaculture antibiotics. However, prolonged exposure to such antibiotics can lead to adverse effects for both the shrimp and the humans consuming them. Additionally, this practice contributes to the global issue of antimicrobial resistance. Curre

Evaluation of a synergistic blend of short- and medium-chain fatty acids as a dietary intervention for the effective management of Vibriosis in shrimp culture.

Vibriosis caused by Vibrio spp. is imposing severe havoc and adverse effects on shrimp culture. Antibiotics are the most widely used therapeutic measures against vibriosis. However, the uncontrolled use of antibiotics may spread the antibiotic residues and increase antibiotic-resistant bacteria. In this study, a product based on a synergistic blend of short- and medium-chain fatty acids (Selacid® GG, herein referred as Sel) was evaluated against vibriosis in shrimp, P. vannamei. The shrimps (n = 30 in triplicate per treatment) were fed with Sel (0.0 %, 0.1 %, 0.2 % or 0.3 %) for 14 days. The s

Transcriptomics reveals different response mechanisms of Litopenaeus vannamei hemocytes to injection of Vibrio parahaemolyticus and WSSV.

As the most important cultural crustacean species worldwide, studies about Pacific white shrimp (Litopenaeus vannamei) have received more attention. It has been well-documented that various pathogens could infect L. vannamei, resulting in huge economic losses. The studies about the responding mechanism of L. vannamei to sole pathogens such as Vibrio parahaemolyticus and white spot virus (WSSV) have been extensively reported, while the studies about the differently responding mechanisms remain unclear. In the present study, we identified the differently expressed genes (DEGs) of L. vannamei hem
