Pentobarbital (PB), a barbiturate anesthetic, is widely used in many clinical treatments, including management of seizures and preoperative sedation. However, its potential role in cancer therapy remains underexplored. The inhibitory effects of PB on breast cancer proliferation were investigated in this study. Breast cancer cell lines MDA-MB-231 and MCF-7 were used to evaluate the effects of PB on cell proliferation. Proliferating cell nuclear antigen (PCNA) overexpression and knockdown models were established to assess its role in PB-mediated proliferation inhibition. Cell proliferation was m

Cerebral metabolic and vascular effects of hypothermia (30 C) and deep pentobarbital anesthesia, separately and combined, were evaluated in 15 mongrel dogs. External cardiovascular support was not used, and mean arterial blood pressures remained greater than 60 torr. Normothermic deep pentobarbital anesthesia, characterized by an electroencephalographic (EEG) frequency of less than 1 Hz, was associated with 30% decreases in cerebral metabolic rates for oxygen (CMRO2) and glucose (CMRG) from lightly anesthetized control values. Hypothermia (30 C) alone caused similar decreases in CMRO2 and CMRG

Administration of pentobarbital inhibits the facilitatory effects of progesterone on the release of gonadotropins. In this experiment facilitatory effects of progesterone on lordosis behavior in guinea pigs were examined with pentobarbital anesthesia. Two major animal groups were subjects: one was short-term ovariectomized (2 weeks) and the other was long-term ovariectomized (several months). All animals received estradiol benzoate (6.6 mug s.c.) followed by progesterone (0.4 mg s.c.) 40 h later. Lordosis behavior was induced by the manual stimulation method of Young et al.29 Sodium pentobarbi

The ventilatory responses to isocapnic hypoxia and hypercapnia were studied in seven chronically tracheostomized dogs awake and during anesthesia with pentobarbital (30 mg/kg, iv), ketamine, or thiopental (10 and 15 mg/kg, respectively, followed by infusion). Isocapnic hypoxic ventilatory drive (HVD) was expressed as the parameter A such that the higher the A, the greater the hypoxic drive. HVD(A) was significantly reduced from 259 +/- 28 (mean +/- SEM) in awake dogs, to 96 +/- 14 after pentobarbital, 161 +/- 27 after thiopental, and 213 +/- 23 after ketamine. Hypercapnic ventilatory drive (HC

AIMS: Safranal (2,6,6-trimethyl-1,3-cyclohexadiene-1-carboxaldehyde, C(10) H(14) O) is an active ingredient in the saffron, which is used in traditional medicine. It has been reported to have sedative and anti-epileptic effects, but its hypnotic effects remain uncertain. The aim of this study was to evaluate effects of safranal on sleep-wake cycle.
METHODS: We established hypnotic-model mice treated with a low dose of pentobarbital 20 mg/kg, and administered different doses of safranal, vehicle, or diazepam. The change of sleep-wake cycle was assessed by sleep recording and c-Fos expression i

This study was performed to investigate whether magnolol enhances pentobarbital-induced sleeping behaviors through the GABAergic systems. Magnolol prolonged the sleeping time induced by pentobarbital. In addition, magnolol increased chloride influx in primary cultured cerebellar granule cells. The expression of the GABA(A) receptor alpha-subunit was increased selectively by magnolol, but magnolol had no effect on the abundance of beta- or gamma-subunits. The expression of glutamic acid decarboxylase (GAD) was not influenced by magnolol. It is suggested that magnolol may enhance pentobarbital-i

PURPOSE: To compare the effectiveness and safety of oral (PO) versus intravenous (IV) pentobarbital sedation for magnetic resonance (MR) imaging and computed tomography (CT) in infants younger than 12 months.
MATERIALS AND METHODS: The institutional review board approved the review of medical records and determined informed consent to be unnecessary. All parents gave informed consent for patient sedation. Prior to MR imaging or CT, infants younger than 12 months were sedated with PO pentobarbital (4-8 mg per kilogram body weight) or IV pentobarbital (2-6 mg/kg), depending on the presence of a

Lipopolysaccharide (LPS)-induced hypotension and impaired aortic contraction to norepinephrine (NE) are thought to be consequent to induction of nitric oxide synthase (iNOS). Anesthesia is often employed in studies of the mechanisms mediating LPS-induced cardiovascular dysfunction in rats. Since sympathetic nervous system activity and compensatory mechanisms can be altered by anesthesia, this study was designed to determine a) if the cardiovascular dysfunction associated with LPS (5 mg/kg, i.v.)-induced endotoxin shock is enhanced in anesthetized compared with conscious male Wistar rats, and b

The possibility that the GABAA receptor is involved in supraspinal morphine analgesia was investigated in rats using pentobarbital and other GABAA receptor-chloride channel ligands. Inhibition of tail-flick was used as an index of analgesia. Pentobarbital almost completely reversed intracerebroventricular (i.c.v.) morphine analgesia, but did not affect intrathecal (i.t.) morphine analgesia. Phenobarbital had a similar effect but was much weaker. Picrotoxin blocked the reversal effect of pentobarbital. Diazepam, when given together with pentobarbital, enhanced the effect of pentobarbital. Furth

High dose barbiturate therapy has been used clinically for about 10 years. However, the hemodynamic consequences of such therapy have not been well defined in humans. We therefore examined the effects of a brief (30-minute), high dose (0.6 mg/kg/minute or 18 mg/kg total) infusion of pentobarbital in nine otherwise healthy patients (aged 20 to 30) scheduled to undergo operation for the removal of large or deeply seated arteriovenous malformations. Monitored variables included intravascular pressures, cardiac output, arterial and mixed venous blood gases and hematocrit, and the electroencephalog

Recent reports have suggested that anesthesia may weaken host defense mechanisms during shock and depress the activities of circulating leukocytes. The purpose of this study was to assay the effects of sodium pentobarbital anesthesia on glucose uptake and survival of leukocytes, and mortality of E coli organisms in vitro. In vitro experiments were performed on canine blood before and after the in vivo administration of anesthesia and the in vitro addition of E coli. Glucose uptake in vitro increased equally in blood obtained from either awake or anesthetized dogs following incubation with orga

The transport efficiency of the dog's pelviureteral system was examined during pacemaker blockade induced by pentobarbital. For an analog representation of transport efficiency, changes in balance level between the input and the output were registered and recorded on a polygraph together with perfusion rate, renal pelvic pressure, ureteral electromyogram and bolus volume. Pentobarbital dramatically blocked the pacemaker activity of the renal pelvis, and the ureter simultaneously became aperistaltic. During the period of pacemaker rest, the balance level was maintained at a high level in all do

Our objective was to describe a case of suspected relay barbiturate intoxication of a dog after ingestion of a rat that had been euthanized and frozen and then later burned in an attempted cremation. This case will be compared to previous reports of relay toxicosis. This report describes a dog that was presented to an emergency and critical care hospital because of lethargy and vomiting after ingesting remains of a rat that had been euthanized 2 wk earlier. The primary-care veterinarian had used a pentobarbital solution for euthanasia. The rat's remains had been frozen until the owner attempte

Gamma-aminobutyric acid (GABA) is one of the inhibitory neurotransmitters with beneficial effects including sedative properties. However, despite various clinical trials, scientific evidence regarding the impact on sleep of orally ingested GABA, whether natural or synthesized through biological pathways, is not clear. GABALAGEN (GBL) is the product of fermented collagen by Lactobacillus brevis BJ20 (L. brevis BJ20) and Lactobacillus plantarum BJ21 (L. plantarum BJ21), enriched with GABA and characterized by low molecular weight. The aim of this study was to investigate the effect of GBL on sle

A fatal concentration of pentobarbital found in a coroner's case where the history had not indicated use of this drug prompted a review of fatalities in Ontario from 2012 to 2015. Coroner's case files, including police and toxicology reports, were reviewed in twenty deaths, in which pentobarbital was identified as the primary cause of death. In all of the deaths (11 females, 9 males), the blood concentration of pentobarbital was greater than 10 mg/L. There were three to eight deaths per year and each was classified as suicide. In 11 cases, there was clear evidence that the drug was purchased o

Withdrawal syndromes following administration of sedative and analgesic infusions represent a significant morbidity that is described increasingly in the pediatric critical care literature. However, there are few descriptions of pentobarbital withdrawal symptoms and their treatment in the pediatric population. We describe an infant receiving multiple sedative and analgesic medications, including pentobarbital, who was resistant to our institution's standard weaning protocol, but whose severe withdrawal symptoms responded dramatically to phenobarbital loading and maintenance.

INTRODUCTION: "Suicide tourism," the practice of traveling to a foreign destination to commit suicide, has been described in the medical literature. Additionally, committing suicide by self-administering veterinary medications has been previously described.
CASE DESCRIPTIONS: We report two successful and one unsuccessful suicide attempts involving border-town travelers utilizing self-administered veterinary pentobarbital over a 1-year period.
DISCUSSION/CONCLUSION: Health care practitioners should be aware of and informed about this phenomenon.
