Effect of parenterally administered ethanol on the tissue distribution of phencyclidine following percutaneous absorption.

At 1, 2, 3, and 4 hr after percutaneous absorption of phencyclidine hydrochloride in the hairless mouse, concentrations of the drug in blood, brain, liver, lung, and heart were much lower in mice that had been pretreated with hypertonic ethanol by intraperitoneal injection than in control mice pretreated with water. These findings are consistent with the hypothesis that hypertonic ethanol inhibits the percutaneous absorption of phencyclidine. The relative pattern of tissue distribution of phencyclidine, however, appeared to be essentially the same for both groups. It appears that the magnitude

Phencyclidine (PCP): effects of acute and chronic administration on EEG activities in the rhesus monkey.

Effects of acute and chronic administration of phencyclidine (PCP) on EEG activities and gross behavior were studied in monkeys with electrodes implanted in the brain. Administration of PCP (2.0 or 4.0 mg/kg i.v.) in monkeys produced a biphasic pattern of inhibition and excitation of behavior during 6-8 h observation period. The inhibitory phase appeared 1-3 min after PCP injection, and was uniquely characterized by high-voltage slow waves with delta waves (0.6-0.8 c/sec) in the parietal lobe and by those with theta waves (4-5 c/sec) in the occipital lobe and hippocampus during behavioral stup

Effect of phencyclidine (PCP) on blood pressure and catecholamine levels in discrete brain nuclei.

PCP (10 mg/kg, s.c.) causes hypertension that is associated with decrease or tendency to decrease the levels of epinephrine and norepinephrine in the hypothalamus and the brainstem regions. If this represents a decreased catecholaminergic activity, then this may enhance sympathetic activity and contribute to the PCP-induced hypertension.

The effect of a single administration of phencyclidine on behavior in the rat over a 21-day period.

The effects of phencyclidine (PCP) (7.0, 11.7, 19.5, 32.6, and 54.4 mg/kg) on locomotor activity, stereotyped behavior (circling, backing up, and weaving frequency), and rotarod performance were evaluated. In addition, the frequency of other PCP-induced abnormal behaviors (head in corner, arched back, and cataleptic freeze) was determined. All doses of PCP produced a significant increase in locomotor activity and stereotyped behavior as well as an impairment of rotarod performance. Both the duration and the time to peak effects (with the exception of rotarod performance) of these PCP-induced b

Effect of phencyclidine (PCP) on adrenergic responses to spinal cord stimulation in pithed rats.

The effect of PCP (1 mg/kg i.a.) on sympathetic and cardiovascular responses were studied in conscious rats and pithed intact and adrenalmedullectomized rats (MEDX). Also, we examined the effect of Phencyclidine (PCP) (3 mg/kg s.c.) on the uptake and disposition of [3H]norepinephrine [3H]NE in rat heart and plasma. In conscious rats, PCP significantly increased blood pressure (PB), heart rate (HR) and plasma catecholamine (CA). Sympathetic stimulation in pithed intact and MEDX rats elicited increases in PB, HR and plasma CA. Administration of PCP (1 mg/kg i.v.) prior to stimulation potentiated

How often do false-positive phencyclidine urine screens occur with use of common medications?

BACKGROUND: Previous reports describe false-positive urine immunoassay screens for phencyclidine (PCP) associated with use of tramadol, dextromethorphan, or diphenhydramine. The likelihood of these false positives is unknown. OBJECTIVE: We sought to find the relative frequency of false-positive PCP screens associated with these medications and to look for any other medications with similar associations. METHODS: In an IRB-approved study, we retrospectively reviewed charts of all ED encounters with positive urine screens for PCP in our hospital from 2007 through 2011, inclusive. Urine samples

Prenatal exposure to phencyclidine produces abnormal behaviour and NMDA receptor expression in postpubertal mice.

Several studies have shown the disruptive effects of non-competitive N-methyl-d-aspartate (NMDA) receptor antagonists on neurobehavioural development. Based on the neurodevelopment hypothesis of schizophrenia, there is growing interest in animal models treated with NMDA antagonists at developing stages to investigate the pathogenesis of psychological disturbances in humans. Previous studies have reported that perinatal treatment with phencyclidine (PCP) impairs the development of neuronal systems and induces schizophrenia-like behaviour. However, the adverse effects of prenatal exposure to PCP

Massive venlafaxine overdose resulted in a false positive Abbott AxSYM urine immunoassay for phencyclidine.

CASE REPORT: A 13-yr-old girl overdosed on 48 x 150 mg venlafaxine (Effexor XR). She was taking venlafaxine regularly for depression. Her only other medications included topical Benzamycin and pyridoxine 50 mg daily for acne. The Abbott AxSYM assay was positive only for phencyclidine, but GC/MS did not confirm the presence of phencyclidine. Toxilab identified only one substance, confirmed by GC/MS as venlafaxine. A serum sample obtained 3 h after her ingestion revealed a venlafaxine concentration of 24460 ng/mL and an O-desmethylvenlafaxine concentration of 3930 ng/mL, confirming the massive a

Phencyclidine-induced cognitive deficits in mice are improved by subsequent subchronic administration of clozapine, but not haloperidol.

This study was undertaken to examine the effects of subsequent administration of antipsychotic drugs (clozapine and haloperidol) on cognitive deficits in mice after repeated administration of phencyclidine (PCP). In the novel object recognition test, repeated administration of PCP (10 mg/kg) significantly decreased exploratory preference in the retention test session but not in the training test session. PCP-induced deficits were significantly improved by subsequent subchronic (2 weeks) administration of clozapine (5 mg/kg), but not haloperidol (0.1 mg/kg). These findings suggest that PCP-indu

[Effects of phencyclidine on rabbit basilar artery in vitro and rabbit cerebral blood flow in vivo].

The effect of phencyclidine [1-(1-phenylcyclohexyl)piperidine, PCP] on rabbit basilar arteries was studied with an in vitro model of ring segment arteries. PCP 0.05-500 mumol.L-1 caused vasoconstriction of basilar arteries in a concentration-dependent manner. Its maximal effect (Emax) was 94 +/- 21 mg and the concentration causing half maximal effect (EC50) was 25 +/- 18 mumol.L-1. PCP 0.01-10 mumol.L-1 also concentration-dependently augmented the vasoconstriction induced by electric stimulation in rabbit basilar arteries. Its Emax was 91 +/- 18 mg and EC50 was 0.27 +/- 0.17 mumol.L-1. The eff

Phencyclidine in CSF and serum: a case of attempted filicide by a mother without a history of substance abuse.

A previously healthy 30-year-old black woman with no history of substance abuse was hospitalized after she attempted to drown her 4-year-old son. She had become progressively confused and delusional after a flu-like illness 2 weeks before. Serum and lumbar CSF samples assayed for phencyclidine (PCP) by gas chromatography-mass spectrometry with d5 PCP as an internal standard were positive. The patient recovered rapidly after treatment with haloperidol and acidification of her urine. Suspicion of PCP abuse should remain high among patients with psychosis, even for those with no history of substa

Effects of barbiturates and other sedative hypnotics in pigeons trained to discriminate phencyclidine from saline.

Pigeons were trained to peck the center key (lighted white) of three response keys to turn off the center keylight and to light one of the side keys with a red keylight and the other side key with a green keylight. Five responses (fixed-ratio component) on either side key relighted the center key. Food was delivered following 10 fixed-ratio components on the red key if 1.5 mg/kg phencyclidine had been given before the session. The position of the red and green keylights on the side keys varied randomly each time they were lighted by a peck on the center key. Subsequently, increasing doses of p

Preincubation with antipsychotic drugs protects against in vitro phencyclidine-mediated spontaneous neuronal network suppression.

Phencyclidine (PCP), a non-competitive N-methyl-D-aspartate receptor antagonist, is known to produce schizophrenia-like psychosis in humans, including positive and negative symptoms as well as cognitive dysfunction. Moreover, acute administration of PCP can emulate corresponding behavioral symptoms in rodents. We investigated the effect of PCP and the possible protective potential of typical and atypical antipsychotic drugs (APDs) in vitro on spontaneously active neuronal networks. To this end, murine primary cortical cells were cultured on microelectrode arrays (MEAs). Concentration-response

Nicotinic receptors and lurasidone-mediated reversal of phencyclidine-induced deficit in novel object recognition.

BACKGROUND: Enhancement of cholinergic function via nicotinic acetylcholine (ACh) receptor (nAChR) agonism is a potential approach for the treatment of cognitive impairment associated with schizophrenia (CIAS). Some atypical antipsychotic drugs (AAPDs) enhance ACh release in rodent brain, indirectly stimulating these receptors. Here, we elucidate which nAChR subtypes mediate novel object recognition (NOR) in normal rats and contribute to the ability of the AAPD, lurasidone, to improve the NOR deficit in sub-chronic (sc) phencyclidine (PCP)-treated rats, a model for CIAS. METHODS: The ability

Phencyclidine: a 5-year retrospective review from the New York City Medical Examiner's Office.

We report here a 5-year retrospective review of autopsy cases from the New York City Medical Examiner's Office that demonstrated phencyclidine (PCP) in the blood. There were a total of 138 cases. There were 52 deaths because of mixed drug intoxication: the blood PCP concentrations in these cases ranged from <1 to 598 ng/mL. There were 80 violent deaths in which PCP was quantified in the blood but was unrelated to the cause of death. There were five nonviolent deaths in which PCP exclusively was detected. In four of these, there were preexisting medical conditions that could also have contribut

Behavioral effects of orally administered glycine in socially housed monkeys chronically treated with phencyclidine.

RATIONALE: Schizophrenia is a major mental disorder. Dissociative anesthetics such as phencyclidine (PCP) produce a syndrome in humans that is clinically indistinguishable from schizophrenia by blocking neurotransmission at N-methyl-D-aspartate (NMDA)-type glutamate receptors. NMDA receptors in brain are modulated by the amino acid glycine (GLY), which reverses neurochemical and behavioral effects of PCP in rodents. The present study investigates GLY effects on PCP-induced behavior in primates. OBJECTIVES: In primates, PCP induces characteristic behavioral symptoms that can be used to model p

Alpha-2 adrenoceptor activation inhibits phencyclidine-induced deficits of spatial working memory in rats.

N-methyl-D-aspartate (NMDA)/glutamate receptor antagonists, such as phencyclidine (PCP), induce behavioral abnormalities (locomotor hyperactivity, sensorimotor gating deficits, impairments of cognition) in animals that are thought to model aspects of schizophrenia. The administration of PCP increases noradrenaline transmission in the rat prefrontal cortex, a brain structure required for normal cognitive processes. Noradrenaline, in turn, works through a set of receptors that have themselves been implicated directly in NMDA antagonist-induced deficits; we recently reported that the alpha-2 agon

Binding studies and photoaffinity labeling identify two classes of phencyclidine receptors in rat brain.

Binding and photoaffinity labeling experiments were employed in order to differentiate 1-(1-phenylcyclohexyl)piperidine (PCP) receptor sites in rat brain. Two classes of PCP receptors were characterized and localized: one class binds [3H]-N-[1-(2-thienyl)cyclohexyl]piperidine [( 3H]TCP) with high affinity (Kd = 10-15 nM) and the other binds the ligand with a relatively low affinity (Kd = 80-100 nM). The two classes of sites have different patterns of distribution. Forebrain regions are characterized by high-affinity sites (hippocampus greater than frontal cortex greater than thalamus greater t

Phencyclidine and dizocilpine induced behaviors reduced by N-acetylaspartylglutamate peptidase inhibition via metabotropic glutamate receptors.

BACKGROUND: N-methyl-D-aspartate (NMDA) receptor open channel blockers phencyclidine (PCP) and dizocilpine (MK-801) elicit schizophrenia-like symptoms in humans and in animal models. Group II metabotropic glutamate receptor agonists reverse the behavioral effects of PCP and MK-801 in animal models. N-acetylaspartylglutamate (NAAG), the third most prevalent neurotransmitter in the mammalian nervous system, is a selective group II metabotropic glutamate receptor agonist. We previously reported that ZJ43, a potent inhibitor of the enzymes that inactivate synaptically released NAAG, reduced motor

Postmortem urine immunoassay showing false-positive phencyclidine reactivity in a case of fatal tramadol overdose.

This is a report of postmortem false-positive reactivity using an enzyme-multiplied urine phencyclidine (PCP) immunoassay (EMIT II+) due to a single-agent fatal tramadol overdose. An autopsy of a 42-year-old male who died alone at home revealed no identifiable lethal anatomic abnormalities, thus leading to toxicologic analysis. Femoral blood was obtained for drug testing by high-performance liquid chromatography (HPLC) and showed a tramadol level of 14.0 mg/L, 2 orders of magnitude greater than the therapeutic range (0.1 to 0.3 mg/L). Urine was also obtained and EMIT II+ immunoassay revealed p
