Identification of cutaneous Leishmaniasis vectors, Phlebotomus papatasi and P. duboscqi using random amplified polymorphic DNA.

Phlebotomus papatasi and P. duboscqi are two closely related, morphologically similar sandfly species that are established vectors of zoonotic cutaneous leishmaniasis in the Old World. We have developed a Random Amplified Polymorphic DNA (RAPD) method to find species-specific DNA profiles of these two species. It was found that using a single 10-mer primer a 'species specific' amplification band of about 0.49 kb was produced in all specimens of P. duboscqi while it was absent in P. papatasi. The 0.49 kb diagnostic band was consistently present in both males and females of P. duboscqi. The suit

A study of phlebotomine sandflies (Diptera:Psychodidae) in Yunnan Province. III. Phlebotomus (Adlerius) fengi sp. nov.

Among the phlebotomine specimens collected from Yunnan, China, in 1984, a new species of the subgenus Adlerius Nitzulescu 1931 was identified. The name Phlebotomus fengi is proposed for this species, in memory of the late medical entomologist, Professor Lan Chow Feng. Although generally similar to P. longiductus, P. fengi had distinct pharyngeal armature, style spines, paramere and aedeagus. Phlebotomus fengi may be a vector of visceral leishmaniasis in southern China.

DNA hybridizations on squash-blotted sandflies to identify both Phlebotomus papatasi and infecting Leishmania major.

Epidemiological field studies on leishmaniasis have been hampered by the laborious, and often inefficient, methods used to assess the rates of infection of sandfly vectors (Diptera; Phlebotominae) by species of the causative disease organisms, protozoal parasites of the genus Leishmania (Kinetoplastida; Trypanosomatidae). We report the rapid and accurate identification of both sandfly vector (Phlebotomus (Phlebotomus) papatasi (Scopoli] and infecting Leishmania major Yakimov & Schokov by DNA hybridizations to squash-blotted sandflies. Large numbers of whole (infected) sandflies can be quickly

In silico assessment of 4 methoxypelargonidin and luteolinidin from Parthenium hysterophorous as acetylcholinesterase inhibitors of Phlebotomus Papatasi.

Phlebotomus papatasi is a vector of various health risk diseases, such as sandfly fever and leishmaniasis. Chemical-based insecticides appear to be effective against the vector but are harmful to non-targeted organisms and the environment. Plant metabolites are an excellent source of natural insecticides that show no adverse effects. To the environment, plant-based flavonoids are emerging compounds that can be used against human disease vectors. In the present investigation, flavonoids were partially isolated from the leaf and flower of Parthenium hysterophorus and characterized using column c

PpSP32-like protein as a marker of human exposure to Phlebotomus argentipes in Leishmania donovani foci in Bangladesh.

Phlebotomus argentipes is a predominant vector of Leishmania donovani, the protozoan parasite causing visceral leishmaniasis in the Indian subcontinent. In hosts bitten by P. argentipes, sand fly saliva elicits the production of specific anti-salivary protein antibodies. Here, we have utilised these antibodies as markers of human exposure to P. argentipes in a visceral leishmaniasis endemic area in Pabna district, Bangladesh. The use of whole salivary gland homogenate as an antigen to detect these antibodies has several limitations, therefore it is being superseded by the use of specific recom

Host preference and Leishmania infantum natural infection of the sand fly Phlebotomus perfiliewi in northern Italy.

The host preference of hematophagous insects is important in determining the cycle of pathogens that they potentially transmit; for example, sand flies are competent vectors of Leishmania parasites. In this work, we evaluated the host preference of sand flies collected in the Emilia-Romagna region of Italy in 2018 and 2019 in an area in which Leishmania infantum circulates actively. Out of about 30,000 sampled sand flies, we obtained 252 engorged females, which were processed to identify the sources of blood meals. Sampling data collected confirmed a positive phototropism of Phlebotomus (Ph.)

Feeding behavior and activity of Phlebotomus pedifer and potential reservoir hosts of Leishmania aethiopica in southwestern Ethiopia.

BACKGROUND: Cutaneous leishmaniasis (CL) is a major public health concern in Ethiopia. However, knowledge about the complex zoonotic transmission cycle is limited, hampering implementation of control strategies. We explored the feeding behavior and activity of the vector (Phlebotomus pedifer) and studied the role of livestock in CL transmission in southwestern Ethiopia. METHODS: Blood meal origins of engorged sand flies were determined by sequencing host DNA. A host choice experiment was performed to assess the feeding preference of P. pedifer when humans and hyraxes are equally accessible. E

High-resolution habitat suitability model for Phlebotomus pedifer, the vector of cutaneous leishmaniasis in southwestern Ethiopia.

BACKGROUND: Phlebotomus pedifer is the vector for Leishmania aethiopica causing cutaneous leishmaniasis (CL) in southwestern Ethiopia. Previous research on the transmission dynamics of CL resulted in recommendations for vector control. In order to target these interventions towards affected areas, a comprehensive understanding of the spatial distribution of P. pedifer at high spatial resolution is required. Therefore, this study determined the environmental predictors that facilitate the distribution of P. pedifer and created a map indicating the areas where conditions are suitable for surviva

Bionomics of Phlebotomus argentipes in villages in Bihar, India with insights into efficacy of IRS-based control measures.

BACKGROUND: Visceral leishmaniasis (VL) is a deadly vector-borne disease. Approximately 90% of Indian VL cases occur in Bihar, where the sand fly, Phlebotomus argentipes, is the principal vector. Sand fly control in Bihar consists of indoor residual spraying (IRS), the practice of spraying the inner walls of village dwellings with insecticides. Prior researchers have evaluated success of IRS-control by estimating vector abundance in village houses, but the number of sampling periods (n = 2-3) were minimal, and outdoor-resting P. argentipes were neglected. We describe a large-scale field study,

Differential ecological traits of two Phlebotomus sergenti mitochondrial lineages in southwestern Europe and their epidemiological implications.

OBJECTIVES: The introduction of leishmaniasis in a new area requires a well-established population of the sandfly vector species of the parasite. No autochthonous cases of anthroponotic cutaneous leishmaniasis have been detected in southwestern Europe, and Leishmania infantum is the only causative agent of leishmaniasis in this area. Phlebotomus sergenti, the main vector of Leishmania tropica, is commonly found in the Iberian Peninsula at sufficient densities to be able to act as a vector. It is characterised by high genetic diversity and classified in four mitochondrial lineages. Our aim was

Impact of urbanization on the sand fly Phlebotomus langeroni nitzulescu in an old focus of visceral leishmaniasis in Egypt.

Changes in the environment often cause changes in disease transmission. Land use change especially urbanization can have a huge impact on transmission of vector-borne diseases. This study investigated the effect of urban development on the abundance of sand flies, in an old endemic focus of infantile visceral leishmaniasis in the north coast of Egypt. Sand fly abundance obtained in this study was compared to those obtained in 1984. In context remote sensing techniques are used to identify landscape features that might have influenced the spatial distribution of the sand fly vector in the area.

Transmission of Leishmania infantum MON-98 to hamsters by the bite of Phlebotomus langeroni Nitzulescu (Diptera: Psychodidae).

The ability of Phlebotomus langeroni to successfully acquire and transmit Leishmania infantum MON-98 to hamsters was demonstrated. Sand flies and Leishmania both originated from an infantile visceral leishmaniasis focus in El Agamy Egypt. P. langeroni females were infected by feeding on lesions of needle-inoculated hamster and on infected blood suspension using a chick-skin membrane apparatus. Infection rate of sand flies fed on membrane was 88% compared to 7.8% for flies fed on leishmanial lesion. The transmission to hamster took place by the bites of infective flies taking a second blood mea

Canine antibody response to Phlebotomus perniciosus bites negatively correlates with the risk of Leishmania infantum transmission.

BACKGROUND: Phlebotomine sand flies are blood-sucking insects that can transmit Leishmania parasites. Hosts bitten by sand flies develop an immune response against sand fly salivary antigens. Specific anti-saliva IgG indicate the exposure to the vector and may also help to estimate the risk of Leishmania spp. transmission. In this study, we examined the canine antibody response against the saliva of Phlebotomus perniciosus, the main vector of Leishmania infantum in the Mediterranean Basin, and characterized salivary antigens of this sand fly species. METHODOLOGY/PRINCIPAL FINDINGS: Sera of do

Repellent efficacy of a combination containing imidacloprid and permethrin against sand flies (Phlebotomus papatasi) in dogs.

Infection in dogs and humans with the protozoan parasite Leishmania are widespread in tropical and subtropical countries around the globe. Sand flies of the order Phlebotomus in the Old World and Lutzomyia in the New World function as the vector of this disease. In dogs, skin lesions are the most prominent signs of canine leishmaniasis, besides other complex underlying manifestations. To prevent dogs from sand fly bites and thus transmission of Leishmania, an imidacloprid 10% w/v / permethrin 50% w/v combination was evaluated for its insecticidal and repellent efficacy.
