ZmYSL2 affects the physiological function of iron deficiency in maize.

Iron deficiency resulted in reduced sensitivity of the enhanced iron tolerance in maize o213 mutants, affecting pigment synthesis, photosynthesis parameters, sugar content, and iron distribution, with variable expression of the YSL gene. Iron (Fe) is a vital trace element for the growth and development of plants. A deficiency of this element results in the formation of yellow leaves. The present study demonstrated that a mutation in the ZmYSL2 gene resulted in reduced sensitivity of o213 mutant maize seedlings to Fe deficiency. The mutation resulted in a reduction in the content of photosynthe

Retention of CdS/ZnS Quantum Dots (QDs) on the Root Epidermis of Woody Plant and Its Implications by Benzo[a]pyrene: Evidence from the in Situ Synchronous Nanosecond Time-Resolved Fluorescence Spectra Method.

The retention of CdS/ZnS QDs on the epidermis has been confirmed to be one of the core procedures during the root uptake process. However, the retention mechanisms of QDs on the epidermis of woody plant were poorly understood for lacking of an appropriate QD quantitative method. In this study, a novel method for in situ determination of CdS/ZnS QDs retained on the root epidermis was established using synchronous nanosecond time-resolved fluorescence spectroscopy. No correlations between Kf values of oleylamine-CdS/ZnS QDs retained on the epidermal tissues and the surface/bulk composition of ma

Competition between gall aphids and natural plant sinks: plant architecture affects resistance to galling.

Gall forming herbivores induce sinks and act as phloem parasites within their host plants. Their performance on the host plant can depend on the sink-source relationship they establish with the plant. Because sink-source relationships within a plant are reflected in its architecture, we examined how architectural differences among cottonwoods might influence the success of the galling aphid, Pemphigus betae. Using cloned cottonwoods in common garden studies, we found three major patterns. First, there is a significant clonal or genetic component to tree architecture; cloned trees grown in a co

SbLOX3 modulates jasmonate-associated flavonoid accumulation, root growth, and drought-induced root hollow formation in Scutellaria baicalensis.

The hollowed root formation and 4'-deoxyflavone accumulation in Scutellaria baicalensis are associated with jasmonate (JAs) signaling, but the upstream regulators integrating these processes with drought tolerance remain unknown. This study investigated whether the 13-lipoxygenase gene SbLOX3 may act as a modulator. Transgenic plants overexpressing SbLOX3 or with RNAi suppression were exposed to polyethylene glycol-simulated and long-term soil drought stress. Metabolite profiling, gene expression analysis, root morphological measurements, and histological observations were conducted. Overexpre

Rice Orange Leaf Phytoplasma Imp Protein Blocks G-M Interaction by Binding Glycoprotein G to Suppress Rice Stripe Mosaic Virus Infection.

Plants frequently encounter multiple pathogens concurrently, yet the molecular bases of pathogen-pathogen interactions during mixed infections remain largely unresolved. Here we show that co-infection of rice by Rice orange leaf phytoplasma (ROLP) and Rice stripe mosaic virus (RSMV) markedly suppresses RSMV proliferation in both host plants and the insect vector. Electron microscopy and confocal imaging reveal that ROLP and RSMV co-inhabit individual rice phloem cells and undergo close membrane interaction, producing swollen RSMV virions with reduced infectivity. We further demonstrate that th

Elevated CO2 and Water Limitation Alter the Primary Metabolite Composition of Mealybug Honeydew, Reducing Parasitoid Fitness.

Honeydew excreted by phloem-feeding insects is a major plant-derived carbohydrate resource in agroecosystems, yet its chemical sensitivity to climate change remains poorly understood. We examined how elevated atmospheric CO2 and water limitation affect honeydew primary metabolites and a honeydew-feeding parasitoid. Using a tritrophic system comprising grapevine (Vitis vinifera), the vine mealybug (Planococcus ficus), and its parasitoid (Anagyrus vladimiri), plants were grown under ambient or elevated CO2 and well-watered or water limitation conditions. Honeydew was quantified, primary metaboli

Methyl salicylate is a critical mobile signal for plant systemic acquired resistance.

In plants, the mobile signal for systemic acquired resistance (SAR), an organism-wide state of enhanced defense to subsequent infections, has been elusive. By stimulating immune responses in mosaic tobacco plants created by grafting different genetic backgrounds, we showed that the methyl salicylate (MeSA) esterase activity of salicylic acid-binding protein 2 (SABP2), which converts MeSA into salicylic acid (SA), is required for SAR signal perception in systemic tissue, the tissue that does not receive the primary (initial) infection. Moreover, in plants expressing mutant SABP2 with unregulate

Whiteflies glycosylate salicylic acid and secrete the conjugate via their honeydew.

During insect feeding, a complex interaction takes place at the feeding site, with plants deciphering molecular information associated with the feeding herbivore, resulting in the upregulation of the appropriate defenses, and the herbivore avoiding or preventing these defenses from taking effect. Whiteflies can feed on plants without causing significant damage to mesophyll cells, making their detection extra challenging for the plant. However, whiteflies secrete honeydew that ends up on the plant surface at the feeding site and on distal plant parts below the feeding site. We reasoned that thi

Poplar Tree Response to Feeding by the Petiole Gall Aphid Pemphigus spyrothecae Pass.

Pemphigus spyrothecae Pass. which is a member of the subfamily Pemphiginae is one of the gall-inducing aphids that occurs on poplar trees. Phloem feeding of a founding mother on leaf petiole results in the formation of a new organ, i.e., the spiral gall. This study documents aphid development inside the galls during the growing season and the effect of their feeding on leaf architecture and physiology of the host plant. In particular, leaf length, width, and area were measured, as well as hydrogen peroxide (H2O2) content, electrolyte leakage (EL), malondialdehyde (MDA) concentration, and the a

Vascular Bundles Mediate Systemic Reactive Oxygen Signaling during Light Stress.

Systemic signaling and systemic acquired acclimation (SAA) are essential for plant survival during episodes of environmental stress. Recent studies highlighted a key role for reactive oxygen species (ROS) signaling in mediating systemic responses and SAA during light stress in Arabidopsis (Arabidopsis thaliana). These studies further identified the RESPIRATORY BURST OXIDASE HOMOLOG D (RBOHD) protein as a key player in mediating rapid systemic ROS responses. Here, we report that tissue-specific expression of RBOHD in phloem or xylem parenchyma cells of the rbohD mutant restores systemic ROS sig

HHO5 orchestrates dose-dependent feedback regulation of organic versus inorganic nitrogen signaling in Arabidopsis.

A major goal in agriculture is to engineer crops to maintain yield with less nitrogen (N) fertilizer. Important regulators of plant N-responses include HRS1 HOMOLOG (HHO) transcription factors (TFs); yet, their redundant repressive mode of action hinders functional analyses. Here, we highlight HHO5 as a unique HHO TF based on its phylogenetic position, phloem specific expression, and dual role in regulating responses to inorganic and organic N-dose signals. Our results support a model whereby HHO5 mediates feedback repression of inorganic nitrate uptake under organic N satiety: (i) Meta-analys

Reveal the lignification of ginkgo after click chemistry by novel monolignol analogs.

Lignin is one of the abundant natural resources in the world, but most of it is discarded as waste. So, understanding the deposition and structure of lignin plays an important role lignin valorization. In order to avoid changing the side chain structure of lignin monomers to make negligible influence on lignin deposition process, propargyl group was introduced at 2/6 position of benzene ring to explore deposition mechanism of lignin in plants by polymerization of alkynyl-labeled lignin precursors (HALK, GALK) in vivo and in vitro. The results showed that HALK and GALK were biocompatibility, co

First Report of Botryosphaeria dothidea Basal Canker of Pistachio Trees in South Africa.

Pistachio (Pistacia vera) cultivation is a relatively new industry in South Africa with tremendous economic potential. However, disease problems could develop in time. The pathogens of pistachio found in other parts of the world plausibly will spread to South Africa and new pathogens never recorded on this host may develop here. In the summer of 1998, 2-year-old rootstocks of P. atlantica and P. integerrima displaying basal cankers and discolored phloem and xylem were observed in the Prieska district of the Northern Cape province. The objective of this study was to identify the causal agent(s)

A serine-rich salivary protein facilitates brown planthopper feeding and survival by mediating salivary sheath formation.

The piercing-sucking pest, brown planthopper (Nilaparvata lugens, BPH), poses a severe and persistent threat to rice in China and across Asia. During feeding, BPHs secrete salivary proteins to form salivary sheaths, which are crucial for successful and sustained phloem sap ingestion. These salivary sheath proteins are crucial for rice-BPH interactions and hold promise as targets for pest control strategies like RNAi. However, the specific functions of salivary proteins in sheath formation remain largely unclear. In this study, we identify NlSRP, a serine-rich salivary protein in BPH, as a nove

Mobile virus-derived siRNAs drive plant antiviral silencing through reiterated 3-4 cell immunization.

The movement of virus-derived small interfering RNAs (vsiRNAs) between plant cells has prompted the widely held assumption that they help immunize as-yet-uninfected tissues. Ascertaining this role is challenging because it requires uncoupling vsiRNA mobility from viral movement while preserving viral virulence. Consequently, the contribution of non-cell-autonomous RNAi to plant antiviral immunity remains essentially unknown. Here, we investigate the naturally phloem-restricted infection of turnip yellows virus (TuYV) in the stereotypical Arabidopsis root system at cellular resolution. We uncov

Excess nitric oxide alters cellular pH to restrict salicylic acid movement and systemic immunity.

Plants carrying a mutation in S-nitrosoglutathione reductase 1 (GSNOR1) accumulate high nitric oxide (NO) and exhibit impaired immunity characterized by reduced pathogen-responsive salicylic acid (SA) accumulation and failure to activate normal SA signaling. We show that this reduced SA responsiveness in gsnor1 plants arises from impaired vascular-associated movement of SA, thereby compromising systemic acquired resistance (SAR). Elevated NO perturbs cellular pH homeostasis, acidifying the apoplast and alkalinizing the cytosol, which likely interferes with phloem-associated SA movement and ren

Dual Defense Strategy of GhRAP1 Gene: Coordinating Antibiosis and Antixenosis to Enhance Aphid Resistance in Plants.

The cotton aphid (Aphis gossypii) is a major pest detrimental to cotton production, and the development of control strategies has been a key research focus. NBS-LRR-type disease resistance genes, the predominant class of R genes, have been found to confer dual resistance against aphids. This study identified 88 resistance to aphid protein (RAP) genes in Gossypium hirsutum (G. hirsutum), other cotton species, and Arabidopsis thaliana (A. thaliana), with a focus on G. hirsutum resistance to aphid protein (GhRAP1). Its expression was upregulated following aphid infestation. Functional analysis de

Generalizability of Enzyme-Based Isolation Approach for Extracellular Vesicles From Traditional Medicinal Plants.

Extracellular vesicles (EVs) from plants, particularly traditional medicinal plants, have garnered therapeutic interest. However, therapeutic utility requires establishing effective extraction and purification protocols for manufacturing. Here, we systemically optimized an enzyme-based EV extraction method and demonstrated the generalizability with different parts from seventeen medicinal plants, fresh, frozen or dried. The enzymatic pulping process (henceforth denoted as Phyto-EVpure) of fresh ginger, Morus alba leaves and Isatis indigotica Fort. roots improved EV yield and purity over grindi

Unraveling the Intertwined Networks of Potato Tuberization: From Epitranscriptomic Signaling to Metabolic Reprogramming.

Potato (Solanum tuberosum L.) tuberization is a complex developmental transition in which a subterranean stolon is converted into a starch-accumulating tuber. This process is governed by long-distance systemic signals, local hormonal dynamics, and extensive metabolic reprogramming. Recent advances have extended our understanding beyond the classical photoperiodic model, revealing the importance of chromatin remodeling, mRNA N6-methyladenosine (m6A) epitranscriptomic modifications, and a developmentally regulated shift in phloem unloading pathways. Here, we synthesize these multilayered regulat

Genome-wide identification, expression profiling, and functional analysis of sugar transporter family genes in Diaphorina citri.

Sugar transporters (STs) are important proteins responsible for the movement of sugar through the cell membrane and play pivotal roles in the growth, development, and osmoregulation of phloem-feeding hemipterans. However, information about STs in the Asian citrus psyllid (ACP), Diaphorina citri Kuwayama (Hemiptera: Liviidae), remains unknown. In this study, a total of 61 STs were identified according to the genome and transcriptome analysis of D. citri. All the ST genes have complete open reading frames (ORFs), and most of them have 12 transmembrane regions. Chromosome localization and phyloge
