Short-term thermal stratification induces hypoxia-related nutrient pulses which reshapes cyanobacterial dynamics in a shallow lake.

The reduction of external nutrient loading is a common target to address lake eutrophication, however internal nutrient dynamics can significantly delay recovery. In shallow systems, the role of episodic thermal stratification in nutrient dynamics and phytoplankton bloom ecology is often overlooked. We studied 45 years of data from the polymictic Lake Müggelsee, where short-lived stratification events strongly altered oxygen conditions, triggered pulses of soluble reactive phosphorus (SRP) and dissolved inorganic nitrogen (DIN) and had consequences for cyanobacterial community dynamics. The DI

Resolving spatiotemporal water quality variability in a river-influenced estuarine system using high-frequency autonomous surface vessel observations.

River-influenced estuarine systems support critical habitats but are vulnerable to water quality degradation driven by freshwater discharge variability, nutrient loading, and climate forcing. Understanding high-frequency variability and spatial heterogeneity remains limited due to reliance on discrete or short-term observations. This study addresses these gaps by analyzing multi-year (2021-2024), high-frequency observations collected by an autonomous surface vessel across eight transects in the Western Mississippi Sound. Ten parameters, including chlorophyll-a, phycocyanin, phycoerythrin, temp

Heat pretreatment alleviates UV-B toxicity in the cyanobacterium Anabaena doliolum: A proteomic analysis of cross tolerance.

This study offers proteomic elucidation of heat pretreatment-induced alleviation of UV-B toxicity in Anabaena doliolum. Heat-pretreated cells exposed to UV-B showed improved activity of PSI, PSII, whole chain, (14)C fixation, ATP and NADPH contents compared to UV-B alone. Proteomic analysis using two-dimensional gel electrophoresis (2-DE), MALDI-TOF MS/MS and reverse transcription polymerase chain reaction (RT-PCR) of UV-B and heat pretreatment followed by UV-B-treated cells exhibited significant and reproducible alterations in nine proteins homologous to phycocyanin-alpha-chain (PC-alpha-chai

Evaluation of Complex Formation between Aminated Polyrotaxane and Proteins Using FRET.

We previously developed an intracellular carrier, an aminated polyrotaxane (amino-PRX), that recognizes and deforms in response to the shape and charge distribution of proteins and nucleic acids. Amino-PRX can deliver various biopharmaceuticals into cells with high efficiency. However, the detailed intracellular behavior of the carrier alone and its complexes in the cells remains unclear. In addition, the usefulness of amino-PRX as a carrier for introducing antibodies into cells is unclear. In this study, we prepared fluorescently capped amino-PRX to visualize the intracellular distribution of

Summer monsoon onset-induced changes of autotrophic pico- and nanoplankton in the largest monsoonal estuary along the west coast of India.

This study presents the response of autotrophic pico- and nanoplankton to southwest monsoon-associated hydrographical transformations in the Cochin backwaters (CBW), the largest monsoonal estuary along the west coast of India. By the onset of the southwest monsoon, the euhaline/mesohaline conditions in the downstream/upstream of CBW usually transform into oligohaline/limnohaline. The flow cytometer analysis revealed the dominance of picoeukaryotes > Synechococcus > nanoautotrophs, with Prochlorococcus either very low or entirely absent. Synechococcus abundance was high during the pre-southwest

Papain- and Trypsin-Derived Spirulina Peptides: Antiproliferative and Antioxidant Profiles.

Enzymatic hydrolysis can release bioactive peptides from spirulina proteins, but the influence of protease type and molecular-weight fraction on antioxidant and cell viability endpoints remains incompletely defined. This study compared papain- and trypsin-derived fractions from a food-grade, C-phycocyanin-rich spirulina preparation. Hydrolysis was monitored by UV‒visible spectroscopy and quantified using the o-phthaldialdehyde degree-of-hydrolysis assay, after which six membrane-derived fractions (P1, P5, P10, T1, T5, and T10) were evaluated by ferric-reducing antioxidant power (FRAP) and 2,2-

Arthrospira platensis cultivation in seawater culture media for CO2 capture and production of valuable compounds.

This study investigated the effects of different culture media (Zarrouk as Control Experiment [ZCE], Zarrouk using Seawater [ZSW], and Seawater with Nutrients [NSW]) on CO2 fixation, biomass growth, and bioproduct composition of Arthrospira platensis, aiming to save the fresh water. Immediate CO2 reduction of up to 50 % was observed with ZCE, whereas ZSW and NSW initially contributed to CO2 production due to natural microorganisms and salinity stress. However, higher CO2 fixation rates were achieved by the end of the experiment after 25 days (86 %). ZSW outperformed ZCE with 80 % CO2 sequestra

Multifunctional nanogel based on carboxymethyl cellulose interfering with cellular redox homeostasis enhances phycocyanobilin photodynamic therapy.

The redox homeostasis defense mechanism of tumor cells is one of the prime reasons for the unsatisfactory effect of photodynamic therapy (PDT). So far, little attention has been paid to this obstacle. In this work, we reported a synthesizing simple yet versatile nanogel (BCPS), synthesized by cystamine dihydrochloride functionalized sodium carboxymethylcellulose (CMC-SS), bovine serum albumin, and Phycocyanobilin self-assembly. The BCPS reduced the levels of glutathione molecules by reacting with glutathione, thereby interfering with intracellular redox homeostasis and enhancing the sensitivit

Artificial complementary chromatic acclimation gene expression system in Escherichia coli.

BACKGROUND: The development of multiple gene expression systems, especially those based on the physical signals, such as multiple color light irradiations, is challenging. Complementary chromatic acclimation (CCA), a photoreversible process that facilitates the control of cellular expression using light of different wavelengths in cyanobacteria, is one example. In this study, an artificial CCA systems, inspired by type III CCA light-regulated gene expression, was designed by employing a single photosensor system, the CcaS/CcaR green light gene expression system derived from Synechocystis sp. Development of pH-Sensitive hydrogel for advanced wound Healing: Graft copolymerization of locust bean gum with acrylamide and acrylic acid.

Wounds pose a formidable challenge in healthcare, necessitating the exploration of innovative tissue-healing solutions. Traditional wound dressings exhibit drawbacks, causing tissue damage and impeding natural healing. Using a Microwave (MW)-)-assisted technique, we envisaged a novel hydrogel (Hg) scaffold to address these challenges. This hydrogel scaffold was created by synthesizing a pH-responsive crosslinked material, specifically locust bean gum-grafted-poly(acrylamide-co-acrylic acid) [LBG-g-poly(AAm-co-AAc)], to enable sustained release of c-phycocyanin (C-Pc). Synthesized LBG-g-poly(AA

Identification of Microcystis aeruginosa Peptides Responsible for Allergic Sensitization and Characterization of Functional Interactions between Cyanobacterial Toxins and Immunogenic Peptides.

BACKGROUND: The cyanobacterium species Microcystis aeruginosa produces microcystin and an array of diverse metabolites believed responsible for their toxicity and/or immunogenicity. Previously, chronic rhinitis patients were demonstrated to elicit a specific IgE response to nontoxic strains of M. aeruginosa by skin-prick testing, indicating that cyanobacteria allergenicity resides in a non-toxin-producing component of the organism. OBJECTIVES: We sought to identify and characterize M. aeruginosa peptide(s) responsible for allergic sensitization in susceptible individuals, and we investigated
