Melanin is omnipresent across diverse taxa in the biosphere, spanning microorganisms to multicellular eukaryotes. This pervasive distribution reflects its multifaceted roles, encompassing pigmentation, radical quenching, photoprotective barriers and immunomodulatory activity. The process of melanin biosynthesis can be simply defined as the conversion of tyrosine into melanin through a complex series of steps in melanosomes within melanocytes. Skin pigmentation results from melanin production by melanocytes in the epidermis. With the development of society, people are becoming increasingly conc

OBJECTIVE: Advances in melanocyte culture techniques have not yet led to reliable clinical methods for treating hypopigmentation disorders. We hypothesized that melanocytes harvested from plucked hair follicles may provide a renewable source of melanocytes for the treatment of hypopigmentation.
METHODS: Hairs with attached cells from the follicles were plucked from Yucatan pigs and implanted in a collagen-glycosaminoglycan matrix for either immediate or delayed implantation into full-thickness excisional porcine wounds. Wounds were allowed to heal and were biopsied at 2 and 4 weeks, respectiv

Daidzin, a prominent isoflavone found in soybeans, Pueraria lobata, and various legumes, has been extensively investigated for its diverse pharmacological activities, which include anticancer, antioxidant, anti-inflammatory, antiepileptic, and alcohol detoxification properties. Previous studies have shown that the dichloromethane fraction of Pueraria lobata stem (DCM-PLS) exhibits significant anti-melanogenic activity, with daidzin identified as the principal active compound. However, the precise role of daidzin in pigmentation remains incompletely understood. This study aimed to investigate t

Melanocytes are specialized neural crest-derived cells present in the epidermal skin. These cells synthesize melanin pigment that protects the genome from harmful ultraviolet radiations. Perturbations in melanocyte functioning lead to pigmentary disorders such as piebaldism, albinism, vitiligo, melasma, and melanoma. Zebrafish is an excellent model system to understand melanocyte functions. The presence of conspicuous pigmented melanocytes, ease of genetic manipulation, and availability of transgenic fluorescent lines facilitate the study of pigmentation. This study employs the use of wild-typ

Cellular homeostasis is an outcome of complex interacting processes with nonlinear feedbacks that can span distinct spatial and temporal dimensions. Skin tanning is one such dynamic response that maintains genome integrity of epidermal cells. Although pathways underlying hyperpigmentation cascade are recognized, negative feedback regulatory loops that can dampen the activated melanogenesis process are not completely understood. In this study, we delineate a regulatory role of IFN-γ in skin pigmentation biology. We show that IFN-γ signaling impedes maturation of the key organelle melanosome by

Abnormal skin pigmentation can cause significant esthetic concerns and impact quality of life. As melanin determines pigmentation, melanogenesis is a key target to manage pigmentation disorders. This study investigated the effects of extracts from the rhizome of Iris ×germanica L. var. florentina Dykes (often called Iris florentina L.) on melanogenesis. Active phytochemicals were identified by combining LC-MS-MS metabolic profiling with subsequent bioassay-directed fractionation of chromatographic eluent collected into 96-well plates. Fractions 41-43 increased melanin and contained germanaism

Cutaneous ageing is an important extrinsic process that modifies the pigmentary system. Because cellular senescence is a fundamental ageing mechanism, we examined the role of senescent cells in ageing pigmentation. Methods: Biopsies obtained from senile lentigo and perilesional normal skin were assayed for a marker of cellular senescence, p16INK4A. To determine the secretory phenotypes of senescent fibroblasts, we performed microarray, RNA sequencing and methylation array analyses in senile lentigo and senescent fibroblasts. To further investigate the impact of senescent cells on ageing-relate

INTRODUCTION AND IMPORTANCE: Cydnidae pigmentation (CP) is a rare and unique pigmentation disorder caused by contact with Cydnidae bug secretions. Only 30 cases of CP have been reported to date, and this is the first case to be reported from Nepal. The unilateral involvement and resistance to alcohol wiping make this case unique, thus adding to the existing literature on CP.
CASE PRESENTATION: A 21-year-old patient, student by occupation, presented with multiple hyperpigmented macules on the hand, observed upon waking during the monsoon season. The patient had no prior history of similar skin

Lower eyelid skin is unique and different from that of other areas. In addition to being an area of high exposure to the sun and elements, there are anatomic considerations and specific histologic characteristics that can cause the skin in this area to be more sensitive. These attributes can readily cause under-eye wrinkling and pigmentation. This review aims to present an updated overview of the current knowledge regarding the clinical characteristics, diagnosis, and management of wrinkles and pigmentation in this area. These disorders are usually caused by different factors, such as genetics

Laugier-Hunziker pigmentation (LHP) is an acquired disorder of hypermelanosis characterized by mucocutaneous hyperpigmentation. LHP may resemble various disorders characterized by mucocutaneous pigmentation. A 58-year-old lady presented with progressively increasing number of variable sized, hyperpigmented macules over the lips, fingers, toes and nails. There was no family history of similar illness. Systemic examination and all relevant investigations were within normal limits. Histopathology of a skin lesion had features consistent with LHP. The diagnosis of LHP must be made only after relev

Facial pigmentation disorders such as ephelides (commonly known as freckles) and solar lentigines are chronic dermatological conditions associated with sun exposure that can impact cosmetic appearance. This case report presents low-level laser therapy (LLLT) as an effective intervention for two different pigmentary disorders in Syrian women: one with ephelides and another with solar lentigines. The first case involved a 34-year-old woman with Fitzpatrick skin type II presenting with facial ephelides, while the second case featured a 49-year-old woman with Fitzpatrick skin type III diagnosed wi

BACKGROUND: Pigmentation-related disorders mainly revolve around abnormal melanogenesis, but the regulatory mechanisms remain unclear. Recent reports have found that COMMD3 plays a role in promoting melanin synthesis.
OBJECTIVES: This study aims to investigate the molecular mechanisms and relation with copper homeostasis of a novel pigment-promoting gene, COMMD3, in skin pigmentation.
METHODS: A UV-induced pigmentation murine model was created by UVB irradiation for examining the function of COMMD3 in acquired hyperpigmentation. Intracellular copper content was measured to determine the effe

INTRODUCTION: Haemophagocytic syndrome (HS) is a common manifestation of several congenital disorders characterised by a disruption of lysosomal secretion, interrupting the cytolytic pathway and triggering a dysfunction in the immune synapse. In this situation, the recognition of certain extra-immunological manifestations may help in the diagnostic process.
PATIENTS AND METHODS: We describe the clinical and biological features present in two brothers with familial haemophagocytic lymphohistiocytosis type 3 (FHL-3), two patients with Griscelli syndrome type 2 (GS-2) and one patient with Chédia

Determining the melanogenic capacity of basal melanocytes is vital when studying skin pigmentation and related disorders. Human skin equivalents (HSEs) can be used as a valuable tool to study pigmentation in vitro, providing in-depth analysis and characterization of molecular mechanisms associated with melanogenesis. Here, we describe a method to both visualize and quantify basal melanocytes within the epidermis of a pigmented HSE. By using an image analysis pipeline, we can determine how melanocyte density changes in response to treatment and how melanogenic activity is altered using immunofl

A 35-year-old Greek woman (Fitzpatrick Skin Type IV) presented to our clinic 4 months after the delivery of her first child with nail changes that were first noted during pregnancy. During the 12th week of pregnancy, homogenous dark brown streaks appeared first on the nails of her right second and third fingers (Figure 1a, b) and later on her right fourth toenail (Figure 2a). During the following weeks, she developed similar changes on the second and fourth toenails (Figure 2b). The streaks were uniformly brown, with distinct margins and width of 2-5 mm. There was no pigmentation in the periun

Induced pluripotent stem (iPS) cells have the ability to differentiate into multiple cell types in the body and have an unlimited growth potential. However, iPS cell-derived melanocytes produced by existing protocols have significant limitations in developing novel strategies for regenerative medicine and cell therapies of pigmentation disorders in humans because they involve culture in media containing fetal bovine serum and nonphysiological agents. In this study, we established an in vitro approach to generate iPS cell-derived human melanocytes that have higher proliferation rates and increa

The mechanisms by which the pigment melanin is transferred from melanocytes and processed within keratinocytes to achieve skin pigmentation remain ill-characterized. Nevertheless, several models have emerged in the past decades to explain the transfer process. Here, we review the proposed models for melanin transfer in the skin epidermis, the available evidence supporting each one, and the recent observations in favor of the exo/phagocytosis and shed vesicles models. In order to reconcile the transfer models, we propose that different mechanisms could co-exist to sustain skin pigmentation unde
