Antigen detection as a point-of-care test for TB: the case of lipoarabinomannan.

The limitations of sputum smear microscopy, routine chest radiology for HIV-associated TB and culture-based diagnosis are well recognized, especially in resource-limited settings. The diagnostic accuracy of a new point-of-care lateral-flow urine strip test for lipoarabinomannan (Determine(®) TB-LAM; Alere, MA, USA), which costs US$3.50 per test strip and provides results within 30 min, was evaluated in a cohort of South African patients for HIV-associated TB before starting anti-retroviral therapy in South Africa. Prevalence of culture-positive TB cases was 17.4%, among which 28.2% had sputum

Internet of medical things (IoMT)-integrated biosensors for point-of-care testing of infectious diseases.

On global scale, the current situation of pandemic is symptomatic of increased incidences of contagious diseases caused by pathogens. The faster spread of these diseases, in a moderately short timeframe, is threatening the overall population wellbeing and conceivably the economy. The inadequacy of conventional diagnostic tools in terms of time consuming and complex laboratory-based diagnosis process is a major challenge to medical care. In present era, the development of point-of-care testing (POCT) is in demand for fast detection of infectious diseases along with "on-site" results that are he

Development of point-of-care testing tool using immunochromatography for rapid diagnosis of human paragonimiasis.

Paragonimiasis, an important food-borne zoonosis, is caused by lung flukes of the genus Paragonimus. Several million people are actually infected or at risk. Paragonimiasis is a re-emerging disease in developing countries. Diagnosis of pulmonary paragonimiasis is made by finding eggs in sputa and/or fecal samples. Eggs are typically not found in ectopic paragonimiasis cases, so diagnosis depends on supportive information, such as a history of eating fresh water crabs or crayfishes, radiographic findings and immunological tests. Here, a new point-of-care-testing (POCT) tool is presented. It use

Point of care testing of phospholipase A2 group IIA for serological diagnosis of rheumatoid arthritis.

Secretory phospholipase A2 group IIA (sPLA2-IIA) was examined as a point of care marker for determining disease activity in rheumatoid (RA) and psoriatic (PsA) arthritis. Serum concentration and activity of sPLA2-IIA were measured using in-house antibodies and a novel point of care lateral flow device assay in patients diagnosed with varying severities of RA (n = 30) and PsA (n = 25) and found to correlate strongly with C-reactive protein (CRP). Levels of all markers were elevated in patients with active RA over those with inactive RA as well as both active and inactive PsA, indicating that sP

Successful Implementation of Hepatitis C Virus Point-of-Care Testing in a Syringe Services Program.

BACKGROUND: Syringe services programs (SSPs) serve many people who use drugs (PWUD) who need treatment for hepatitis C virus (HCV). The need for laboratory-based HCV RNA testing can present a barrier to diagnosis, care, and treatment. We evaluated the receipt of HCV test results and initiation of the treatment evaluation process among PWUD who received a point-of-care (POC) HCV RNA testing in an SSP. METHODS: We conducted an observational, prospective study of clients of a health department-run SSP in Seattle, Washington. Participants completed a one-step HCV testing algorithm using POC HCV R

Advancing Prostate Cancer Diagnostics: The Potential of Paper-Based Biosensors for Point-of-Care Testing.

Prostate cancer remains one of the most frequently diagnosed malignancies among men and continues to be a major cause of cancer-related deaths worldwide. Early and accurate detection is crucial for improving patient survival and enabling timely and appropriate treatment. Although the prostate-specific antigen (PSA) test is widely used for screening, its limited specificity-particularly within the diagnostic "gray zone"-often results in unnecessary biopsies, overdiagnosis, and missed detection of early-stage disease. Consequently, growing attention has turned to novel biomarkers with greater se

Systematic bias in point-of-care INR testing: comparative analysis of Abbott and Roche devices.

OBJECTIVES: Point-of-care (POC) International Normalised Ratio (INR) testing provides rapid anticoagulation monitoring. Still, it is susceptible to systematic bias, particularly at supratherapeutic levels, which can lead to inappropriate warfarin dose adjustments and increased thrombotic risk. This study evaluated bias trends between Abbott and Roche POC INR devices. METHODS: Bias trends were assessed using Royal College of Pathologists of Australasia Quality Assurance Programs survey data (2023-2025), in-house verification, and patient sample comparisons against laboratory instrument testing

Extracellular vesicles-based point-of-care testing for the diagnosis and monitoring of Alzheimer's disease.

Alzheimer's disease (AD) is a debilitating condition that affects millions of people worldwide. One promising strategy for detecting and monitoring AD early on is using extracellular vesicles (EVs)-based point-of-care testing; however, diagnosing AD using EVs poses a challenge due to the low abundance of EV-biomarkers. Here, we present a fully integrated organic electrochemical transistor (OECT) that enables high accuracy, speed, and convenience in the detection of EVs from AD patients. We incorporated self-aligned acoustoelectric enhancement of EVs on a chip that rapidly propels, enriches, an

Point-of-Care Testing in Patients with Hereditary Disorders of Primary Hemostasis: A Narrative Review.

Inherited disorders of primary hemostasis, such as von Willebrand disease and congenital platelet disorders, can cause extensive, typically mucocutaneous bleeding. Assays to diagnose and monitor these disorders, such as von Willebrand factor activity assays and light transmission aggregometry, are performed in specialized hemostasis laboratories but are commonly not available in local hospitals. Due to the complexity and relative scarcity of these conventional assays, point-of-care tests (POCT) might be an attractive alternative in patients with hereditary bleeding disorders. POCTs, such as th

Virus Detection: From State-of-the-Art Laboratories to Smartphone-Based Point-of-Care Testing.

Infectious virus outbreaks pose a significant challenge to public healthcare systems. Early and accurate virus diagnosis is critical to prevent the spread of the virus, especially when no specific vaccine or effective medicine is available. In clinics, the most commonly used viral detection methods are molecular techniques that involve the measurement of nucleic acids or proteins biomarkers. However, most clinic-based methods require complex infrastructure and expensive equipment, which are not suitable for low-resource settings. Over the past years, smartphone-based point-of-care testing (POC

An all-in-one point-of-care testing device for multiplexed detection of respiratory infections.

The impact of the COVID-19 pandemic has reinforced the need for rapid, cost-effective, and reliable point-of-care testing (POCT) devices for massive population screening. The co-circulation of SARS-CoV-2 with several seasonal respiratory viruses highlights the need for multiplexed biosensing approaches. Herein, we present a fast and robust all-in-one POCT device for parallel viral antigen and serological analysis. The biosensing approach consists of a functionalized polycarbonate disc-shaped surface with microfluidic structures, where specific bioreagents are immobilized in microarray format,

Specific lateral flow detection of isothermal nucleic acid amplicons for accurate point-of-care testing.

For point-of-care testing (POCT), coupling isothermal nucleic acid amplification schemes (e.g., recombinase polymerase amplification, RPA) with lateral flow assay (LFA) readout is an ideal platform, since such integration offers both high sensitivity and deployability. However, isothermal schemes typically suffers from non-specific amplification, which is difficult to be differentiated by LFA and thus results in false-positives. Here, we proposed an accurate POCT platform by specific recognition of target amplicons with peptide nucleic acid (PNA, assisted by T7 Exonuclease), which could be dir

The impact of antenatal syphilis point of care testing on pregnancy outcomes: A systematic review.

BACKGROUND: Mother-to-child transmission of syphilis remains a leading cause of neonatal death and stillbirth, disproportionally affecting women in low-resource settings where syphilis prevalence rates are high and testing rates low. Recently developed syphilis point-of-care tests (POCTs) are promising alternatives to conventional laboratory screening in low-resource settings as they do not require a laboratory setting, intensive technical training and yield results in 10-15 minutes thereby enabling both diagnosis and treatment in a single visit. Aim of this review was to provide clarity on th

Integrating Fluorescent Copper Nanoclusters into Hydrogel for Onsite Point-of-Care Testing of Cr(VI).

Copper nanoclusters (Cu NCs) have promising applications in fluorescence sensors due to their excellent optical properties. However, their efficient application in point-of-care testing (POCT) remains a challenge due to the limited development of solid-state fluorescence platforms. In this study, a POCT platform for Cr(Ⅵ) was fabricated based on a Cu NCs hydrogel by assisting of a smartphone. The Cu NCs hydrogel was facilely synthesized by integrating cysteine (Cys)-Cu NCs into agarose- hydrogel via a self-assembly process. The prepared Cu NCs hydrogels display more stronger emission at 600 nm

A Nested PCR-Based Point of Care Testing of Multiplex Pathogens Associated with Bloodstream Infection.

Bloodstream infections (BSI) carry high mortality, but traditional blood culture is too slow for urgent clinical needs. This study aims to develop a rapid point-of-care testing assay based on one-tube multiplex nested polymerase chain reaction (PCR) (OM-NPCR-POCT) for early diagnosis of three pathogens in bloodstream infection patients: Escherichia coli (ECO), Klebsiella pneumoniae (KPN), and Streptococcus pneumoniae (SPN). The analytical sensitivity of the one-tube multiplex nested PCR (OM-NPCR) was evaluated using recombinant bacterial plasmids. The analytical sensitivity of the OM-NPCR-POCT
