The causal web of foetal alcohol spectrum disorders: a review and causal diagram.

Foetal alcohol spectrum disorders (FASDs) are a leading cause of developmental disability. Prenatal alcohol use is the sole necessary cause of FASD, but it is not always sufficient. Multiple factors influence a child's susceptibility to FASD following prenatal alcohol exposure. Much of the FASD risk factor literature has been limited to discussions of association, rather than causation. While knowledge of predictor variables is important for identifying who is most at risk of FASD and for targeting interventions, causal knowledge is important for identifying effective mechanisms for prevention

Pharmacologic Implications of Marijuana Use During Pregnancy.

Marijuana is the most commonly used recreational drug in the United States, including among women of childbearing age and women who are pregnant. Changing legal statutes that allow for the use of medical marijuana and the decriminalization of marijuana for personal use reflect more permissive societal views on the use of this drug. Active compounds in marijuana cross the placenta rapidly and are excreted in breast milk. Results of studies of the effects of marijuana on a developing fetus and neonate are conflicting, but researchers have identified chronic marijuana exposure as a risk factor fo

Prenatal Stress Enhances Susceptibility to Allergic Diseases of Offspring.

BACKGROUND AND OBJECTIVES: Maternal prenatal stress (MPS) may result in a range of longterm consequences in the offspring among which there is susceptibility to allergic diseases. This review presents the current knowledge on the pathways through which MPS may affect the fetus, and the existing evidence regarding the association between MPS and development of allergy in the offspring. RESULT AND DISCUSSION: A pivotal mediator triggered in response to stress is the release of glucocorticoids (GC). GC may affect gene expression through binding to GC receptors, thus affecting fetal development i

Polycystic ovary syndrome: Understanding the role of the brain.

Polycystic ovary syndrome (PCOS) is a prevalent endocrine disorder and the leading cause of anovulatory infertility. Characterised by hyperandrogenism, menstrual dysfunction and polycystic ovaries, PCOS is a broad-spectrum disorder unlikely to stem from a single common origin. Although commonly considered an ovarian disease, the brain is now a prime suspect in both the ontogeny and pathology of PCOS. We discuss here the neuroendocrine impairments present in PCOS that implicate involvement of the brain and review evidence gained from pre-clinical models of the syndrome about the specific brain

Hypospadias: a transgenerational effect of diethylstilbestrol?

BACKGROUND: In 2002, an increased risk of hypospadias was reported for sons of women exposed to diethylstilbestrol (DES) in utero, suggesting transgenerational effects of DES. The aim of this study was to further assess the association between parental DES exposure and hypospadias in a case-referent study. METHODS: Cases with hypospadias were retrieved from the hospital information system. Referents were recruited via the parents of cases. Both parents completed postal questionnaires. Associations were estimated by odds ratios (OR) with 95% confidence intervals (CI). Additionally, conditional

Maternal smoking during pregnancy and the risk of childhood brain tumors: a meta-analysis of 6566 subjects from twelve epidemiological studies.

OBJECTIVE: Prior epidemiological studies suggest a possible association between maternal smoking during pregnancy and risk of childhood brain tumors. A meta-analysis was performed statistically pooling all available observational studies on this topic in order to evaluate this suspected association. METHODS: Using previously described methods, a protocol was developed for a meta-analysis examining the association between maternal smoking during pregnancy and subsequent development of primary brain tumors in their offspring. Literature search techniques, study inclusion criteria and statistica

The effect of intrauterine vascular compromise on further development of corpus spongiosum and urethra.

An in-utero experimental study was performed to evaluate the effects of intrauterine vascular compromise on further development of corpus spongiosum and male urethra. Thirty time-mated pregnant New Zealand white rabbits on their twenty-third day of gestation were used. Deterioration of the blood supply of the corpus spongiosum and urethra was attempted by electrocauterizing the perineum adjacent to the root of the phallus without damaging the corpus spongiosum and urethra, under the operating microscope. A bipolar cautery was carried out using a specially designed forceps having 100 microns in

Immediate and long-term renal effects of fetal exposure to gentamicin.

Aminoglycoside antibiotics, like gentamicin, given to pregnant females cross the placenta and accumulate in the fetal kidney, which, like the adult kidney, was found to be the major site of deposition. In young guinea-pigs whose mothers were given gentamicin during the week following nephrogenesis in the fetus, nephron growth was found to be retarded temporarily. In rats whose mothers were given gentamicin during the period of fetal nephrogenesis, the final number of nephrons was reduced by about 20%. In both cases, renal development was impaired, although the concentration of gentamicin in th

Pregnancy after gamete intrafallopian transfer in a woman with primary infertility and in utero exposure to diethylstilbestrol. A case report.

In utero exposure to diethylstilbestrol (DES) has an adverse effect on reproductive performance and may be associated with infertility. Gamete intrafallopian transfer (GIFT) is a new reproductive technique that has been advocated as an alternative to in vitro fertilization in women with at least one normally functioning fallopian tube. The process involves the translaparoscopic placement of oocytes and sperm into the fallopian tube. The technique has been successful in treating infertility due to endometriosis, male factors and immunologic factors as well as unexplained infertility. We accompl

Maternal cocaine use and genitourinary tract malformations.

A specific fetal disruption syndrome associated with maternal cocaine use has not as yet been documented. In the first 23 pregnancies evaluated in our program for chemically dependent women, one infant with prune belly syndrome and one infant with hypospadias were delivered to cocaine-using women. In the present study, all infants delivered in our program received a renal ultrasound at 2-3 days of life. Fifty infants born to cocaine-using women (group I) and 30 born to polydrug, noncocaine-using women (group II) were evaluated. The two groups of women were similar for age, gravidity, and race.

Maternal transfer of photoperiodic information influences the photoperiodic response of prepubertal Djungarian hamsters (Phodopus sungorus sungorus).

Daylengths during the spring are repeated in reverse order in the autumn. For some photoperiodic species, a given photoperiod may be stimulatory for reproduction in the spring and inhibitory in the autumn. The mechanisms regulating this type of seasonal response have, until recently, remained a mystery. Horton (1984a) showed in Microtus montanus that the photoperiod experienced by the mother influences the gonadal development of her young after weaning. To determine if this phenomenon is characteristic of other photoperiodic rodents, adult Djungarian hamsters were paired on 16L:8D, 14L:10D, or

[Metrifonate-induced congenital tremor in piglets].

Congenital tremor was experimentally induced in pigs. Eight sows of the Comborough breed that had farrowed for the first time were used, having a good body condition and a liveweight of 115 to 120 kg. Other 96 sows of the same breed and body weight were used as controls. The test animals were given metrifonate orally in the prescribed therapeutic and prophylactic doses (50 mg/kg) four times during pregnancy. Both test and control sows were fed a standard mixture No 16 during pregnancy, and were vaccinated against swine fever, leptospirosis, Aujeszky's disease, and erysipelas. Congenital tremor

Environmental and developmental origins of ovarian reserve.

BACKGROUND Oocyte number is established early in life before a gradual loss of this ovarian reserve during reproductive life until oocyte availability becomes limiting at the menopause. Although there is a large genetic component to the ovarian reserve achieved before birth, other influences including the maternal endocrine and nutritional milieu, and environmental factors may represent important developmental determinants. Environmental and nutritional factors may also modify the downward trajectory of ovarian reserve in adult life. The combination of these early and later life influences has

County-Level Environmental Quality and Risk of Hypospadias in Arkansas.

BACKGROUND: Hypospadias is one of the most common genital defects in boys, affecting about 1 in 125 live-born boys. Several studies have examined the association between a single environmental exposure and risk of hypospadias, but few have examined the possible effects of multiple environmental exposures simultaneously. Thus, we aimed to address this gap by examining the association between cumulative maternal prenatal environmental exposures and hypospadias occurrence in offspring. METHODS: We conducted a retrospective cohort study using data from 2006 to 2010 Arkansas environmental quality
