Propofol and bradycardia: causation, frequency and severity.

As part of the development of a model for the study of adverse events, we have investigated the risk of bradycardia with propofol. A systematic search for any type of report, published and unpublished, was made to review the evidence that propofol increases the risk of bradycardia, asystole and death from bradycardic events. Quantitative and qualitative analyses of data with different strengths of evidence were performed. Sixty-five published and 187 spontaneous reports to drug monitoring centres described with different strength of evidence a biological basis for propofol-induced bradycardia,

Physicochemical compatibility of propofol with thiopental sodium.

The physicochemical compatibility of propofol and thiopental sodium when mixed together in various ratios and stored was studied. Mixtures of propofol and thiopental sodium in five volume ratios from 1:3 to 3:1 were refrigerated (4 degrees C) for up to seven days and then centrifuged at 2000g for two hours. Droplet sizes were determined at intervals by optical microscopy and laser diffraction, and chemical stability of the 1:1 mixture was evaluated by high-performance liquid chromatography (HPLC). Optical microscopy and laser diffraction indicated negligible changes in droplet size within 48 h

Infectious risks associated with the use of propofol.

BACKGROUND: Investigations of the Centers for Disease Control into postoperative infections have implicated extrinsically contaminated propofol. METHODS: To evaluate the infectious risk associated with intravenous anaesthetic agents, we surveyed the literature from 1971 to 1995 using the Medline database. Papers covering infections related to intravenous anaesthetic agents were included. RESULTS: The review of the literature on infections associated with propofol and other intravenous anaesthetics shows that this infectious risk is minimal and often caused by breakdowns in aseptic techniques

Evaluation of propofol as a general anesthetic for horses.

This study provides baseline information on the potential use of propofol as a general anesthetic for horses. Using a Latin square design, propofol (2, 4, and 8 mg/kg) was administered intravenously on three separate occasions to six mature horses. Information about anesthetic induction, duration, and recovery was recorded along with results of rectal temperature, heart rate, respiratory rate, pHa, PaCO2 and PaO2. Statistical analysis included a mixed model analysis of variance, a general linear model analysis and least square means test for post hoc comparisons. A P < .05 was considered signi

The effect of propofol on hematoma formation in rhytidectomy.

Significant surgical advances in face-lift surgery have evolved over the years, including wider flap undermining, the development of the superficial musculoaponeurotic system as a separate layer, and appropriate platysmal and coronal procedures. These more radical concepts offer improved results, but they may be associated with more complications, the most frequent being hematoma formation. A retrospective study of 386 patients was undertaken over a period of 5 years. Patients were separated into two groups: those that were sedated with a drug combination of diazepam, meperidine hydrochloride,

Pharmacodynamic stability of a mixture of propofol and alfentanil.

We studied 40 patients undergoing body surface surgery in a double-blind manner to compare the pharmacodynamic stability of either mixed or separate infusions of propofol and alfentanil. No differences were found between the two groups in respiratory or cardiovascular variables during operation, or in requirements for analgesia after operation. The power of the study to determine a difference of 10 mm Hg in mean arterial pressure with a probability of 0.05 was 0.82 and for a difference of 1 kPa in end-tidal carbon dioxide partial pressure 0.89. We conclude that propofol and alfentanil may be a

Perioperative arrhythmias with a propofol-based anesthetic.

Propofol was used for the induction and maintenance of anesthesia in a patient undergoing a laparoscopic tubal ligation. This new anesthetic has not been associated with postoperative ventricular arrhythmias. This report demonstrates the occurrence of supraventricular tachycardia deteriorating to ventricular tachycardia in a patient who received propofol. Included is a discussion of the possible causes of this event.

[Anesthesia for cardioversion. A comparison of propofol and etomidate].

Anaesthesia for elective direct current cardioversion (DCC) was induced with propofol (Diprivan) 1.2 mg/kg in 28 patients and with 0.2 mg/kg etomidate (Hypnomidate) in 20 patients. These mostly high risk patients (NYHA class II to III) were successfully treated with defibrillation. Blood pressure and heart rate were recorded before and after induction and at 2 minutes intervals up to 20 minutes after DCC. Both anaesthetic agents caused mild hypotension. Heart rate did not change significantly after induction but fell significantly after DCC from the mean value of 124 +/- 26 bpm and 122 +/- 37

Total intravenous anaesthesia with propofol and buprenorphine.

A combination of propofol infusion and two bolus doses of buprenorphine, 2.5 or 5.0 micrograms/kg were evaluated in a total intravenous anaesthesia technique in 36 patients of ASA grade 1 or 2 undergoing cholecystectomy. Additional boluses of propofol were given intravenously if needed. Systolic blood pressure after tracheal intubation increased significantly only in those who received the smaller dose of buprenorphine. Patients in both groups remained haemodynamically stable throughout surgery with minimal side effects. Recovery was fast even with prolonged infusions and without major side ef

Propofol: effects on indices of cerebral ischemia.

The effects of propofol given before and during a period of profound hypotension that caused incomplete global cerebral ischaemia were investigated in anaesthetised cats. Cortical cerebral blood flow, extracellular fluid pH, potassium and calcium ion activities, and electroencephalogram were recorded. Neuropathological outcome was also assessed. Propofol-treated animals had higher cerebral blood flows than control animals after the period of hypotension (p <0.05); they also had better correction of extracellular fluid acidosis and hyperkalaemia (p <0.01) and a late improvement in calcium ion a

Propofol, the newest induction agent of anesthesia.

Propofol is a rapidly acting intravenous anesthetic agent which has many advantageous kinetic properties explaining its usefulness by bolus dose for induction of anesthesia or for administration by continuous intravenous infusion. It is rapidly distributed in the body with a half-life of only around 2 min and has an efficient hepatic and extrahepatic clearance (total body clearance may exceed liver blood flow). Premedication has little effect on the already good induction characteristics of propofol. The incidence of cardiorespiratory depression appears to be higher than that of other inductio

Propofol as an intravenous anaesthetic agent in dogs.

Studies in dogs with an emulsion formulation of the intravenous anaesthetic, propofol, showed that induction of anaesthesia was smooth and it was possible to maintain anaesthesia by intermittent injection. The mean dose for induction of anaesthesia in unpremedicated dogs was 5.95 mg/kg body-weight. When no premedication was administered anaesthesia was maintained by a total dose of approximately 0.806 mg/kg/minute. Premedication with between 0.02 and 0.04 mg/kg of acepromazine reduced the mean induction dose by about 30 per cent and the maintenance dose by more than 50 per cent. In 68 unpremed

Nurse-Administered Propofol Sedation Training Curricula and Propofol Administration in Digestive Endoscopy Procedures: A Scoping Review of the Literature.

Although efficacy and safety of nonanesthesiologist administration of propofol and nurse-administered propofol sedation practices have been amply demonstrated in patients at low American Society of Anesthesiologists physical status risk, they are still severely limited. To date, it is quite difficult to find a protocol or a shared training program. The aim of the study was to verify requirements, types of training, and operating methods described in the literature for the administration of propofol by a nurse. A scoping review of the literature was conducted in accordance with the PRISMA-ScR g

Comparison of Ketofol (Combination of Ketamine and Propofol) and Propofol Anesthesia in Aneurysmal Clipping Surgery: A Prospective Randomized Control Trial.

BACKGROUND: The maintenance of hemodynamic stability is of pivotal importance in aneurysm surgeries. While administering anesthesia in these patients, the fluctuations in blood pressure may directly affect transmural pressure, thereby precipitating rupture of aneurysm and various other associated complications. We aimed to compare the effects of ketofol with propofol alone when used as an induction and maintenance anesthetic agent during surgical clipping of intracranial aneurysms. MATERIALS AND METHODS: Forty adult, good-grade aneurysmal subarachnoid hemorrhage patients posted for aneurysm n
