Systematic evaluation of blood contamination in nanoparticle-based plasma proteomics.

Circulating blood proteomics enables minimally invasive biomarker discovery. Nanoparticle-based circulating plasma proteomics studies have reported varying number of proteins (ca 2000-7000), but it remains unclear whether a higher protein number is more informative. Here, we first develop OmniProt-a silica-nanoparticle workflow optimized through a systematic evaluation of nanoparticle types and protein corona formation parameters. Next, we present an Astral spectral library for 10,109 protein groups. Using the Astral with 60 sample-per-day throughput, OmniProt identifies ca 3000 to 6000 protei

Biofluid-Programmed Biological Identity of Nanocarriers: Consequences for Drug Targeting and Reproducible Delivery.

Nanocarriers are commonly designed and evaluated based on their physicochemical properties, including size, morphology, surface charge, composition, ligand density, drug loading, and release behavior. After administration, however, nanocarrier surfaces are rapidly remodeled by proteins, lipids, metabolites, enzymes, mucins, salts, and immune components present in systemic, mucosal, luminal, and disease-associated biofluids, thereby generating an acquired biological identity. Rather than focusing predominantly on plasma protein coronas or individual exposure environments, this review integrates

Dual Stealth Functional Immuno-magnetic Nanoparticles for High-Performance Isolation of Circulating Tumor Cells.

As a biomarker of hepatocellular carcinoma (HCC) biopsy, circulating tumor cells (CTCs) are often used in the diagnosis of cancer and treatment guidance. For CTCs detection, immuno-magnetic nanoparticles (IMNs) are one of the most commonly used platforms. However, the nonspecific adsorption of proteins and non-tumor cells weakens the performance of IMNs to capture CTCs. In this work, we developed an IMNs platform which was constructed by a biomimetic protein corona precoating and a polyethylene glycol (PEG) spacer to form the PEG and corona-coated IMNs (IP-CMNs). Due to the dual stealth effect

Nature-Inspired Alternatives to PEG for Next-Generation Long-Circulating Nanocarriers.

Polyethylene glycol (PEG) has long been considered as the gold standard for imparting stealth properties to nanocarriers. However, the recent emergence of anti-PEG antibodies has spurred innovation toward nature-inspired alternatives. By mimicking the structural and functional architectures of biological systems such as cell membranes and protein corona, nanocarriers can be engineered to achieve prolonged circulation, superior biocompatibility, and active targeting capabilities. This perspective presents nature-inspired alternatives to PEG, including cell membranes and the protein corona, for

Mitigating the toxic effects of CdSe quantum dots towards freshwater alga Scenedesmus obliquus: Role of eco-corona.

The extensive use of semiconducting nanoparticles such as quantum dots in biomedical and industrial products can lead to their inadvertent release into the freshwater system. Natural exudates in the aquatic system comprising extracellular polymeric substance (EPS) and protein-rich metabolites can eventually adsorb onto the quantum dots (QDs) surface and form an eco-corona. The alterations in the physio-chemical and toxicological behavior of CdSe/ZnS QDs under the influence of eco-corona in the freshwater system have not been explored yet. In the present study, lake water medium conditioned wit

Stealth Nanoparticles with a "Self-Consuming" Shell for Long-Term Blood Vessel Imaging.

The development of lanthanide-doped upconversion nanoparticle (UCNP)-based imaging with minimal autofluorescence and improved penetration depth is important in medical applications. Exogenous nanocarriers readily adsorb plasma proteins following intravenous administration (<0.5 min), resulting in the formation of a protein corona on the fixed surface. The protein corona facilitates UCNP interception by the immune system, preventing targeted delivery to disease sites. In this study, we report a novel surface-camouflaging strategy using lanthanide hydroxyl carbonate that is slowly dissolved by p

Ligand conformational entropy as a regime-switching descriptor of the electrostatic-uptake relationship in nanocarrier-immune cell interactions.

Surface charge is widely regarded as a primary determinant of nanoparticle-immune cell interactions, yet nanocarriers with similar physicochemical profiles often exhibit markedly different biological outcomes. Here, we show that ligand conformational diversity, quantified through an information-theoretic descriptor (Hligand), is strongly associated with a regime transition in the relationship between surface charge and cellular uptake. Analysis of 105 gold nanoparticles from a benchmark protein corona dataset identifies a critical threshold at Hligand*≈1.76 bits (F = 47.3, p < 10-14): below th

Zwitterionic Poly(ethylene glycol) Nanoparticles Minimize Protein Adsorption and Immunogenicity for Improved Biological Fate.

We report the assembly of poly(ethylene glycol) nanoparticles (PEG NPs) and optimize their surface chemistry to minimize the formation of protein coronas and immunogenicity for improved biodistribution. PEG NPs cross-linked with disulfide bonds are synthesized utilizing zeolitic imidazolate framework-8 NPs as the templates, which are subsequently modified with PEG molecules with different end groups (carboxyl, methoxy, or amino) to vary the surface chemistry. Among the modifications, the amino and residual carboxyl groups form a pair of zwitterionic structures on the surface of PEG NPs, which

Toxicokinetics and Toxicological Implications of Eco- and Bio-Corona Formation on Micro- and Nanoplastics in Aquatic Systems.

Microplastics and nanoplastics are environmentally transformable interfaces rather than static particulate debris. After release into aquatic systems, photo-oxidation, mechanical abrasion, fragmentation, and biofilm-associated aging modify their surface chemistry, morphology, polarity, and reactivity. Simultaneously, natural organic matter, humic substances, extracellular polymeric substances, proteins, lipids, and microbial products adsorb onto plastic surfaces, forming eco-coronas that regulate aggregation, colloidal stability, contaminant partitioning, and desorption behavior. Discovering nanoparticle corona ligands for liver macrophage capture.

Liver macrophages capture circulating nanoparticles and reduce their delivery to target organs. Serum proteins adsorb to the nanoparticle surface after administration. However, the adsorbed serum proteins and their cognate cell receptors for removing nanoparticles from the bloodstream have not been linked. Here we use a multi-omics strategy to identify the adsorbed serum proteins binding to specific liver macrophage receptors. We discovered six absorbed serum proteins that bind to two liver macrophage receptors. Nanoparticle physicochemical properties can affect the degree of the six serum pro

Patient-Derived Organoids as a Model to Understand Tumor Microenvironment-Driven Nano-Bio Interactions - A Framework Towards Improved Nanomedicine Translation.

Nanomedicines that perform well in conventional cell cultures often fail to translate into patients, in part due to the inability to reproduce the protein corona, and hence the biological identity, that nanoparticles acquire within the tumor microenvironment (TME). Here, we establish patient-derived colonic organoids (PDCOs) as a platform for profiling nano-bio interactions under physiologically tumor-relevant conditions using graphene quantum dots (GQD) and Silicon Quantum Dots (SiQD) as material-distinct probes. By combining spectral flow cytometry, confocal imaging, dynamic light scattering

Regulated PEGylation of cationic nanogels for enhanced siRNA delivery and orthotopic ovarian tumor therapy.

Cationic nanogels represent an alternative yet promising class of polymeric vectors for siRNA delivery. However, their intrinsic surface charges inevitably lead to the formation of a protein corona, which often compromises delivery efficiency. In this study, we propose a regulated PEGylation strategy to address this obstacle. Based on a recently developed electrostatic templated polymerization (ETP) method, we prepared cationic nanogels with precisely regulated PEG length and density, and thoroughly investigated their impact on protein corona resistance and siRNA delivery efficiency. The ident

Clinically-Driven Rapidly Developed Nanoparticle Corona for Label-Free Cerebrospinal Fluid Leakage Detection.

Rapid diagnosis of cerebrospinal fluid (CSF) leaks is critical as endoscopic endonasal skull base surgery gains global prominence. Current clinical methods such as endoscopic examination with and without intrathecal injection of fluorescent dye are invasive and rely on subjective judgment by physicians, highlighting the clinical need for label-free point-of-care (POC). However, a viable solution remains undeveloped due to the molecular complexity of CSF rhinorrhea mixed with nasal discharge and the scarcity of specific biomarkers, delaying sensor development. In this study, we accelerated the

Mechanical Programming of Hollow DNA Nanocapsules Rewires In Vivo Trafficking.

DNA nanostructures are programmable, biocompatible platforms for biomedicine; however, their mechanical properties are coupled to sequence and structural dimensions, limiting control over stiffness. Here, we report a paradigm shift from static design to mechanical engineering of DNA nanostructures. Using a nanoemulsion interfacial-confined coordination assembly strategy, hollow DNA nanocapsules (HDCs) were constructed with mechanical properties decoupled from chemical identity, without hard templates. By tuning the nanoemulsion-to-DNA-solution ratio, HDCs with sizes (∼160 nm) and surface charg

Optimization of seminal extracellular vesicle isolation protocols for cancer biomarker analysis using proteomic profiling.

Seminal extracellular vesicles (EVs) hold promise as a source of biomarkers for urogenital cancers. In this study, small seminal EVs were isolated from healthy donors by size-exclusion chromatography (SEC) and three ultracentrifugation (UC) protocols, with or without filtration. EV samples prepared by UC had a higher abundance of typical EV-related molecules such as CD9, CD81, and Alix, compared to SEC preparations. However, ribosomal and ER-related proteins were more enriched in SEC samples. Furthermore, the level of several nonvesicular proteins and/or EV-corona proteins was higher in SEC sa

Overcoming hepatic tropism: Precision engineering of lipid nanoparticles for extrahepatic RNA delivery.

Lipid nanoparticles (LNPs) are effective carriers for RNA and have become the standard of care; however, their natural affinity for the liver restricts their use elsewhere. To address this limitation, the "biological identity" of LNPs must be reprogrammed by modifying the three interconnected factors of lipid composition, physicochemical properties and protein corona modulation. Many previous studies have tended to evaluate individual regulatory factors in isolation; therefore, building on prior research, this review establishes a comprehensive conceptual framework that integrates these three

The fate of calcium carbonate nanoparticles administered by oral route: absorption and their interaction with biological matrices.

BACKGROUND: Orally administered particles rapidly interact with biological fluids containing proteins, enzymes, electrolytes, and other biomolecules to eventually form particles covered by a corona, and this corona potentially affects particle uptake, fate, absorption, distribution, and elimination in vivo. This study explored relationships between the biological interactions of calcium carbonate particles and their biokinetics. METHODS: We examined the effects of food grade calcium carbonates of different particle size (nano [N-Cal] and bulk [B-Cal]: specific surface areas of 15.8 and 0.83 m

Biomolecular Corona of Extracellular Vesicles: Report on Isev Workshop.

The concept of the "biomolecular corona" (BC) was first introduced to describe the spontaneous absorption of molecules, mainly proteins, onto the surface of synthetic nanoparticles upon exposure to biological fluids. More recently, the same term has been used to describe biomolecules that dynamically interact with the surface of extracellular vesicles (EVs) during their biogenesis, secretion, or upon exposure to intracellular or extracellular milieus, including biological fluids. The EV biomolecular corona is a key determinant of EV identity, shaping their cellular uptake, biodistribution, and

Rationally Designed Zwitterionic Peptides Improve siRNA Delivery of Cationic Diblock Copolymer-Based Nanoparticle Drug-Delivery Systems.

Nanoparticle drug delivery systems (NP DDS) have proven to be tremendously impactful for delivering therapeutic agents in cancer treatments, vaccinations, gene therapy, diagnostics, and enabled new agents such as RNA therapeutics. However, the exposure of NP DDS to biological milieus leads to the rapid adsorption of proteins and other molecules, forming a proteinaceous corona that obscures NP surface characteristics and controls the biological interactions of the NP DDS. This corona often potentiates phagocytic uptake, hindering the efficacy of targeting groups, and reducing NP drug delivery e
