We investigated the behavioral responses of the slime mold Physarum polycephalum to Escherichia coli expressing human small GTPases to elucidate how internalized proteins affect its actomyosin-based motility machinery. We found that prey bacteria expressing hKRAS WT or hRhoA triggered a deterministic "conflict" behavior, in which P. polycephalum failed to complete ingestion despite physical contact. Notably, the oncogenic hKRAS G12C mutation induced a transition to stochastic switching among predation, conflict, and sampling-mediated rejection, whereas hRac1 expression showed a tendency toward

Breast cancer is the most frequently diagnosed malignancy in women, and safer, more effective therapies are urgently needed. Inspired by the prenylated coumarin scaffolds of Ferulin C and Miliusol, we rationally designed and synthesized a series of novel 4-hydroxycoumarin derivatives (A, B, and PB series) to develop potent anti-breast cancer agents. Among them, lead compound PB1-3 (7-methoxy, geranyl-substituted) exhibited the most potent antiproliferative activity against MCF-7 (IC₅₀ = 3.13 μM) and 4 T1 (IC₅₀ = 4.28 μM) cells, with high selectivity over normal cells. Structure-activity relati

Dehydrodolichyl diphosphate synthase (DHDDS) is a ubiquitously expressed enzyme that catalyzes cis-prenyl chain elongation to produce the poly-prenyl backbone of dolichol. It appears in all tissues including the nervous system and it is a highly conserved enzyme that can be found in all animal species. Individuals who have biallelic missense mutations in the DHDDS gene are presented with non-syndromic retinitis pigmentosa with unknown underlying mechanism. We have used the Drosophila model to compromise DHDDS ortholog gene (CG10778) in order to look for cellular and molecular mechanisms that,

Alzheimer's disease (AD) is a neurodegenerative disease characterized by excessive deposition of β amyloid (Aβ), hyperphosphorylation of tau protein, and neuronal cell death. Recent studies have shown that myelin cell damage, which leads to cognitive dysfunction, occurs before AD-related pathological changes. Here, we examine the effect of icariin (ICA), a prenylated flavonol glycoside, in improving cognitive function in AD model mice. ICA has been reported to exhibit cardiovascular protective functions and antiaging effects. In this study, we used 3 × Tg-AD mice as an AD model. The Morris wat

Rab38 is a new member of the Rab small G protein family that regulates intracellular vesicle trafficking. Rab38 is expressed in melanocytes and it has been clarified that a point mutation in the postulated GTP-binding domain of Rab38 is the gene responsible for oculocutaneous albinism in chocolate mice. However, basic information regarding recombinant protein production, intracellular location, and tissue-specific expression pattern has not yet been reported. We produced recombinant Rab38 using a baculovirus/insect cell-protein expression system. A combination of Triton X-114 phase separation

The increasing resistance of phytopathogenic bacteria to chemical pesticides has underscored the urgent need for novel bactericidal agents. Sophora flavescens produces a wide range of secondary metabolites with a wide range of biological activities. Here, eight prenylated flavonoids were evaluated for the activity against phytopathogenic bacteria. Among the compounds tested, sophoraflavanone G (SFG) demonstrated broad-spectrum activity, showing a MIC50 of 1.56 μg/mL against Xanthomonas oryzae pv oryzae (Xoo). Subsequently, the mechanism of SFG on Xoo was initially explored. SFG inhibited biofi

The integral membrane zinc metalloprotease ZMPSTE24 plays a key role in the proteolytic processing of farnesylated prelamin A, the precursor of the nuclear scaffold protein lamin A. Failure of this processing step results in the accumulation of permanently farnesylated forms of prelamin A which cause the premature aging disease Hutchinson-Gilford Progeria Syndrome (HGPS), as well as related progeroid disorders, and may also play a role in physiological aging. ZMPSTE24 is an intriguing and unusual protease because its active site is located inside of a closed intramembrane chamber formed by sev

Bisphosphonates (BPs) are the most widely used anti-resorptive agents and first-line drugs for managing bone-related diseases, such as osteoporosis, Paget disease of bone, and bone metastatic cancer. BPs are known to inhibit osteoclasts' functions, and recent studies have highlighted the importance of macroautophagy/autophagy in osteoclasts. However, the involvement of autophagy in BP-mediated inhibition of osteoclast functions remains unclear. In this study, we showed that BPs inhibit the bone resorptive functions of osteoclasts by blocking autophagy. At the non-apoptotic doses, zoledronic ac

INTRODUCTION: Biofilm formation by Streptococcus agalactiae (S. agalactiae) is a major driver of chronic bovine mastitis that resists conventional antibiotic treatment, yet targeted strategies that disarm its biofilm machinery remain limited. Sophora flavescens (S. flavescens) is a medicinal plant rich in prenylated flavonoids with antimicrobial properties, but the antibiofilm mechanisms of these compounds against S. agalactiae remain unexplored.
METHODS: The antibacterial activity of 15 prenylated flavonoids from S. flavescens was evaluated against major mastitis-associated bacteria, and min

Statins, traditionally used for managing hypercholesterolemia, have emerged as promising agents for cancer therapy. By targeting the mevalonate pathway-a cornerstone of cellular metabolism and tumorigenesis-statins disrupt critical processes for cancer cell survival and proliferation. Some of these processes include cholesterol biosynthesis, protein prenylation, and post-translational modifications. This review discusses repurposing statins for cancer treatment given their anti-tumoral effects across many cancers, including breast, prostate, colorectal and hepatocellular carcinoma. Despite sta

Choroideremia, an X-linked retinal dystrophy causing progressive vision loss, arises from deficient Rab escort protein-1 (REP1), critical for the prenylation of Rab proteins. The precise mechanisms linking REP1 dysfunction to retinal degeneration remain unclear. Here, we generated conditional REP1 knockout mouse to model choroideremia and dissect REP1's role in retinal homeostasis. Histological analysis revealed severe photoreceptor (PR) layer thinning by postnatal day 30, accompanied by disrupted synaptic architecture in the outer plexiform layer. Electroretinography revealed significant visu

Closely related Aspergillus section Nigri species are difficult to resolve taxonomically, while short-read comparisons may be influenced by reference-genome choice. We applied reciprocal genome projection to 31 whole-genome sequencing libraries representing South African clinical section-Nigri isolates, Japanese and USA A. niger-labeled isolates, and A. welwitschiae-labeled isolates. Reads were aligned independently to A. niger ATCC 1015 and A. welwitschiae Aspwel1, followed by reference-specific differential-representation and KEGG analyses. Species-level classification showed substantial ove

Despite the success of BCR-ABL1 tyrosine kinase inhibitors in patients with chronic myeloid leukemia (CML), resistance to tyrosine kinase inhibitors remains a therapeutic challenge. One strategy used to overcome resistance is combination of existing BCR-ABL1 tyrosine kinase inhibitors with agents that target alternative pathways. We report that inhibition of isoprenylcysteine carboxylmethyltransferase (Icmt), a key enzyme in the protein prenylation pathway, with the selective inhibitor cysmethynil enhances the effect of BCR-ABL1 tyrosine kinase inhibitors in killing CML cells. Cysmethynil augm

Isoprenylation is an important post-transcriptional modification of small GTPases required for their activation and function. Isoprenoids, including farnesyl pyrophosphate (FPP) and geranylgeranyl pyrophosphate, are indispensable for isoprenylation by serving as donors of a prenyl moiety to small G proteins. In the human body, isoprenoids are mainly generated by the mevalonate pathway (also known as the cholesterol-synthesis pathway). The hydroxymethylglutaryl coenzyme A reductase catalyzes the first rate-limiting steps of the mevalonate pathway, and its inhibitor (statins) are widely used as

WecA is an integral membrane protein that initiates the biosynthesis of enterobacterial common antigen and O-antigen lipopolysaccharide (LPS) by catalyzing the transfer of N-acetylglucosamine (GlcNAc)-1-phosphate onto undecaprenyl phosphate (Und-P) to form Und-P-P-GlcNAc. WecA belongs to a large family of eukaryotic and prokaryotic prenyl sugar transferases. Conserved aspartic acids in putative cytoplasmic loops 2 (Asp90 and Asp91) and 3 (Asp156 and Asp159) were targeted for replacement mutagenesis with either glutamic acid or asparagine. We examined the ability of each mutant protein to compl

The prenylated protein carboxyl methyltransferase (PPMT) catalyzes the posttranslational methylation of isoprenylated C-terminal cysteine residues found in many signaling proteins such as the small monomeric G proteins and the gamma subunits of heterotrimeric G proteins. Here we report that both membrane-bound PPMT from rat kidney and the recombinant bacterially expressed form of the enzyme required divalent cations for catalytic activity. Unlike EDTA and EGTA, the metal chelator 1,10-phenanthroline strongly inhibited the PPMT activity of kidney intracellular membranes in a dose- and time-depe

Advances in basic immunology in 2016 included studies that further characterized the role of different proteins in the differentiation of effector T and B cells, including cytokines and proteins involved in the actin cytoskeleton. Regulation of granule formation and secretion in cytotoxic cells was also further described by examining patients with familial hemophagocytic lymphohistiocytosis. The role of prenylation in patients with mevalonate kinase deficiency leading to inflammation has been established. We reviewed advances in clinical immunology, as well as new approaches of whole-genome se

Bone-resorbing osteoclasts are highly dependent on vesicular trafficking pathways that are regulated by Rab GTPases. In particular, polarised transport of acidic vesicles of the endocytic/lysosomal pathway is required for formation of the ruffled border, the resorptive organelle of the osteoclast. The breakdown products of resorption are then transported through the osteoclast by transcytosis, enabling their excretion. In this review, we summarise these trafficking routes, highlight the emerging evidence that the bone disease osteopetrosis results from defects in vesicular trafficking in osteo

Polycystic ovary syndrome (PCOS) is the most common endocrinopathy affecting reproductive-aged women. The hyperandrogenemia associated with the syndrome is a result of excessive growth and steroidogenic activity of theca-interstitial tissues in response to various factors, including elevated gonadotropins, hyperinsulinemia, and oxidative stress. PCOS frequently coexists with other cardiovascular risk factors, such as dyslipidemia and systemic inflammation. Statins inhibit the synthesis of mevalonate, the key precursor to cholesterol biosynthesis, and reduce cardiovascular morbidity and mortali

The phenotype and antigen-specificity of T cells expanded by mitogenic stimulation from intra-ocular fluid (IOF) samples of affected eyes of six Behçet's disease (BD) patients, and seven patients with other uveitis entities, were determined. High numbers of gammadelta T cells, predominantly Vgamma9Vdelta2 T cells, were only detected in the IOF-derived TCL of three BD patients. Whereas no TCL responded to heat shock protein (HSP) 65 kDa, reactivity to isopentyl pyrophosphate (IPP) and related non-peptide prenyl pyrophosphates (PPP) was restricted to the gammadelta T cell containing TCL.
