Extensive Necrotic Skin Lesions Due to Post-varicella Protein S Deficiency.

Postvaricella protein S deficiency is a rare and severe disease. We report a case of extensive necrotic skin lesions of acute onset 7 days after varicella in a 4-year-old girl. Protein S antigen and activity were <10%, and antiprotein S antibodies were detected. She was treated with anticoagulation, plasmapheresis and fresh frozen plasma. She survived but required leg amputation.

Protein S deficiency complicated pregnancy in women with recurrent pregnancy loss.

This prospective study aimed to evaluate pregnancy outcome and complications in women with recurrent pregnancy loss (RPL) and protein S (PS) deficiency, who received low dose aspirin (LDA) or LDA plus heparin (LDA/H) therapies. Clinical characteristics, pregnancy outcome and complications of 38 women with two or more RPL and <60% of plasma free PS antigen were compared among three groups: antiphospholipid antibody (aPL)-negative women who received LDA (group A), aPL-negative women who received LDA/H (group B) and aPL-positive women who received LDA/H (group C). Gestational weeks (GW) at delive

Low cut-off values increase diagnostic performance of protein S assays.

Conflicting data have been reported on the accuracy of protein S (PS) assays for detection of hereditary PS deficiency. In this study we assessed the diagnostic performance of two total PS antigen assays, four free PS assays and three PS activity assays in a group of 28 heterozygous carriers of mutations in PROS1 and 165 control subjects. Several control groups were formed, one of healthy volunteers and - because PS levels are influenced by oral contraception and pregnancy, and assays measuring PS activity may be influenced by the presence of the factor V Leiden mutation -, we also investigate

GOSPEL: a neuroprotective protein that binds to GAPDH upon S-nitrosylation.

We recently reported a cell death cascade whereby cellular stressors activate nitric oxide formation leading to S-nitrosylation of GAPDH that binds to Siah and translocates to the nucleus. The nuclear GAPDH/Siah complex augments p300/CBP-associated acetylation of nuclear proteins, including p53, which mediate cell death. We report a 52 kDa cytosolic protein, GOSPEL, which physiologically binds GAPDH, in competition with Siah, retaining GAPDH in the cytosol and preventing its nuclear translocation. GOSPEL is neuroprotective, as its overexpression prevents NMDA-glutamate excitotoxicity while its

Tuberous sclerosis with portal vein thrombosis, protein C and S deficiency.

A 29-year-old lady with a bad obstetric history and portal vein thrombosis, presented to the Skin OPD for facial lesions. On examination, angiofibromas on face, shagreen patch and periungual fibromas were observed. She also had dental pits and a retinal hamartoma. Investigations revealed hamartomas in the brain and kidney. Hematological work-up showed protein C and S deficiency with Factor V Leiden positivity. Except for the cutaneous symptoms, the patient did not have any clinical manifestations in other organs affected by tuberous sclerosis. A similar association of tuberous sclerosis with p

Cerebral venous thrombosis in pregnancy: the role of protein S deficiency.

We report a 20 year-old, 28 week-pregnant woman, who developed superior sagittal sinus thrombosis, associated with a decreased free protein S level. Her father, who had been a stroke victim, had also significantly lower level of protein S. In very rare cases, a congenital or acquired protein S deficiency leads to cerebral venous thrombosis. The detection of such abnormalities has major practical consequences for the long-term management of patients to prevent further thrombotic episodes.

S-100 protein in the diagnosis of tuberculoid/borderline tuberculoid leprosy.

BACKGROUND: A definitive diagnosis of tuberculoid and borderline tuberculoid leprosy is based on a demonstration of either acid-fast bacilli or nerve elements within the granulomas. On routine hematoxylin and eosin stains, the nerve fibers are not easily identifiable. In this study, we used S-100 protein to highlight the nerve elements and to count their numbers in leprosy and non-leprosy granulomas. MATERIALS AND METHODS: Skin biopsy specimens from 15 cases of tuberculoid/borderline tuberculoid leprosy and 14 cases belonging to other granulomatous diseases of the skin were stained with S-100

Warfarin skin necrosis in a postpartum woman with protein S deficiency.

BACKGROUND: Warfarin-induced skin necrosis is a rare and dangerous complication affecting 0.01-0.1% of patients on warfarin. Deficiencies in protein C or protein S in association with other factors have been implicated in its etiology. No report has described this disorder in the immediate postpartum period in patients with protein S deficiency. CASE: A 1-week postpartum woman with known protein S deficiency presented with skin necrosis after a previously uneventful course of warfarin. CONCLUSION: Reduced levels of free protein S during the antepartum and immediate postpartum periods predisp

Bone mineral density and its association with inherited protein S deficiency.

Two unrelated children with thrombotic disease associated with inherited protein S deficiency and osteopenia have been identified. Measurement of protein S yielded markedly reduced levels of free protein S (less than 12.5%) in both propositi, a normal level of total protein S (79%) in propositus #1, and markedly reduced level of total protein S (34%) in propositus #2. Bone densitometry measurements of the two children revealed trabecular vertebral bone mineral content below two standard deviations. This defect is associated with vertebral body compression fractures in propositus #2. Therefore,

S-100 protein content of dermal and oral mucosal blue nevi.

Three common blue nevi from skin of the trunk and upper extremities, and 5 from oral mucosae were studied using an immunoperoxidase stain for S-100 protein. The skin lesions were uniformly negative whereas all of the oral mucosal lesions contained numerous positively stained dendritic cells. This difference may, in part, be explained by the different embryologic origins of the connective tissue stromas; the connective tissues of the head and neck are thought to be of neural crest origin whereas the connective tissues in the rest of the body are of mesodermal origin. These findings strengthen a

Detection of S-100 protein in melanocytic and neurogenic cutaneous tumors.

Quantitative determination of S-100 protein content on human melanoma tissues and in sera of melanoma patients was made possible through radioimmunoassay (RIA). Melanoma tissue extracts prepared from metastatic sites in both skin and lymph nodes contained 0.08-2.80% of S-100 out of the total extractable proteins, and it was also noticed that the tumors obtained from lymph-node metastasis possessed slightly higher levels of S-100 as compared with those obtained from the skin lesions. Serum S-100 levels of 12 melanoma patients with Stage 1 to 3 diseases were also examined by means of RIA, but on

Unleashing the potential of S-layer proteins for engineered living materials.

The field of engineered living materials (ELMs) aims to create self-regenerative, self-assembled, and multifunctional materials that mimic natural biomaterials. Novel ELMs can be produced by engineering biomolecules that are naturally secreted and displayed on bacterial cell surfaces. Surface-layer (S-layer) proteins are a class of proteins that form a two-dimensional paracrystalline lattice on the surface of many prokaryotes. These proteins provide a secretion, surface-anchoring, and high-density display platform that can be exploited for material formation. In this review, we discuss two str

Mechanisms of NO-Mediated Protein S-Nitrosylation in the Lens-Induced Myopia.

BACKGROUND: Myopia is a chronic ocular disease, emerging as the most common type of refractive error. This study intends to preliminarily explore the roles of protein S-nitrosylation of nitric oxide (NO) in the regulation of myopia by detecting the expression of neuronal nitric oxide synthase (nNOS) and downstream S-nitrosylation, using the animal model of lens-induced myopia (LIM) in mice. METHODS: The 3-week-old C57BL/6 J mice were divided into three groups: group I, lens-induced 0-week group (take eyeballs at the age of 3 weeks); group II, self-control eyes of experimental group (take eyeb

Use of the "tag-switch" method for the detection of protein S-sulfhydration.

Protein S-sulfhydration (i.e., converting protein cysteines -SH to persulfides -SSH) is a redox-based posttranslational modification. This reaction plays an important role in signaling pathways mediated by hydrogen sulfide or other reactive sulfane sulfur species. Recently, our laboratories developed a "tag-switch" method which can be used to selectively label and detect protein S-sulfhydrated residues. In this chapter, we provide a comprehensive summary of this method, including the design of the method, preparation of the reagents, validation on small-molecule substrates, as well as applicat

Anaesthetic management of the parturient with protein S deficiency and lumboperitoneal shunt.

Protein S, a vitamin K-dependent plasma protein, is a cofactor for protein C, an essential component of the regulatory system of coagulation. We describe the anesthetic management of a parturient with protein S deficiency and an indwelling lumboperitoneal shunt, which was placed following transverse and sagittal sinus thrombosis. She was treated with aspirin and enoxaparin during pregnancy and underwent caesarean section under general anaesthesia without complication.

Distinct interactions of GTP, UTP, and CTP with G(s) proteins.

Early studies showed that in addition to GTP, the pyrimidine nucleotides UTP and CTP support activation of the adenylyl cyclase (AC)-stimulating G(s) protein. The aim of this study was to elucidate the mechanism by which UTP and CTP support G(s) activation. As models, we used S49 wild-type lymphoma cells, representing a physiologically relevant system in which the beta(2)-adrenoreceptor (beta(2)AR) couples to G(s), and Sf9 insect cell membranes expressing beta(2)AR-Galpha(s) fusion proteins. Fusion proteins provide a higher sensitivity for the analysis of beta(2)AR-G(s) coupling than native sy

Idiopathic osteonecrosis in an adult with familial protein S deficiency and hyperhomocysteinemia.

We describe a 36-year-old man with familial protein S deficiency and homozygosity to the methylene tetrahydrofolate reductase (MTHFR) thermolabile variant who had a stroke followed by an episode of idiopathic osteonecrosis that was successfully managed by surgical core decompression. The patient's symptomatic thrombophilia, as well as that of several of his first-degree relatives who also had thrombotic events, raises the possibility that the thrombophilia was a contributing factor to the development of his osteonecrosis.

[Cerebrovascular accident and aortic thrombosis in a patient with protein S deficiency].

The authors report the case of a 50 year old man admitted to hospital with a right hemiplegia and aphasia of sudden onset in whom embolic fragments were found in the left mid and anterior cerebral artery territories at left carotid angiography : transoesophageal echocardiography demonstrated a protrusive plaque of atherosclerosis in the ascending aorta and a pediculated thrombosis in the descending aorta. Biological investigations revealed a protein S level of 3% (normal : 70-140%). This case illustrates the acute development of a thromboembolic phenomenon originating from the aortic arch in a

Conformational and hydrophobic properties of rat and bovine S-100 proteins.

The binding of Ca2+ to rat or bovine S-100 proteins, in the absence of ligands, showed a dissociation constant (in 60 mM K+) of 0.5 to 1.0 mM as measured by the effects of Ca2+ on binding of S-100 to phenyl-Sepharose, reactivity of sulfhydryl groups, and difference spectra for PHE, TYR, and TRP residues. Binding of the ligands, "Stainsall" and chlorpromazine lowered the dissociation constant of S-100 for Ca2+ by 2- to 10-fold as measured by the same parameters. The conformational change, in response to Ca2+ binding, probably occurs by exposure to solvent of the hydrophobic region of alpha and
