Pseudouridimycin Biosynthesis: Biochemical Characterization of the Glucose-Methanol-Choline (GMC) Family Oxidoreductase, PumI.

Pseudouridimycin (PUM) is a C-nucleoside antibiotic that selectively inhibits bacterial RNA polymerase (RNAP) with remarkable potency. It binds to the nucleoside triphosphate (NTP) entry region in the RNAP active site by mimicking uridine-5'-triphosphate (UTP), thus blocking RNA synthesis in bacteria. Since PUM does not inhibit human RNAP, it presents a highly selective scaffold for clinical applications. Besides its unique mode of action, PUM's peptidyl C-nucleoside structure comprises a rare pseudouridine (PU) moiety linked to an N-hydroxylated dipeptide, which is crucial for binding interac

Urinary metabolomics of HCV patients with severe liver fibrosis before and during the sustained virologic response achieved by direct acting antiviral treatment.

Hepatitis C virus (HCV) infection induces a long-term inflammatory response and oxidative-stress in the liver microenvironment, leading to hepatic fibrosis and metabolic alterations. Direct-acting-antiviral-agents (DAAs) induce HCV-clearance, even though liver damage is only partially restored. In this context, understanding the impact of viral-eradication on liver metabolic activities could allow optimizing the metabolic care of the patient. The present prospective longitudinal study aims at characterizing the urinary metabolic profile of HCV-induced severe liver fibrosis and the metabolic ch

A noninflammatory mRNA vaccine for treatment of experimental autoimmune encephalomyelitis.

The ability to control autoreactive T cells without inducing systemic immune suppression is the major goal for treatment of autoimmune diseases. The key challenge is the safe and efficient delivery of pharmaceutically well-defined antigens in a noninflammatory context. Here, we show that systemic delivery of nanoparticle-formulated 1 methylpseudouridine-modified messenger RNA (m1Ψ mRNA) coding for disease-related autoantigens results in antigen presentation on splenic CD11c+ antigen-presenting cells in the absence of costimulatory signals. In several mouse models of multiple sclerosis, the dis

Insights into serum metabolic biomarkers for early detection of incident diabetic kidney disease in Chinese patients with type 2 diabetes by random forest.

Diabetic kidney disease (DKD) is a leading cause of end-stage renal disease (ESRD) worldwide. Early detection is critical for the risk stratification and early intervention of progressive DKD. Serum creatinine (sCr) and urine output are used to assess kidney function, but these markers are limited by their delayed changes following kidney pathology, and lacking of both sensitivity and accuracy. Hence, it is essential to illustrate potential diagnostic indicators to enhance the precise prediction of early DKD. A total of 194 Chinese individuals include 30 healthy participants (Stage 0) and 164

E2-based mRNA vaccine encapsulated in lipid nanoparticles protects pigs against classical swine fever virus.

UNLABELLED: Classical swine fever (CSF), caused by the classical swine fever virus (CSFV), remains a significant threat to the global pig industry. Recent advances in mRNA vaccines offered a promising platform for combating CSFV. In this study, we designed and evaluated three lipid nanoparticle (LNP)-encapsulated mRNA vaccine candidates encoding the ectodomain of E2 glycoprotein (E2_EX), E2_EX fused with the transmembrane (TM) region of the PEDV S protein (E2tm), and E2_EX fused with the TM region of the influenza virus HA protein (E2tm-HA). Among these, the E2tm mRNA vaccine induced the most

Comparison of uridine and N1-methylpseudouridine mRNA platforms in development of an Andes virus vaccine.

The rodent-borne Andes virus (ANDV) causes a severe disease in humans. We developed an ANDV mRNA vaccine based on the M segment of the viral genome, either with regular uridine (U-mRNA) or N1-methylpseudouridine (m1Ψ-mRNA). Female mice immunized by m1Ψ-mRNA developed slightly greater germinal center (GC) responses than U-mRNA-immunized mice. Single cell RNA and BCR sequencing of the GC B cells revealed similar levels of activation, except an additional cluster of cells exhibiting interferon response in animals vaccinated with U-mRNA but not m1Ψ-mRNA. Similar immunoglobulin class-switching and

Urine metabolomic changes in cats with renal disease or calcium oxalate uroliths.

Two common problems in the urinary system of cats are renal disease (RD) and calcium oxalate (CaOx) stones. The objective of this study was to assess urine metabolomic parameters of cats with these diseases to determine metabolic abnormalities and differences between the groups. The urine metabolic profile for each cat was determined, along with their lifetime history of stone incidence and renal disease. In order to reduce the data for analysis, factor analysis/factor loading was used allowing statistical hypothesis testing and the selection of significant metabolites from 488 identified meta

RNA Modifications and Epigenetics in Modulation of Lung Cancer and Pulmonary Diseases.

Lung cancer is the leading cause of cancer-related mortality worldwide, and its tumorigenesis involves the accumulation of genetic and epigenetic events in the respiratory epithelium. Epigenetic modifications, such as DNA methylation, RNA modification, and histone modifications, have been widely reported to play an important role in lung cancer development and in other pulmonary diseases. Whereas the functionality of DNA and chromatin modifications referred to as epigenetics is widely characterized, various modifications of RNA nucleotides have recently come into prominence as functionally imp

Atmospheric pressure ionization mass spectrometry of purine and pyrimidine markers of inherited metabolic disorders.

Purines and pyrimidines are of interest owing to their significance in processes in living organisms. Mass spectrometry is a promising analytical tool utilized in their analysis. Two atmospheric pressure ionization (API) methods (electrospray ionization (ESI) and atmospheric pressure chemical ionization (APCI)) in both negative and positive modes applied to selected purine and pyrimidine metabolites (markers of inherited metabolic disorders) were studied. APCI is less sensitive to alkali metal cations present in a sample and offers higher response than ESI for studied compounds. Both of the te

Nucleoside Modifications and Poly(A) Tail Length Greatly Influence Protein Expression from In Vitro-Transcribed mRNA in a Salmonid Cell Line.

BACKGROUND/OBJECTIVES: It is challenging to develop efficient vaccines against intracellular pathogens such as viruses, and since viral infections are one of the main challenges for farmed salmon, a novel vaccine strategy is needed. mRNA vaccines are optimized and approved for humans, but for fish, the mRNA technology is new, and optimization is required to ensure efficient protein expression. We made an mRNA tailored to salmon and studied the effect of modified nucleosides and the length of the poly(A) tail on protein expression from in vitro-transcribed mRNA in CHSE-214 cells, using enhanced

Plasma metabolites and lipids associate with kidney function and kidney volume in hypertensive ADPKD patients early in the disease course.

BACKGROUND: Autosomal dominant polycystic kidney disease (ADPKD) is the most common hereditary kidney disease and is characterized by gradual cyst growth and expansion, increase in kidney volume with an ultimate decline in kidney function leading to end stage renal disease (ESRD). Given the decades long period of stable kidney function while cyst growth occurs, it is important to identify those patients who will progress to ESRD. Recent data from our and other laboratories have demonstrated that metabolic reprogramming may play a key role in cystic epithelial proliferation resulting in cyst gr

Urinary excretion of purine and pyrimidine metabolites in the neonate.

Random urine samples from 614 neonates were screened for metabolites of purine and pyrimidine metabolism using an adapted column chromatographic method. A restricted number of metabolites and a number of unidentified peaks appeared on the chromatograms. No inborn errors of this metabolism were found. The chromatograms were identical in term and in premature or dysmature neonates, except for the presence of more unidentified peaks in the latter group. The pattern was not influenced by the type of feeding or i.v. nutrition. Metabolites of different medications were identified. One female neonate

[Clinical significance and pathogenic mechanisms of insulin resistance in chronic renal insufficiency (part II): pathogenic factors of insulin resistance in chronic renal insufficiency].

INTRODUCTION: Factors implicated in the pathogenesis of insulin resistance in chronic renal failure are: uremic toxins, exercise tolerance, metabolic acidosis, secondary hyperparathyroidism, vitamin D deficiency. Many of them may contribute, but are not the main cause of insulin resistance in uremia. The aim of this review is to debate about each, separately. UREMIC TOXINS: Hippurate and pseudouridine are specific for uremia and inhibit glucose utilization at concentrations found in sera of uremic subjects. Partially purified toxins from uremic sera, after hemodialysis therapy, ameliorate bet

RNA modifications in organ fibrosis: bibliometric evolution, convergent mechanisms, and emerging pharmacological targets.

Fibrosis is a common pathological outcome of chronic tissue injury, yet effective antifibrotic pharmacotherapies remain scarce. RNA modifications are covalent chemical alterations of the ribose sugar or nitrogenous bases of RNA without changing the RNA sequence. This study aimed to map the evolution of research linking RNA modifications to fibrosis and to integrate the bibliometric signals with mechanistic and pharmacological evidence. We searched the Web of Science Core Collection on June 3, 2026. After restriction to English-language articles and reviews and exclusion of two retracted public

Optimized Reverse Transfection Protocol for Telomerase mRNA Delivery to Early-Senescent Human Fibroblasts.

Cellular senescence is associated with profound alterations in cellular physiology, including reduced membrane fluidity, impaired endosomal trafficking, diminished endocytic capacity, and increased extracellular RNase activity, all of which hinder efficient mRNA delivery. These barriers have limited the application of RNA-based approaches in senescent cells, particularly for delivering large therapeutic transcripts. This protocol describes an optimized reverse-transfection method for the efficient delivery of modified messenger RNA (mRNA) into senescent human fibroblasts. Although human telome
