Successful management of colonic pythiosis in two dogs in Thailand using antifungal therapy.

Gastrointestinal pythiosis is a severe, progressive and often a fatal disease, which is caused by the aquatic pathogen Pythium insidiosum. Treatment is challenging due to the disease's resistance to antifungal drugs. Surgical resection is frequently attempted in cases of pythiosis; however, it can be technically challenging. This report presents two dogs with decreased appetite, abdominal pain, progressive haematochezia, tenesmus and significant weight loss. With the medical histories of both being young canines, living in areas with access to natural water resources and with the main chronic

Antifungal Activity of Plant-Derived Essential Oils on Pathogens of Pulse Crops.

Pulse crops such as chickpeas, lentils, and dry peas are grown widely for human and animal consumption. Major yield- and quality-limiting constraints include diseases caused by fungi and oomycetes. The environmental and health concerns of synthetic fungicides used for disease management, emergence of fungicide-resistant pathogens, and demand for organic pulse crop products necessitate the search for effective alternatives. Safe and environmentally friendly plant-derived essential oils (EOs) have been reported effective against some pathogenic fungi. Growth on EO-amended growth medium and an in

Linezolid in Combination With Azoles Induced Synergistic Effects Against Candida albicans and Protected Galleria mellonella Against Experimental Candidiasis.

The incidence of resistant Candida isolates has increased continuously in recent decades, especially Candida albicans. To overcome this resistance, research on antifungal sensitizers has attracted considerable attention. Linezolid was found to inhibit the growth of Pythium insidiosum and synergize with amphotericin B against Cryptococcus neoformans. The objective of this study was to determine the interactions of linezolid and azoles against C. albicans in vitro and in vivo. In vitro, linezolid combined with azoles induced synergistic effects not only against some susceptible C. albicans isola

Brassinosteroids antagonize gibberellin- and salicylate-mediated root immunity in rice.

Brassinosteroids (BRs) are a unique class of plant steroid hormones that orchestrate myriad growth and developmental processes. Although BRs have long been known to protect plants from a suite of biotic and abiotic stresses, our understanding of the underlying molecular mechanisms is still rudimentary. Aiming to further decipher the molecular logic of BR-modulated immunity, we have examined the dynamics and impact of BRs during infection of rice (Oryza sativa) with the root oomycete Pythium graminicola. Challenging the prevailing view that BRs positively regulate plant innate immunity, we show

Genetic and phenotypic diversity of plant growth promoting rhizobacteria isolated from sugarcane plants growing in pakistan.

Bacteria were isolated from roots of sugarcane varieties grown in the fields of Punjab. They were identified by using API20E/NE bacterial identification kits and from sequences of 16S rRNA and amplicons of the cpn60 gene. The majority of bacteria were found to belong to the genera of Enterobacter, Pseudomonas, and Klebsiella, but members of genera Azospirillum, Rhizobium, Rahnella, Delftia, Caulobacter, Pannonibacter, Xanthomonas, and Stenotrophomonas were also found. The community, however, was dominated by members of the Pseudomonadaceae and Enterobacteriaceae, as representatives of these ge

Beta-cyanoalanine synthase as a molecular marker for induced resistance by fungal glycoprotein elicitor and commercial plant activators.

ABSTRACT The biocontrol agent Pythium oligandrum produces glycoprotein elicitor in the cell wall fraction, designated CWP, and induces resistance to a broad range of pathogens. To understand the mechanism of CWP-induced resistance to pathogens, gene expression at the early stage of CWP treatment in tomato roots was analyzed using a cDNA array. At 4 h after CWP treatment, 144 genes were up-regulated and 99 genes were down-regulated. In the 144 up-regulated genes, nine genes exhibited about eightfold increased expression. Analysis of the response of these nine genes to three commercial plant act

SOBIR1 Integrates Pathogen Recognition and Hormonal Metabolism in Nicotiana tabacum.

The receptor-like kinase SOBIR1 (SUPPRESSOR OF BIR1) is a core component of immune receptor complexes mediating plant responses to extracellular immunogenic signals. While its role in pattern-triggered immunity is well defined in model species, its broader physiological functions remain largely unexplored. Here, we generated CRISPR-Cas9-derived double knockouts of two homeologous SOBIR1 genes in allotetraploid Nicotiana tabacum. The resulting Nt sobir1 double mutants failed to trigger elicitor-induced cell death in response to elicitins from Phytophthora and Pythium spp. but retained an attenu

Therapeutic penetrating keratoplasty for recalcitrant microbial keratitis: An update.

Therapeutic penetrating keratoplasty (TPK) is a surgical procedure for salvaging medically refractory infectious keratitis. By replacing diseased, infected host corneal tissue with a full-thickness donor cornea, TPK aims to eliminate infection, preserves globe integrity and maintain vision. However, recurrence of infection in the graft remains a significant concern, particularly in cases caused by resistant organisms such as Acanthamoeba, Pythium, microsporidia, and certain bacterial and fungal pathogens. Although often curative, TPK carries a higher risk of endothelial rejection, reduced graf

Targeting microbial pathogens by expression of new recombinant dermaseptin peptides in tobacco.

Dermaseptin B1 (DrsB1), an antimicrobial cationic 31 amino acid peptide, is produced by Phyllomedusa bicolor. In an attempt to enhance the antimicrobial efficacy of DrsB1, the DrsB1 encoding 93 bp sequence was either fused to the N or C terminus of sequence encoding chitin-binding domain (CBD) of Avr4 gene from Cladosporium fulvum. Tobacco leaf disk explants were inoculated with Agrobacterium rhizogenes harboring pGSA/CBD-DrsB1 and pGSA/DrsB1-CBD expression vectors to produce hairy roots (HRs). Polymerase chain reaction (PCR) was employed to screen putative transgenic tobacco lines. Semi-quant

Physical and microbial root-zone factors underlying tomato wilt in long-term biofloc aquaponic systems.

Biofloc aquaponics presents a sustainable approach to soilless crop production, but its long-term reliability is often compromised by root-zone disorders such as root rot and wilt. This study evaluated four media-based biofloc aquaponic designs over two years (encompassing two full fish production cycles and four tomato growing seasons) to investigate system failures, including plant wilting, necrosis, and the underlying physical and microbial stressors. In the first year, plant wilt was primarily linked to poor drainage and sludge accumulation in coupled systems, resulting in root-zone hypoxi

Inhibition of Oomycetes by the Mixture of Maleic Acid and Copper Sulfate.

Since the protective activity of the Bordeaux mixture against plant disease caused by oomycetes was discovered, copper compounds have been used for more than a century as an effective plant protection strategy. However, the application of excessive copper can cause adverse effects through long-term heavy metal accumulation in soils. Therefore, it is necessary to develop new strategies to reduce or replace copper in pesticides based on organic and low-input farming systems. Organic acids are eco-friendly. In this study, we tested the antifungal and anti-oomycete activity of maleic acid (MA) and

Uncoupled defense gene expression and antimicrobial alkaloid accumulation in elicited opium poppy cell cultures.

Treatment of opium poppy (Papaver somniferum L.) cell cultures with autoclaved mycelial homogenates of Botrytis sp. resulted in the accumulation of sanguinarine. Elicitor treatment also caused a rapid and transient induction in the activity of tyrosine/dopa decarboxylase (TYDC, EC 4.1.1.25), which catalyzes the conversion of L-tyrosine and L-dopa to tyramine and dopamine, respectively, the first steps in sanguinarine biosynthesis. TYDC genes were differentially expressed in response to elicitor treatment. TYDC1-like mRNA levels were induced rapidly but declined to near baseline levels within 5

Development of Resistance to Damping-Off in Rice, Oryza sativa L., Using CRISPR/Cas9.

Damping-off disease hinders rice seedling growth and reduces yield. Current control methods, such as seed or soil sterilization, rely on chemicals that cause environmental pollution and promote pathogen resistance. As a sustainable alternative, we targeted the damping-off resistance-related gene OsDGTq1 using CRISPR/Cas9. Field experiments first verified OsDGTq1's significance in resistance. The CRISPR/Cas9 system, delivered via Agrobacterium-mediated transformation, was used to edit OsDGTq1 in rice cultivar Ilmi. Lesions from major damping-off pathogens, Rhizoctonia solani and Pythium gramini

Simplified Point-of-Care Testing for Human Pythiosis: Development of a Whole-Blood-Based Lateral Flow Immunoassay.

Background/Objectives: Human pythiosis, caused by Pythium insidiosum, is associated with severe morbidity and high mortality when diagnosis is delayed. Culture-based diagnosis is time-consuming and may be insensitive in clinical specimens, highlighting the need for rapid point-of-care serodiagnostic tools. Here, we developed and clinically evaluated the Anti-Pin Antibody Test Strip, a whole-blood-compatible lateral flow immunoassay (LFIA) for detecting anti-P. insidiosum antibodies. Methods: Secretory protein antigens of P. insidiosum were prepared and conjugated to gold nanoparticles for LFIA
