Discovery and characterization of quinoxaline and quinoline carboxamides as allosteric cannabinoid receptor modulators.

Cannabinoid (CB) receptors type 1 (CB1) and type 2 (CB2) are Gi protein-coupled receptors that play important roles in brain function and immune regulation. While orthosteric CB1 and CB2 receptor agonists and antagonists have been widely studied, only few allosteric modulators are known. Screening of a proprietary compound library led to the discovery of quinoxaline-6-carboxamides as novel allosteric modulators of CB receptors. N-(4-Ethylphenyl)quinoxaline-6-carboxamide (3j, PSB-18579) acted as a negative allosteric CB1 receptor modulator as determined in β-arrestin-2 recruitment assays (IC50

Effective degradation of quinoline by catalytic ozonation with MnCexOy catalysts: performance and mechanism.

Quinoline inevitably remains in the effluent of coking wastewater treatment plants due to its bio-refractory nature, which might cause unfavorable effects on human and ecological environments. In this study, MnCexOy was consciously synthesized by α-MnO2 doped with Ce3+ (Ce:Mn = 1:10) and employed as the ozonation catalyst for quinoline degradation. After that, the removal efficiency and mechanism of quinoline were systematically analyzed by characterizing the physicochemical properties of MnCexOy, investigating free radicals and monitoring the solution pH. Results indicated that the removal ra

Microbial Efflux Pump Inhibitors: A Journey around Quinoline and Indole Derivatives.

Antimicrobial resistance (AMR) is a complex threat to human health and, to date, it represents a hot topic in drug discovery. The use of non-antibiotic molecules to block resistance mechanisms is a powerful alternative to the identification of new antibiotics. Bacterial efflux pumps exert the early step of AMR development, allowing the bacteria to grow in presence of sub-inhibitory drug concentration and develop more specific resistance mechanisms. Thus, efflux pump inhibitors (EPIs) offer a great opportunity to fight AMR, potentially restoring antibiotic activity. Based on our experience in d

Molecular targets and anticancer activity of quinoline-chalcone hybrids: literature review.

α,β-Unsaturated chalcone moieties and quinoline scaffolds play an important role in medicinal chemistry, especially in the identification and development of potential anticancer agents. The multi-target approach or hybridization is considered as a promising strategy in drug design and discovery. Hybridization may improve the affinity and potency while simultaneously decreasing the resistance and/or side effects. The conjugation of quinolines with chalcones has been a promising approach to the identification of potential anticancer agents. Most of these hybrids showed anticancer activities thro

Synthesis and biological evaluation of novel quinoline-piperidine scaffolds as antiplasmodium agents.

The parasitic disease malaria places almost half of the world's population at risk of infection and is responsible for more than 400,000 deaths each year. The first-line treatment, artemisinin combination therapies (ACT) regimen, is under threat due to emerging resistance of Plasmodium falciparum strains in e.g. the Mekong delta. Therefore, the development of new antimalarial agents is crucial in order to circumvent the growing resistance. Chloroquine, the long-established antimalarial drug, still serves as model compound for the design of new quinoline analogues, resulting in numerous new act

1,2,4-Triazole-quinoline/quinolone hybrids as potential anti-bacterial agents.

Bacterial infections, caused predominately by Gram-positive and Gram-negative organisms, are proliferation of harmful strains of bacteria on or inside the body. Hospital-acquired and community-acquired bacterial infections have already put a heavy burden on the global health system. Antibiotics can disrupt the processes necessary for bacterial cell growth and proliferation, are effective weapons to fight against bacterial infections. However, the overuse and misuse of antibiotics have led to a rise in antibiotic resistance, creating an urgent need to develop novel antibiotics. 1,2,4-Triazole-q

The arrhythmogenic cardiotoxicity of the quinoline and structurally related antimalarial drugs: a systematic review.

BACKGROUND: Several quinoline and structurally related antimalarial drugs are associated with cardiovascular side effects, particularly hypotension and electrocardiographic QT interval prolongation. A prolonged QT interval is a sensitive but not specific risk marker for the development of Torsade de Pointes-a potentially lethal polymorphic ventricular tachyarrhythmia. The increasing use of quinoline and structurally related antimalarials in mass treatments to eliminate malaria rapidly highlights the need to review their cardiovascular safety profiles. METHODS: The primary objective of this sy

Molecularly imprinted polymers for selective adsorption of quinoline: theoretical and experimental studies.

The effects of solvent on the synthesis of molecularly imprinted polymers (MIPs) for the selective adsorption of quinoline were evaluated in this work. The MIPs were synthesized by the "bulk" method using the quinoline molecule (IQ) as a template in different solvents, such as toluene (MIPT) and chloroform (MIPC). The adsorbents were characterized by thermogravimetric analysis (TGA), Fourier transform infrared (FT-IR) spectroscopy, scanning electron microscopy (SEM), and N2 adsorption/desorption measurements. The influences of time, adsorbate concentration, and temperature on the adsorption of

Antileishmanial activity of new hybrid tetrahydroquinoline and quinoline derivatives with phosphorus substituents.

Heterocyclic compounds, such as hybrid tetrahydroquinoline and quinoline derivatives with phosphorated groups, have been prepared by multicomponent cycloaddition reaction between phosphorus-substituted anilines, aldehydes and styrenes. The antileishmanial activity of these compounds has been evaluated on both promastigotes and intramacrophagic amastigotes of Leishmania infantum. Good antileishmanial activity of functionalized tetrahydroquinolines 4a, 5a, 6b and quinoline 8b has been observed with similar activity than the standard drug amphotericin B and close selective index (SI between 43 an

Sulforaphane prevents quinolinic acid-induced mitochondrial dysfunction in rat striatum.

Quinolinic acid (QA) triggers striatal neuronal death by an excitotoxic cascade that involves oxidative stress, which in turns is tightly linked to mitochondria. Mitochondrial dysfunction is a molecular feature described in several brain pathologies. In this work, we determined whether the sulforaphane-neuroprotective effect in the rodent experimental model of Huntington's disease induced by QA is associated with mitochondrial function preservation. We found that QA impaired mitochondrial function within 24 h post-lesion. Sulforaphane effectively disrupted the mitochondrial dysfunction by prev

Indoleamine 2,3 dioxygenase and quinolinic acid immunoreactivity in Alzheimer's disease hippocampus.

The present immunohistochemical study provides evidence that the kynurenine pathway is up-regulated in Alzheimer's disease (AD) brain, leading to increases in the excitotoxin quinolinic acid (QUIN). We show that the regulatory enzyme of the pathway leading to QUIN synthesis, indoleamine 2,3 dioxygenase (IDO) is abundant in AD compared with controls. In AD hippocampus, both IDO- and QUIN-immunoreactivity (-IR) was detected in cortical microglia, astrocytes and neurones, with microglial and astrocytic expression of IDO and QUIN highest in the perimeter of senile plaques. QUIN-IR was present in g

A critical review of quinoline hybrids as privileged scaffolds for anti-malarial therapy.

Malaria remains a life-threatening parasitic disease of global concern, with the emergence and spread of drug-resistant Plasmodium strains necessitating the development of novel, potent, and safe anti-malarial agents. As a privileged pharmacophore with inherent anti-malarial potential, quinoline exerts its anti-malarial effects primarily by inhibiting hemozoin crystallization, interfering with parasite metabolic pathways, disrupting DNA replication and protein synthesis, and targeting key enzymes involved in Plasmodium survival. Of note, quinoline hybrids exhibit prominent advantages, includin
