The key sensor of energy status in mammalian cells, AMP-activated protein kinase (AMPK), can also be activated by the AMP analog aminoimidazolecarboxamide nucleoside monophosphate (ZMP) generated directly from aminoimidazolecarboxamide ribonucleoside (AICAR) or from inhibition of purine synthesis by the antifolate pemetrexed (PTX), a drug used extensively in the treatment of lung cancers. Despite this common mechanism, signaling downstream of AMPK activated by PTX or AICAR differed. AICAR-activated AMPK inhibited mTORC1 both directly by phosphorylation of the mTORC1 subunit Raptor and indirect

BACKGROUND: Skeletal muscle is a plastic tissue that can adapt to different stimuli. It is well established that Mammalian Target of Rapamycin Complex 1 (mTORC1) signalling is a key modulator in mediating increases in skeletal muscle mass and function. However, the role of mTORC1 signalling in adult skeletal muscle homeostasis is still not well defined.
METHODS: Inducible, muscle-specific Raptor and mTOR k.o. mice were generated. Muscles at 1 and 7 months after deletion were analysed to assess muscle histology and muscle force.
RESULTS: We found no change in muscle size or contractile proper

The mammalian target of rapamycin complex 1(mTORC1) integrates diverse signals to control cell growth, proliferation, survival, and metabolism. Role of reactive oxygen species (ROS) on mTORC1 signaling remains obscure and mechanisms through which ROS modulate mTORC1 are not known. We demonstrate that low doses ROS exposure stimulate mTORC1 while high concentrations or long-term ROS treatment decrease mTORC1 activity in vivo and in a variety of cell lines. The dose/time needed for inhibition or activation are cell type dependent. In HEK293 cells hydrogen peroxide (H(2)O(2)) stimulates phosphoryl

During animal migration, ephemeral communities of taxa at all trophic levels co-occur over space and time. The interactions between predators and prey along migration corridors are ecologically and evolutionarily significant. However, these interactions remain understudied in terrestrial systems and warrant further investigations using novel approaches. We investigated the predator-prey interactions between a migrating avivorous predator and ephemeral avian prey community in the fall migration season. We tested for associations between avian traits and prey selection and hypothesized that prey

Liver is a central organ for lipid metabolism. Disruption of lipid homeostasis leads to lipid accumulation in hepatocytes, which is a feature of metabolic dysfunction-associated steatotic liver disease (MASLD). Mechanical force and mechanosensitive proteins have been found to play a crucial role in energy metabolism. However, their role in hepatic lipid metabolism remains unclear. In this study, mechanosensitive ion channel PIEZO1 is detected in hepatocytes, and downregulated in the liver of MASLD patients and high-fat diet (HFD)-induced MASLD mouse model. Under HFD feeding, mice with hepatocy

Traumatic brain injury (TBI) often leads to enduring axonal damage and persistent neurological deficits. While PTEN's role in neuronal growth is recognized, its long-term activation changes post-TBI and its effects on sensory-motor circuits are not well understood. Here, it is demonstrated that the neuronal knockout of PTEN (PTEN-nKO) significantly enhances both structural and functional recovery over the long term after TBI. Importantly, in vivo, DTI-MRI revealed that PTEN-nKO promotes white matter repair post-TBI. Additionally, calcium imaging and electromyographic recordings indicated that

Dysfunction of the mechanistic target of rapamycin (mTOR) signaling pathway is implicated in neuropsychiatric disorders including depression and anxiety. Most studies have been focusing on neurons, and the function of mTOR signaling pathway in astrocytes is less investigated. mTOR forms two distinct complexes, mTORC1 and mTORC2, with key scaffolding protein Raptor and Rictor, respectively. The ventral tegmental area (VTA), a vital component of the brain reward system, is enrolled in regulating both depression and anxiety. In the present study, we aimed to examine the regulation effect of VTA a

Targeted deletion of Raptor, a component of mechanistic target of rapamycin complex 1 (mTORC1), reveals an essential role for mTORC1 in initiation/maintenance of leukemia in a CLL model, resulting from a failure for haemopoietic stem/progenitor cells (HSPCs) to commit to the B cell lineage. Induction of Raptor-deficiency in NSG mice transplanted with Mx1-Raptor CLL progenitor cells (PKCα-KR-transduced HSPCs) after disease establishment revealed a reduction in CLL-like disease load and a significant increase in survival in the mice. Interestingly in an aggressive CLL-like disease model, rapamyc

BACKGROUND: Chemotherapy resistance and relapse in acute myeloid leukemia (AML) are associated with poor prognosis. Bone marrow mesenchymal stem cells (MSCs) show enhanced adipogenic differentiation in AML, which may contribute to chemoresistance; however, the underlying mechanisms remain incompletely understood.
AIMS: To investigate how adipogenic differentiation of MSCs from chemotherapy-resistant AML patients (CR-AML-MSCs) promotes chemoresistance, focusing on the roles of the fat mass and obesity-associated protein (FTO) and the mTORC1 pathway.
STUDY DESIGN: Experimental study.

BACKGROUND: Catheter kinking during coronary angiography is a common complication, particularly in patients with tortuous vascular anatomy. Traditional retrieval methods may be unsuccessful in such scenarios.
CASE SUMMARY: We report a case of a 73-year-old man presenting with cardiogenic shock secondary to severe mitral regurgitation due to posteromedian papillary muscle rupture. During coronary angiography, the guide catheter kinked in the common femoral artery. Conventional untwisting and snaring techniques failed. A novel approach utilizing a Raptor grasping device facilitated successful c

Captive breeding has contributed to successful restoration of many species of birds of prey. Avicultural techniques pioneered by raptor breeders include double clutching, direct fostering, cross-fostering, hatch and switch, hacking, imprinting male and female falcons for semen collection, and artificial insemination techniques. However, reproductive failure occurs related to management problems, including hygiene measures, food quality issues, breeding flock structure, or individual health issues of breeding birds. These may result in non-egg laying females, low-quality eggs, or infertile eggs

There are few studies evaluating safe and effective sedatives for wild birds. Most sedatives are injectable drugs that are commonly administered intramuscularly (IM) or subcutaneously (SC); however, needle injuries occasionally occur from the bird struggling during the injection. Previous research in parrots and other bird species explored the effects of intranasal (IN) midazolam compared with IM administration; however, this delivery method has not been well studied in wild birds, such as raptors. To our knowledge, there is only 1 study that has been published evaluating midazolam IN in a rap

The mechanistic target of rapamycin (mTOR) is elevated in prostate cancer, making this protein attractive for tumor treatment. Unfortunately, resistance towards mTOR inhibitors develops and the tumor becomes reactivated. We determined whether epigenetic modulation by the histone deacetylase (HDAC) inhibitor, valproic acid (VPA), may counteract non-responsiveness to the mTOR inhibitor, temsirolimus, in prostate cancer (PCa) cells. Prostate cancer cells, sensitive (parental) and resistant to temsirolimus, were exposed to VPA, and tumor cell growth behavior compared. Temsirolimus resistance enhan

This study provides a taphonomic analysis of prey accumulations of crowned hawk-eagles (Stephanoaetus coronatus) from Ngogo, Kibale National Park, Uganda, collected over 37 months from below nests of two eagle pairs. Crowned hawk-eagles are powerful predators capable of killing animals much larger than themselves, and are significant predators of cercopithecoid monkeys in forest habitats throughout sub-Saharan Africa. At Ngogo, 81% of the individuals in the kill sample are monkeys. Redtail monkeys (Cercopithecus ascanius) are particularly well represented in the sample, making up 66% of monkey

The mechanistic target of rapamycin complex 1 (mTORC1) is a critical sensor for bone homeostasis and bone formation; however, the role of mTORC1 in osteoclast development and the underlying mechanisms have not yet been fully established. Here, we found that mTORC1 activity declined during osteoclast precursors differentiation in vitro and in vivo. We further targeted deletion of Raptor (mTORC1 key component) or Tsc1 (mTORC1 negative regulator) to constitutively inhibit or activate mTORC1 in osteoclast precursors (monocytes/macrophages), using LyzM-cre mice. Osteoclastic formation was drastical

Tendons transmit contractile forces between musculoskeletal tissues. Whereas the biomechanical properties of tendons have been studied extensively, the molecular mechanisms regulating postnatal tendon development are not well understood. Here we examine the role of mTORC1 signaling in postnatal tendon development using mouse genetic approaches. Loss of mTORC1 signaling by removal of Raptor in tendons caused severe tendon defects postnatally, including decreased tendon thickness, indicating that mTORC1 is necessary for postnatal tendon development. By contrast, activation of mTORC1 signaling in

The mammalian target of rapamycin (mTOR) is a serine-threonine kinase that has been shown to be essential for the differentiation and function of various immune cells. Earlier in vitro studies showed that mTOR signalling regulates B-cell biology by supporting their activation and proliferation. However, how mTOR signalling temporally regulates in vivo germinal centre B (GCB) cell development and differentiation into short-lived plasma cells, long-lived plasma cells and memory cells is still not well understood. In this study, we used a combined conditional/inducible knock-out system to investi

Haemosporidians, including Leucocytozoon spp., Plasmodium spp., and Haemoproteus spp., are vector-borne parasites that cause avian malaria and malaria-like diseases in birds. These infections can result in anaemia, reduced body condition, impaired reproductive success, and mortality, thereby acting as selective agents within host populations. Climate change is expected to influence dynamics of some haemosporidians, particularly in northern regions, by altering vector distributions and extending transmission seasons. In this study, tawny owls (Strix aluco; n = 27) from central Norway, were scre

Avian trichomonosis is a world-wide parasitic disease caused by the protozoan Trichomonas gallinae. Although several degrees of pathogenicity have been described on the basis of the clinical signs in birds, there are few reports concerning the genetic characterization of the parasite and its relationship with pathogenicity. The parasite usually appears apathogenic but is occasionally responsible for outbreaks of the disease in avian populations, particularly affecting nestlings of ornithophagous raptors. For 3 years, cultures of oropharingeal samples from 612 wild and domestic pigeons (Columba
