Hemoglobin messenger RNA from human bone marrow. Isolation and translation in homozygous and heterozygous beta-thalassemia.

A method for isolating human hemoglobin messenger RNA (mRNA) from bone marrow cells was developed to investigate the molecular basis for the defect in globin synthesis in beta thalassemia. Active mRNA was isolated from the bone marrow cells and peripheral reticulocytes of patients with homozygous beta thalassemia, heterozygous beta thalassemia, sickle cell trait, double heterozygosity for beta thalassemia and sickle cell trait, as well as from a patient with normal hemoglobin synthesis but with an elevated reticulocyte count secondary to hereditary spherocytosis. The mRNA was prepared for assa

Distribution of maternal red cell antibodies and the risk of severe alloimmune haemolytic disease of the foetus in a Chinese population: a cohort study on prenatal management.

BACKGROUND: Haemolytic disease of the foetus and newborn (HDFN) is the most common aetiology of haemolytic anaemia and hyperbilirubinaemia in foetuses and neonates. Studies on the distribution of antibodies that cause haemolytic disease of the foetus (HDF) in China are limited, and the effects of multiple antibodies on the severity of HDF need further evaluation. METHODS: An observational cohort study from January 2005 to December 2019 was conducted in two hospitals affiliated with Sun Yat-sen University. Maternal red cell alloimmunization was identified by the Guangzhou Blood Centre. In tota

Low bone mass density is associated with hemolysis in Brazilian patients with sickle cell disease.

OBJECTIVES: To determine whether kidney disease and hemolysis are associated with bone mass density in a population of adult Brazilian patients with sickle cell disease. INTRODUCTION: Bone involvement is a frequent clinical manifestation of sickle cell disease, and it has multiple causes; however, there are few consistent clinical associations between bone involvement and sickle cell disease. METHODS: Patients over 20 years of age with sickle cell disease who were regularly followed at the Hematology and Hemotherapy Center of Campinas, Brazil, were sorted into three groups, including those w

Developmental changes in delta-aminolevulinic acid dehydratase (ALAD) activity and blood reticulocyte percent in the developing rat. A brief note.

A correlation between blood reticulocyte percent and the activity of erythrocytic delta-aminolevulinic acid dehydratase (ALAD) in the developing rat has been established during the progression from a state of macrocytic hypochromic anemia at birth to a normocytic normochromic state at puberty, and finally to a state just prior to adulthood at which the reticulocyte percent had stabilized. Both reticulocyte percent and erythrocytic ALAD activity was found to decrease with age, rapidly at first until a normocytic normochromic state was reached at puberty and then more slowly until just before ad

[Silent forms of hereditary spherocytosis].

We report 7 cases of "silent" form of hereditary spherocytosis observed among members of 4 different families. Silent form of hereditary spherocytosis occurred in 5.4% of all patients with hereditary spherocytosis treated in our institute. The patients with silent form featured normal Hb level and red blood cell count, normal or slightly elevated reticulocyte count and bilirubin level. Osmotic resistance of red blood cells was decreased and autohemolysis was increased, nevertheless, the differences from normal range were less prominent than in patients with manifest form of the disease. [Pyruvate kinase deficiency. I. Clinical and hematological observations (author's transl)].

Clinical and hematological studies were performed on ten homozygous and seven heterozytous individuals with pyruvate kinase deficiency, aged between 2 and 71 years. Five of the homozygotes were splenectomized. With the exception of a decreased enzyme activity between 41 and 55 per cent and minor changes in their red cell metabolism the heterozygotes showed no abnormal results. In the homozygotes the following results could be demonstrated: 1. Pyruvate kinase activity was decreased to 11 to 35 per cent of normal enzyme activity. 2. There is no relation between the severity of hemolysis and the

Enhancing diagnostic accuracy for iron deficiency in pregnant women through mean reticulocyte volume.

BACKGROUND AND OBJECTIVES: Women are more prone to iron deficiency (ID) anemia when pregnant. The diagnostic use of mean reticulocyte volume (MRV) in identifying ID anemia during pregnancy has not been thoroughly investigated. The objective of this study is to evaluate the effectiveness of MRV in diagnosing ID in pregnant women. METHODS AND STUDY DESIGN: Firstly, MRV of 20 healthy female volunteers (healthy group) was measured on specific days for one month. Subsequently, clinical data from 724 pregnant women were thoroughly examined. These women were divided into two groups: 282 with ID (res

Reticulocyte analysis in iron deficiency anemia and hemolytic anemia.

Reticulocyte analysis was studied in 28 anemic patients, 15 with iron deficiency anemia (IDA), and 13 with hemolytic anemia including 9 glucose 6 phosphate dehydrogenase deficiency (G6PD def.), and 4 with G6PD def. combined with HbE trait or alpha thalassemia trait (alpha thal trait). The reticulocyte analysis among these patients showed the increased number of reticulocyte percentage with low degree of maturation in both IDA and G6PD def. patients. The significantly decreased reticulocyte hemoglobin content (CHr) was found in IDA (CHr = 21.74 +/- 4.61 pg in IDA vs 28.41 +/- 1.34 pg in normal;

Derivated fetal haemoglobin as a marker for red cell age in the human fetus reflecting stimulated or impaired red blood cell production.

We have determined whether derivated fetal haemoglobin (dHbF, consisting of glycated and acetylated HbF) can be used as a cell age marker for fetal red blood cells (RBCs). Cord blood was obtained between 19 and 39 weeks of gestation from 28 alloimmunised anaemic fetuses (23 RhD+ and 5 Kell) and from 20 non-anaemic fetuses and newborns (controls). Density gradient centrifugation was applied to 36 samples (20 RhD+, 15 controls and 1 Kell) to obtain fractions of increasing cell age. Blood samples were used for measurements of mean cellular volume (MCV), mean cell haemoglobin (MCH), mean corpuscul

Procainamide-associated pancytopenia.

A 42-year-old male developed thrombocytopenia and anemia along with agranulocytosis during treatment with procainamide. Bone marrow hypoplasia was evident on biopsy, and the patient was without evidence of systemic lupus erythematosis. He had prompt marrow recovery on drug withdrawal. While procainamide-induced agranulocytosis has been reported on a number of occasions, this is only the second case of pancytopenia.

Discrimination of Iron Deficiency, Alpha and Beta Thalassemia on the Basis of Red Cell Distribution Width and Reticulocyte Indices.

BACKGROUND: The two major causes of microcytic anemia are iron deficiency and thalassemia. In clinical practice, in some cases, differentiation of microcytic and hypochromic anemia become difficult on the basis of traditional red cell parameters. This study primarily aimed to evaluate the usefulness of red cell distribution width (RDW), immature reticulocyte fraction (IRF), and reticulocyte hemoglobin equivalent (Ret-He) in the discrimination of alpha thalassemia, beta thalassemia, and latent iron deficiency. METHODS: In this retrospective study laboratory data including complete blood counts

Examination of Reticulocytosis among Chronically Transfused Children with Sickle Cell Anemia.

Sickle cell anemia (SCA) is an inherited hemolytic anemia with compensatory reticulocytosis. Recent studies have shown that increased levels of reticulocytosis during infancy are associated with increased hospitalizations for SCA sequelae as well as cerebrovascular pathologies. In this study, absolute reticulocyte counts (ARC) measured prior to transfusion were analysed among a cohort of 29 pediatric SCA patients receiving chronic transfusion therapy (CTT) for primary and secondary stroke prevention. A cross-sectional flow cytometric analysis of the reticulocyte phenotype was also performed. M

Fascioliasis: similarities of the anemia in rats to that produced by infused proline.

Fasciola hepatica releases large amounts of proline into the bile of its host. Significant increases in the levels of other amino acids in the bile also occur. The present investigation examines whether proline and these other amino acids may play a role in inducing the anemia that frequently accompanies fascioliasis. In experiments, rats were infused intra-abdominally for 2 weeks with 1 of 3 solutions: 20.0 mM proline in saline, a mixture of 8 amino acids (excluding proline) each at 2.0 mM in saline, or saline only. At the end of the experiments 2 cc of blood was removed from each rat, via ca

Perillyl alcohol-induced hematological modulations: insights into human blood cells damage, eryptosis, and systemic perturbations.

Perillyl alcohol (POH) is a natural monoterpene with established antitumor activity. Although its anticancer efficacy is well-documented, its impact on normal blood cells, particularly red blood cells (RBCs), remains underexplored. Given the importance of RBC integrity in maintaining homeostasis, this study aims to investigate the hematological effects of POH, focusing on hemolysis, eryptosis, and systemic blood parameters. Erythrocytes collected from June-August 2023 were treated with POH (0.5-2.5 mM) for 24 hr at 37°C. Hemolysis was assessed using photometric assays. Eryptosis was detected b

Large Hemoglobin Differences at Birth in Monochorionic Twins with a Placental Chorangioma and Delayed Cord Clamping.

We report a case of a monochorionic diamniotic twin with an uncomplicated pregnancy, but with an unexpected large intertwin hemoglobin (Hb) difference at birth. Twin 1 was delivered vaginally and had an uneventful neonatal course. The umbilical cord of Twin 1 was clamped approximately 5 min after birth. After the birth of Twin 1, Twin 2 developed severe bradycardia and showed limited cardiac output on ultrasound, for which an emergency cesarean section was performed. A full blood count revealed an Hb of 20.1 g/dL for Twin 1 and 10.2 g/dL for Twin 2 (intertwin difference 9.9 g/dL). Reticulocyte

[Hematological parameters in characterization of anemia in leptospirosis].

Blood samples from 46 patients with leptospirosis were analyzed on a Cobas Vega (ABX, France) device over the course of disease. Hemoglobin content, erythrocyte count, mean volume of erythrocytes, mean content and mean concentration of hemoglobin in an erythrocyte, anisocytosis index, reticulocyte count, and content of free hemoglobin (FH) were analyzed. Anemia (normocytic normochromic hyperregeneratory) is a typical manifestation of leptospirosis during the second week of disease. The leading pathogenetic mechanism of anemia associated with leptospirosis is erythrocyte hemolysis, whose intens

Red blood cell homeostasis in children and adults with and without asymptomatic malaria infection in Burkina Faso.

Asymptomatic malaria infections may affect red blood cell (RBC) homeostasis. Reports indicate a role for chronic hemolysis and splenomegaly, however, the underlying processes are incompletely understood. New hematology analysers provide parameters for a more comprehensive analysis of RBC hemostasis. Complete blood counts were analysed in subjects from all age groups (n = 1118) living in a malaria hyperendemic area and cytokines and iron biomarkers were also measured. Subjects were divided into age groups (<2 years, 2-4, 5-14 and ≥15 years old) and clinical categories (smear-negative healthy su
