Inherited retinal dystrophies (IRDs) are a heterogeneous group of rare genetic disorders that affect multiple retinal cell types, including photoreceptors, Müller glia, bipolar cells, retinal ganglion cells (RGCs), and the retinal pigment epithelium (RPE). Recombinant adeno-associated viruses (rAAVs) have emerged as the leading vectors for gene delivery in the retina due to their safety profile and ability to drive long-term expression. However, achieving precise cell targeting and appropriate transgene regulation remains a key challenge. Recent advances in capsid engineering and the discovery

High fat diets (HFD) have been utilized in rodent models of visual disease for over 50 years to model the effects of lipids, metabolic dysfunction, and diet-induced obesity on vision and ocular health. HFD treatment can recapitulate the pathologies of some of the leading causes of blindness, such as age-related macular degeneration (AMD) and diabetic retinopathy (DR) in rodent models of visual disease. However, there are many important factors to consider when using and interpreting these models. To synthesize our current understanding of the importance of lipid signaling, metabolism, and infl

BACKGROUND: Notch signaling, especially via the Notch1 receptor, is involved in normal eye development by maintaining retinal progenitor cell state before their differentiation, although the regulatory mechanism of fine-tuned Notch1 expression is unclear.
METHODS: We re-analyzed publicly available human fetal retina single-cell RNA/ATAC-seq data at days 59, 74, and 78, using Seurat/Signac pipelines.
RESULTS: We observed NOTCH1 expression in retinal progenitor cells, which diminished at later stages of differentiation. Integration of single-cell RNA-seq and ATAC-seq revealed a decrease in chr

Homeostatic plasticity stabilizes input and activity levels during neural development, but whether it can restore connectivity and preserve circuit function during neurodegeneration is unknown. Photoreceptor degeneration is the most common cause of blindness in the industrialized world. Visual deficits are dominated by cone loss, which progresses slowly, leaving a window during which rewiring of second-order neurons (i.e., bipolar cells) could preserve function. Here we establish a transgenic model to induce cone degeneration with precise control and analyze bipolar cell responses and their ef

Retinal on-pathway dysfunction is implicated in human complete-type congenital stationary night blindness (CSNB1), a Mendelian genetic condition that results from mutations in the NYX gene encoding the protein nyctalopin. We probed cone pathway dysfunction in four human genotyped CSNB1 affected males by electroretinogram (ERG) recordings elicited with photopic sinusoidal and rapid-on/off-ramp flicker stimuli that are reputed to elicit on/off-pathway activity selectively. Results were analyzed in relation to ERG abnormalities created in anesthetized non-human primates by intravitreal applicatio

Retina and optic nerve of eyes enucleated due to an iris ring melanoma or a malignant melanoma of the ciliary body were studied to investigate the correlation between corpora amylacea count and glaucoma stages. Ten eyes from ten patients [age: 62.3 +/- 13.0 (mean +/- SD); range 43-84 years] were used for the present study. Eight eyes had shown elevated intraocular pressure readings preoperatively. The corpora amylacea count was correlated with histomorphometric changes of the intra- and parapapillary region. The corpora amylacea count of the retinal ganglion cells decreased significantly with

Age-related neuronal adaptations are known to help maintain function. This study aims to examine gross age-related in vivo retinal functional adaptations (using electroretinography) in young and middle aged C57BL/6J and Thy1-YFPh mice and to relate this to in vivo retinal structure (using optical coherence tomography). Electroretinography responses were generally larger in Thy1-YFPh mice than in C57BL/6J mice, with similar in vivo retinal layer thicknesses except for longer inner/outer photoreceptor segment in Thy1-YFPh mice. Relative to 3-month-old mice, 12-month-old mice showed reduced photo

Diabetic retinopathy (DR) is the most common complication of diabetes and remains one of the major causes of blindness in the world; infants born to diabetic mothers have higher risk of developing retinopathy of prematurity (ROP). While hyperglycemia is a major risk factor, the molecular and cellular mechanisms underlying DR and diabetic ROP are poorly understood. To explore the consequences of retinal cells under high glucose, we cultured wild type or E2f1-/- mouse retinal explants from postnatal day 8 with normal glucose, high osmotic or high glucose media. Explants were also incubated with

INTRODUCTION: Cognitive impairment (CI) in bipolar disorder (BD) significantly impacts overall functioning and quality of life. A better understanding of the neurobiological mechanisms associated with CI is needed. Studies on neurodegenerative diseases, such as Alzheimer's and Parkinson's diseases, have revealed promising findings related to retinal thickness alterations using optical coherence tomography (OCT). Similarly, retinal differences between healthy controls and individuals with BD or schizophrenia have been described. This study explores the utility of OCT in discerning retinal chang

Stem cell-derived retinal organoid (RO) transplantation is a promising approach for treating retinal degenerative diseases such as retinitis pigmentosa. Photoreceptors (PRs) in RO sheets form synaptic connections with host bipolar cells (BCs) and improve visual function in animal models of advanced retinal degeneration. However, the contribution of horizontal cells (HCs), which are critical for PR synapse formation in the developing retina, to host-graft synapse formation remains unclear. In this study, we used HC-depleted retinal degeneration mice (rd1) and Islet1-/- genome-edited ROs (gROs)

Retinal prostheses aim at restoring sight to patients blinded by atrophy of photoreceptors using electrical stimulation of the inner retinal neurons. Bipolar cells can be targeted using subretinal implants, and their responses are then relayed to the central visual pathways via the retinal neural network, preserving many features of natural signal processing. Epiretinal implants stimulate the output retinal layer-ganglion cells-and encode visual information directly in spiking patterns.Several companies and academic groups have demonstrated that electrical stimulation of the degenerate retina

In the pupillary light response (PLR), increases in ambient light constrict the pupil to dampen increases in retinal illuminance. Here, we report that the pupillary reflex arc implements a second input-output transformation; it senses temporal contrast to enhance spatial contrast in the retinal image and increase visual acuity. The pupillary contrast response (PCoR) is driven by rod photoreceptors via type 6 bipolar cells and M1 ganglion cells. Temporal contrast is transformed into sustained pupil constriction by the M1's conversion of excitatory input into spike output. Computational modeling

Information that defines the depth of activation of retinal neurons is useful in considering strategies for stimulation with a retinal prosthesis, or interpreting the results from human studies that have previously been performed. The purpose of this study was to test the assertion that electrical pulse durations >0.5 msec preferentially stimulate retinal neurons deep to the ganglion cell layer. Thirteen Dutch-belted rabbits (1.2-2.0 kg) were used in this study. A Goldmann-like dome was used to deliver photic stimuli to the retina to measure the electroretinogram (ERG) and the light-induced co

Glutamate, a key neurotransmitter in the vertebrate retina, acts via ionotropic and metabotropic receptors. Retina expresses mRNA for all metabotropic glutamate receptors and proteins for all but mGluR3. Every retinal cell class expresses one or more of these receptors. In general, these receptors are present presynaptically and serve to modulate synaptic transmission. While mGluRs on the photoreceptor terminal act as autoreceptors to titer glutamate levels, those on horizontal cell processes seem to shape the light response. Similarly, autoreceptors on bipolar axon terminals modulate glutamat

Optogenetic gene therapy has emerged as a mutation-agnostic therapeutic modality for vision restoration in advanced retinal degenerative diseases, where conventional gene replacement strategies are no longer applicable. By introducing light-sensitive opsins into surviving retinal neurons using viral vectors, this approach restores photosensitivity after photoreceptor loss. Advances in opsin engineering, vector design, and cell-specific targeting have accelerated the translation of optogenetics from proof-of-concept studies to an expanding clinical pipeline. This review summarizes recent progre

Pathologic myopia is a major cause of irreversible visual impairment worldwide and is characterized by excessive axial elongation accompanied by progressive retinal degeneration. Whether vision loss results primarily from passive retinal stretching or selective neurodegeneration remains unclear, hindering the development of effective neuroprotective and regenerative therapies. Here, we investigated retinal neuronal, vascular, and glial alterations in retinal pigment epithelium (RPE)-specific Lrp2 knockout (Best1-Cre/Lrp2fl/fl conditional knockout, CKO) model of pathologic myopia. The CKO mice

PURPOSE: Mutations in GPR179, which encodes the G protein-coupled receptor 179, lead to autosomal recessive complete (c) congenital stationary night blindness (CSNB), which is characterized by an ON-bipolar retinal cell dysfunction. This study further defined the exact site of Gpr179 expression and its protein localization in human retina and elucidated the pathogenic mechanism of the reported missense and splice site mutations.
METHODS: RNA in situ hybridization was performed with mouse retinal sections. A commercially available antibody was validated with GPR179-overexpressing COS-1 cells a

Protein MAP1B was recently reported to link GABA(C) receptors to the cytoskeleton at neuronal synapses. This interaction was demonstrated in the mammalian retina, where GABA(C) receptors were thought to be exclusively expressed in bipolar cells. Our previous studies on cultured photoreceptors suggested however the presence of GABA(C) receptors in cones. To further investigate GABA(C) receptor expression in cones, we measured GABA responses in mammalian photoreceptors in situ, and we examined the distribution of the receptor and that of protein MAP1B in the mammalian outer retina. Photoreceptor

Accumulating evidence indicates that retinal ischemia constitutes a pivotal pathological trigger of visual impairment. The cerebral arterial circle (circle of Willis), formed by the vertebral and internal carotid arteries, provides cerebral blood perfusion. As a direct branch originating from the internal carotid artery (ICA), the ophthalmic artery supplies blood to the ocular globe. Based on the anatomical features of the ocular vasculature, a retinal ischemic injury model was successfully generated in C57BL/6 mice via selective left ICA ligation. Morphological and vascular analyses demonstra

The retina is a vital component of the visual system, converting light into neural signals processed by the brain. Its structure includes photoreceptors, bipolar cells, and ganglion cells, each contributing to vision. Genetics play a critical role in retinal development and health, with mutations in over 270 genes linked to inherited retinal diseases, such as retinitis pigmentosa (RP), age-related macular degeneration (AMD), and Leber congenital amaurosis (LCA). These conditions range from mild visual impairment to complete blindness, reflecting the genetic diversity underlying retinal disease
