On October 13, 2021, a tick infestation occurred in a home in rural area of Nanchang city, China, and we were asked to inspect the tick infestation. Ticks were collected in the largest number on courtyard door jambs, followed by living room and bedroom door jambs. Ticks were identified morphologically as Rhipicephalus sanguineus adults. The 16S rRNA analysis effectively distinguished the ticks in this study from other Rhipicephalus species, including R. sanguineus south-east, temperate and tropical lineages and identified genetically as R. sanguineus south China lineage. Tick samples were subj

Rhipicephalus microplus ticks are vectors for multiple pathogens infecting animals and humans. Although the medical importance of R. microplus has been well-recognized and studied in most areas of China, the occurrence of tick-borne Rickettsiales has seldom been investigated in Guizhou Province, Southwest China. In this study, we collected 276 R. microplus ticks from cattle (209 ticks) and goats (67 ticks) in three locations of Guizhou Province. The Rickettsia, Anaplasma, and Ehrlichia were detected by targeting the 16S rRNA gene and were further characterized by amplifying the key genes. One

The cattle tick Rhipicephalus microplus is one of the most important ectoparasites in the tropical and subtropical regions of the world. Synthetic pyrethroids are widely used to control this tick, and the selection of resistant populations is a huge problem worldwide. The activity of thymol, a natural monoterpene, free or in combination with other compounds, has been demonstrated against different species of ticks. However, the mode of action is not fully understood. The present study aimed to evaluate the efficacy and the potential mode of action of the combination of cypermethrin and thymol

The prevalence, resistance ratios and factor associated with Rhipicephalu microplus populations resistant to amitraz, flumethrin, coumaphos, and ivermectina (IVM) in Mexico were studied. Field tick populations were collected from 54 farms in 15 different states of Mexico. The dose-response bioassays were carried out using the larval immersion test (amitraz and IVM) and the modified larval packet test (flumethrin and coumaphos) against R. microplus. Mortality data were subjected to probit analysis to calculate lethal concentrations at 50%. A logistic regression model was used to evaluate the re

Serpins are evolutionarily conserved serine protease inhibitors found in many organisms. In arthropods, serpins are involved in feeding, development, oviposition, anti-coagulation and innate immune responses. We characterized of 11 serpins in the tick Rhipicephalus haemaphysaloides. These serpins have orthologous genes in other ticks, as indicated by phylogenetic analysis. Analysis of the reactive center loop and hinge regions of the protein sequences indicated that RHS7 encodes proteins that may lack proteinase inhibitor activity. All R. haemaphysaloides serpins had high amino acid sequence i

Attempts to eliminate Rhipicephalus microplus from Uruguay have been unsuccessful, and, currently, the country is divided into two areas: a tick-free area and a tick-infested area. In the tick-infested area, different farms face different situations. Some farms are in regions where, due to environmental conditions or a lack of infrastructure, it is difficult to eliminate R. microplus, and the only option is to control it. In contrast, other farms can attempt complete removal. Before deciding whether a farmer should attempt to eliminate R. microplus, the probability of reintroduction must be ev

Rhipicephalus microplus causes high economic losses for Uruguay; the country is divided into a R. microplus-free area and an endemic area. The aim of this research was to evaluate the R. microplus and tick fever situation in Uruguay and the feasibility of implementing R. microplus control or elimination programs to reduce the number of infested farms. A representative survey was performed to determine the number of infested farms, the number of farms with outbreaks of tick fever and the control and preventive measures being used. Control or elimination programs using generational treatment wer

Rhipicephalus microplus is an ectoparasite responsible for transmissions of babesiosis and anaplasmosis causing large losses to livestock production. To survive R. microplus tick produces several active molecules, such as protease inhibitors. This ectoparasite has been described as a rich source of serine protease inhibitors most of them are Kunitz-BPTI members named BmTIs which have no clear function yet. In the present work, we described the expression and functional characterization of rBmTI-A which showed to be similar to the native BmTI-A, a double-headed Kunitz-BPTI inhibitor, capable to

The capacity of a topical combination of imidacloprid and permethrin (Advantix(®)) to prevent transmission of Ehrlichia canis was studied in two groups of six dogs. One group served as controls, whereas the other group was treated. All dogs were exposed to E. canis-infected Rhipicephalus sanguineus ticks on Days 7, 14, 21 and Day 28 post acaricidal treatment. The adult R. sanguineus ticks were released into the individual kennels of the dogs to simulate natural tick exposure. In situ tick counts were conducted on Day 9, 16 and 23 and any remaining ticks were counted and removed on Day 30. The

Babesia bigemina infections were investigated in four genetic groups of beef cattle and in Rhipicephalus (Boophilus) microplus engorged female ticks. Blood samples and engorged female ticks were collected from 15 cows and 15 calves from each of the following genetic groups: Nelore, Angus x Nelore, Canchim x Nelore, and Simmental x Nelore. Microscopic examination of blood smears and tick hemolymph revealed that merozoites of B. bigemina (6/60) as well as kinetes of Babesia spp. (9/549) were only detected in samples (blood and ticks, respectively) originated from calves. PCR-based methods using

The objective of this study was to evaluate essential oils (EOs) as an alternative control method for Rhipicephalus microplus and Amblyomma mixtum larvae. The EOs used were obtained by steam distillation from the leaves of cinnamon (Cinnamomum verum), mexican mint (Plectranthus amboinicus), lemongrass (Cymbopogon citratus), peppercorns (Pimenta dioica), and peruvian pepper tree (Schinus molle). To evaluate the acaricidal effect of EOs against the larvae of two tick species, a larval immersion test (LIT) was performed using six concentrations (10 mg/mL, 5 mg/mL, 2.5 mg/mL, 1.87 mg/mL, 1.25 mg/m

BACKGROUND: The cattle tick Rhipicephalus microplus is a major constraint to livestock production in tropical regions, causing substantial economic losses and driving intensive acaricide use. Despite its importance, the role of pasture composition in shaping off-host tick populations remains poorly understood. This study evaluated whether forage grass species create distinct microenvironments that influence the seasonal abundance and persistence of R. microplus larvae in a tropical region.
RESULTS: Larval abundance was higher overall in the dry season than in the rainy season, with a signific

The resistance of Rhipicephalus microplus to the main commercial acaricides has stimulated research for new antiparasitic agents, including natural products as an alternative to eco-friendly control. Among the main classes of secondary metabolites of plants stand out the alkaloids, which are characterized by a variety of biological activities such as enzymatic inhibition. The enzyme Glutathione-S-transferase (GST) is an important catalyst of xenobiotic detox reactions in arthropods and represents a promising target in the acaricide development. This work aimed to associate in silico and in vit

The cattle tick Rhipicephalus microplus affects animal production economically by reducing weight gain and milk production and causing diseases, such as babesiosis and anaplasmosis. Using synthetic acaricides to reduce their incidence has caused the emergence of resistant tick populations. The present study aimed to assess the in vitro acaricidal activity of combinations of essential oils (EOs) from Ocimum americanum, Ocimum gratissimum, and Lippia multiflora against R. microplus larvae. In fact, numerous biological properties have been reported on EOs from these three plants, including acaric

Climate change ranks among the most important issues globally, affecting geographic distributions of vectors and pathogens, and inducing losses in livestock production among many other damaging effects. We characterized the potential geographic distribution of the ticks Rhipicephalus (Boophilus) microplus, an important vector of babesiosis and anaplasmosis globally. We evaluated potential geographic shifts in suitability patterns for this species in two periods (2050 and 2070) and under two emissions scenarios (RCPs 4.5 and 8.5). Our results anticipate increases in suitability worldwide, parti

BACKGROUND: The tick vector Rhipicephalus microplus which transmits Babesia spp. and rickettsial pathogens has not been reported in Kenya since 1998. More recently, the pathogenic Babesia bovis has been detected in cattle blood DNA. The status of R. microplus in Kenya remains unknown. This study employed morphological and molecular tools to characterize R. microplus originating from Kenya and assess the genetic relationships between Kenyan and other African R. microplus genotypes.
METHODS: Ticks were collected in south-eastern Kenya (Kwale County) from cattle and characterized to investigate

Bartonella henselae is a fastidious intraerythrocytic, gram-negative bacteria that causes cat scratch disease in humans. Ixodes ricinus has been confirmed to be a competent vector of B. henselae, and some indirect evidences from clinical cases and epidemiological studies also suggested that some other tick species, including Rhipicephalus sanguineus, may transmit the bacteria. B. henselae has been detected in R. sanguineus but no experimental investigations have been performed to evaluate the vector competency of this tick species regarding B. henselae transmission. To this end, this work aime

A new genotype phylogenetically close to Ehrlichia canis named as Ehrlichia minasensis was identified infecting cattle and deer in Canada, as well as Rhipicephalus microplus ticks and cattle in Brazil. Although it was detected in R. microplus, little is known about the epidemiology of this ehrlichiosis, especially in other tick species. This study evaluated the minimum infection rate of E. minasensis in Amblyomma sculptum and R. microplus ticks from locations where naturally infected cattle were previously detected. Overall, 45 engorged R. microplus ticks after molting [43 pools of adults (13.

Permethrin is a commonly used acaricide for tick control on domestic animals and in residential environments, while fipronil use is restricted to on-animal treatment. Following widespread reports of permethrin and fipronil application failures to control indoor infestations of Rhipicephalus sanguineus (Latreille), the brown dog tick, 31 tick populations were obtained from Florida and Texas for acaricide resistance screening. These field-collected ticks from kennels and residential facilities were challenged with technical grade permethrin and fipronil to create dose response curves that were c

In most parts of the world, ticks are rapidly developing resistance to commonly used acaricides thus rendering control difficult. This constraint is further compounded by the introduction of new species in areas where they did not exist before. Such is the case with the introduction into and rapid spread of Rhipicephalus (Boophilus) microplus in some countries of West Africa. With the looming threat of its further spread in the region, the objective of the present study was to update knowledge on cattle ticks in Cameroon. Among 19,189 ticks collected monthly from 60 animals in 5 herds from Mar
