Valorization of turkey cruor through peptic hydrolysis: peptidomic and machine learning approaches for the identification of antimicrobial peptides.

The slaughterhouse blood remains an underexploited by-product with promising potential for producing natural antimicrobial agents. This study investigated the hydrolysis of turkey cruor with pepsin under varying durations, as well as the impact of discoloration on the peptide population and antimicrobial activities. The discoloration at pH 4 effectively removed heme and enhanced antifungal and anti-yeast activities, particularly against Rhodotorula mucilaginosa, Paecilomyces spp., and Mucor racemosus. Comparative peptidomics combined with partial least squares-discriminant analysis enabled the

Structural basis of stereospecific reduction by quinuclidinone reductase.

Chiral molecule (R)-3-quinuclidinol, a valuable compound for the production of various pharmaceuticals, is efficiently synthesized from 3-quinuclidinone by using NADPH-dependent 3-quinuclidinone reductase (RrQR) from Rhodotorula rubra. Here, we report the crystal structure of RrQR and the structure-based mutational analysis. The enzyme forms a tetramer, in which the core of each protomer exhibits the α/β Rossmann fold and contains one molecule of NADPH, whereas the characteristic substructures of a small lobe and a variable loop are localized around the substrate-binding site. Modeling and mut

Antifungal non-invasive soak under general anaesthetic to treat fungal rhinitis in an Australian Quarter Horse gelding at pasture.

BACKGROUND: Mycotic rhinitis is a rare disease in horses, with few cases reported worldwide and none reported in Australia. Fungal infection of the upper respiratory tract can occur in all species, but its prevalence in horses is considerably lower than in canines or humans. The disease is linked to a variety of pathogens and the clinical signs are associated with subsequent upper respiratory tract damage. CASE REPORT: A 6-year-old Quarter Horse gelding in Queensland, Australia, was presented with chronic nasal discharge and dyspnoea following previous unsuccessful medical treatment of 9 mont

Synthesis of functionalized 3-, 5-, 6- and 8-aminoquinolines via intermediate (3-pyrrolin-1-yl)- and (2-oxopyrrolidin-1-yl)quinolines and evaluation of their antiplasmodial and antifungal activity.

(3-Pyrrolin-1-yl)- and (2-oxopyrrolidin-1-yl)quinolines were prepared via cyclization of diallylaminoquinolines and 4-chloro-N-quinolinylbutanamides, respectively, as novel synthetic intermediates en route to N-functionalized 3-, 5-, 6- and 8-aminoquinolines with potential biological activity. (3-Pyrrolin-1-yl)quinolines were subjected to bromination reactions, and the reactivity of (2-oxopyrrolidin-1-yl)quinolines toward lithium aluminum hydride and methyllithium was assessed, providing an entry into a broad range of novel functionalized (pyrrolidin-1-yl)- and (hydroxyalkylamino)quinolines. Development of a serum-free system for the in vitro cultivation of Brugia malayi infective-stage larvae.

Over the past several years, numerous attempts have been made to culture the infective-stage (L3) larvae of the human filarial parasite Brugia malayi in an in vitro system that promotes molting to the fourth larval stage (L4). Although there have been reports in the literature of successful L3 to L4 development in vitro, all of these systems have required serum supplementation. The complexity of serum as a culture supplement has made reproducibility of results and identification of specific factors necessary for L3 development problematic. We have developed a serum-free in vitro system consist

Exploring novel antifungal peptides from peptic hydrolysis of chicken cruor protein via regression-based machine learning approach.

There is a growing interest in natural preservatives driven by consumer demand for clean-label products. In Canada, approximately 48 million liters of blood are produced annually during chicken slaughter, offering an opportunity to valorize cruor, the solid blood component rich in hemoglobin, for use in food preservation. This study investigated the hydrolysis of chicken cruor with pepsin at pH 2, 3, 4, and 5 for 180 min to produce antimicrobial peptides. The highest degree of hydrolysis (11.70 ± 0.77 %) was observed at pH 2, similar to pH 3 where the enzyme exhibited a zipper mechanism. Hydro

Fungemia in children infected with the human immunodeficiency virus: new epidemiologic patterns, emerging pathogens, and improved outcome with antifungal therapy.

We characterized 27 episodes of fungemia in 22 children infected with the human immunodeficiency virus (HIV). Fungemia in these patients presented as a community-acquired infection in the setting of outpatient total parenteral nutrition or intravenous antibiotic therapy through a chronically indwelling central venous catheter (CVC). Fungemia developed only in patients with CVCs (P < .001). Non-albicans Candida species, Torulopsis glabrata, Rhodotorula rubra, and Bipolaris spicifera constituted 52% of all causes. Fungemia was detected early, within a median of 2.4 days after the onset of new fe

Habitat-structured fungal mycobiomes at the water-gill interface of farmed red tilapia in Central Thailand: An internal transcribed spacer rRNA amplicon sequencing study.

BACKGROUND AND AIM: Tilapia aquaculture is rapidly expanding across Southeast Asia and plays a critical role in regional food security. While bacterial microbiomes of farmed fish have been widely investigated, the fungal component of aquatic microbial communities remains poorly characterized, particularly at the biologically important interface between rearing water and fish gills. Fungi may influence fish health, environmental microbial ecology, and occupational exposure risks within aquaculture systems. This study aimed to characterize fungal mycobiomes associated with rearing water and gill

Cryptococcus neoformans isolation from swallow (Hirundo rustica) excreta in Iran.

Cryptococcus neoformans is an encapsulated yeast that can cause cryptococcosis, a life-threatening infection that mainly occurs in immunocompromised patients. The major environmental sources of C. neoformans have been shown to be soil contaminated with avian droppings. In the present study, we evaluated the isolation of C. neoformans from swallow (Hirundo rustica) excreta in two northern cities of Iran. Ninety-seven swallow droppings were evaluated and 498 yeast-like colonies were isolated and identified as Rhodotorula spp. (62.8%), Candida spp. (28.5%)and C. neoformans (8.7%). Cryptococcus ne

Changes of Composition and Nutrition Value of Some Gluten Free (Pseudo)cereals by Addition of Carotenogenic Yeast Cell Homogenate.

Gluten-free cereals and pseudocereals such as oats, quinoa, and buckwheat are widely used as bases for functional foods due to their protein quality, minerals, fiber, and polyphenols. However, they contain relatively low levels of some vitamins. This study evaluated the enrichment of cereal porridges with a cell-free homogenate of the carotenogenic yeast Rhodotorula toruloides (RT) as a natural strategy to enhance their nutritional value. Model porridges prepared from gluten-free oats, quinoa, buckwheat, and their blends were supplemented with 0%, 5%, or 10% R. toruloides homogenate (RTh). Sam
