Characterization and Comparison of Microbiota in the Gastrointestinal Tracts of the Goat (Capra hircus) During Preweaning Development.

Bacterial communities in gastrointestinal tracts (GIT) play an important role in animal health and performance. Despite its importance, little information is available on the establishment of microbial populations in the goat GIT or on changes occurring during early development. Therefore, this study investigated the bacterial community dynamics of the rumen, duodenum, jejunum, ileum, cecum, and colon in 15 goats at five developmental stages (0, 14, 28, 42, and 56 days old) by using 16S rDNA sequencing and quantitative real-time PCR technology. 940 genera were found to belong to 44 phyla distr

Mucin-degrading gut bacteria: context-dependent roles in intestinal homeostasis and disease.

Akkermansia muciniphila, Bacteroides thetaiotaomicron, Mediterraneibacter (formerly Ruminococcus) gnavus, and other mucin-degrading (MD) bacteria play pivotal roles in shaping gut microbial ecosystems, maintaining gut barrier function, and mediating host-microbiota crosstalk. These bacteria influence intestinal homeostasis by modulating epithelial cell differentiation, immune responses, and gut microbiota composition through mucin degradation and the production of bioactive metabolites. Their abundance and functional activities fluctuate dynamically in response to dietary components, host immu

Microbiota of Chickens and Their Environment in Commercial Production.

Chickens in commercial production are subjected to constant interaction with their environment, including the exchange of microbiota. In this review, we therefore focused on microbiota composition in different niches along the whole line of chicken production. We included a comparison of microbiota of intact eggshells, eggshell waste from hatcheries, bedding, drinking water, feed, litter, poultry house air and chicken skin, trachea, crop, small intestine, and cecum. Such a comparison showed the most frequent interactions and allowed for the identification of microbiota members that are the mos

Associations of ruminal microbiota with susceptibility to subacute ruminal acidosis in dairy goats.

Long-term feeding of high-concentrate diets (HCD) to ruminants often leads to subacute ruminal acidosis (SARA). The fact that some ruminants adapt to HCD without developing SARA suggests that the ruminal microbiota communities may play a role in the susceptibility to this disorder. To address this hypothesis, 20 dairy goats were fed an HCD consisting of 30 % forage and 70 % concentrate for 10 weeks. The dairy goats were divided into a SARA susceptible group (SS, pH < 5.8 for more than 3 h within 12 h) and a SARA tolerant group (ST, pH < 5.8 for not more than 3 h within 12 h) according to rumin

Gut microbial dysbiosis correlates with stroke severity markers in aged rats.

BACKGROUND: An imbalanced gut microbial community, or dysbiosis, has been shown to occur following stroke. It is possible that this dysbiosis negatively impacts stroke recovery and rehabilitation. Species level resolution measurements of the gut microbiome following stroke are needed to develop and test precision interventions such as probiotic or fecal microbiota transplant therapies that target the gut microbiome. Previous studies have used 16S rRNA amplicon sequencing in young male mice to obtain broad profiling of the gut microbiome at the genus level following stroke, but further investig

Metabolic and inflammatory perturbation of diabetes associated gut dysbiosis in people living with and without HIV infection.

BACKGROUND: Gut dysbiosis has been linked with both HIV infection and diabetes, but its interplay with metabolic and inflammatory responses in diabetes, particularly in the context of HIV infection, remains unclear. METHODS: We first conducted a cross-sectional association analysis to characterize the gut microbial, circulating metabolite, and immune/inflammatory protein features associated with diabetes in up to 493 women (~ 146 with prevalent diabetes with 69.9% HIV +) of the Women's Interagency HIV Study. Prospective analyses were then conducted to determine associations of identified meta

Analysis of gut microbiota characteristics in osteoarthritis patients.

OBJECTIVE: Osteoarthritis (OA) is a chronic and debilitating disease that represents a leading cause of disability among middle-aged and elderly individuals. With the aging population, the incidence of OA and its associated socioeconomic burden is increasing; however, the underlying pathogenesis remains unclear. Based on the "gut-bone axis" theory, this study systematically characterized the gut microbiota of patients with OA to identify potential biological targets for clinical diagnosis and treatment. METHODS: Fecal samples from 32 OA patients and 26 healthy controls were analyzed for gut b

Altered Gut Microbiota Mediates the Association between APOE Genotype and Amyloid-β Accumulation in Middle-Aged Adults.

IMPORTANCE: The apolipoprotein E ( APOE ) ε4 allele is the strongest genetic risk factor for Alzheimer's disease (AD), yet the mechanisms linking APOE to amyloid-β (Aβ) pathology remain incompletely understood. Emerging evidence suggests that the gut microbiome may modulate neurodegeneration; however, its role as a mediator in the APOE-Aβ relationship remains unclear. OBJECTIVE: To evaluate whether specific microbial taxa mediate APOE-related effects on brain Aβ burden in an established population-based study of middle-aged adults. DESIGN SETTING AND PARTICIPANTS: This cross-sectional study

Dysfunction of cecal microbiota and CutC activity in mice mediating diarrhea with kidney-yang deficiency syndrome.

OBJECTIVE: Previous studies have indicated that diarrhea with kidney-yang deficiency syndrome leads to a disorder of small intestine contents and mucosal microbiota. However, the relationship of TMA-lyase (CutC) activity and TMAO with diarrhea with kidney-yang deficiency syndrome remains unexplored. Therefore, this study explores the relationship between cecal microbiota and choline TMA-lyase (CutC) activity, as well as the correlation between trimethylamine oxide (TMAO), inflammatory index, and CutC activity. METHOD: Twenty SPF-grade male KM mice were randomly divided into the normal group (

Restoration of the gut microbiota is associated with a decreased risk of hepatic encephalopathy after TIPS.

BACKGROUND & AIMS: Hepatic encephalopathy (HE) is a major complication after transjugular intrahepatic portosystemic shunt (TIPS) and is primarily influenced by the gut microbiota. We aimed to evaluate alterations in the microbiota after TIPS and the association between such alterations and HE. METHODS: We conducted a prospective longitudinal study of 106 patients with cirrhosis receiving TIPS. Faecal samples were collected before and after TIPS, and the gut microbiota was analysed by 16S ribosomal RNA sequencing. RESULTS: Among all patients, 33 developed HE (HE+ group) within 6 months after

Stability of equol production capability is associated with the diversity of the gut microbiota of the host: a prospective cohort study.

BACKGROUND: Equol, a gut microbiota–derived metabolite of the soy isoflavone daidzein that acts mainly via estrogen receptor β, has been linked to several health benefits, including relief of menopausal symptoms and prevention of arteriosclerosis. However, little is known about how equol production capability changes over time and how these changes relate to the gut microbiota. METHODS: We analyzed data from 498 community-dwelling adults who participated annually in a health checkup between 2016 and 2019. Equol production was assessed each year using urinary equol and daidzein concentrations.

Exposure to household furry pets influences the gut microbiota of infant at 3-4 months following various birth scenarios.

BACKGROUND: Early-life exposure to household pets has the capacity to reduce risk for overweight and allergic disease, especially following caesarean delivery. Since there is some evidence that pets also alter the gut microbial composition of infants, changes to the gut microbiome are putative pathways by which pet exposure can reduce these risks to health. To investigate the impact of pre- and postnatal pet exposure on infant gut microbiota following various birth scenarios, this study employed a large subsample of 746 infants from the Canadian Healthy Infant Longitudinal Development Study (C

Metagenomics and culturomics reveal the dual role of the gut microbiome in the development of immune-related toxicities and the efficacy of immune checkpoint inhibitors in cancer.

BACKGROUND: Despite their major impact on cancer treatment, immune checkpoint inhibitors (ICI) are frequently associated with immune-related adverse events (irAE). Growing evidence suggests that the occurrence of irAE may be correlated with enhanced ICI efficacy, although the underlying mechanisms remain unknown. Most studies investigating the role of the gut microbiome in oncology have relied on sequencing approaches, particularly shotgun metagenomics. Although microbiome profiling revealed strong associations between specific bacterial taxa and clinical outcomes, it has limitations, includin

Enzymatic Production of Prebiotic Xylooligosaccharides Using a Bacillus pumilus GH30_8 Glucuronoxylanase: Structural Basis of Glucuronoxylan Recognition and Hydrolysis.

Transformation of agro-industrial products into value-added products, such as prebiotic oligosaccharides, is a key element of the emerging bioeconomy. Here, we characterized a new GH30_8 glucuronoxylanase from Bacillus pumilus (BpXyn30_8A) for its potential in producing xylooligosaccharides (XOS). BpXyn30_8A showed tolerance to ethanol and NaCl and released both linear and branched XOS containing MeGlcA at the penultimate nonreducing end residue. Its X-ray structure, determined at 2.16 Å resolution, revealed high similarity to other glucuronoxylanases. Furthermore, BpXyn30_8A achieved higher x

L-Theanine affects intestinal mucosal immunity by regulating short-chain fatty acid metabolism under dietary fiber feeding.

To investigate the effects of l-Theanine (LTA) on intestinal mucosal immunity and the regulation of short-chain fatty acid (SCFA) metabolism under dietary fiber feeding, a 28-day feeding experiment was performed in Sprague-Dawley rats. The results show that LTA increased the proportion of Prevotella, Lachnospira, and Ruminococcus while increasing the total SCFA, acetic acid, propionic acid, and butyric acid contents in the feces. LTA also increased IgA, IgE, and IgG levels in the ileum, and increased villi height and crypt depth. Moreover, LTA upregulated the mRNA and protein expression of ace

Structural Characterization and In Vitro Fermentation Properties of Polysaccharides from Polygonatum filipes.

In this study, a homogeneous polysaccharide, designated as PFP-80, was isolated from the dried root of Polygonatum filipes using enzymatic extraction combined with graded ethanol precipitation. Structural characterization suggested that PFP-80 was a fructan polysaccharide with a molecular weight of 4.06 kDa. The analysis with gas chromatograph-mass spectrometer (GC-MS) and nuclear magnetic resonance (NMR) further confirmed that PFP-80 consisted of →1)-β-D-Fruf-(2→ and →1,6)-β-D-Fruf-(2→ linkages, with branching occurring at the O-6 position. After 48 h of fermentation, the pH was decreased whi

Protective effects of carboxymethyl chitosan-dialdehyde glucan/polydopamine carrier-delivered Bacillus subtilis on the intestinal tract of mice infected with Salmonella.

This study investigated the protective effects of carboxymethyl chitosan-dialdehyde glucan/polydopamine carrier-delivered Bacillus subtilis (PBS) on Salmonella-induced intestinal inflammation in mice. The results indicated that PBS significantly lowered the organ index and inflammatory factor levels while increasing the levels of oxidase in mice. Interestingly, after PBS intervention, the mRNA expression levels of mucin, tight junction proteins, immune proteins, and transcription factors that constitute the intestinal barrier in mice exhibited a significant increase compared to those in the mo

Bioactivity and influence on colonic microbiota of polyphenols from noni (Morinda citrifolia L.) fruit under simulated gastrointestinal digestion.

Noni (Morinda citrifolia L.) is a tropical fruit rich in bioactive compounds. Little is known about its polyphenol composition at different ripeness levels and digestive characteristics. Here, we studied changes in polyphenols and antioxidant activity as noni ripened. Rutin and kaempferol-3-O-rutinoside were found in high amounts in noni, with antioxidant capacity increasing as it ripened. Under simulated digestion, polyphenols were gradually released from the oral to gastrointestinal phases, partially decomposing in the small intestine due to their instability. Conversely, fiber-bound phenols
