Mesenchymal stromal cell-derived secretome-based therapy for neurodegenerative diseases: overview of clinical trials.

BACKGROUND: Over the past few years, mesenchymal stromal cells (MSCs) have attracted a great deal of scientific attention owing to their promising results in the treatment of incurable diseases. However, there are several concerns about their possible side effects after direct cell transplantation, including host immune response, time-consuming cell culture procedures, and the dependence of cell quality on the donor, which limit the application of MSCs in clinical trials. On the other hand, it is well accepted that the beneficial effects of MSCs are mediated by secretome rather than cell repla

Conditioned medium and secretome from epididymal epithelial cell cultures improve sperm kinetics and capacitation.

BACKGROUND AND AIM: Sperm maturation occurs in the epididymis through interactions with existing molecules inside the lumen. However, the mechanism of epididymis molecular transfer is currently unclear. This study was aimed to determine the necessity of the epididymal epithelial cells (EECs) in the process of sperm maturation in terms of sperm kinetics and tyrosine phosphorylation. MATERIALS AND METHODS: A true experimental research design was used in this study. The medium tested was a primary culture of mice caput epididymal cells (cells and culture medium), conditioned medium (CM) (superna

A liver secretome gene signature-based approach for determining circulating biomarkers of NAFLD severity.

Non-invasive biomarkers of non-alcoholic fatty liver disease (NAFLD) supporting diagnosis and monitoring disease progression are urgently needed. The present study aimed to establish a bioinformatics pipeline capable of defining and validating NAFLD biomarker candidates based on paired hepatic global gene expression and plasma bioanalysis from individuals representing different stages of histologically confirmed NAFLD (no/mild, moderate, more advanced NAFLD). Liver secretome gene signatures were generated in a patient cohort of 26 severely obese individuals with the majority having no or mild

Stem cell secretome derived from human amniotic fluid affords neuroprotection in an ischemic model.

Human amniotic fluid stem cells (hAFSCs) are growing in interest; yet, little is understood about their secretome and neuroprotective actions in different diseases, including stroke. When stem cells are grown in vitro, they release an array of cytokines and growth factors that can stimulate neuroprotective processes. Furthermore, administering secretome rather than cells may be a safer route for patients who are at risk for rejection, promoting innate restorative processes. Current literature implicates that the miRNA contents of such secretome, more specifically exosomes, may regulate the eff

Hypoxia-inducible factor 1-alpha enhances the secretome to rejuvenate adult cardiosphere-derived cells.

OBJECTIVE: After cardiac injury, endogenous repair mechanisms are ineffective. However, cell-based therapies provide a promising clinical intervention based on their ability to restore and remodel injured myocardium due to their paracrine factors. Recent clinical trials have demonstrated that adult cardiosphere-derived cell therapy is safe for the treatment of ischemic heart failure, although with limited regenerative potential. The limited efficiency of cardiosphere-derived cells after myocardial infarction is due to the inferior quality of their secretome. This study sought to augment the th

A Computational Model for Investigating Tumor Apoptosis Induced by Mesenchymal Stem Cell-Derived Secretome.

Apoptosis is a programmed cell death that occurs naturally in physiological and pathological conditions. Defective apoptosis can trigger the development and progression of cancer. Experiments suggest the ability of secretome derived from mesenchymal stem cells (MSC) to induce apoptosis in cancer cells. We develop a hybrid discrete-continuous multiscale model to further investigate the effect of MSC-derived secretome in tumor growth. The model encompasses three biological scales. At the molecular scale, a system of ordinary differential equations regulate the expression of proteins involved in

Characterization of Gaucher disease bone marrow mesenchymal stromal cells reveals an altered inflammatory secretome.

Gaucher disease causes pathologic skeletal changes that are not fully explained. Considering the important role of mesenchymal stromal cells (MSCs) in bone structural development and maintenance, we analyzed the cellular biochemistry of MSCs from an adult patient with Gaucher disease type 1 (N370S/L444P mutations). Gaucher MSCs possessed a low glucocerebrosidase activity and consequently had a 3-fold increase in cellular glucosylceramide. Gaucher MSCs have a typical MSC marker phenotype, normal osteocytic and adipocytic differentiation, growth, exogenous lactosylceramide trafficking, cholester

Crosstalk between visceral adipose tissue secretome and tumoral microenvironment in obesity-induced colorectal cancer.

BACKGROUND: Currently, colorectal cancer (CRC) remains a major global health challenge, with a rising incidence linked to a sedentary lifestyle and metabolic diseases, the main player being obesity (OB). Visceral adipose tissue (VAT) emerged as a key mediator of this association, functioning as a highly active endocrine-immune organ, rather than a mere energy reservoir. In OB, VAT undergoes pathological remodeling, including adipocyte hypertrophy and hypoxia, leading to a state of chronic low-grade inflammation. This review explores the mechanisms by which the dysfunctional VAT secretome orche

TP53 deficiency in AML induces resistance to T-cell engagers through an immunosuppressive secretome.

Bispecific T-cell engagers (BiTE® molecules) have transformed the treatment of B-cell malignancies, yet clinical activity in AML has been modest. Resistance is driven in part by the genetic heterogeneity of AML, most notably TP53 mutations, present in 10-15% of de novo and up to 25% of therapy-related AML. Thus, we hypothesized that TP53 aberrations in AML contribute to cell-intrinsic and extrinsic resistance against T-cell-based immunotherapy. Cytotoxicity against TP53-deleted (DEL) primary AML cells and TP53-knockdown (KD) AML cell lines was reduced in co-cultures with T cells stimulated wit

Exosome-Rich Mesenchymal Stem Cell Secretome Improves Symptoms From Parkinson's Disease: A Case Series.

BACKGROUND: Parkinson's disease (PD) is a progressive neurological condition that primarily affects the central nervous system. It causes neurons to eventually degrade, leading to muscle tremors, rigidity, bradykinesia, impaired balance, and mask-like facies, among other symptoms. A combination of levodopa and carbidopa is the most common treatment for PD, though they are also given separately. These treatments have significant side effects, including headache, dizziness, nausea, somnolence, loss of appetite, diarrhea, constipation, and dyskinesia, which further exacerbate the already present

Secretome-Assisted Regenerative Therapy for Hyaluronic Acid Filler-Induced Vascular Occlusion: A Case Report.

Hyaluronic acid (HA) fillers are widely used for non-surgical facial contouring, but vascular occlusion remains one of the most feared complications due to its potential to cause necrosis, functional impairment, and permanent deformities. Conventional management focuses on high-dose hyaluronidase and supportive therapy, which may not always prevent long-term sequelae. In recent years, regenerative approaches such as secretome and mesenchymal stem cell (MSC) therapies have emerged as promising alternatives. We report the case of a 29-year-old woman who developed nasal vascular occlusion followi

The role of secretome from mesenchymal stromal cells in promoting nerve regeneration after neurotmesis.

BACKGROUND: Neurotmesis, remains a significant clinical challenge due to limited intrinsic regenerative capacity and suboptimal outcomes of current therapies. Mesenchymal stromal cells (MSCs) secretome has emerged as a promising cell-free alternative, providing neurotrophic and immunomodulatory factors to support nerve repair. This study aimed to evaluate the regenerative efficacy of primed adipose-derived MSC secretome in a rat model of sciatic nerve neurotmesis. METHODS: Human and rat adipose-derived MSCs were cultured and primed under hypoxic and inflammatory conditions. Secretomes were ch

Secretome of SHED Sheets Facilitates Mandibular Bone Defect Repair by Shifting Macrophages Inflammatory State.

OBJECTIVE: The inflammatory response, especially the inflammatory status of macrophages, is crucial for bone graft integration and regeneration. Cell sheets from various stromal cells exhibit immunomodulatory and pro-osteogenic traits. However, the therapeutic potential of the secretome from human exfoliated deciduous teeth stromal cell (SHED) sheets remains unclear. This study aimed to explore whether SHED sheet-derived secretome (SS-E) aids bone defect repair via macrophage immunomodulation and to clarify the underlying mechanisms. METHODS: SS-E was collected from osteogenically induced SHE

Peripheral blood mononuclear cell secretome from patients with autoimmune encephalitis promotes seizures in vitro.

OBJECTIVE: Autoimmune encephalitis (AE) is characterized by inflammatory processes in the central nervous system and frequently presents with seizures. Even though an ictogenic potential has been shown for some antibodies against neuronal surface antigens (NSAbs), AE pathophysiology is complex, and NSAbs-independent mechanisms are likely to contribute to seizures. We investigated whether the secretome released by peripheral blood mononuclear cells (PBMCs) from AE patients contributes to seizure generation independently of NSAbs. METHODS: PBMCs were isolated from 19 patients with AE (including

The Genes-Stemness-Secretome Interplay in Malignant Pleural Mesothelioma: Molecular Dynamics and Clinical Hints.

MPM has a uniquely poor somatic mutational landscape, mainly driven by environmental selective pressure. This feature has dramatically limited the development of effective treatment. However, genomic events are known to be associated with MPM progression, and specific genetic signatures emerge from the exceptional crosstalk between neoplastic cells and matrix components, among which one main area of focus is hypoxia. Here we discuss the novel therapeutic strategies focused on the exploitation of MPM genetic asset and its interconnection with the surrounding hypoxic microenvironment as well as

Cell-Free Blood Cell Secretome (BCS) Counteracts Skin Aging and Restores Firmness and Elasticity.

BACKGROUND: Blood Cell Secretome, BCS (also Autologous Conditioned Serum, ACS) is efficacious in treatment of musculoskeletal disorders. It contains inflammation resolving cytokines, growth factors, exosomes, and lipid mediators. Skin aging is associated with reduced TGF-ß signaling and collagen synthesis and chronic (sub-acute) inflammation, among other factors. Pre-clinically, BCS counteracts these mechanisms, suggesting it as a treatment against cutaneous aging. OBJECTIVE: This 24-week study evaluated the effects of deep dermal to immediate sub-dermal micropuncture injections of cell

Systemic sclerosis biomarkers detection in the secretome of TGFβ1-activated primary human lung fibroblasts.

TGFβ1 is a profibrotic mediator that contributes to a broad spectrum of pathologies, including systemic sclerosis-associated pulmonary fibrosis (SSc-PF). However, the secretome of TGFβ1-stimulated primary human normal lung (NL) fibroblasts has not been well characterized. Using fluorescent 2-dimensional gel electrophoresis (2D-PAGE) and differential gel electrophoresis (DIGE) followed by Mass Spectrometry, we identified 37 differentially secreted proteins in the conditioned media of TGFβ1-activated NL fibroblasts and generated a protein-protein association network of the TGFβ1 secretome using

The Functional Parasitic Worm Secretome: Mapping the Place of Onchocerca volvulus Excretory Secretory Products.

Nematodes constitute a very successful phylum, especially in terms of parasitism. Inside their mammalian hosts, parasitic nematodes mainly dwell in the digestive tract (geohelminths) or in the vascular system (filariae). One of their main characteristics is their long sojourn inside the body where they are accessible to the immune system. Several strategies are used by parasites in order to counteract the immune attacks. One of them is the expression of molecules interfering with the function of the immune system. Excretory-secretory products (ESPs) pertain to this category. This is, however,

Lung Cancer Cell-Derived Secretome Mediates Paraneoplastic Inflammation and Fibrosis in Kidney in Mice.

Kidney failure is a possible but rare complication in lung cancer patients that may be caused by massive tumor lysis or a paraneoplastic effect. Clinical case reports have documented pathological characteristics of paraneoplastic syndrome in glomeruli, but are short of molecular details. When Lewis lung carcinoma 1 (LLC1) cells were implanted in mice lungs to establish lung cancer, renal failure was frequently observed two weeks post orthotopic xenograft. The high urinary albumin-to-creatinine ratio (ACR) was diagnosed as paraneoplastic nephrotic syndrome in those lung cancer mice. Profiling t

RGC Neuroprotection Following Optic Nerve Trauma Mediated By Intranasal Delivery of Amnion Cell Secretome.

PURPOSE: Intranasally delivered ST266, the biological, proteinaceous secretome of amnion-derived multipotent progenitor cells, reduces retinal ganglion cell (RGC) loss, optic nerve inflammation, and demyelination in experimental optic neuritis. This unique therapy and novel administration route delivers numerous cytokines and growth factors to the eye and optic nerve, suggesting a potential to also treat other optic neuropathies. Thus, ST266-mediated neuroprotection was examined following traumatic optic nerve injury. METHODS: Optic nerve crush injury was surgically induced in C57BL/6J mice.
