Overlapping ATP2C1 and ASTE1 genes in human genome: implications for SPCA1 expression?

The ATP2C1 gene encodes for the secretory pathway calcium (Ca2+)-ATPase pump (SPCA1), which localizes along the secretory pathway, mainly in the trans-Golgi. The loss of one ATP2C1 allele causes Hailey-Hailey disease in humans but not mice. Examining differences in genomic organization between mouse and human we speculate that the overlap between ATP2C1 and ASTE1 genes only in humans could explain this different response to ATP2C1 dysregulation. We propose that ASTE1, overlapping with ATP2C1 in humans, affects alternative splicing, and potentially protein expression of the latter. If dysregula

AKT2-mediated lysosomal dysfunction promotes secretory autophagy in retinal pigment epithelium (RPE) cells.

Age-related macular degeneration (AMD) is the leading cause of blindness in the elderly, with the non-neovascular or atrophic form being the most common. Current treatment options are limited, emphasizing the urgent need for new therapeutic strategies. Our key finding is that increased levels of AKT2 in the RPE cells impair lysosomal function and trigger secretory autophagy; a non-canonical macroautophagy/autophagy pathway where cellular materials are released via the plasma membrane rather than being degraded by lysosomes. We showed that this process involves a protein complex, AKT2-SYTL1-TRI

Cellular Release of Infectious Hepatitis C Virus Particles via Endosomal Pathways.

Hepatitis C virus (HCV) is a positive-sense, single-stranded RNA virus that causes chronic hepatitis, liver cirrhosis and hepatocellular carcinoma. The release of infectious HCV particles from infected hepatocytes is a crucial step in viral dissemination and disease progression. While the exact mechanisms of HCV particle release remain poorly understood, emerging evidence suggests that HCV utilizes intracellular membrane trafficking and secretory pathways. These pathways include the Golgi secretory pathway and the endosomal trafficking pathways, such as the recycling endosome pathway and the e

Ghrelin as a GH-releasing factor.

Growth hormone (GH) is secreted by the pituitary gland in a pulsatile manner. It is accepted that this pulsatility is primarily controlled by the hypothalamus, although this secretion can also be modulated by factors from GH-targeted organs, the pituitary and other regions of the central nervous systems, or by factors arriving from peripheral organs. In mammals, hypothalamic control of GH pulsatility is classically regulated by the interplay of two opposing hormones, stimulatory GHRH and inhibitory somatostatin (SS). Recognition of the gastric ghrelin peptide as the natural ligand for GH secre

Hepatitis C virus replication and Golgi function in brefeldin a-resistant hepatoma-derived cells.

Recent reports indicate that the replication of hepatitis C virus (HCV) depends on the GBF1-Arf1-COP-I pathway. We generated Huh-7-derived cell lines resistant to brefeldin A (BFA), which is an inhibitor of this pathway. The resistant cell lines could be sorted into two phenotypes regarding BFA-induced toxicity, inhibition of albumin secretion, and inhibition of HCV infection. Two cell lines were more than 100 times more resistant to BFA than the parental Huh-7 cells in these 3 assays. This resistant phenotype was correlated with the presence of a point mutation in the Sec7 domain of GBF1, whi

Endoplasmic reticulum stress: implications for inflammatory bowel disease pathogenesis.

PURPOSE OF REVIEW: To provide an overview of the emerging role of cellular stress responses in inflammatory bowel disease (IBD). RECENT FINDINGS: The unfolded protein response (UPR) is a primitive cellular pathway that is engaged when responding to endoplasmic reticulum stress and regulates autophagy. Highly secretory cells such as Paneth cells and goblet cells in the intestines are particularly susceptible to endoplasmic reticulum stress and are exceedingly dependent upon a properly functioning UPR to maintain cellular viability and homeostasis. Primary genetic abnormalities within the compo

Calcium transport and the secretory ameloblast.

The distribution of calcium in the enamel organ of the rat incisor was investigated using potassium pyroantimonate for ultrastructural localization of calcium. Substantial amounts of precipitate occurred in the intercellular compartment of the enamel organ and modest deposits were observed in specific organelles of the secretory ameloblast. Mitochondria, nuclei granular endoplasmic reticulum, Golgi vesicles and secretory granules consistently contained small deposits of pyroantimonate. Complexing of calcium by the pyroantimonate was confirmed by EGTA decalcification and scanning electron micro

Proteome analysis of Legionella vacuoles purified by magnetic immunoseparation reveals secretory and endosomal GTPases.

Legionella pneumophila, the causative agent of Legionnaires' disease, replicates in macrophages and amoebae within 'Legionella-containing vacuoles' (LCVs), which communicate with the early secretory pathway and the endoplasmic reticulum. Formation of LCVs requires the bacterial Icm/Dot type IV secretion system. The Icm/Dot-translocated effector protein SidC selectively anchors to LCVs by binding the host lipid phosphatidylinositol-4-phosphate (PtdIns(4)P). Here, we describe a novel and simple approach to purify intact vacuoles formed by L. pneumophila within Dictyostelium discoideum by using m

A KDEL Retrieval System for ER-Golgi Transport of Japanese Encephalitis Viral Particles.

Evidence has emerged that RNA viruses utilize the host secretory pathway for processing and trafficking mature viral particles and for exiting the infected cells. Upon completing the complex assembly process, the viral particles take advantage of the cellular secretory trafficking machinery for their intracellular trafficking toward the Golgi organelle and budding or export of virions. In this study, we showed that Japanese encephalitis virus (JEV)-induced extracellular GRP78 contains no KDEL motif using an anti-KDEL-specific antibody. Overexpression of the KDEL-truncated GRP78 in the GPR78 kn

Brucella modulates secretory trafficking via multiple type IV secretion effector proteins.

The intracellular pathogenic bacterium Brucella generates a replicative vacuole (rBCV) derived from the endoplasmic reticulum via subversion of the host cell secretory pathway. rBCV biogenesis requires the expression of the Type IV secretion system (T4SS) VirB, which is thought to translocate effector proteins that modulate membrane trafficking along the endocytic and secretory pathways. To date, only a few T4SS substrates have been identified, whose molecular functions remain unknown. Here, we used an in silico screen to identify putative T4SS effector candidate proteins using criteria such a

PACAP and acetylcholine cause distinct Ca2+ signals and secretory responses in chromaffin cells.

The adrenomedullary chromaffin cell transduces chemical messages into outputs that regulate end organ function throughout the periphery. At least two important neurotransmitters are released by innervating preganglionic neurons to stimulate exocytosis in the chromaffin cell-acetylcholine (ACh) and pituitary adenylate cyclase activating polypeptide (PACAP). Although PACAP is widely acknowledged as an important secretagogue in this system, the pathway coupling PACAP stimulation to chromaffin cell secretion is poorly understood. The goal of this study is to address this knowledge gap. Here, it is

Redirecting intracellular trafficking and the secretion pattern of FSH dramatically enhances ovarian function in mice.

FSH and luteinizing hormone (LH) are secreted constitutively or in pulses, respectively, from pituitary gonadotropes in many vertebrates, and regulate ovarian function. The molecular basis for this evolutionarily conserved gonadotropin-specific secretion pattern is not understood. Here, we show that the carboxyterminal heptapeptide in LH is a gonadotropin-sorting determinant in vivo that directs pulsatile secretion. FSH containing this heptapeptide enters the regulated pathway in gonadotropes of transgenic mice, and is released in response to gonadotropin-releasing hormone, similar to LH. FSH

Secretory type II phospholipase A2 and the generation of intrauterine signals.

Recent advances in the understanding of the metabolism of cell membrane glycerophospholipids and the subsequent generation of biologically active metabolites have provided new insight into their role in normal spontaneous-onset labour and delivery at term, and in preterm labour. In particular, multiple isozymes of the lipolytic enzyme phospholipase A2 (PLA2) have been identified in human gestational tissue during late pregnancy and at the time of labour. Specific isozymes have been identified that are active intracellularly (e.g. cytosolic PLA2) or extracellularly (e.g. Type II and Type IV sec

Antagonism of the final common pathway of mast cell histamine secretion by arylalkylamines.

The antagonism of histamine secretion from rat peritoneal mast cells by a range of drugs (e.g., antihistamines, local anesthetics, tranquilizers, antidepressants, adrenergics and antiadrenergics) and arylalkylamines was studied. Simple arylalkylamines, those without quaternary ammonium, phenolic or anionic groups, uniformly inhibited histamine secretion induced via three major secretory pathways of mast cells (compound 48/80, A23187 and concanavalin A). That is, each simple arylalkylamine nonselectively inhibited all three secretagogues at about the same concentration (avg. range = 2.2). This

Analysis of copper toxicity by western blotting of secretory proteins translocated via the Sec61 channel.

As a trace element, copper plays a vital role in regulating cellular homeostasis by serving as a cofactor for various enzymes. Therefore, maintaining an adequate copper concentration inside the cell is crucial. Even moderate dysregulation of homeostasis can lead to cytotoxicity, protein aggregation, resulting in cell death. One of the significant cellular consequences in Saccharomyces cerevisiae is the inhibition of Sec61-mediated protein translocation in the secretory pathway, resulting in the accumulation of the cytosolic form of precursor secretory proteins, such as Gas1 and CPY, in the cyt

Evidence for basal secretion in the subcommissural organ of the chicken (Gallus gallus).

The basal region of the subcommissural organ (SCO) was studied in the chicken Gallus gallus. The presence of concentric rough endoplasmic reticulum associated with lipid droplets in the basal processes of the ependymal cells, along with the increase of the lysosomal population, can be interpreted as evident signs of degeneration of the basal processes. The nearly absence of secretory processes ending on the meningeal blood vessels, suggests that the leptomeningeal route is missing. On the contrary, the presence of basal ependymal processes in contact with the hypendymal vessels could indicate

Epigenetic maneuvering: an emerging strategy for mycobacterial intracellular survival.

Mycobacterium tuberculosis (Mtb) has elaborated numerous mechanisms for its pathogenesis. Mtb manipulates host signaling pathways to interfere with the immune response and cell death pathways. By employing virulence factors - of which secretory proteins are emerging as significant components - it ensures successful survival in the host. In this review, we discuss advances made on the largely unexplored secretory modifiers of Mtb that alter the host epigenome to impact host pathways for the pathogen's advantage. We highlight the findings on the Mtb-encoded modification enzymes and their role in

Tubular secretion in chronic kidney disease: from established physiology to clinical utility.

In recent years, growing clinical evidence has renewed interest in tubular secretion as a distinct and underappreciated domain of kidney function in chronic kidney disease (CKD). Secretory clearance may provide information beyond estimated glomerular filtration rate and albuminuria, especially for kidney function decline, CKD progression, mortality, and selected adverse events. However, the evidence remains fragmented. Available studies differ in solute selection, sampling methods, analytical platforms, and outcome definitions. It also remains unclear how secretion measures should be translate

Protein Sorting in Plasmodium Falciparum.

Plasmodium falciparum is a unicellular eukaryote with a very polarized secretory system composed of micronemes rhoptries and dense granules that are required for host cell invasion. P. falciparum, like its relative T. gondii, uses the endolysosomal system to produce the secretory organelles and to ingest host cell proteins. The parasite also has an apicoplast, a secondary endosymbiotic organelle, which depends on vesicular trafficking for appropriate incorporation of nuclear-encoded proteins into the apicoplast. Recently, the central molecules responsible for sorting and trafficking in P. falc
