Injectable Silk-Based Hydrogel as an Alternative to Cervical Cerclage: A Rabbit Study.

Background: Preterm birth is a common cause of morbidity and mortality in newborn infants. Cervical insufficiency (CI) is a significant cause of preterm birth. The treatment for CI is cerclage, which is a suture placed around the cervix to provide mechanical support. Cerclage, however, is associated with limited efficacy. Here we present an injectable, silk-based hydrogel as an alternative to cerclage. Objective: Pregnant rabbits were used as an animal model of pregnancy to study the mechanical properties, biocompatibility, and degradation of the hydrogel after cervical injection. Study Design

Dyeing and Functionalization of Wearable Silk Fibroin/Cellulose Composite by Nanocolloidal Array.

A wearable silk fibroin/cellulose composite is reported. It is structurally dyed and functionalized by embedding three-dimensional (3D) or two-dimensional poly(methyl methacrylate) and polystyrene nanocolloidal arrays to form opal and inverse opal silk methylcellulose photonic crystal films (SMPCF). The brilliant color of SMPCF is utilized for naked-eye detection of humidity and a trace amount (0.02%) of H2O content in organic solvents. Volatile organic compounds gases of 5 types were detected. By alternately exposed to organic solvents of methanol, acetonitrile, acetone, ethanol, isopropanol,

Effect of different cocoon stifling methods on the properties of silk fibroin biomaterials.

Stifling treatments are applied to silk cocoons in order to kill the pupae, preventing the emergence of moths and allowing to preserve the silk during long periods of time. All of them involve the application of aggressive steps, such as sun exposure, hot steam from boiling water or hot air, during hours or even days. None of the scientific articles related to silk fibroin biomaterials has previously taken into account this fact in its section of materials and methods. In this work, the consequences of the stifling treatments most commonly used by the silk producing countries and companies are

Evaluation of the Spectral Response of Functionalized Silk Inverse Opals as Colorimetric Immunosensors.

Regenerated silk fibroin is a high molecular weight protein obtained by purifying the cocoons of the domesticated silkworm, Bombyx mori. This report exploits the aqueous processing and tunable β sheet secondary structure of regenerated silk to produce nanostructures (i.e., inverse opals) that can be used as colorimetric immunosensors. Such sensors would enable direct detection of antigens by changes in reflectance spectra induced by binding events within the nanostructure. Silk inverse opals were prepared by solution casting and annealing in a humidified atmosphere to render the silk insoluble

Behcet's Disease: Is There Geographical Variation? A Review Far from the Silk Road.

Behcet's Disease (BD) is a systemic vasculitis characterized by the triad of recurrent mouth and genital ulcers with eye involvement. To date there are no laboratory tests specific for the disease and diagnosis continues to remain on clinical grounds. Multiple criteria have been created as guides for diagnosis; however, given the wide spectrum of organ involvement, some cases remain undiagnosed. The diagnosis of Behcet's Disease may only be made over time as the clinical manifestations emerge sometimes separated by months and even years. With an increased recognition of this disease it has bec

The biocompatibility of silk fibroin and acellular collagen scaffolds for tissue engineering in the ear.

Recent experimental studies have shown the suitability of silk fibroin scaffold (SFS) and porcine-derived acellular collagen I/III scaffold (ACS) as onlay graft materials for tympanic membrane perforation repair. The aims of this study were to further characterize and evaluate the in vivo biocompatibility of SFS and ACS compared with commonly used materials such as Gelfoam and paper in a rat model. The scaffolds were implanted in subcutaneous (SC) tissue and middle ear (ME) cavity followed by histological and otoscopic evaluation for up to 26 weeks. Our results revealed that SFS and ACS were w

Engineering of fluorescent emission of silk fibroin composite materials by material assembly.

This novel materials assembly technology endows the designated materials with additional/enhanced performance by fixing "functional components" into the materials. Such functional components are molecularly recognized and accommodated by the designated materials. In this regard, two-photon fluorescence (TPF) organic molecules and CdTe quantum dots (QDs) are adopted as functional components to functionalize silk fibers and films. TPF organic molecules, such as, 2,7-bis[2-(4-nitrophenyl) ethenyl]-9,9-dibutylfluorene (NM), exhibit TPF emission quenching because of the molecular stacking that lead

Silk fibroin conduits: a cellular and functional assessment of peripheral nerve repair.

Silk fibroin conduits were designed with appropriate porosity for peripheral nerve repair. The aim of this work was to use these conduits to examine cell inflammatory responses and functional recovery in a sciatic nerve defect model. A total of 45 randomized Lewis rats were used to create an 8-mm defect bridged by a silk guide, commercial collagen guide, or an autograft. After 1, 4, and 8 weeks, macrophage recruitment, percentage of newly formed collagen, number of myelinated axons, and gastrocnemius muscle mass were evaluated. Following 8 weeks, ED1+ cells in autograft and silk conduits decre

Topology-Constrained Load Transfer Reprograms Silk Fiber Deformation beyond β-Sheet Order.

The mechanical response of silk fibers is commonly associated with β-sheet crystallinity and molecular orientation, yet how these molecular descriptors are transmitted across the fibrillar hierarchy remains less clear. Here, we compare Hyphantria cunea silk with Bombyx mori silk to examine how nanofibril-level load-transfer descriptors are associated with deformation when β-sheet fraction and axial orientation are comparable. Peptide profiling and single-fiber synchrotron infrared microspectroscopy show that H. cunea silk contains β-sheet-compatible sequence motifs and reaches β-sheet content

Silk Fibroin Counteracts Fibroblast Senescence to Restore ECM Homeostasis in Aged Skin.

Skin aging is characterized by a progressive decline in regenerative capacity, primarily driven by fibroblast senescence, oxidative stress, chronic inflammation, and the degradation of type I/III collagen, culminating in an extracellular matrix (ECM) imbalance. Current injectable fillers-such as hyaluronic acid, collagen, and PLLA-provide temporary structural support but fail to address the underlying cellular senescence or restore ECM homeostasis, highlighting the need for regenerative biomaterials. Silk fibroin (SF), a natural protein, self-assembles into a β-sheet-rich scaffold that structu

Polycationic silk fibroin chelating nano-hydroxyapatite promotes angiogenesis for burn wound healing.

The regeneration of deep burn wounds, which cause dermal tissue necrosis, remains a significant clinical challenge. Herein, we report a skin dressing fabricated by integrating nano-hydroxyapatite (nHA) with a polycationic silk fibroin (CSF) to promote burn wound healing via enhanced angiogenesis. The nHA was firmly incorporated into the CSF matrix through metal-phenolic coordination, improving the mechanical robustness and stability of the composite. The CSF-nHA dressing significantly promoted the proliferation of NIH-3T3 fibroblasts and enhanced the recruitment and migration of human umbilica

Bioorthogonally Active Silk Fibroin Patch Facilitate Immunomodulatory Prodrug Activation for Tissue Repair.

Current silk fibroin (SF)-based tissue repair patches often exhibit limited immunomodulatory efficacy, exacerbating inflammatory responses and compromising functional tissue integration. To address this limitation, we developed a bioorthogonal prodrug activation strategy within an SF patch via tetrazine functionalization (SF-Tz). This platform enables the spatiotemporally controlled release of trans-cyclooctene (TCO)-caged dexamethasone (TCO-Dex) via inverse electron-demand Diels-Alder reactions. The SF-Tz platform was engineered through optimized carboxylation and tetrazine conjugation protoc

Flexural performance and microstructural correlation of natural silk fabric-reinforced epoxy composites.

This study investigates the flexural behaviour and microstructure-property relationships of natural silk fabric-reinforced epoxy composites fabricated using a cast moulding route. Three rectangular specimens (100 × 20 × 5 mm) were tested under three-point bending in accordance with ASTM D790 with a 60 mm support span. The composites were produced at moderate fibre volume fractions (approximately 20-40 vol%), and their density and void content were evaluated using mass-volume measurements and optical image analysis. The measured flexural strength ranged between 100 and 130 MPa, with results rep

Engineered silk fibroin dendritic cell backpacks to boost antitumor immunity following radiotherapy.

Radiotherapy initiates an antitumor immune response by releasing tumor antigens; however, its efficacy is often limited by the subsequent quantitative and qualitative dysfunction of dendritic cells within the tumor microenvironment. This dysfunction leads to impaired antigen presentation and suboptimal T cell activation. To address this, we developed a biomaterial-based cellular engineering strategy using silk fibroin-based dendritic cell backpacks. Silk fibroin backpacks offer structural stability and aqueous processability, enabling the incorporation of CpG under mild conditions. The easy fu

Macroscopic assembly behavior of silk/lignin-based adhesive hydrogels: elementary macroscopic assembly.

Macroscopic assembly has been explored as a strategy to construct appealing materials. Yet, an open question remains: does macroscopic assembly always occur one by one? In this study, a new kind of silk-based hydrogel was developed through incorporating lignin-related compounds via 1-ethyl-3-(3-pyl)-carbodiimide/N-hydroxysuccinimide (EDC/NHS)-mediated coupling chemistry. Thanks to the hydrogen-bond, hydrophobic, and π-π interactions, the hydrogels possessed the capability of adhesive and macroscopic assembly, which could be further tuned by introducing iron ions, as Fe3+ can form coordination
