Unsupervised learning reveals rapid gait adaptation after leg loss and regrowth in spiders.

Many invertebrates voluntarily lose (autotomize) limbs during antagonistic encounters, and some regenerate functional replacements. Because limb loss can have severe consequences on individual fitness, it is likely subject to significant selective pressures, making this an excellent phenomenon with which to investigate biomechanical robustness. Spiders frequently autotomize one or more legs. We investigated the time course of locomotor recovery after leg loss and regeneration in juvenile tarantulas (Arachnida: Araneae) naive to autotomy. We recorded high-speed video of spiders running with all

On small huntsman spiders (Araneae, Philodromidae) occurring in Guizhou and Hubei provinces, China.

Spiders of the family Philodromidae Thorell, 1869 from Guizhou and Hubei provinces, China are studied. A total of three genera and seven species are reported and illustrated, comprising Sinodromuslanyue sp. nov. (newly recorded genus for Hubei), and all known species from both provinces: Philodromusauricomus L. Koch, 1878, P.guiyang Long & Yu, 2022, P.subaureolus Bösenberg & Strand, 1906, P.paiki Jang, Lee, Yoo & Kim, 2024 (the previously records of P.spinitarsis Simon, 1895 from Guizhou and Hubei are presumed to be misidentifications, and should belong to P.paiki), P.rufus Walckenaer, 1826 (n

Beyond venomous fangs: Uloboridae spiders have lost their venom but not their toxicity.

BACKGROUND: Venom, one of nature's most potent secretions, has played a crucial role in the evolutionary success of many animal groups, including spiders. However, Uloboridae spiders appear to lack venom and capture their prey, unlike venomous spiders, by extensive silk-wrapping and regurgitation of digestive fluids onto the entire prey package. A prevailing hypothesis posits that toxins may have been reallocated from the venom to alternative secretions, like silk or digestive fluids. Yet, whether uloborids have retained venom toxins and the mechanisms underlying prey immobilisation remain unr

A trade-off in evolution: the adaptive landscape of spiders without venom glands.

BACKGROUND: Venom glands play a key role in the predation and defense strategies of almost all spider groups. However, the spider family Uloboridae lacks venom glands and has evolved an adaptive strategy: they excessively wrap their prey directly with spider silk instead of paralyzing it first with toxins. This shift in survival strategy is very fascinating, but the genetic underpinnings behind it are poorly understood. RESULTS: Spanning multiple spider groups, we conducted multiomics analyses on Octonoba sinensis and described the adaptive evolution of the Uloboridae family at the genome lev

Grab it or not? Measuring avoidance of spiders with touchscreen-based hand movements.

BACKGROUND AND OBJECTIVES: Previous studies have shown that humans, particularly those with high levels of spider fear, show automatic avoidance of spiders. However, most tasks used in these studies employ symbolic approach-avoidance movements instead of naturalistic movements. METHODS: The current study employed a touchscreen-based approach-avoidance task in which 86 female participants made hand and arm movements that mimic the real-life touching, grasping, and moving of spiders. Using a large touchscreen, they had to reach for pictures of spiders and neutral objects (leaves), touch them, a

Where cone snails and spiders meet: design of small cyclic sodium-channel inhibitors.

A 13 aa residue voltage-gated sodium (NaV) channel inhibitor peptide, Pn, containing 2 disulfide bridges was designed by using a chimeric approach. This approach was based on a common pharmacophore deduced from sequence and secondary structural homology of 2 NaV inhibitors: Conus kinoshitai toxin IIIA, a 14 residue cone snail peptide with 3 disulfide bonds, and Phoneutria nigriventer toxin 1, a 78 residue spider toxin with 7 disulfide bonds. As with the parent peptides, this novel NaV channel inhibitor was active on NaV1.2. Through the generation of 3 series of peptide mutants, we investigated

Adaptation of the spiders to the environment: the case of some Chilean species.

Spiders are small arthropods that have colonized terrestrial environments. These impose three main problems: (i) terrestrial habitats have large fluctuations in temperature and humidity; (ii) the internal concentration of water is higher than the external environment in spiders, which exposes them continually to water loss; and (iii) their small body size determines a large surface/volume ratio, affecting energy exchange and influencing the life strategy. In this review we focus on body design, energetic, thermal selection, and water balance characteristics of some spider species present in Ch

Trophic specialisation in a predatory group: the case of prey-specialised spiders (Araneae).

Predators appear to be less frequently specialised (i.e. adapted to restricted diet) on their prey than herbivores, parasites or parasitoids. Here, we critically evaluate contemporary evolutionary hypotheses that might be used to explain the evolution of specialised foraging in predators. We propose a unifying concept within which we define four types of trophic categories using ecological (diet breadth) and evolutionary (degree of adaptations) contexts. We use data on spiders (Araneae), the most diversified order of terrestrial predators, to assess applicability of frameworks and evolutionary

Approach behavior can mitigate predominately univalent negative attitudes: evidence regarding insects and spiders.

Three experiments tested whether disliking of predominately univalently negative attitude objects could be reduced by a procedure pairing approach behaviors with subliminally presented images of the objects. Experiment 1 demonstrated that participants who approached images of insects rated insects less negatively than participants who did not approach insect pictures. Experiment 2 extended this effect to spiders and used an implicit measure of spider attitudes. Experiment 3 examined the consequences of an approach induction for affect during actual approach behavior in a sample of individuals

The role of fear and expectancies in capture of covert attention by spiders.

Fear-related stimuli are often prioritized during visual selection but it remains unclear whether capture by salient objects is more likely to occur when individuals fear those objects. In this study, participants with high and low fear of spiders searched for a circle while on some trials a completely irrelevant fear-related (spider) or neutral distractor (butterfly/leaf) was presented simultaneously in the display. Our results show that when you fear spiders and you are not sure whether a spider is going to be present, then any salient distractor (i.e., a butterfly) grabs your attention, sug

Taxonomy of Australian Funnel-web spiders using rp-HPLC/ESI-MS profiling techniques.

The complex nature of venom from spider species offers a unique natural source of potential pharmacological tools and therapeutic leads. The increased interest in spider venom molecules requires reproducible and precise identification methods. The current taxonomy of the Australian Funnel-web spiders is incomplete, and therefore, accurate identification of these spiders is difficult. Here, we present a study of venom from numerous morphologically similar specimens of the Hadronyche infensa species group collected from a variety of geographic locations in southeast Queensland. Analysis of the c

Differential PFAS transfer through the terrestrial and aquatic food web to predatory spiders.

The environmental persistence of Per- and polyfluoroalkyl substances (PFAS) results in prolonged exposure of organisms, which in turn leads to bioaccumulation and trophic transfer. However, if trophic transfer is studied at all, different ecosystems are generally examined in isolation. The aim of this study was therefore to unravel the PFAS transfer through a contaminated terrestrial and aquatic food web, with spiders as top predators. To this end, organisms from the aquatic and terrestrial compartments of a PFAS-contaminated ecosystem were collected and screened for 44 PFAS. The trophic posit

Beyond species-level differences: A comprehensive review of intraspecific venom plasticity in spiders.

Spider venoms represent a diverse array of biological cocktails, crucial for predator-prey dynamics and ecological interactions. While differences in venom composition between species (interspecific variation) are well-characterized, the extent and drivers of intraspecific venom plasticity remain a critical, underexplored area. This comprehensive review synthesizes current research on venom variation within species, emphasizing the multifaceted factors that govern this dynamic phenotype. Venom composition, yield, and bioactivity are significantly influenced by both endogenous and exogenous ele

Assessment of local pain and analgesia in envenoming by wandering spiders (Phoneutria spp.).

Phoneutrism (bites by wandering spiders of the genus Phoneutria) frequently results in local pain. We describe a retrospective cohort study of a case series of phoneutrism admitted to our Emergency Department (ED), in which we used the Numeric Pain Rating Scale (NPRS 0-10) to assess the intensity of local pain upon admission, and recorded the analgesic measures used to control this pain. Other criteria for inclusion were: (1) An age ≥8 years, (2) Treatment exclusively at our ED, and (3) Visualization or photographing the spider at the time of the bite and/or bringing the spider for identificat

Brown Spiders' Phospholipases-D with Potential Therapeutic Applications: Functional Assessment of Mutant Isoforms.

Phospholipases-D (PLDs) found in Loxosceles spiders' venoms are responsible for the dermonecrosis triggered by envenomation. PLDs can also induce other local and systemic effects, such as massive inflammatory response, edema, and hemolysis. Recombinant PLDs reproduce all of the deleterious effects induced by Loxosceles whole venoms. Herein, wild type and mutant PLDs of two species involved in accidents-L. gaucho and L. laeta-were recombinantly expressed and characterized. The mutations are related to amino acid residues relevant for catalysis (H12-H47), magnesium ion coordination (E32-D34) and

Searching for the toxic potential of Loxosceles amazonica and Loxosceles willianilsoni spiders' venoms.

The Loxosceles genus belongs to the Sicariidae family and it comprises species whose venom can cause accidents with potentially fatal consequences. We have previously shown that SMase D is the enzyme responsible for the main pathological effects of Loxosceles venom. Despite the severity of accidents with Loxosceles, few species are considered to be of medical importance. Little is known about the venom of non-synanthropic species that live in natural environments. To contribute to a better understanding about the venom's toxicity of Loxosceles genus, the aim of this study was to (i) characteri

Bugs as drugs, part two: worms, leeches, scorpions, snails, ticks, centipedes, and spiders.

In this second of a two-part series analyzing the evidence for the use of organisms as medicine, the use of a number of different "bugs" (worms, leeches, snails, ticks, centipedes, and spiders) is detailed. Several live organisms are used as treatments: leeches for plastic surgery and osteoarthritis and the helminths Trichuris suis and Necator americanus for inflammatory bowel disease. Leech saliva is the source of a number of anticoagulants, including the antithrombin agent hirudin and its synthetic analogues, which have been approved for human use. A rational nomenclature for naming peptide toxins from spiders and other venomous animals.

Molecular toxinology research was initially driven by an interest in the small subset of animal toxins that are lethal to humans. However, the realization that many venomous creatures possess a complex repertoire of bioactive peptide toxins with potential pharmaceutical and agrochemical applications has led to an explosion in the number of new peptide toxins being discovered and characterized. Unfortunately, this increased awareness of peptide-toxin diversity has not been matched by the development of a generic nomenclature that enables these toxins to be rationally classified, catalogued, and

Speeded detection and increased distraction in fear of spiders: evidence from eye movements.

Anxiety patients exhibit attentional biases toward threat, which have often been demonstrated as increased distractibility by threatening stimuli. In contrast, speeded detection of threat has rarely been shown. Therefore, the authors studied both phenomena in 3 versions of a visual search task while eye movements were recorded continuously. Spider-fearful individuals and nonanxious control participants participated in a target search task, an odd-one-out search task, and a category search task. Evidence for disorder-specific increased distraction by threat was found in all tasks, whereas speed
