Centromere protein CEP170 is dispensable for mouse spermatogenesis and male fertility.

Spermatogenesis is crucial for male fertility and involves highly coordinated cellular differentiation, which depends on mitosis, meiosis, and cellular transformation processes that all require dynamic remodeling of the microtubule cytoskeleton. Microtubule anchoring, spindle assembly, and ciliary control all depend on centromere protein 170 (CEP170), which is found in the subdistal pole of the centrosome. CEP170 is regarded as a promising option for controlling male fertility because of its high expression in the testes and interactions with meiotic proteins. Given the embryonic lethality ass

Cdc1 removes the ethanolamine phosphate of the first mannose of GPI anchors and thereby facilitates the integration of GPI proteins into the yeast cell wall.

Temperature-sensitive cdc1(ts) mutants are reported to stop the cell cycle upon a shift to 30°C in early G2, that is, as small budded cells having completed DNA replication but unable to duplicate the spindle pole body. A recent report showed that PGAP5, a human homologue of CDC1, acts as a phosphodiesterase removing an ethanolamine phosphate (EtN-P) from mannose 2 of the glycosylphosphatidylinositol (GPI) anchor, thus permitting efficient endoplasmic reticulum (ER)-to-Golgi transport of GPI proteins. We find that the essential CDC1 gene can be deleted in mcd4∆ cells, which do not attach EtN-P

Chemical induction of nondisjunction in drosophila.

Tests for chemically induced nondisjunction and loss of the sex chromosomes in Drosophila were performed. Of 31 compounds tested four gave rise only to an increase of XO exceptions, indicating the induction of chromosome loss. Six compounds, all known spindle inhibitors (colchicine, organic mercury, lead, and tin compounds) gave rise to an increase both of XXY and XO or of only XXY. The effect by metalloorganic compounds of which methylmercury was studied particularly closely, follows a peculiar pattern. In females with structurally normal X chromosomes only an increase of XX gametes is obtain

Meiosis and the Neo-XY system of Dichroplus vittatus (Melanoplinae, Acrididae): a comparison between sexes.

The origin of neo-XY sex systems in Acrididae is usually explained through an X-autosome centric fusion, and the behaviour of the neo-sex chromosomes has been solely studied in males. In this paper we analysed male and female Dichroplus vittatus. The karyotype comprises 2n = 20 chromosomes including 9 pairs of autosomes and a sex chromosome pair that includes a large metacentric neo-X and a small telocentric neo-Y. We compared the meiotic behaviour of the sex bivalent between both sexes. Mean cell autosomal chiasma frequency was low in both sexes and slightly but significantly higher in males

Identifies microtubule-binding protein CSPP1 as a novel cancer biomarker associated with ferroptosis and tumor microenvironment.

Centrosome and spindle pole-associated protein (CSPP1) is a centrosome and microtubule-binding protein that plays a role in cell cycle-dependent cytoskeleton organization and cilia formation. Previous studies have suggested that CSPP1 plays a role in tumorigenesis; however, no pan-cancer analysis has been performed. This study systematically investigates the expression of CSPP1 and its potential clinical outcomes associated with diagnosis, prognosis, and therapy. CSPP1 is widely present in tissues and cells and its aberrant expression serves as a diagnostic biomarker for cancer. CSPP1 dysregul

The Transformation of the Centrosome into the Basal Body: Similarities and Dissimilarities between Somatic and Male Germ Cells and Their Relevance for Male Fertility.

The sperm flagellum is essential for the transport of the genetic material toward the oocyte and thus the transmission of the genetic information to the next generation. During the haploid phase of spermatogenesis, i.e., spermiogenesis, a morphological and molecular restructuring of the male germ cell, the round spermatid, takes place that includes the silencing and compaction of the nucleus, the formation of the acrosomal vesicle from the Golgi apparatus, the formation of the sperm tail, and, finally, the shedding of excessive cytoplasm. Sperm tail formation starts in the round spermatid stag

Exploring the tumor-promoting function and prognostic value of ESPL1 in prostate adenocarcinoma.

BACKGROUND: Extra spindle pole bodies-like 1 (ESPL1), a gene encoding the Separase protein, has been implicated in cancer development across multiple tumor types. However, its precise role in prostate adenocarcinoma (PRAD) remains underexplored. This study investigates ESPL1's contribution to PRAD progression and evaluates its utility as a prognostic indicator and potential therapeutic target. METHODS: We analyzed ESPL1 expression using datasets from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO). The prognostic significance of ESPL1 was evaluated through survival analysis.

SPC25 promotes gastric cancer cells G1/S cell cycle transition and tumorigenesis by enhancing SLC1A5-mediated glutamine metabolism.

Although spindle pole body component 25 (SPC25) has been implicated in various cancers, its precise role in GC remains unexplored. This study aimed to investigate the role and mechanism of SPC25 in GC progression. SPC25 expression was analyzed using TCGA database and 40 paired GC clinical specimens. RT-qPCR, Western blot and immunohistochemistry (IHC) staining were performed to detect the expression of SPC25 in GC samples. Flow cytometry was conducted to assess the cell cycle, and commercial kits were used to measure the levels of glutamine, glutamic acid, α-Ketoglutaric acid (α-KG) and NADPH/

Connecting the dots: a centrosome-associated RAC1 signaling network links centrosome amplification with autophagy.

In dividing cells, centrosomes are the primary microtubule-organizing centers and typically duplicate only once per cell division cycle, ensuring that there is just a single centrosome at each mitotic spindle pole. Centrosome amplification presents a paradox: although supernumerary centrosomes can induce chromosomal instability and promote invasive behavior, they may also impede cell proliferation by activating the Hippo-LATS pathway and the PIDDosome-p53 axis. Therefore, cells with supernumerary centrosomes must either tolerate the abnormal centrosome numbers or restore centrosome homeostasis

The dynamic pool of Rec8-cohesin is crucial for meiotic recombination and transcription regulation in the yeast Saccharomyces cerevisiae.

Cohesin is a ring-shaped protein complex that mediates sister-chromatid cohesion (SCC) to ensure accurate chromosome segregation during mitosis and meiosis. In Saccharomyces cerevisiae, cohesin consists of four core subunits-Smc1, Smc3, Scc1/Mcd1, and Scc3. During meiosis, the mitotic α-kleisin Scc1/Mcd1 is replaced by the meiosis-specific α-kleisin Rec8. Rec8-containing cohesin is essential for multiple meiotic processes, including chromosome morphogenesis, homologous recombination, axis and synaptonemal complex formation, SCC, and transcriptional regulation. While stable association of Rec8-

Extraciliary roles of the ciliopathy protein JBTS17 in mitosis and neurogenesis.

OBJECTIVE: JBTS17 is a major gene mutated in ciliopathies such as Joubert syndrome and oral-facial-digital syndrome type VI. Most patients with loss of function mutations in JBTS17 exhibit cerebellar vermis hypoplasia and brainstem malformation. However, some patients with JBTS17 mutations show microcephaly and abnormal gyration. We examined potential roles of JBTS17 in neurogenesis to understand the pathological mechanism of JBTS17-related cortical abnormalities. METHODS: We examined subcellular localization and cell-cycle-dependent expression of JBTS17 proteins using anti-JBTS17 antibodies

The primary cilium as a multiple cellular signaling scaffold in development and disease.

Primary cilia, single hair-like appendage on the surface of the most mammalian cells, were once considered to be vestigial cellular organelles for a past century because of their tiny structure and unknown function. Although they lack ancestral motility function of cilia or flagella, they share common ground with multiciliated motile cilia and flagella on internal structure such as microtubule based nine outer doublets nucleated from the base of mother centrioles called basal body. Making cilia, ciliogenesis, in cells depends on the cell cycle stage due to reuse of centrioles for cell division

T cell activation by terminal complex of complement and immune complexes.

T cell hyperactivation and complement consumption are prominent features of the immunopathology of systemic lupus erythematosus. Although complement activation is secondary to autoantibodies that form immune complexes (ICs), the trigger for alterations in human peripheral blood T cells is poorly understood. To study the impact (on T cells) of several types of preformed ICs and terminal complement complex, also referred to as C5b-9, we incubated these immune reactants with peripheral blood naive CD4(+) T cells as well as Jurkat cells and analyzed their effects on cellular behavior. We first ass

New markers for cervical dysplasia to visualise the genomic chaos created by aberrant oncogenic papillomavirus infections.

Extensive research over the past 20 years provided strong evidence that persistent infections with high risk type human papillomaviruses (HR-HPVs) cause cervical cancer. However, depending on their age, more than 20% of normal women are infected with these viruses and only very few develop clinically relevant dysplastic lesions or even cancer. During an acute HPV infection, expression of viral genes, in particular the viral E6 and E7 oncogenes is restricted to differentiated epithelial cells, which lost the capability to replicate their genomes and are therefore at no further risk for acquirin

Morphology of sympathetic preganglionic neurons in the thoracic spinal cord of the cat: an intracellular horseradish peroxidase study.

Horseradish peroxidase was intracellularly injected into sympathetic preganglionic neurons (SPN) of the third thoracic segment in cats. Seven neurons were reconstructed from serial horizontal or parasagittal sections of the spinal cord. The cell bodies of all neurons were located in the n. intermediolateralis pars principalis (ILp). They were spindle-shaped with the long axis in craniocaudal direction or large and multipolar or small and oval in shape. Preferentially on the cranial and caudal pole of the cell body, five to eight primary dendrites arose from the cell body. Dendritic branches we

Adnexal Endometrioid Carcinomas With Sex Cord-Like Morphology are Frequently PAX8-Negative, SOX17-Positive, and Enriched for CTNNB1 Alterations.

Sex cord-like morphology is now a well-known feature of occasional examples of endometrioid carcinoma of the ovary or fallopian tube. Recently, we have observed that these tumors are frequently negative for PAX8, prompting us to review their morphologic, immunohistochemical, and genomic spectrum. Twenty tumors (17 ovarian, 3 fallopian tube) with available tissue were identified in patients ranging from 32 to 78 (median 60) years. Sex cord-like patterns included cords/trabeculae (n=17), small tubular glands (n=16), and "granulosa-like" nests (n=12). In addition, 1 had ependymoma-like features w

Molecular Characterization of Vascular Transformation of Lymph Node Sinuses via Oncomine Comprehensive Assay.

Vascular transformation of lymph node sinuses (VTSs) is defined as phenotypic transformation of the lymph node sinusoidal lining from lymphatic to vascular endothelium. The pathogenesis of VTS remains unclear. Thus far, VTS has not been characterized at the molecular level. Herein, we shed some light on VTS pathogenesis while providing a detailed description of its clinical, histopathologic, immunophenotypic, and molecular features. Our specimens involved 2 women and 3 men, aged 36 to 66 years (mean 49 years old). The lymph nodes were in the right inguinal (n = 2), left axillary (n = 1), right

Renal schwannoma appears on imaging as renal cell carcinoma: a case report.

BACKGROUND: Renal schwannomas are exceedingly rare benign tumors originating from Schwann cells, with fewer than 50 cases reported in the literature. The "ancient" variant refers to long-standing schwannomas that develop degenerative changes, which may mimic the aggressive radiological features of renal cell carcinoma. This diagnostic overlap often leads to preoperative misdiagnosis and can significantly impact surgical decision-making, often resulting in radical rather than nephron-sparing approaches.
