The pathogenesis of joint infections is not well understood. In particular, we do not know why these infections respond poorly to antibiotic treatment. Here we show that methicillin-resistant Staphylococcus aureus, a major cause of joint infections, forms exceptionally strong biofilmlike aggregates in human synovial fluid (SF), to an extent significantly exceeding biofilm formation observed in growth medium or serum. Screening a transposon bank identified bacterial fibronectin- and fibrinogen-binding proteins as important for the formation of macroscopic clumps in SF, suggesting an important r

We report a previously healthy 36-year-old mole who developed a pulmonary syndrome caused by Hantavirus infection. He was admitted to an intensive care unit in shock and respiratory failure and died four hours after admission. Blood cultures were positive for Staphylococcus aureus. This patient could be an index case that shows, for the first time, an association of Hantavirus Pulmonary Syndrome with Staphylococcus aureus infection similar to what happens with other viruses.

UNLABELLED: Late onset of neonatal infection could have been transmitted in prenatal period, but it is usually secondary to a postnatal transmission.
CASE REPORT: A premature neonate developed staphylococcal pneumonia at 18 days of life. Genomic typing of the strains of Staphylococcus aureus obtained from the patient and from his mother (found in the endocervix culture 48 h before delivery) was identical. These strains were different from those isolated in other neonates colonised by S. aureus in the unit during at that moment.
CONCLUSION: The observed case of staphylococcal pneumonia may co

Tonsillar and pharyngeal smears revealed growth of pathogenic microorganisms, mainly of Staphylococcus aureus, in 58.0 +/- 3.7% of 174 patients with duodenal ulcer and gastroduodenitis during remission or unstable remission, in 40.0 +/- 5.3% of 85 patients with other gastroenterological diseases and in 17.5 +/- 6.0% of 40 subjects constituting control group. Urease activity ratio (biological marker of Helicobacter pylori) was found to be approximately the same in every group of patients vs 11.1 +/- 6.2% of 27 healthy persons. The relationship between enhanced cell-mediated immunity (CMI) to staphylococcal antigens, expressed as delayed hypersensitivity (DH), and the development of catarrhal infiltrates at the limbus in the rabbit has been explored by others. This DH is required for infiltrates to develop in the rabbit cornea when it is exposed to conjunctival inoculation with live Staphylococcus aureus cells. Similar investigations have not been pursued in the human, although St. aureus has been isolated from lids of patients with sterile marginal ulcers. We have tested 69 patients with blepharitis, eleven with and

Children have frequent staphylococcal infections, and many lack antibody to TSST-1, a toxin associated with the toxic shock syndrome (TSS). To determine why there have been no nonmenstrual cases of TSS reported in children in Utah, the authors tested S. aureus isolated from children for TSST-1 by radial immunodiffusion and sera from other hospitalized children by radioimmunoassay for antibody to TSST-1. TSST-1 was produced by 25% of S. aureus. Fifty-two children had infections with toxin producing strains. None had TSS. The prevalence of presumably protective levels of antibody (greater than o

Sixteen children with "scalded skin" due to staphylococcal infections are described [six cases of staphylococcal scarlet fever, 6 cases of bullous impetigo and 4 cases of toxic epidermal necrolysis (Lyell's disease--of which Ritter's disease is only the neonatal manifestation)]. The clinical features of each of these conditions are described, the common feature being the severity of the pathological changes. The role of the exfoliating toxin secreted by the pathogenic staphylococci mostly belonging to phagegroup II, phagetype 3A, 3N, 3C, 55 or 71) is emphasized. Treatment should be aimed at re

Necrotizing soft tissue infections (NSTIs) are critical clinical conditions characterized by extensive necrosis of any layer of the soft tissue and systemic toxicity. Group A streptococci (GAS) and Staphylococcus aureus are two major pathogens associated with monomicrobial NSTIs. In the tissue environment, both Gram-positive bacteria secrete a variety of molecules, including pore-forming exotoxins, superantigens, and proteases with cytolytic and immunomodulatory functions. The present review summarizes the current knowledge about streptococcal and staphylococcal toxins in NSTIs with a special

Staphylococcus aureus and other staphylococci continue to cause life-threatening infections in both hospital and community settings. They have become increasingly resistant to antibiotics, especially β-lactams and aminoglycosides, and their infections are now, in many cases, untreatable. Here we present a non-antibiotic, non-phage method of treating staphylococcal infections by engineering of the highly mobile staphylococcal pathogenicity islands (SaPIs). We replaced the SaPIs' toxin genes with antibacterial cargos to generate antibacterial drones (ABDs) that target the infecting bacteria in t

Auranofin, (AF), a gold(I) complex in clinical use for the therapy of rheumatoid arthritis, is reported here to produce remarkable bactericidal effects in vitro against Staphylococcus sp. Noticeably, a similar antimicrobial action and potency are also noticed toward a few methicillin-resistant Staphylococcus aureus strains but not toward Escherichia coli. The time and concentration dependencies of the antimicrobial actions of AF have been characterized through recording time kill curves, and a concentration dependent profile highlighted. Overall, the present results point out that auranofin mi

PURPOSE: Colonization with Staphylococcus aureus is considered a risk factor for the rising incidence of pediatric community-acquired skin and soft tissue infections (CA-SSTIs), and intrafamily spread is thought to be the source of colonization.
METHODS: A prospective study was conducted to determine skin and nasal staphylococcal colonization rates among the caretakers of CA-SSTI patients and those of nonabscess controls. A questionnaire regarding risk factors was administered to all participants. Fisher's Exact test and the chi(2) test were used for statistical analysis.
RESULTS: Forty-six

Bacterial peritonitis is a serious complication of peritoneal dialysis patients and of patients after abdominal surgery. Especially episodes due to Staphylococcus aureus can harm the peritoneum severely, resulting in peritoneal fibrosis. Human peritoneal mesothelial cells play a critical role in maintaining the integrity of the peritoneum, as they release components of the fibrinolytic system and regulate the influx of immune cells by expressing chemokines and adhesion molecules. Using cultured human peritoneal mesothelial cells (HMCs) and blood mononuclear cells, we analyzed the effect of dif

The authors have evaluated the efficacy and tolerance of teicoplanin against Gram-positive infections in 8 patients (four with endocarditis, one with cholangitis, one with purulent arthritis, one with gathered fistula and one with general pyoderma), arose both in the immunocompetent and immunodepressed subjects. All patients received IM teicoplanin at a dosage of 200-400 mg/die, according to the severity of the infectious process. Teicoplanin proved to be an effective and well-tolerated drug in the treatment of staphylococcus infections, both for immunocompetent and with serious deficit of the

Two hundred and forty-four children were evaluated in a study comparing clindamycin hydrochloride, erythromycin, and phenoxymethyl penicillin for the treatment of streptococcal pyoderma. Similar numbers of patients were followed during (day 7) and after (day 14) therapy in each of the three treatment groups. All patients had skin lesions positive for group A streptococci with or without staphylococci; the percentage rate of pure and mixed cultures was similar for the three groups of patients. Both clindamycin and erythromycin proved somewhat superior to penicillin on the basis of effecting ear

Staphylococcal disease represents a universal burden including acute, life-threatening infections as well as chronic infections usually associated with foreign materials. Infections occur notably in permanent carriers of Staphylococcus aureus. To date, all the attempts to develop an efficacious vaccine against S. aureus have failed. Failures in vaccine clinical trials might be related to a focus on single targets and development of humoral-based vaccines rather than vaccines with a combination of antigens stimulating both humoral and cellular immunity. The end points of these unsuccessful tria

BACKGROUND: Methicillin-resistant Staphylococcus aureus (MRSA) poses a significant challenge in clinical environments due to its resistance to standard antibiotics. Staphylococcal Protein A (SpA), a crucial virulence factor of MRSA, undermines host immune responses, making it an attractive target for vaccine development. This study aimed to identify potential epitopes within SpA that could elicit robust immune responses, ultimately contributing to the combat against multidrug-resistant (MDR) MRSA.
METHODS: The SpA protein sequence was retrieved from the UniProt database, with various bioinfor

BACKGROUND: Methicillin-resistant Staphylococcus remains a serious problem in the treatment of periprosthetic joint infection (PJI). Higher failure rates were reported when vancomycin was used in 2-stage exchange arthroplasty. Therefore a better therapeutic drug is needed to treat PJI caused by methicillin-resistant organisms. The purpose of the study was to evaluate the safety and efficacy of daptomycin when administered in bone cement combined with systemic use for methicillin-resistant Staphylococci PJI.
METHODS: We conducted a retrospective study from January 2010 to December 2012. Twenty

In case of periprosthetic joint infections, the antibiotic treatment administered intraoperatively entails consequences on bacterial ecology with potential secondary effects. This study evaluates the rapid detection of methicillin-resistant staphylococci (MRS) by Xpert® technology directly on intraoperative samples. Xpert® technology was compared to conventional culture for 104 clinical specimens performed on 30 patients. The performance of the test expressed in terms of sensitivity, specificity, positive predictive value, and negative predictive value was, respectively, 87.1%, 100%, 100%, and
