Estimating the initial relative infection rate for a stochastic epidemic model.

Consider the problem of making inference about the initial relative infection rate of a stochastic epidemic model. A relatively complete analysis of infectious disease data is possible when it is assumed that the latent and infectious periods are non-random. Here two related martingale-based techniques are used to derive estimates and associated standard errors for the initial relative infection rate. The first technique requires complete information on the epidemic, the second only the total number of people who were infected and the population size. Explicit expressions for the estimates are

Radiation risk to patients from nuclear medicine in Poland (1981).

This paper presents the method and results of an estimation of radiation risk to the patients subjected to in vivo diagnostic radio-isotopic procedures in Poland in 1981. The qualitative and quantitative structure of these examinations is also presented. The estimation of an expected number of the stochastic effects caused by the internal exposure to ionizing radiation from the administered radionuclides have been performed for the patients according to ICRP recommendations. The conclusion was that 33 fatal radiation induced cancers, caused mainly by thyroid procedures with 131I, may be expect

Dynamic analysis of an HIV stochastic time-delay differential equations incorporating two infection pathways and CTL immune response.

We propose a five-dimensional stochastic time-delay differential equation model that includes virus-to-cell infection, silent and active cell-cell transmissions and CTL immune response. Using mathematical methods, the non-degenerate five-dimensional stochastic delay differential equation model is transformed into a degenerate eight-dimensional stochastic differential equation model. The existence of a unique global positive solution is demonstrated. Then, by establishing a suitable Lyapunov function, we obtain the existence of a stationary Markov process when the stochastic CTL-activated repro

Novel insights for a nonlinear deterministic-stochastic class of fractional-order Lassa fever model with varying kernels.

Lassa fever is a hemorrhagic virus infection that is usually spread by rodents. It is a fatal infection that is prevalent in certain West African countries. We created an analytical deterministic-stochastic framework for the epidemics of Lassa fever employing a collection of ordinary differential equations with nonlinear solutions to identify the influence of propagation processes on infected development in individuals and rodents, which include channels that are commonly overlooked, such as ecological emergent and aerosol pathways. The findings shed light on the role of both immediate and sub

BRAINCELL modelling platform for stochastic nanoscale organisation and dynamic extracellular signalling among neurons and glia.

Biophysical cell models have been central to understanding signal processing in brain cells and their networks, yet important limitations remain. First, the rich repertoire of nanoscale structures, such as dendritic spines and thin astrocyte processes, has been difficult to incorporate into whole-cell models because of their number and complexity. BRAINCELL addresses this by generating stochastic populations of morphological and physiological features constrained by empirical statistics. Second, brain-cell activity depends on dynamic interactions with the extracellular environment, traditional

Optimising pandemic response through vaccination strategies using neural networks.

Epidemic risk assessment poses inherent challenges, with traditional approaches often failing to balance health outcomes and economic constraints. This paper presents a data-driven decision support tool that models epidemiological dynamics and optimises vaccination strategies to control disease spread while minimising economic losses. The proposed economic-epidemiological framework comprises three phases: modelling, optimising, and analysing. First, a stochastic SVEI3RD compartmental model with eight state variables captures epidemic dynamics, stratifying infections by severity to enable detai

Multivariate measures of critical transitions in a bistable gene regulatory network.

Developing concise gene regulatory networks that accurately encode cellular decision-making processes remains a key area of research across biological dynamics. Here, this challenge is addressed by investigating the behavior of a minimal gene regulatory network comprising three transcription factors within deterministic and stochastic frameworks. The components of the network interact through self-activation of a single transcription factor, mutual inhibition between a pair, and additional cross-inhibition exerted by the third. Following a brief mathematical analysis of the deterministic model

Optimal control for stochastic neural oscillators.

This study develops an event-based, energy-efficient control strategy for desynchronizing coupled neuronal networks using optimal control theory. Inspired by phase resetting techniques in Parkinson's disease treatment, we incorporate stochasticity of the system's dynamics into deterministic models to address neural system intrinsic noise. We use an advanced computational solver for nonlinear stochastic partial differential equations to solve the stochastic Hamilton-Jacobi-Bellman equation via level set methods for a single neuron model; this allows us to find control inputs which drive the dyn

Analysis of drug-resistant tuberculosis transmission dynamics in China using fractional stochastic model.

This study investigates the dynamics of the drug-resistant tuberculosis model through a fractional stochastic modeling framework. The model employs fractional-order derivatives to capture the memory effects in disease transmission, while Brownian motion is introduced to represent the random disturbances, thereby providing a more realistic description of the disease dynamics. First, a fractional deterministic model based on the Atangana-Baleanu-Caputo derivative was developed, and its optimal parameter values were obtained from the actual data from the case of drug-resistant tuberculosis in Chi

Superspreading events in the transmission dynamics of SARS-CoV-2: Opportunities for interventions and control.

Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), the etiological agent of the Coronavirus Disease 2019 (COVID-19) disease, has moved rapidly around the globe, infecting millions and killing hundreds of thousands. The basic reproduction number, which has been widely used-appropriately and less appropriately-to characterize the transmissibility of the virus, hides the fact that transmission is stochastic, often dominated by a small number of individuals, and heavily influenced by superspreading events (SSEs). The distinct transmission features of SARS-CoV-2, e.g., high stochasticity

Extinction pathways and outbreak vulnerability in a stochastic Ebola model.

A zoonotic disease is a disease that can be passed from animals to humans. Zoonotic viruses may adapt to a human host eventually becoming endemic in humans, but before doing so punctuated outbreaks of the zoonotic virus may be observed. The Ebola virus disease (EVD) is an example of such a disease. The animal population in which the disease agent is able to reproduce in sufficient number to be able to transmit to a susceptible human host is called a reservoir. There is little work devoted to understanding stochastic population dynamics in the presence of a reservoir, specifically the phenomena

Hamiltonian Analysis of Subcritical Stochastic Epidemic Dynamics.

We extend a technique of approximation of the long-term behavior of a supercritical stochastic epidemic model, using the WKB approximation and a Hamiltonian phase space, to the subcritical case. The limiting behavior of the model and approximation are qualitatively different in the subcritical case, requiring a novel analysis of the limiting behavior of the Hamiltonian system away from its deterministic subsystem. This yields a novel, general technique of approximation of the quasistationary distribution of stochastic epidemic and birth-death models and may lead to techniques for analysis of t

Comparative Risks of Cancer from Drywall Finishing Based on Stochastic Modeling of Cumulative Exposures to Respirable Dusts and Chrysotile Asbestos Fibers.

Sanding joint compounds is a dusty activity and exposures are not well characterized. Until the mid 1970s, asbestos-containing joint compounds were used by some people such that sanding could emit dust and asbestos fibers. We estimated the distribution of 8-h TWA concentrations and cumulative exposures to respirable dusts and chrysotile asbestos fibers for four worker groups: (1) drywall specialists, (2) generalists, (3) tradespersons who are bystanders to drywall finishing, and (4) do-it-yourselfers (DIYers). Data collected through a survey of experienced contractors, direct field observation

First pilot trial of the STAR-Liege protocol for tight glycemic control in critically ill patients.

Tight glycemic control (TGC) has shown benefits in ICU patients, but been difficult to achieve consistently due to inter- and intra- patient variability that requires more adaptive, patient-specific solutions. STAR (Stochastic TARgeted) is a flexible model-based TGC framework accounting for patient variability with a stochastically derived maximum 5% risk of blood glucose (BG) below 72 mg/dL. This research describes the first clinical pilot trial of the STAR approach and the post-trial analysis of the models and methods that underpin the protocol. The STAR framework works with clinically speci

A new type of stochastic dependence revealed in gene expression data.

Modern methods of microarray data analysis are biased towards selecting those genes that display the most pronounced differential expression. The magnitude of differential expression does not necessarily indicate biological significance and other criteria are needed to supplement the information on differential expression. Three large sets of microarray data on childhood leukemia were analyzed by an original method introduced in this paper. A new type of stochastic dependence between expression levels in gene pairs was deciphered by our analysis. This modulation-like unidirectional dependence

A stochastic model for the aggregation of Onchocerca volvulus in nodules.

A model is presented which describes the aggregation of female Onchocerca volvulus in nodules and their distribution in the human population. The basic model is based on a single parameter, the formation probability q, which represents the probability with which incoming larvae form a new nodule. This parameter describes parasite behaviour which cannot easily be recognized in available data without modelling. The estimate for the average formation probability of muq = 0.39 suggests an attraction of the invading infective larvae to already existing nodules or resident worms with probability 0.6

Environmental tobacco smoke deposition in the human respiratory tract: differences between experimental and theoretical approaches.

Total deposition of environmental tobacco smoke (ETS) particles was measured in a group of 15 nonsmokers who inhaled ETS of count median diameter of 0.2 microm and geometric standard deviation of 1.6. A total deposition of 56.0 +/- 15.9% was observed for nasal breathing and 48.7 +/- 11.5% for oral breathing. In contrast, our stochastic deposition model predicted a total deposition of only 17.9% (male) and 15.7% (female) for nose breathing, and 13.4% (male) and 10.7% (female) for mouth breathing, if based on standard breathing conditions. Consideration of individual lung volumes and breathing p

Medical aspects of the hazardous waste problem.

Although no one knows exactly how much toxic material continues to be released into our environment, most observers believe the amount is substantial. In the last few years, in the state of Massachusetts alone, 22 communities have had their municipal water supplies seriously compromised by chemical contamination, (1) causing alarm and dismay among water users. Nation-wide, public concern has reached the point that in some opinion polls, hazardous waste ranks second only behind inflation as a cause of serious worry. Despite widespread anxiety, shared by public health officials, few studies have

The effect of demographic stochasticity on Zika virus transmission dynamics: Probability of disease extinction, sensitivity analysis, and mean first passage time.

Viral infections spread by mosquitoes are a growing threat to human health and welfare. Zika virus (ZIKV) is one of them and has become a global worry, particularly for women who are pregnant. To study ZIKV dynamics in the presence of demographic stochasticity, we consider an established ZIKV transmission model that takes into consideration the disease transmission from human to mosquito, mosquito to human, and human to human. In this study, we look at the local stability of the disease-free and endemic equilibriums. By conducting the sensitivity analysis both locally and globally, we assess t

Stochasticity and the limits to confidence when estimating R0 of Ebola and other emerging infectious diseases.

Dynamic models - often deterministic in nature - were used to estimate the basic reproductive number, R0, of the 2014-5 Ebola virus disease (EVD) epidemic outbreak in West Africa. Estimates of R0 were then used to project the likelihood for large outbreak sizes, e.g., exceeding hundreds of thousands of cases. Yet fitting deterministic models can lead to over-confidence in the confidence intervals of the fitted R0, and, in turn, the type and scope of necessary interventions. In this manuscript we propose a hybrid stochastic-deterministic method to estimate R0 and associated confidence intervals
