Streptococcus suis serotype 2 (SS2) has been reported to be a highly invasive pathogen in swine and a zoonotic agent for humans. Although many bacterial virulence factors have been identified, our an insightful understanding of SS2 pathogenicity is lacking. The gene nadR, encoding nicotinamide-nucleotide adenylyltransferase, was first reported as a regulator and transporter of the intracellular NAD synthesis pathway in Salmonella typhimurium. In this study, we constructed a mutant strain of nadR (ΔnadR) to test the phenotypic and virulence variations between the deletion mutant and the wild-ty

Deafness is a common and often permanent neurological sequel of Streptococcus (S.) suis meningitis. Suppurative labyrinthitis, rather than direct auditory nerve infection, has been found to be the site responsible for deafness. Neuroimaging is important to localise the site involved in hearing loss and to assess the feasibility of a cochlear implantation. S. suis is very sensitive to penicillin. Although a relapse of S. suis meningitis is uncommon, it can occur despite an adequate duration of appropriate antibiotic therapy. We describe a patient with S. suis meningitis, who developed permanent

Infections caused by Streptococcus suis are considered a global problem in the swine industry. In this animal species, S. suis is associated with septicemia, meningitis, endocarditis, arthritis and, occasionally, other infections. Moreover, it is an agent of zoonosis that afflicts people in close contact with infected pigs or pork-derived products. Although sporadic cases of S. suis infection in humans have been reported, a large outbreak due to S. suis serotype 2 emerged in the summer of 2005 in Sichuan, China. A similar outbreak was observed in another Chinese province in 1998. Symptoms repo

Seventy clinically normal 13-day-old crossbred pigs from 10 litters from a Streptococcus suis-infected herd were randomly assigned by litter and weight to 7 groups of 10 pigs each to determine whether different antibiotic regimens would eliminate the tonsillar carrier state of S. suis. Six antimicrobial regimens were tested: penicillin intramuscularly (IM) once daily (s.i.d.) for 3 consecutive days; penicillin IM s.i.d. for 5 consecutive days; ampicillin IM s.i.d. for 5 consecutive days; ampicillin per os s.i.d. for 5 consecutive days; ampicillin intranasally s.i.d. for 5 consecutive days; and

Streptococcus pasteurianus is an opportunistic pathogen affecting various animals and is an underreported zoonotic threat. It is also a causative agent of swine streptococcosis and can be co-detected with Streptococcus suis, another significant pig and zoonotic pathogen. However, the dynamics of co-colonization between these pathogens, along with the genomic features and antibiotic resistance profiles of S. pasteurianus, remain poorly understood. In this study, we developed a multiplex PCR (mPCR) assay to detect S. pasteurianus and S. suis in 827 tonsil samples from healthy pigs, with 81 sampl

Streptococcus suis serotype 2 (S. suis 2) is an important zoonotic pathogen, with its capsular polysaccharide (CPS) serving as a key virulence factor. However, the regulatory mechanisms of CPS biosynthesis remain unclear. This study aimed to prepare and characterize the capsular synthesis regulatory factor Cps2D from S. suis 2 and investigate its enzymatic activity. Bioinformatics analysis using ProtParam and Alpha Fold revealed Cps2D as a hydrophilic, stable protein of 242 amino acids, with a molecular weight of 28 kDa and a pI of 8.69. It was annotated as a tyrosine protein phosphatase, prim

BACKGROUND: Antimicrobial resistance in Streptococcus suis represents an increasing global health concern, particularly in Thailand, where human and swine infections remain common and severe. METHODS: A descriptive study was conducted in northeastern Thailand from January 2020 to December 2024 to examine the association between clinical characteristics and penicillin susceptibility (susceptible, intermediate, resistant) among S. suis isolates. In vitro antibiotic interactions were assessed by checkerboard microdilution in 23 drug-resistant isolates. RESULTS: Among 373 culture-confirmed S. suis

A high prevalence of acquired bleomycin resistance has been reported in both clinical and non-clinical settings. Streptococcus suis is a zoonotic swine pathogen capable of causing a spectrum of clinical disease outcomes in both pigs and humans and contributes to significant economic losses to the swine industry worldwide. A recent report evaluating the genomic diversity of S. suis isolates obtained from within the U.S. identified a ble gene predicted to encode a bleomycin resistance protein in all isolates evaluated. The goal of this study was to compare the amino acid sequence similarity amon

The phagolysosomes of macrophages play a crucial role in eradicating pathogenic microorganisms, but bacteria have evolved sophisticated mechanisms to survive in the acidic environment of phagolysosomes, leading to host infection and subsequent dissemination. However, it is largely unknown how bacteria sense the extracellular stimuli and regulate their acid tolerance capacity to resist the killing by host immune cells. Here, we report the new substrate FadR of the serine/threonine kinase (STK) in Streptococcus suis serotype 2 (SS2) and demonstrate that the phosphorylation site is Thr230. Notabl

The increasing prevalence of antimicrobial resistance against zoonotic bacteria, including Streptococcus (S.) suis, highlights the need for new therapeutical strategies, including the repositioning of drugs. In this study, susceptibilities of bacterial isolates were tested toward ten different 3-amidinophenyalanine (Phe(3-Am)) derivatives via determination of minimum inhibitory concentration (MIC) values. Some of these protease inhibitors, like compounds MI-432, MI-471, and MI-476, showed excellent antibacterial effects against S. suis. Their drug interaction potential was investigated using h

Streptococcus suis is a porcine and zoonotic pathogen that causes severe systemic infection in humans and pigs. The treatment of S. suis infection relies on antibiotics; however, antimicrobial resistance (AMR) is an urgent global problem, pushing research attention on the surveillance of antibiotic-resistant S. suis to the fore. This study investigated the antimicrobial susceptibility of 246 S. suis strains isolated from diseased pigs in Thailand from 2018-2020. The major sources of S. suis strains were lung and brain tissues. PCR-based serotyping demonstrated that the most abundant serotype w

Streptococcus suis is an emerging zoonotic pathogen causing streptococcal toxic shock like syndrome (STSLS), meningitis, septicemia, and even sudden death in human and pigs. Serious septicemia indicates this bacterium can evade the host complement surveillance. In our previous study, a functionally unknown protein SntA of S. suis has been identified as a heme-binding protein, and contributes to virulence in pigs. SntA can interact with the host antioxidant protein AOP2 and consequently inhibit its antioxidant activity. In the present study, SntA is identified as a cell wall anchored protein th

Streptococcus suis is an emerging zoonotic pathogen causing severe infections in pigs and humans. Thirty-three serotypes of S. suis have been identified using serum agglutination. The capsular polysaccharides synthesis (cps) locus is usually conserved among different strains of the same serotype. The cps loci of 15 serotypes have been sequenced, while the loci of the other serotypes remain unknown. In the present study, two to six serotype-specific genes of each of eight serotypes, i.e., serotypes 3, 4, 5, 8, 10, 19, 23, and 25, were identified using cross-hybridization with 93 nucleic acid pr

Streptococcus suis is an important pathogen for both swine and humans. In this study, we genotyped 105 S. suis isolates from porcine endocarditis in East Japan on the basis of profiles of capsular serotype-specific, virulence-associated and pilus-associated genes. The most common genotype was cps2J/mrp+/epf-/sly-/sbp2-/sep1-/sgp1+ (76.19%), followed by nt(non-typeable)/mrp+/epf-/sly-/sbp2-/sep1-/sgp1+ (7.62%) and cps2J/mrp+/epf+/sly+/sbp2+/sep1-/sgp1- (7.62%). The representative isolates of mrp+/epf-/sly-/sbp2-/sep1-/sgp1+ were classified into ST28 complex, a clonal complex previously referred

The antimicrobial susceptibility of 151 clinical Streptococcus suis strains isolated from diseased pigs in Spain was determined by a microdilution method. Isolates were mostly susceptible to beta-lactam antimicrobials, aminoglycosides, enrofloxacin, novobiocin and spectinomycin. More than 87% of the S. suis isolates were resistant to tetracyclines, sulphonamides, macrolides and clindamycin. Strains of serotype 9 were significantly more resistant than strains of serotype 2 (P<0.05) to tylosin (94% versus 77%) and clindamycin (94% versus 64%). Eighty-seven percent of the S. suis isolates were re
