Pancreatic tumors frequently develop in nutrient-poor microenvironments due to impaired perfusion, forcing cancer cells to engage in metabolic adaptation for survival. This study identified the upregulation of transketolase-like 1 (TKTL1) as a nutrient-stress-responsive factor associated with metabolic adaptation of pancreatic cancer cells under amino-acid starvation, particularly glutamine scarcity. In particular, we found that TKTL1 induction was specifically observed in cancer cells and not in normal pancreatic ductal cells. Functional assays showed that TKTL1 overexpression increased proli

Three groups of Sprague-Dawley rats were fed a thiamine deficient diet, which was supplemented by daily subcutaneous injections of a minimum requirement of thiamine, and treated with lead(II) acetate in different molar ratios to thiamine (1:1, 2:1, 10:1) for 5 and 9 months, respectively. The prolonged administration of lead(II) acetate decreases the thiamine level in lead-treated rats and diminishes the enzymatic activity of pyruvate dehydrogenase as well as that of transketolase. The thiamine level in the liver decreased by 30 to 40% compared with a reference group and the activity of the ery

Thiamine helps transketolase in removing toxic metabolites, counteracting high glucose-induced damage in microvascular cells, and progression of diabetic retinopathy/nephropathy in diabetic animals. Diabetic subjects show reduced thiamine levels. Hyperglycemia and reduced thiamine availability concur in impairing thiamine transport inside the blood-retinal barrier, with thiamine transporter-2 (THTR2) primarily involved. Here, we examined the behavior of thiamine transporter-1 (THTR1), THTR2, and their transcription factor Sp1 in response to high glucose and altered thiamine availability in ren

In thirteen preterm infants, 45 full-term infants, and their mothers thiamin was analyzed in plasma from maternal veins, umbilical arteries, umbilical veins, and placental tissue. The blood flow in the umbilical veins was determined by pulsed Doppler ultrasonography. Thiamin-dependent transketolase was measured in erythrocytes from full-term infants and their mothers. Plasma thiamin concentrations in umbilical veins from preterm infants (227.0 +/- 85.0 nmol/L) and full-term infants (121.3 +/- 103.3 nmol/L) were seven times greater than maternal concentrations (p < 0.005). Three family members are reported with functional symptoms considered to be caused by intracellular deficiency of thiamin. Persistence of desaturation of erythrocyte transketolase in the face of megadose thiamin hydrochloride (THCl), accompanied by a balanced multivitamin and mineral formula, suggested a familial thiamin dependency state. Each of three individuals responded clinically to the administration of thiamin tetrahydrofurfuryl disulfide (TTFD), and erythrocyte transketolase (TKA) became fully saturated with thiamin pyrophosphate (TPP). Dysautonomic symptoms observed are compared with

Thiaminase from bicuspid mollusc, after a single administration into mice (25 or 50 un, subcutaneously), caused a decrease in the activity of transketolase and alpha-ketoglutaric acid dehydrogenase in the animal tissues within 3 hrs. The maximal antivitaminous effect of thiaminase was manifested within 2-5 days and slightly decreased within 7 days (at a dose of 25 un). Amount of thiamin was decreased in mice liver tissue from 6.54+/-0.36 mcg/g to 4.84+/-0.23 mcg/g within the second day after the thiaminase administration. C-6 glioma and C-1300 neuroblastoma cells were cultured in thiamine deficient and control media. Thiamine levels, transketolase and pyruvate decarboxylase activities, and high energy phosphate metabolites were all measured in deficient and control cells. Thiamine levels in the deficient cells were found to be below the level of detectability. Pyruvate decarboxylase activity was more susceptible to thiamine deficiency in both cell lines than transketolase. In spite of the large decrease in pyruvate decarboxylase activity, high energy phosphate metabolites were not decreased in either cell line.

Three shipments of sheep being exported live by sea were examined to determine their thiamine status. Measurements were made of thiamine concentration in liver and ruminal contents, transketolase activity in erythrocytes and thiaminase activity in ruminal liquor. Sheep that died or were clinically ill and euthanased had significantly lower hepatic and ruminal thiamine concentrations than clinically healthy control sheep. A high proportion had thiamine concentrations comparable to those found in sheep that die with polioencephalomalacia. Thiamine concentrations decreased with increasing time th

In the course of human adaptation to the climatic and geographic factors, there was a stable reduction of thiamine concentration in blood and urine. The activity of red cell transketolase did not change or slightly rose. The changes in TDF effect were insignificant thus pointing to the absence of hypovitaminosis. Intensive vitamin therapy did not lead to an essential increase in thiamine blood concentration which made up 2.0--2.5 micrograms % and as usual remained lower than generally accepted rates. The main amount of thiamine was excreted with urine. TDF effect did not exceed 4--8%. The resu

Thiamine deficiency is a continuing problem leading to beriberi and Wernicke's encephalopathy. The symptoms of thiamine deficiency develop in the heart, brain and neuronal tissue. Yet, it is unclear how rapid thiamine deficiency develops and which organs are prone to development of thiamine deficiency. We investigated these issues in a thiamine deficient animal model. Twenty-four male Lewis rats were fed a thiamine deficient diet, which contained 0.04% of normal thiamine intake. Six control rats were fed 200 μg of thiamine per day. Every week a group of six rats on the thiamine-deficient diet

The effect of 5-fluorouracil on the thiamin status of normal female adult rats has been investigated. Pre-treatment of the animals with the cytotoxic drug daily for 3 successive days resulted in a significant decrease in hepatic concentrations of thiamin concomitant with a decrease in thiamin-dependent transketolase enzyme activity and an increase in thiamin-pyrophosphate-(TPP-)stimulating effect in whole blood when compared with those of pair-fed control animals. The TPP effect of transketolase enzyme activity was also increased by 5-fluorouracil in vitro. Furthermore, the treatment with 5-fl

This study was conducted to examine the effect of dietary thiamin, ranging from deficient to excessive supplies, on thiamin status of lactating rats and their offspring, and the thiamin level in milk. Therefore, after parturition, rat dams were divided into eight groups of 10 each, and were fed diets with 0, 2, 4, 6, 7, 40, 350 and 3500 mg/kg thiamin over a total of 13 days during lactation. Milk for determining the thiamin concentration was obtained from day 6 and 13 of lactation. At day 14 of lactation rat dams and their offspring were used to ascertain the thiamin status including transketo

The relative roles of alcohol per se, thiamine deficiency, and liver disease in the pathogenesis of alcohol-related brain damage have not been fully elucidated. In particular, the extent to which alterations of brain thiamine metabolism contribute to cognitive dysfunction in alcoholism in the absence of Wernicke's encephalopathy has not been established. In the present study, thiamine diphosphate-dependent enzymes were measured using standard spectrophotometric techniques in homogenates of brain tissue obtained at autopsy from eight alcoholic patients, all of whom died in hepatic coma without

Turnover of [14C]thiamin was studied in mice with Ehrlich ascites carcinoma fed a thiamin-deficient diet and injected with 5 or 2 micrograms/mouse of the labeled vitamin. The process of conversion of [14C]thiamin to thiamin pyrophosphate (TPP) was monitored by measuring the activities of transketolase, pyruvate dehydrogenase, and oxoglutarate dehydrogenase. The amount of coenzyme-unsaturated apotransketolase was assessed by measuring the TPP effect--determining transketolase activity with and without the addition of TPP in vitro. Tumor growth was accompanied by thiamin deficiency, manifested i

The erythrocytes transketolase activity and the TDP-effect were determined in persons residing in the regions beyond the polar circle and these demonstrated a moderate thiamine deficiency. The daily thiamine intake with food in the examined did not exceed 1.5 mg, the thiamine-diphosphate of the epoenzyme amounting to 90 per cent. Following introduction of various pharmacological preparations of vitamin B1 for a space of 2 weeks a somewhat greater transketolase activity and a diminution of the TDP-effect were observed. Administration of cocarboxylase and vitamin B1 with Mg and gelatin more favo

Mice were made deficient by thiamin deprivation. Thiamin pyrophosphokinase and thiamin pyrophosphatase activities were measured, and their changes were compared to transketolase (TK), pyruvate and alpha-ketoglutarate dehydrogenase (DG) activities and thiamin pyrophosphate (TPP) level in the same tissues. Thiamin pyrophosphokinase activity was increased on day 10 of thiamin deficiency and remained unchanged within the next period of investigation. Thiamin pyrophosphatase activity decreased after 5 days and was enhanced in 10, 15 and 20 days of deficiency. This fact may be related to the regulat

To clarify the effect of polychlorinated biphenyls (PCB), and dichlorodiphenyltrichloroethane (DDT) and other organochloride compounds on thiamine metabolism, rats were separated into six groups and placed on different admixed diets. Group 1 was fed a normal diet; group 2, a PCB-supplemented diet; group 3, a DDT-supplemented diet; group 4, a benzenehexachloride-supplemented (BHC) diet. After 50 days of these dietary regimens all animals were sacrificed and various studies were done on the excised tissues. In PCB-treated rats, thiamine levels in blood, liver and sciatic nerve decreased, transke

50 chronic alcoholics reporting to the medical emergency ward of Basle University Hospital with alcohol-related illness were examined with respect to thiamine nutritional status by means of the transketolase activation test of erythrocytes (ETK). 46% of the chronic alcoholics, compared to only 2% of the control population (1152 healthy adults), had transketolase activation quotients indicating a strong probability of thiamine deficiency (alphaETK greater than 1.25). The most important symptoms associated with the biochemical parameters of thiamine deficiency were: anemia, pathologic liver func
