Animal performance and biochemical parameters are sex-dependent in peripubertal rats exposed to deoxynivalenol.

Deoxynivalenol (DON), a mycotoxin produced mainly by Fusarium graminearum and F. culmorum commonly contaminates food commodities across the globe. Due to this, exposure to DON might pose potential health hazards to humans and animals. Biological factors like sex and age can influence the toxicity of DON. However, in toxicological studies involving DON, the sex and age-dependent response has been often overlooked. Thereby, the objective of this study was to evaluate if sex differences are evident in DON's systemic effects in peripubertal rats. Juvenile animals (n = 24) with 28 days postnatal da

Effects of Fusarium metabolites beauvericin and enniatins alone or in mixture with deoxynivalenol on weaning piglets.

The impact of the Fusarium-derived metabolites beauvericin, enniatin B and B1 (EB) alone or in combination with deoxynivalenol (DON) was investigated in 28-29 days old weaning piglets over a time period of 14 days. The co-application of EB and DON (EB + DON) led to a significant decrease in the weight gain of the animals. Liver enzyme activities in plasma were significantly decreased at day 14 in piglets receiving the EB + DON-containing diet compared to piglets receiving the control diet. All mycotoxin-contaminated diets led to moderate to severe histological lesions in the jejunum, the liver

Comparative Microbial Conversion of Deoxynivalenol and Acetylated Deoxynivalenol in Different Parts of the Chicken Intestine as Detected In Vitro and Translated to the In Vivo Situation.

To assess potential differences in the intestinal microbial metabolism of deoxynivalenol (DON) and its acetylated forms 3-Ac-DON and 15-Ac-DON, in vitro anaerobic incubations with intestinal contents from chickens were conducted. Quantitative microbiota characterization was obtained by 16S rRNA sequencing. The data showed substantial differences in the level of different toxin conversions by the microbiota from the different intestinal segments. The transformation rate of DON to its metabolite DOM-1 decreased in the order of cecum > ileum > jejunum, and caecum contents could completely transfo

Deoxynivalenol-induced cell apoptosis monitoring using a cytochrome c-specific fluorescent probe based on a photoinduced electron transfer reaction.

Deoxynivalenol (DON) is considered a mycotoxin that is toxic to the agricultural environment and human body. It is necessary to study the pathophysiological mechanism of DON toxicity at the cellular level. Cytochrome c (Cyt c), as an important biomarker of DON-induced apoptosis that may lead to a bipartite 'point-of-no return' event, is of great significance to be detected using cell imaging. Herein, we synthesized a DON-deactivated emission fluorescent probe, the molecularly imprinted polymer-coated quantum dots (CdTe@MIP), for monitoring the Cyt c level with a photoinduced electron transfer s

Deoxynivalenol in the Diet Impairs Bone Mineralization in Broiler Chickens.

Deoxynivalenol (DON) is one of the most abundant and important trichothecene mycotoxins produced by Fusarium species. In chickens, DON intake causes feed refusal, impairs performance, gut barrier function, and immunity, and raises oxidative stress. To determine the effect of DON on bone mineralization and serum calcium and phosphorus, 80 newly-hatched chickens were fed 4 diets with 0, 2.5, 5, and 10 mg DON/kg feed in this pilot study. In week 5, chickens were euthanized, femur and tibiotarsus bones were separated from the meat, and after incineration ash composition, as well as serum calcium a

Sick building syndrome. III. Stachybotrys chartarum.

Increasingly, physicians are being asked to evaluate patients with putative environmentally associated illnesses. These can include a variety of problems, including infectious illnesses (Legionnaire's disease), chemical exposure in the workplace, and sick building syndromes. The latter has been an issue particularly in asthma because of the association of mold and increased bronchial responsiveness. Recently, attention has been focused on the mold Stachybotrys in human disease. Stachybotrys was first identified more than 60 years ago following an epidemic of stomatitis, rhinitis, conjunctiviti

Evolutionary Adaptability Coupled with Computation-Driven Engineering for Thermostability Enhancement of a Deoxynivalenol-Detoxifying Fusion Enzyme.

Deoxynivalenol (DON), a trichothecene mycotoxin commonly found in cereal grains and their derived products, poses significant risks to human and animal health. In previous work, a fusion enzyme composed of the dehydrogenase DADH and the aldo-keto reductase AKR13B3 was engineered to convert DON into the non-toxic 3-epi-DON in a single step. However, the poor thermal stability of this fusion enzyme limited its industrial application. In this study, EVcouplings and the GRAPE-WEB platform were utilized to identify key amino acid residues governing the thermal stability of the fusion enzyme AKR13B3

T-2 toxin-induced testicular impairment by triggering oxidative stress and ferroptosis.

T-2 toxin is a trichothecene mycotoxin of significant danger to humans and animals. Its impact on reproductive toxicity is attributed to oxidative stress, which ultimately leads to cell death. Ferroptosis is a programmed cell death that characterized by lipid peroxidation. This study aimed to investigate the toxic effects of T-2 toxin on mouse testis and the potential mechanism of T-2 toxin-induced ferroptosis. T-2 toxin significantly altered the morphology of the testis and decreased testosterone level, sperm concentration, and increased sperm malformation rate, as well as induced oxidative d

Application of PEG-CdSe@ZnS quantum dots for ROS imaging and evaluation of deoxynivalenol-mediated oxidative stress in living cells.

Deoxynivalenol (DON), a trichothecene mycotoxin, has attracted global attention due to its prevalence and substantial effects on animal and human health. DON induces the upregulation of intracellular reactive oxygen species (ROS) by disrupting the normal mitochondrial functionality, which causes oxidative stress, cell apoptosis, and even severe disorders. The aim of present work is to develop a simple, convenient, and in situ method for monitoring ROS and evaluating DON-mediated oxidative stress. Herein, polyethylene glycol-modified CdSe@ZnS quantum dots (QDs) were employed as simple and conve

Haemorrhagic syndrome of cattle associated with the feeding of sweet vernal (Anthoxanthum odoratum) hay containing dicoumarol.

An outbreak of a haemorrhagic diathesis in cattle fed home produced hay is described. A similar syndrome was reproduced experimentally in calves by feeding them the hay. The experimental disease was characterised by increased prothrombin and partial thromboplastin times while the leucocyte and erythrocyte counts remained normal until the terminal haemorrhage. The calves ate well and grew well until the rapid onset of progressive weakness, stiff gait, mucosal pallor, tachycardia, tachypnoea and haematomata ending in sudden death. The absence of blood coagulation was seen at necropsy while petec

Protective effects of baicalin against deoxynivalenol-induced oxidative and inflammatory damage in chicken-derived hepatic 3D cell cultures.

Deoxynivalenol (DON) is a trichothecene mycotoxin often contaminating grains used in poultry feed production and causing several adverse effects in farm animals. Therefore, it is important to investigate compounds that can be potential candidates to mitigate these effects, such as baicalin. The effects of DON and baicalin were investigated in chicken-derived 3D hepatic cell cultures, and cell viability, LDH activity, oxidative parameters (NRF-2, 8-OHdG) and inflammatory parameters (IL-6, IL-8, IFN-γ) were monitored for 24 and 48 h. Our results suggest that DON reduced cellular metabolic activi

Betulinic acid attenuates T-2 toxin-induced lung injury by activating Nrf2 signaling pathway and inhibiting MAPK/NF-κB signaling pathway.

T-2 toxin, a type-A trichothecene mycotoxin, exists ubiquitously in mildewed foods and feeds. Betulinic acid (BA), a pentacyclic triterpenoid derived from plants, has the effect of relieving inflammation and oxidative stress. The purpose of this study was to investigate whether BA mitigates lung impairment caused by T-2 toxin and elucidate the underlying mechanism. The results indicated that T-2 toxin triggered the inflammatory cell infiltration, morphological alterations and cell apoptosis in the lungs. It is gratifying that BA ameliorated T-2 toxin-caused lung injury. The protein expression

Direct activation of ribosome-associated double-stranded RNA-dependent protein kinase (PKR) by deoxynivalenol, anisomycin and ricin: a new model for ribotoxic stress response induction.

Double-stranded RNA (dsRNA)-activated protein kinase (PKR) is a critical upstream mediator of the ribotoxic stress response (RSR) to the trichothecene deoxynivalenol (DON) and other translational inhibitors. Here, we employed HeLa cell lysates to: (1) characterize PKR's interactions with the ribosome and ribosomal RNA (rRNA); (2) demonstrate cell-free activation of ribosomal-associated PKR and (3) integrate these findings in a unified model for RSR. Robust PKR-dependent RSR was initially confirmed in intact cells. PKR basally associated with 40S, 60S, 80S and polysome fractions at molar ratios

Organ-specific responses of age-dependent vulnerability to deoxynivalenol in female mice.

The responses underlying heightened sensitivity of young populations to deoxynivalenol (DON) toxicity remain uncharacterized in translational murine models. This study aimed to fill this gap by providing the first systematic, multi-organ (intestine, liver, and kidney) comparison of DON toxicity, contrasting the responses of prepubertal and adult mice. Prepubertal (21-day-old) and adult (65-day-old) Swiss mice (n = 10/group) were fed control or 10 mg/kg DON-contaminated diets for 15 or 28 days, respectively. Growth performance was monitored, and intestine, liver, and kidney were subsequently co

Enniatin B attenuates deoxynivalenol-induced toxicity in liver cells and modulates lipid metabolism-related pathways.

Mycotoxin contamination of food and feed is an increasing global health concern, particularly due to Fusarium toxins such as the regulated deoxynivalenol (DON) and the emerging enniatin B (ENNB). However, the combined mechanisms of action of these toxins remain poorly understood. This study aimed to assess their toxic effects, alone and in combination, using the HepaRG liver cell model. Toxicity, individually and in combination at concentrations equivalent to those used in single exposures (5:1 DON to ENNB ratio), was assessed in HepaRG cells. Several endpoints were investigated, including via

A novel mycotoxin-degrading enzyme complex can biodegrade AFB1, DON, and ZEN co-contamination in both in vitro and in vivo experiments.

A novel mycotoxin-degrading enzyme complex (MDE), developed via a microcapsule coating process and containing three degrading enzymes, exhibits the ability to biodegrade aflatoxin B1 (AFB1), deoxynivalenol (DON) and zearalenone (ZEN). This study aims to evaluate the efficacy of MDE against AFB1, DON, and ZEN through in vitro and in vivo experiments. In vitro simulated digestion experiments revealed that the MDE degraded the concentrations of AFB1, DON, and ZEN by 76.27 %, 74.04 %, and 60.77 %, respectively. In vivo experiments were conducted using 39 one-day-old male Cobb broilers allocated in

Lactiplantibacillus plantarum JM113 alleviates mitochondrial dysfunction induced by deoxynivalenol in the jejunum of broiler chickens.

Mitochondria are primary targets of deoxynivalenol (DON), which play a pivotal role in maintaining intestinal health. It has been suggested that Lactiplantibacillus plantarum JM113 (L. plantarum JM113) exhibits protective effects against the enterotoxicity of DON in broilers. However, the changes in mitochondrial homeostasis during this process remain unclear. A total of 144 one-day-old Arbor Acres broilers were randomly assigned to 3 groups, including the CON group (fed a basal diet and gavaged with 0.5 mL PBS), the DON group (supplemented with 5 mg/kg DON based on the CON group) and the DJ g

Deoxynivalenol triggers pyroptosis in mouse hippocampal neurons by activating oxidative stress and interfering with neurotransmitter release.

As a prevalent natural pollutant, deoxynivalenol (DON) exhibits high toxicity, affects multiple organs, can penetrate the blood-brain barrier and induce neurotoxicity, thereby threatening food safety. In this study, we investigated the effects of DON on hippocampal neuronal injuries using both in vivo and in vitro models to evaluate its potential risks. Results showed that subchronic exposure of DON exposure (500 ng/mL in cells and 1 mg/kg.bw in mice) increased excessive ROS production, activated the NF-κB pathway, and upregulated NLRP3 and ASC expression levels. Consequently, this triggered C

Deoxynivalenol Exposure Leads to Abnormal Renal Tubular Autophagy Flow.

Deoxynivalenol (DON) is one of the common mycotoxins contaminating feed. Renal tubular epithelial cells may experience oxidative stress as a result of consuming DON over time. However, little is known about the biological mechanisms through which DON results in kidney damage. This study employed proteomics, atomic force microscope (AFM), transmission electron microscope (TEM), and fluorescent probes, among other techniques, to reveal that DON exposure caused disturbances in renal autophagy flux both in vivo and in vitro. Mechanistically, this study found that DON exposure led to impaired forma
