Mouse models are frequently used to develop treatments for human cancer. However, the degree to which their tumor microenvironments (TMEs) are synonymously assembled is particularly poorly characterized. Through systematic immunoprofiling of 15 commonly used mouse models, we found that most murine TMEs recapitulate the composition of poorly infiltrated human tumors, extensively biased toward high macrophage densities. We discovered substantial species-specific biases of chemokine expression networks known to drive TMEs assembly, together with discoordinated frequencies of T and myeloid cell su

Immune checkpoint inhibitors (ICI) have improved outcomes for asymptomatic patients with melanoma brain metastases (MBM) but have delivered limited benefit in glioblastoma (GBM). Defining context-dependent features of ICI response is critical for tailoring effective therapeutic strategies and improving GBM patient outcomes. Multiparameter flow cytometry analyses of immune composition and activation status were performed on 110 tumor cell suspensions obtained from cavitron ultrasonic surgical aspirates (CUSA) specimens (n = 40), and surgically resected GBM (n = 21) and MBM (n = 49) tumors. GBM

Sézary syndrome is an aggressive leukemic variant of cutaneous T-cell lymphomas, characterized by erythroderma, lymphadenopathy, and peripheral blood involvement by CD4+ malignant T-cells. The pathogenesis of Sézary syndrome is not fully understood. However, the course of the disease is strongly influenced by the tumor microenvironment, which is altered by a combination of cytokines, chemokines, and growth factors. The crosstalk between malignant and reactive cells affects the immunologic response against tumor cells causing immune dysregulation. This review focuses on the interaction of malig

Erythropoietin (EPO), the primary cytokine of erythropoiesis, stimulates both proliferation and differentiation of erythroid progenitors and their maturation to red blood cells. Basal EPO levels maintain the optimum levels of circulating red blood cells. However, during hypoxia, EPO secretion and its expression is elevated drastically in renal interstitial fibroblasts, thereby increasing the number of erythroid progenitors and accelerating their differentiation to mature erythrocytes. A tight regulation of this pathway is therefore of paramount importance. The biological response to EPO is com

Ovarian cancer is the deadliest gynecological disease among U.S. women. Poor 5-year survival rates (<30%) are due to presentation of most women at diagnosis with advanced stage disease with widely disseminated intraperitoneal metastasis. However, when diagnosed before metastatic propagation the overall 5-year survival rate is >90%. Metastasizing tumor cells grow rapidly and aggressively attach to the mesothelium of all organs within the peritoneal cavity, including the parietal peritoneum and the omentum, producing secondary lesions. In this review, the involvement of mesothelin (MSLN) in the

The microenvironment of follicular lymphoma (FL) is composed of tumor-infiltrating CD8(+) T cells, follicular regulatory T cells, lymphoma-associated macrophages and mast cells, follicular helper T cells, follicular dendritic cells, and follicular reticular cells, all of which have been reported to have relevance in the prognosis of FL patients. In addition, some of these cells play a role in the histologic transformation of FL. Macrophages contribute to a poor prognosis in FL patients treated in the pre-rituximab era, but are associated with good prognosis in those treated in the rituximab er

Gastric carcinoma (GC) is the 4(th) most prevalent cancer and has the 2(nd) highest cancer-related mortality rate worldwide. Despite the incidence of GC has decreased over the past few decades, it is still a serious health problem. Chronic inflammatory status of the stomach, caused by the infection of Helicobacter pylori (H. pylori) and through the production of inflammatory mediators within the parenchyma is suspected to play an important role in the initiation and progression of GC. In this review, the correlation between chronic inflammation and H. pylori infection as an important factor fo

BACKGROUND AND OBJECTIVES: Colorectal cancer (CRC) ranks as the third most common cancer globally and is the second leading cause of cancer-related deaths. While some CRC patients with deficient mismatch repair (dMMR) or high microsatellite instability (MSI-H) respond to immune checkpoint inhibitors (ICIs), many exhibit resistance, partly due to polymorphonuclear myeloid-derived suppressor cells (PMN-MDSCs). APE1/Ref-1, a bifunctional protein involved in DNA repair and redox regulation, has an inhibitor, APX3330, which targets its redox function without affecting DNA repair. This study aims to

INTRODUCTION: Antibody-drug conjugates (ADCs) have revolutionized oncology, yet tumor microenvironment (TME) heterogeneity remains a major barrier. TME-targeted ADCs offer a paradigm shift by specifically modulating immunosuppressive cellular crosstalk and stromal components to overcome tumor evasion.
AREAS COVERED: This review summarizes design strategies and clinical progress of TME-targeted ADCs. Literature was searched via PubMed and Embase (2015-2025), focusing on early-phase trials, safety profiles, and translational challenges in advanced solid tumors.
EXPERT OPINION: The next decade

Ameloblastoma is a benign odontogenic tumor with an aggressive growth phenotype orchestrated by a complex and heterogeneous tumor microenvironment. This review addresses how tumor cells, cancer-associated fibroblasts, mesenchymal stem cells, endothelial cells, and immune cells interact with non-cellular elements especially the extracellular matrix and hypoxic niches to drive invasive growth and recurrence. Several genetic changes associated with ameloblastoma activate mitogen-activated protein kinase (MAPK), Hedgehog (HH), Wnt/β-catenin, and less commonly PI3K/AKT signaling pathways. These pat

Melanoma has one of the highest propensities to metastasize to the brain, and despite major advances with immune checkpoint inhibitors, brain metastases remain a leading cause of morbidity and mortality. The biological mechanisms governing brain colonization, immune evasion, and resistance to therapy are incompletely understood. This review summarizes recent advances in the understanding of the tumor microenvironment and signaling pathways involved in melanoma brain metastasis, with emphasis on mechanisms that may provide novel therapeutic opportunities. We performed a review of preclinical an

The character of the tumor microenvironment is a relevant prognostic and predictive biomarker across a wide range of malignancies. The composition, density, and functional capacity of tumor-infiltrating immune cells are especially crucial for selecting suitable immunotherapy. Head and neck squamous cell carcinomas are considered immunologically hot tumors, with high numbers of tumor-infiltrating effector and regulatory T cells. Higher T cell counts, along with a better prognosis, were observed in patients with head and neck squamous cell carcinomas associated with human papillomavirus infectio

INTRODUCTION: Immunotherapy, especially anti-PD-1 antibodies (α-PD-1), has revolutionized the landscape of cancer treatment. However, the response rate of α-PD-1 for Gastric Cancer (GC) remains relatively low. The acidic immunosuppressive Tumor Microenvironment (TME) greatly hinders the efficacy of α-PD-1. Thus, therapeutic strategies targeting the acidic TME in GC are highly desired.
OBJECTIVES: This study aimed to investigate the effect of FX-11@PEG-Ce6 on the therapeutic efficacy of photoimmunotherapy for gastric cancer.
METHODS: We developed FX-11 encapsulated with PEG-Ce6 nanoparticles

BACKGROUND: The tumor microenvironment (TME), consisting of tumor-associated stroma and tumor-infiltrating lymphocytes (TIL), is crucial for prognostic information in gastric cancer (GC). Despite its potential, routine clinical adoption remains limited.
METHODS: In a study of 320 GC patients, virtual staining and image processing were applied to hematoxylin & eosin-stained slides. This method quantified the tumor-stroma ratio (TSR) and TIL, leading to a TME-based prediction model (TMEPATH) using a scoring system derived from univariate Cox regression. Subgroups were categorized to predict GC

There is an urgent need for appropriate methods to identify patients who are sensitive to neoadjuvant therapy (NAT) for precise stratified treatment. Based on clinical proteomics screening and further validation in patient cohorts, we reported that soluble glucose-regulatory protein 78 (sGRP78) could be a serological biomarker to infer the poor chemotherapy response in breast cancer. Consequently, an "sGRP78 index" was established to accurately predict patient responses to NAT. When exploring the linked mechanisms on how sGRP78 confer the resistance, it was found that in high sGRP78 index tumo

Mesothelial cells (MCs) constitute a dynamic mesothelium in which their numerous crucial functions synergistically interact with other cells to maintain serosal integrity and homeostasis. Previous studies have confirmed the crucial role of interactions between MCs and tumor cells in tumorigenesis and progression in the tumor microenvironment (TME). However, recent research has found that MCs can induce an immunosuppressive microenvironment by secreting various cytokines, chemokines, and exosomes which recruit immunosuppressive cells or interact with immune cells, thus contributing to tumor pro

Gliomas are aggressive and heterogeneous brain tumors with limited treatment options. While oncogenic mutations in gliomas have been well-characterized, their impact on the tumor microenvironment remains poorly understood. To investigate how genomic alterations may influence the glioma microenvironment, we performed an integrative multiomic and spatial transcriptomic analysis of 93 glioma samples (46 IDH-mutant gliomas and 47 IDH-wildtype glioblastoma) from 69 patients representing both primary and recurrent stages. Using whole-genome sequencing, chromatin conformation capture (Hi-C), RNA-seq,

Gemcitabine (GEM), a nucleoside analog chemotherapy agent, has been widely used in the treatment of various cancers. In recent years, there has been growing interest in understanding the immunomodulatory or immunosuppressive effects of GEM. The immunomodulatory roles of GEM could influence the anti-tumor immune responses via several mechanisms, such as modulation of antigen presentation, cytokine production, and immune cell population. Furthermore, there is evidence that GEM enhances the therapeutic efficacy of immunotherapies, including oncolytic viruses, immune checkpoint inhibitors, CAR T-c

To address the challenges of insufficient hydrogen peroxide (H2O2) levels, rapid Fe3+ precipitation, and a slow Fenton reaction cycle, tumor-activated, self-accelerating CDT nanocatalysts are synthesized, comprising poly (lactic-co-glycolic acid) (PLGA)-encapsulated Ca-Fe peroxide clusters and polyarginine (R). Nanocatalysts are camouflaged with cancer cell membranes (CCM) to enhance tumor targeting. Additionally, polyarginine tailored the PLGA responsiveness to low H2O2 levels (50-100 µm). H2O2 triggered the degradation of PLGA, releasing CaFe clusters to produce Fe3+/Fe2+ and additional H2O2

Most chemotherapeutic and bioimaging agents struggle with inadequate bioavailability, primarily due to their limited biocompatibility and lack of specificity in targeting, leading to low or decreased anticancer efficacy and inaccurate imaging. To surmount these obstacles, the development of stimuli-responsive polymer delivery platforms, predominantly leveraging the tumor microenvironment (TME), has emerged as a promising strategy. Therapeutic and diagnostic agents can be released controllably at the tumor site by virtue of the bond cleavage or hydrophobic to hydrophilic transformation of TME-s
