The potential contamination of human blood or plasma with prions, such as variant Creuftzfelt-Jacob disease (vCJD), is becoming a serious problem. In this study, we established a Western blot-based detection method for PrP(Sc) (263K) spiked in plasma. Although plasma contains a large amount of protein, specific detection of a small amount of 263K in plasma could be specifically detected only after ultra-centrifugation followed by heat-denaturation of plasma proteins included in the resulting precipitate, before the digestion with proteinase K.

Mutations in the gene for the lysosomal enzyme glucocerebrosidase (GCase) cause Gaucher disease and are the most common risk factor for Parkinson disease (PD). Analytical ultracentrifugation of 8 μM GCase shows equilibrium between monomer and dimer forms. However, in the presence of its co-factor saposin C (Sap C), only monomer GCase is seen. The interactions of the parasympatholytic drugs adiphenin, adiphenin H and propanthelin and the structurally related compounds, diphenhydramine, D-propoxyphene and methadone with bovine serum albumin (BSA) are studied by equilibrium dialysis and ultracentrifugation. The results show that BSA has a few binding sites (n less than 10) for the mentioned drugs. The association between the compounds under study and BSA are relatively weak. The binding constants are in the range of 10(3) l/Mol.

We succeeded in the separation of human serum high density lipoprotein into their subfractions (HDL2 and HDL3) by high performance liquid chromatography with gel permeation columns (1, 2). The cholesterol contents in HDL2 and HDL3 for normal men and women (n = 42) and for the patients with atherosclerotic heart disease (n = 20) were measured by this separation method. The results in our experiment agree closely with those reported by other investigators using the ultracentrifugal method.

Low density lipoproteins (LDL) and four subfractions of very low density lipoprotein (VLDL) were isolated by means of density gradient preparative ultracentrifugation from blood serum of patients with chronic renal failure. Constituents of the fractions were analyzed and physico-chemical characteristics of the lipoprotein particles were studied. These data showed that there are pronounced changes in the composition of VLDL subfractions containing small particles, thus suggesting the impaired catabolism of the particles in blood.

The properties of the preparations of the total typhoid antigens isolated by various methods, purified and concentrated by ultrafiltrations, were studied, and the technique of the preservation of their high solubility on drying (the neutralization and removal of disperse components prior to drying) was developed. Preparations obtained by ultracentrifugation were characterized by lower ash content and, when compared with alcohol tryptic preparations, had better antigenic properties, inducing the synthesis of not only O- and Vi-, but also H-antibodies in high titers.
