Mechanisms and quality variations of non-volatile and volatile metabolites in black tea from various ages of tea trees: Insights from metabolomics analysis.

The sensory quality of black tea (BT) influenced by various factors, among which tree age is particularly significant. People prefer BT produced by fresh leaves from old tea trees, yet the correlation between tree age and tea quality has not been thoroughly investigated. In this study, we analyzed the quality of BT from young trees (H-JYH) and old trees (H-OJYH) using e-tongue technology and sensory evaluation. Our findings revealed that H-OJYH had stronger sweetness and sourness, richer flavor, and diminished bitter-astringency compared to H-JYH. 1231 non-volatile metabolites and 504 volatile

Synergistic anti-Campylobacter jejuni activity of fluoroquinolone and macrolide antibiotics with phenolic compounds.

The increasing resistance of Campylobacter to clinically important antibiotics, such as fluoroquinolones and macrolides, is a serious public health problem. The objective of this study is to investigate synergistic anti-Campylobacter jejuni activity of fluoroquinolones and macrolides in combination with phenolic compounds. Synergistic antimicrobial activity was measured by performing a checkerboard assay with ciprofloxacin and erythromycin in the presence of 21 phenolic compounds. Membrane permeability changes in C. jejuni by phenolic compounds were determined by measuring the level of intrace

Blackthorn juice from central Italy exhibits selective anticancer activity by inducing apoptosis and autophagy in gastric carcinoma cells.

Blackthorn (Prunus spinosa L.) juice, a polyphenol-rich blend derived from the Marche region of Italy, was chemically characterized and evaluated for its selective effects against gastric cancer cells. The juice contained a mix of polyphenols, including neochlorogenic acid, cyanidin-3-glucoside, rutin, and smaller molecules - such as gallic, caffeic, and vanillic acids - characterized by high absorption rates. In vitro, the juice significantly reduced the viability of AGS and KATO III gastric cancer cells, while sparing non-tumorigenic GES-1 cells. In malignant cells, the juice activated Caspa

The impact of chemical structure on redox behavior and biological activity of hydroxybenzoic acids.

Hydroxybenzoic acids (HBAs) are dietary phenolics with recognized antioxidant potential, yet the links between their chemical structure and biological activity remain unclear. Four HBAs - gallic (GA), gentisic (GE), protocatechuic (PR), and vanillic (VA) acids - were compared. Antioxidant efficiency correlated with hydroxyl density, with GA showing the highest activity and VA the weakest. In the Cellular Antioxidant Activity assay, GA again exhibited the strongest effect, whereas PR, GE, and VA showed lower activities, reflecting differences in bioavailability. GA reduced the viability of both

Exploring the Selective Demethylation of Aryl Methyl Ethers with a Pseudomonas Rieske Monooxygenase.

Biocatalytic dealkylation of aryl methyl ethers is an attractive reaction for valorization of lignin components, as well as for deprotection of hydroxy functionalities in synthetic chemistry. We explored the demethylation of various aryl methyl ethers by using an oxidative demethylase from Pseudomonas sp. HR199. The Rieske monooxygenase VanA and its partner electron transfer protein VanB were recombinantly coexpressed in Escherichia coli and they constituted at least 25 % of the total protein content. Enzymatic transformations showed that VanB accepts NADH and NADPH as electron donors. The Van

Polyphenol Oxidase Activity on Guaiacyl and Syringyl Lignin Units.

The natural heterogeneity of guaiacyl (G) and syringyl (S) compounds resulting from lignin processing hampers their direct use as plant-based chemicals and materials. Herein, we explore six short polyphenol oxidases (PPOs) from lignocellulose-degrading ascomycetes for their capacity to react with G-type and S-type phenolic compounds. All six PPOs catalyze the ortho-hydroxylation of G-type compounds (guaiacol, vanillic acid, and ferulic acid), forming the corresponding methoxy-ortho-diphenols. Remarkably, a subset of these PPOs is also active towards S-compounds (syringol, syringic acid, and si

A trypanosome trifecta: an independently tunable triple inducible system for genetic studies in Trypanosoma brucei.

Trypanosoma brucei is a model unicellular parasite for cellular and molecular genetic studies, but tools for more multiplexed experiments are limited. Tetracycline (Tet)-inducible gene regulation has been a long-standing and effective approach for overexpression, RNAi, and genome-wide screens and has been foundational for studying essential genes that are required for biological processes and identifying potential drug targets. To achieve greater flexibility in experimental design, we capitalized upon previously described dual inducer systems that combined vanillic acid (Van) or cumate (Cym) w

Anti-Hair Loss Effect of Veratric Acid on Dermal Papilla Cells.

The activation of hair follicle dermal papilla cells (HFDPCs), a critical target of hair loss relief, can be achieved through the upregulation of proliferation, the stimulation of hair inducibility, and the inhibition of cellular senescence. Veratric acid (VA) is a major benzoic acid found in fruits and vegetables. The biological activity of VA on HFDPCs remains to be elucidated. In this study, we investigated the capacity of VA for hair loss mitigation. An MTT assay, Ki67 staining, quantitative RT-PCR (qRT-PCR), and a Western blot analysis were performed to confirm the proliferative effect of

Effects of solute characteristics and concentration on a lyotropic liquid crystal: solute-induced phase change.

We investigated the effects of increased concentrations of the solutes, salicylic acid, benzoic acid, and o-, m-, and p-methoxy benzoic acids, on the anisotropic properties of a liquid crystal solvent. The lamellar liquid crystal was composed of 37% polyoxyethylene (20) isohexadecyl ether in aqueous buffer of pH 1. Phase change, transition temperature, refractive index, and specific resistance of the mesophase were studied in the presence of solutes. Transfer rates of the solutes from the bulk mesophase into aqueous buffer across a lipoidal barrier were used to determine their apparent permeab

Phytochemical profiling of Laurus nobilis L. leaf extracts with direct analysis in real-time mass spectrometry and HPLC-DAD.

This work focuses on the extraction and analysis of polyphenols, more precisely flavonoid aglycones, from Algerian Laurus nobilis L. leaf extracts. The extraction of these bioactive substances was carried out according to a routine method with some modifications. Two main extraction steps were carried out. Hence, the first step was to solubilize flavonoids in methanol, followed by liquid-liquid extraction using ethyl acetate. The extract thus obtained was separated into six fractions by column chromatography. The identification of constituents was carried out by high-performance liquid chromat

Tissue-specific antibacterial and anti-biofilm activities of Penthorum chinense Pursh: integration of metabolomics and Random Forest machine learning.

Penthorum chinense Pursh is a traditional medicinal and edible plant rich in flavonoids and complex polyphenols. Evaluating its aqueous extracts against foodborne pathogens via biomass assay, microplate spread tests and scanning electron microscopy revealed distinct tissue-specific efficacies: leaf and stem extracts exhibited strong bactericidal effects (16-128 mg/mL), notably against Bacillus cereus, whereas flower extracts demonstrated potent biofilm inhibition (IC50 < 1 mg/mL). Broad-coverage UPLC-ESI-Q-TOF-MS metabolite profiling annotated 93 compounds, and integration with Random Forest n

In vitro Effect of Chlorquine and Picroliv on Plasmodium Berghei Induced Alterations in the Activity of Adenosine Triphosphatase, Aryl Hyrocarbon Hydroxylase Enzymes and Malondialdehyde in Spleen Explant Culture.

BACKGROUND: In recent years, great progress has been made in reducing the high level of malaria suffering worldwide. There is a great need to evaluate drug resistance reversers and consider new medicines against malaria. There are many approaches to the development of antimalarial drugs. Specific concerns must be taken into account in these approaches, in particular the requirement for inexpensive and simple new therapies and the need to limit drug discovery expenses. Important ongoing efforts are the optimisation of treatment with available medications, including the use of combination therap

Shell thickness controlled hydrophilic magnetic molecularly imprinted resins for high-efficient extraction of benzoic acids in aqueous samples.

Hydrophilic magnetic molecular imprinted resins (MMIRs) were firstly fabricated by one-pot copolycondensation of resorcinol, melamine, and formaldehyde in the pores of Fe3O4 @mSiO2. The porous structure of mSiO2 limited copolycondensation. After the removal of mSiO2, controllable imprinting shell thickness (about 5 nm) was formed, reducing the probability of embedded sites, favoring the efficient adsorption. Moreover, the reaction time (3 h) was just 1/5-1/8 of those synthesized by conventional heating method. The transmission electron microscopy (TEM), Fourier transform infrared (FT-IR), vibr

Electron transfer from plant phenolates to carotenoid radical cations. Antioxidant interaction entering the Marcus theory inverted region.

β-Carotene, lycopene, and zeaxanthin are maximally regenerated by plant phenolates from their radical cations formed during laser flash photolysis in 9:1 (v/v) chloroform/methanol for a driving force corresponding to the reorganization energy according to the Marcus theory. For β-carotene, the reorganization energy has values of 0.41 ± 0.04 and 0.40 ± 0.04 eV for the plant phenols in the presence of 1 and 2 equiv of base, respectively, at 23 °C. For a driving force lower than the reorganization energy, regeneration of the carotenoids is less efficient as is seen for m-hydroxybenzoic acid, vani
