Complete and irreversible unilateral vestibular loss: A novel rat model of vestibular pathology.

BACKGROUND: Both basic and applied studies on the pathophysiology of vestibular disorders are currently impaired by the lack of animal models of controlled vestibular damages. NEW METHODS: In the present study, we describe the procedure to achieve a surgical unilateral vestibular neurectomy (UVN) in the rat and evaluate its functional consequences. This procedure is suitable for reproducing a unilateral, sudden and definitive vestibular areflexia. RESULTS: Proper induction of a UVN induces a severe vestibular syndrome, which mimics vestibular disorders encountered in humans. This model is al

Aged mice are less susceptible to motion sickness and show decreased efferent vestibular activity compared to young adults.

INTRODUCTION: The efferent vestibular system (EVS) is a feedback circuit thought to modulate vestibular afferent activity by inhibiting type II hair cells and exciting calyx-bearing afferents in the peripheral vestibular organs. In a previous study, we suggested EVS activity may contribute to the effects of motion sickness. To determine an association between motion sickness and EVS activity, we examined the effects of provocative motion (PM) on c-Fos expression in brainstem efferent vestibular nucleus (EVN) neurons that are the source of efferent innervation in the peripheral vestibular organ

The nucleus magnocellularis in the red-eared turtle, Chrysemys scripta elegans: eighth nerve endings and neuronal types.

Eighth nerve endings and neuronal types in the nucleus magnocellularis were analyzed in the red-eared turtle (Chrysemys scripta elegans). One group of turtles had horseradish peroxidase (HRP) injected into the surgically exposed inner ear. Following injection the animals survived for 3-5 days and then their 8th nerve fibers and endings were analyzed. A second group of turtles were impregnated by the Golgi-Kopsch technique. This also demonstrated 8th nerve endings and neurons in the nucleus magnocellularis. A third group had brains stained with cresyl violet to demonstrate normal morphology. Tw

Intrinsic and synaptic plasticity in the vestibular system.

The vestibular system provides an attractive model for understanding how changes in cellular and synaptic activity influence learning and memory in a quantifiable behavior, the vestibulo-ocular reflex. The vestibulo-ocular reflex produces eye movements that compensate for head motion; simple yet powerful forms of motor learning calibrate the circuit throughout life. Learning in the vestibulo-ocular reflex depends initially on the activity of Purkinje cells in the cerebellar flocculus, but consolidated memories appear to be stored downstream of Purkinje cells, probably in the vestibular nuclei.

[Sudden onset vertigo: imaging work-up].

Vertigo and dysequilibrium are a frequent cause of medical consultation. Clinical evaluation is essential. Some cases of vertigo are diagnosed clinically while others require imaging, sometimes emergently (suspected stroke). MRI is the imaging modality of choice to assess the labyrinth (labyrinthitis? labyrinthine hemorrhage?), internal auditory canal (vestibular schwannoma? other tumor?…) and brain parenchyma including all structures of the auditory pathways: vestibular nuclei, vestibulocerebellar tract, tracts involved with ocular motricity, vestibular cortex… Multiple central etiologies exi

[Benign paroxysmal vertigo in childhood].

Benign paroxysmal vertigo of childhood is a clinical disorder that usually begins at age 4. The child's otoneurological examination is normal in the intercrisis and the brief episodes of true vertigo are typical. In this paper we describe six new cases that had a follow-up of at least one year after diagnosis. Only a few of the patients had demonstrated vasomotor migraine, but all the children had a family history of migraine, which supports the hypothesis that this disorder is a migraine equivalent. The pathogenesis may be related with a transitory vascular disturbance that produces ischemia

[So-called internuclear ophthalmoplegias. Electro-oculographic study].

A study is presented of three patients suffering from internuclear ophthalmoplegia without diplopia. The components of the nystagmus and the dysmetry of the oculomotor changes, as well as the difference in the respective speeds during abduction and adduction toward the side opposite to the lesion were recorded. A dysmetric component was found in the saccadic low amplitude movements (reading movements).

In Vivo Localization of the Human Velocity Storage Mechanism and Its Core Cerebellar Networks by Means of Galvanic-Vestibular Afternystagmus and fMRI.

Humans are able to estimate head movements accurately despite the short half-life of information coming from our inner ear motion sensors. The observation that the central angular velocity estimate outlives the decaying signal of the semicircular canal afferents led to the concept of a velocity storage mechanism (VSM). The VSM can be activated via visual and vestibular modalities and becomes manifest in ocular motor responses after sustained stimulation like whole-body rotations, optokinetic or galvanic vestibular stimulation (GVS). The VSM has been the focus of many computational modelling ap

Short-term galvanic vestibular stimulation promotes functional recovery and neurogenesis in unilaterally labyrinthectomized rats.

Current experimental research on the therapeutic effects of galvanic vestibular stimulation (GVS) has mainly focused on neurodegenerative disorders. However, it primarily stimulates the vestibular nuclei and could be potentially effective in modulating imbalance between them in the case of unilateral labyrinthectomy (UL). Fifty male Wistar rats (180-220g) were used in 5 groups of 10: intact, sham, right-UL (RUL; without intervention), and two other right-UL groups with GVS intervention [one group treated with low rate GVS (GVS.LF; 6-7Hz), and the other treated with high rate GVS (GVS.HF; 17-18

Abnormal oculovestibular response.

In normal individuals, the visual and vestibular systems interact through a common subcortical center located near the vestibular nuclei. When the patient is healthy, this interaction allows appropriate integration of what might otherwise be conflicting information regarding environmental motion and moving within the environment. In patients with active peripheral vestibular lesions, such as those seen in Meniere's disease, this delicate interaction loses effectiveness. In such patients, optokinetic stimuli are capable of precipitating episodes of incapacitating vertigo with its associated veg

Laterality in the vestibulo-cerebellar mossy fiber projection to flocculus and caudal vermis in the rabbit: a retrograde fluorescent double-labeling study.

The vestibular nuclear input into the flocculus and the uvula and nodulus of the caudal vermis was studied in rabbits by means of retrograde transport of the fluorescent tracers Fast Blue and Diamidino Yellow. Through simultaneous injection of the tracers in the homonymous lobules at either side of the midline, the distribution, the preference in laterality and the degree of collateralization of vestibulo-cerebellar neurons could be studied. The nucleus prepositus hypoglossi was included in the analysis. In the nucleus prepositus hypoglossi and in the medial vestibular nucleus 6% of the number

Organization of vestibular inputs to nucleus tractus solitarius and adjacent structures in cat brain stem.

The vestibular system is involved in maintaining stable blood pressure and respiration during changes in posture and is essential for eliciting motion sickness-related vomiting. Because the nucleus tractus solitarius (NTS) participates in the regulation of sympathetic and inspiratory outflow and the triggering of emesis, we tested the hypothesis that this region receives vestibular inputs in cats. In one set of experiments, microinjections of the tracer Phaseolus vulgaris leucoagglutinin into the medial and inferior vestibular nuclei labeled projections to the middle and lateral regions of the

A pilot study on early microgliogenesis following unilateral vestibular neurectomy: A key player in vestibular compensation?

Following unilateral vestibular damage, several behavioral deficits arise, referred to as vestibular syndrome. At the central level the vestibular syndrome is associated with an imbalance in neuronal activity between the two vestibular nuclei (VN). Its recovery is correlated with a rebalancing of the electrophysiological activity between both VN, known as vestibular compensation. Key plasticity mechanisms within the VN involved in this mechanism include, among others, neurogliogenesis, modulation of neuronal excitability and increased histamine release. In this study, we aimed to characterize

Acute unilateral loss of vestibular function.

Sudden unilateral loss of vestibular function is the most severe condition that can occur in the vestibular system. The clinical syndrome is caused by the physiologic properties of the vestibulo-ocular reflex (VOR) arc. In the normal situation, the two peripheral vestibular end organs are connected to a functional unit in coplanar pairs of semicircular canals working in a push-pull mode. "Push-pull" mode means that, when one side is excited, the other side is inhibited, and vice versa due to two mechanisms. First, first-order vestibular afferents are bipolar cells. They have a tonic firing rat

Characterization of direct Purkinje cell outputs to the brainstem.

Purkinje cells (PCs) primarily project to cerebellar nuclei but also directly innervate the brainstem. Some PC-brainstem projections have been described previously, but most have not been thoroughly characterized. Here, we use a PC-specific cre line to anatomically and electrophysiologically characterize PC projections to the brainstem. PC synapses are surprisingly widespread, with the highest densities found in the vestibular and parabrachial nuclei. However, there are pronounced regional differences in synaptic densities within both the vestibular and parabrachial nuclei. Large optogenetical

A nonlinear model of the neural integrator improves detection of deficits in the human VOR.

A nonlinear model has been proposed to describe the set-point-dependent characteristics of the neural integrator (NI) in the oculomotor system. It was shown to yield improved prediction of slow-phase eye position in the vestibulo-ocular reflex (VOR) of normal subjects, when compared to the classical linear model of the NI. In this paper, we compare the parameters of this nonlinear NI model fitted to VOR data from: 1) compensated subjects diagnosed with vestibular deficiencies such as vestibular neuronitis and Meniere's disease and 2) normal (symptom-free) subjects. The identified models exhibi

The vestibulocochlear nerve (VIII).

The vestibulocochlear nerve (8th cranial nerve) is a sensory nerve. It is made up of two nerves, the cochlear, which transmits sound and the vestibular which controls balance. It is an intracranial nerve which runs from the sensory receptors in the internal ear to the brain stem nuclei and finally to the auditory areas: the post-central gyrus and superior temporal auditory cortex. The most common lesions responsible for damage to VIII are vestibular Schwannomas. This report reviews the anatomy and various investigations of the nerve.
