Epidemiology & molecular biology of Vibrio cholerae O139 Bengal.

The emergence of Vibrio cholerae O139 Bengal as the second aetiologic agent of epidemic cholera in October 1992 in the south Indian coastal city of Madras has shattered the long-held notion that only V. cholerae belonging to serogroup O1 are capable of causing epidemic (and pandemic) cholera. Within months of its appearance in Madras, V. cholerae O139 engulfed the entire Indian subcontinent in a series of outbreaks of cholera. It also spread to several neighbouring countries in Asia. Several western countries also reported imported cholera cases due to this organism. In the regions of the Indi

Analysis of antimicrobial resistance and virulence of Vibrio parahaemolyticus.

Vibrio parahaemolyticus poses dual threats to marine ecosystems and human health through seafood-borne transmission. This study reveals critical links between antibiotic resistance and virulence phenotypes in 95 V. parahaemolyticus isolates recovered from 284 retail seafood samples and 12 aquaculture samples (9 diseased shrimp and 3 water samples) collected from multiple coastal regions in Eastern and Southern China. All isolates exhibited multidrug resistance, with ubiquitous resistance to critically important antibiotics including polymyxins (100 %) and sulfonamides (100 %). Phenotypic analy

It's time to act: Understanding and combating Vibrio vulnificus.

Vibrio vulnificus is a highly pathogenic marine bacterium that can cause life-threatening conditions such as septicemia and necrotizing fasciitis; in medically fragile individuals, mortality rates can exceed 50%. Its virulence factors, including the MARTX toxin and hemolysin VvhA, function to disrupt host cells, evade immune responses, and facilitate bacterial dissemination. Climate warming has dramatically expanded the geographic range of V. vulnificus, doubling infection risks in high-latitude northern waters, while microplastic pollution and extreme weather events further enhance its ecolog

The role of morphological adaptability in Vibrio cholerae's motility.

UNLABELLED: Vibrio cholerae, the causative agent of cholera, displays remarkable adaptability to diverse environmental conditions through morphological changes that enhance its pathogenicity and influence the global epidemiology of the disease. This study examines the motility differences between filamentous and comma-shaped forms of the V. cholerae O1 strain under various viscosity conditions. Utilizing the El Tor strain, we induced filamentous transformation and conducted a comparative analysis with the canonical comma-shaped morphology. Our methodology involved assessing motility patterns,

[Vibrio vulnificus, an increasing threat of sepsis in Germany?].

BACKGROUND: The prevalence of Vibrio vulnificus heavily depends on the temperature and salinity of the sea water. In the course of climate change an increase in cases of fatal sepsis caused by V. vulnificus at the German Baltic Sea coast could be detected. OBJECTIVE: To generate awareness for a life-threatening infection with increasing incidence in Germany. MATERIAL AND METHODS: This article presents an overview of the current state of the literature followed by an exemplary description of cases with V vulnificus sepsis caused by contact with water in the Baltic Sea, which were treated at t

Vibrio vulnificus meningoencephalitis in a patient with thalassemia and a splenectomy.

Vibrio vulnificus usually causes wound infection, gastroenteritis, and septicemia. However, it is a rare conditional pathogen causing meningoencephalitis. We report a case of a young, immunocompromised man presenting with severe sepsis after exposure to sea water and consumption of seafood. The patient subsequently developed meningoencephalitis, and Vibrio vulnificus was isolated from his blood culture. The sequence was confirmed by Next-generation sequencing of a sample of cerebrospinal fluid, as well as from a bacteria culture. After the pathogen was detected, the patient was treated with ce

Molecular tools in understanding the evolution of Vibrio cholerae.

Vibrio cholerae, the etiological agent of cholera, has been a scourge for centuries. Cholera remains a serious health threat for developing countries and has been responsible for millions of deaths globally over the past 200 years. Identification of V. cholerae has been accomplished using a variety of methods, ranging from phenotypic strategies to DNA based molecular typing and currently whole genomic approaches. This array of methods has been adopted in epidemiological investigations, either singly or in the aggregate, and more recently for evolutionary analyses of V. cholerae. Because the ne

Environmental reservoirs and mechanisms of persistence of Vibrio cholerae.

It is now well accepted that Vibrio cholerae, the causative agent of the water-borne disease cholera, is acquired from environmental sources where it persists between outbreaks of the disease. Recent advances in molecular technology have demonstrated that this bacterium can be detected in areas where it has not previously been isolated, indicating a much broader, global distribution of this bacterium outside of endemic regions. The environmental persistence of V. cholerae in the aquatic environment can be attributed to multiple intra- and interspecific strategies such as responsive gene regula

The complexity of ToxT-dependent transcription in Vibrio cholerae.

Vibrio cholerae is the causative agent of the disease cholera, characterized by profuse watery diarrhoea. Two of the main virulence factors associated with the disease are cholera toxin (CT) and toxin-coregulated pilus (TCP). Expression of CT and TCP is regulated via a complex cascade of factors that respond to environmental signals, but ultimately ToxT is the direct transcriptional activator of the genes encoding CT and TCP. Recent studies have begun to unveil the mechanisms behind ToxT-dependent transcription. We review current knowledge of transcriptional activation by ToxT and the environm

A model for Vibrio cholerae colonization of the human intestine.

Vibrio cholerae is a strict human pathogen that causes the disease cholera. It is an old-world pathogen that has re-emerged as a new threat since the early 1990s. V. cholerae colonizes the upper, small intestine where it produces a toxin that leads to watery diarrhea, characterizing the disease (Kahn et al., 1988). The dynamics of colonization by the bacteria of the intestines are largely unknown. Although a large initial infectious dose is required for infection, data suggests that only a smaller sub-population colonizes a portion of the small bowel leading to disease. There are many barriers

[A case of Vibrio vulnificus infection presenting with invagination].

A 73-year-old man with advanced lung cancer ate raw fish the day before being admitted to our hospital with severe abdominal pain, fever and vomiting. Soon afterward he fell into shock and disseminated intravenous coagulation. A CT scan revealed invagination and emergency surgery was performed. On the first day after surgery, Vibrio vulnificus was cultured from his ascites and blood. The infection was controlled by antibiotic treatment and he was discharged 23 days after the surgery. Vibrio vulnificus infection is an opportunistic infection that develops mostly in patients with liver cirrhosis

Environmental reservoirs of Vibrio cholerae and their role in cholera.

In the aquatic environment, Vibrio cholerae has been reported to be associated with a variety of living organisms, including animals with an exoskeleton of chitin, aquatic plants, protozoa, bivalves, waterbirds, as well as abiotic substrates (e.g. sediments). Most of these are well-known or putative environmental reservoirs for the bacterium, defined as places where the pathogen lives over time, with the potential to be released and to cause human infection. Environmental reservoirs also serve as V. cholerae disseminators and vectors. They can be responsible for the start of an epidemic, may b

Role of FlgT in anchoring the flagellum of Vibrio cholerae.

Flagellar motility has long been regarded as an important virulence factor. In Vibrio cholerae, the single polar flagellum is essential for motility as well as for proper attachment and colonization. In this study, we demonstrate that the novel flagellar protein FlgT is involved in anchoring the flagellum to the V. cholerae cell. A screen for novel colonization factors by use of TnphoA mutagenesis identified flgT. An in-frame deletion of flgT established that FlgT is required for attachment, colonization, and motility. Transmission electron microscopy revealed that while the flgT mutant is cap

Vibrio2009: the third international conference on the biology of Vibrios.

Vibrio2009, the third international conference on the biology of Vibrios, was held in Rio de Janeiro, Brazil, in November 2009. This conference united researchers studying various aspects of pathogenesis, symbiosis and environmental persistence of this diverse group of marine bacteria. Through many of the presentations, it became apparent how horizontal gene transfer and genetic flexibility has driven the incredible diversity of these microbes. Interestingly, unifying themes of behaviour could be seen in the interaction(s) of Vibrios with other organisms, such as with other bacteria, corals, i

A fatal case of Vibrio vulnificus cellulitis with septicaemia.

Vibrio vulnificus is a gram-negative marine bacterium that may cause local wound infection, distinctive soft tissue infection, gastroenteritis and septicaemia with a high mortality rate. A healthy man presented with severe abdominal pain, diarrhoea and fever followed by development of multiple blisters, cellulitis and necrotizing fasciitis of the lower limbs, who progressed rapidly to fulminant sepsis caused by this organism. Vibrio vulnificus septicaemia should be suspected in the presence of sepsis and progressive soft-tissue infection with recent history of raw seafood consumption.

Heme, an iron supply for vibrios pathogenic for fish.

One of the main mechanisms present in gram-negative bacterial pathogens to obtain iron is the utilization of free heme or heme proteins from the host tissues. Vibrio anguillarum, the etiological agent of vibriosis in fish, and Photobacterium damselae subsp. piscicida, the causative agent of fish pasteurellosis, can acquire iron from free heme or heme-containing proteins present in the host tissues by a siderophore-independent mechanism. Similarly to other animal and human pathogens, the general mechanism for heme uptake in these two species consists in the presence of an outer membrane recepto
