Q10-triggered facial vitiligo.

BACKGROUND: Generation and accumulation of reactive oxygen/nitrogen species in the epidermis of patients with vitiligo has been widely documented. Moreover, semiquinone radical-mediated sensitivity has been shown in blood lymphocytes of these patients. OBJECTIVES: To determine the possible mechanism behind Q10-induced facial vitiligo. METHODS: This was a clinical assessment supported by in vivo Fourier transform-Raman spectroscopy and repigmentation. RESULTS: Topical Q10 application generated hydrogen peroxide (H2 O2 ) leading in turn to facial vitiligo in susceptible individuals. Proof of

Vitiligo-like primary melanoma.

Depigmented or hypopigmented macules and patches have been described in association to primary and metastatic melanoma. Primary melanoma presenting at the onset as a vitiliginous patch without histopathological evidence of regression seems to be a rare event. We report 2 patients with melanoma in situ that expressed clinically as vitiligo-like patches and review the possible immunologic mechanisms responsible for this unusual presentation.

Generalized vitiligo after erythroderma.

We report a 65-year-old black man whose onset of an erythroderma was followed in 6 weeks by a generalized vitiligo. The etiology of the erythroderma was unclear, although a prostate adenocarcinoma was found on evaluation. This is to our knowledge the first patient reported with a vitiligo following exfoliative dermatitis, and the first report of two uncommon markers of internal malignancy (erythroderma and vitiligo) occurring with prostate adenocarcinoma in the same patient.

Vitiligo Treatment Impact score (VITs): development and validation of a vitiligo burden of treatment questionnaire using the ComPaRe Vitiligo e-cohort.

BACKGROUND: Vitiligo management is challenging and requires long-term adherence of patients who often complain of the burden associated with treatment. OBJECTIVE: To develop and validate a patient reported measurement of the burden of treatment in vitiligo. METHODS: The study was nested within the ComPaRe Vitiligo e-cohort, an online e-cohort of vitiligo patients in France. Items were derived from a literature review and from the qualitative analysis of a survey using open-ended questions of 204 patients with Vitiligo. Construct validity of the resulting instrument was assessed by comparing

Vitiligo Area Scoring Index and Vitiligo European Task Force assessment: reliable and responsive instruments to measure the degree of depigmentation in vitiligo.

BACKGROUND: Vitiligo is a common skin disorder causing depigmented macules that can impair a patient's quality of life. Currently, there are no standardized outcome measures to assess the degree of depigmentation. Moreover, there is limited knowledge on the measurement properties of outcome measures in vitiligo. OBJECTIVES: To assess the reliability and responsiveness of the Vitiligo Area Scoring Index (VASI) and the Vitiligo European Task Force assessment (VETFa), two well-described clinician-reported outcomes. METHODS: We included three vitiligo patient groups. In one group of 31 patients,

Genetic Susceptibility to Vitiligo: GWAS Approaches for Identifying Vitiligo Susceptibility Genes and Loci.

Vitiligo is an autoimmune disease with a strong genetic component, characterized by areas of depigmented skin resulting from loss of epidermal melanocytes. Genetic factors are known to play key roles in vitiligo through discoveries in association studies and family studies. Previously, vitiligo susceptibility genes were mainly revealed through linkage analysis and candidate gene studies. Recently, our understanding of the genetic basis of vitiligo has been rapidly advancing through genome-wide association study (GWAS). More than 40 robust susceptible loci have been identified and confirmed to

Genetics of Vitiligo: A Review.

Vitiligo is a common acquired depigmentation skin disease with obvious family aggregation. About 25-50% of patients have positive family history, which belongs to polygenetic disease. In recent years, through candidate genes and genome-wide association studies, multiple susceptibility gene loci have been found, and studies also show that there is genetic heterogeneity among different populations. Environmental factors can also interact with genetic factors to trigger diseases through various mechanisms. The risk assessment model based on genetic and environmental factors provides a new directi

Genetics and epigenetics in vitiligo.

Vitiligo, a complex autoimmune disorder characterized by melanocyte destruction, arises from an intricate interplay of genetic, epigenetic, immune, and environmental factors. Genome-wide association studies (GWAS) have identified over 50 susceptibility loci, including key genes within the MHC region and those involved in immunity, oxidative stress, and melanogenesis. Concurrently, epigenetic research has unraveled regulatory networks critical to vitiligo pathogenesis, with a focus on DNA methylation and non-coding RNAs (e.g., microRNAs, long non-coding RNAs, and circular RNAs). These advanceme

Epigenetic Modifications in Vitiligo.

Vitiligo is an autoimmune depigmenting skin disorder and can affect the mental health of the patients. Current research suggests that the development of vitiligo involves a combination of genetic susceptibility, immune imbalance, and oxidative stress. However, its pathogenesis has not been fully elucidated. Epigenetic modification has gained increasing attention as an emerging way to regulate gene expression at the transcriptional or post-transcriptional level. Currently known modes of epigenetic modification include the regulation of non-coding RNAs, DNA methylation, and histone modification.

Mechanisms of pyroptosis in vitiligo.

Vitiligo is a common depigmentation disorder characterized by the loss of skin pigmentation, resulting in white patches on the skin. The pathogenesis of vitiligo involves complex interactions between genetic, immunological and environmental factors. Recent studies have implicated pyroptosis, a form of programmed cell death, in the development and progression of vitiligo. Pyroptosis is characterized by the activation of inflammatory caspases, such as caspase-1, leading to cell lysis and the release of inflammatory cytokines. This process has been linked to the melanocytes and other cells within

The Patient Journey in Vitiligo.

BACKGROUND: Vitiligo is a chronic, non-life-threatening skin disorder characterized by depigmented macules caused by the selective loss of melanocytes. Although its pathogenesis is multifactorial, encompassing autoimmune, genetic, and oxidative stress-related factors, it remains underdiagnosed, and patients frequently face delays in appropriate care. The visible nature of vitiligo, especially when it affects exposed areas like the face or hands, significantly impacts patients' psychological well-being and quality of life, often leading to social stigma, anxiety, and reduced self-esteem. A comp

Vitiligo: are microbes to blame?

Vitiligo is a prevalent acquired depigmenting disease that is distinguished by the depletion of functional melanocytes and epidermal melanin. Despite significant advancements in comprehending vitiligo, the precise etiology and pathogenesis of the condition remain elusive. So far, the treatment of vitiligo is still one of the most difficult dermatological challenges. Thus, developing a better understanding of vitiligo pathogenesis to develop more effective treatments is very important. Vitiligo has long been acknowledged as an autoimmune disorder, and microbes serve as crucial regulators of the
